Evaluation on Early-maturing Korean Japonica Cultivars for High-temperature Tolerance during Grain Filling Stage (국내 육성 조생종 벼 품종들에 대한 등숙기 고온내성 평가)

  • Early-maturing Korean Japonica cultivars and Jungmo1024 were used as plant materials to evaluate hightemperature tolerance during grain filling stage. National Institute of Crop Science (NICS) in Korea developed Jungmo1024, a mutant line from Namil (wild type) treated by using sodium azide (SA) as mutagen. To evaluate high-temperature tolerance, all cultivars were exposed to high-temperature (day $31.5{\pm}2.5^{\circ}C$/ night $27.5{\pm}1.3^{\circ}C$) and ordinary temperature (day $27.5{\pm}1.8^{\circ}C$/ night $24.7{\pm}1.6^{\circ}C$) during grain filling stage. In these conditions, we performed compared evaluation of grain shape such as length and width and grain quality by using a 1625 Cervitec grain inspector. High-temperature during grain filling stage caused decrease of grain shape. In grain shape such as length and width, the decrease rate of width (average 6.3%) was higher than the decrease rate of length (average 1.3%). Hence, high-temperature affected width of grain than length of grain. In addition, high-temperature showed a decided difference in rate of head rice between ordinary temperature (average 76.3%) and high-temperature (average 13.3%).As a result, Taebong, Ungwang, Manan, and Jungmo1024 seemed relatively a decent high-temperature tolerance than other cultivars. Especially, Jungmo1024 seemed remarkable rate of head rice (average $34.4{\pm}6.2%$) than other cultivars under high-temperature. It is considered that a genetic trait of Jungmo1024 can be useful to improve breeding for high-temperature tolerance.

  • In order to introduce bivoltine races in a tropical country like India, it is necessary to have stability in cocoon crop under high temperature environments. Unlike any temperate country like Japan, the rearing conditions/environment, climatic conditions, quality of mulberry leaf and incidence of diseases are unpredictable in India. Geneticists and breeders of all the sericultural countries have experienced the influence of environment during the process of breeding. in order to select efficiently the breeds with high temperature tolerance, it is verb important to analyse clearly the heritability nature of high temperature tolerance. In light of the above, the present study was undertaken to determine the effect of high temperature treatment of (A) $35{\pm}1^{\circ}C$ and 85${\pm}$5% RH for 24 hrs continuously, (B) $35{\pm}1^{\circ}C$ and 85${\pm}$5% RH for 48 hrs continuously and (C) the control (25${\pm}1^{\circ}$ and 65${\pm}$5% RH in the normal rearing condition from the $3^{rd}of5^{th}$ instar on the pure races such as Moria, Nl37 and Cl46 as well as their Fl hybrids. The overall performance indicate that the hybrids are mare tolerant than the pure races. it was also observed that the overall performance declined in those batches where 48 hrs treatment was given. The most interesting observation noticed in this study was that there was maternal effect regarding temperature tolerance as evident from the better performance of those hybrids where the female parent used was more tolerant as pure race.

  • To improve the fermentation characteristics(such as starch-degradability, ethanol tolerance, sugar and high-temperature tolerance) of recombinant haploid yeast Saccharomyces diastaticus K114, hybridization technique was used. The hybridization partner was S. diastaticus 1177 which had good glucoamylase activity and fermentabi- lity. The best hybrid HH64 showed improved ethanol tolerance, sugar and high-temperature tolerance. Especia- lly, the starch-fermentability was significantly improved, since the hybrid produced 1.60% (w/v) ethanol from 4% (w/v) starch, while the recombinant haploid K114 produced 1.30% (w/v) ethanol. The optimum temperature and pH for the starch-fermentation by the hybrid HH64 was 30$\circ$C and 5, respectively. The hybrid yeast HH64 produced 7.5% (w/v) ethanol directly from 20% (w/v) starch.

  • Angular contact ball bearings in a high speed spindles are under the extreme conditions, such as high temperature, big centrifugal force and thrust cutting forces. So, the assembly contacts between spindle shaft and inner ring bearings, bearing housing and outer ring of bearings are occasionally unstable at high speed revolution. Furthermore, the ball contact angle of a bearing, which influence stiffness and lifetime of bearings, are changed according to loads and rotational speed. To analyze internal forces of a bearing under high speed revolution, the ball contact are calculated using nonlinear equations in consideration of rotational speed, thrust loads and raceway form. Diameter increase of inner and outer ring by influence factors, such as internal forces to inner and outer ring, centrifugal force and temperature of inner and outer rings are calculated to establish stable state in bearing assembly in high speed spindle. Finally, contribution ratio of influence factor to assembly design tolerance of inner and outer rings are shown and the stable assembly design tolerance are proposed.

  • It was measured the energy expenditure in each season, the cold tolerance in winter and the heat tolerance in summer. Farmers' adaptability to the change of environment was compared with those of city-dwellers such as indoor workers and street cleaners to determine the effect of living environment, especially living temperature, on the health of human body. It turned out that farmers had experienced wide range of temperature that was higher in summer and lower in winter than indoor workers. Farmers and street cleaners showed seasonal adaptation in energy expenditure, which was high in winter and low in summer. However, indoor workers did not show seasonal changes. Energy expenditure had an inverse correlation with the temperature in work place where subjects spend the longer time in a day except in female indoor workers in Seoul. And It was proved that farmers and street cleaners had stronger cold tolerance and heat tolerance than indoor workers.

A Trifloxystrobin Fungicide Induces Systemic Tolerance to Abiotic Stresses

  • Trifloxystrobin is a strobilurin fungicide, which possesses broad spectrum control against fungal plant diseases. We demonstrated that pre-treating red pepper plants with trifloxystrobin resulted in increased plant growth and leaf chlorophyll content compared with those in control plants. Relative water content of the leaves and the survival rate of intact plants indicated that plants acquired systemic tolerance to drought stress following trifloxystrobin pre-treatment. The recovery rate by rehydration in the drought treated plant was better in those pre-treated with trifloxystrobin than that in water treated plants. Induced drought tolerance activity by trifloxystrobin was sustained for 25 days after initial application. The trifloxystrobin treated red pepper plants also had induced systemic tolerance to other abiotic stresses, such as frost, cold, and high temperature stresses. These findings suggest that applying the chemical fungicide trifloxystrobin induced systemic tolerance to certain abiotic stresses in red pepper plants.

Behavioral responses and tolerance limits of wild goldeye rockfish Sebastes thompsoni to high temperature exposure (고 수온 노출에 따른 자연산 불볼락 Sebastes thompsoni의 행동반응 및 내성 한계)

  • To investigate the tolerance limit and critical thermal maximum (CTM), behavioral responses of wild goldeye rockfish Sebastes thompsoni according to exposure to high water temperature were observed using a continuous behavior tracking system. As a result, behavioral index (BI) of S. thompsoni in each temperature (20.0, 25.0, and 30.0℃) showed a significant difference (p<0.05) when compared with the value measured in a stable condition of 15.0℃. The activity level of S. thompsoni exposed to 25.0℃ decreased sharply after 20 hours. Their rest time at the bottom of experiment chamber increased, and their normal swimming and metabolic activities were disturbed. In addition, at a high water temperature of 30.0℃, S. thompsoni reached the limit of resistance and showed a sub-lethal reaction of swimming behavior, with energy consumption in the body increased and all test organisms died. In conclusion, the eco-physiological response of S. thompsoni to water temperature varied greatly depending on the fluctuation range of the exposed temperature and the exposure time. In addition, the tolerance limit of S. thompsoni to high water temperature was predicted to be 25.0-30.0℃. The maximum critical thermal that had a great influence on the survival of this species was found to be around 30.0℃.

Parental inheritance of heat stress tolerance during grain filling period in wheat

  • Wheat (Triticum asetivum L.) is one of the major grain crops worldwide. The reduced productivity ascribed by adverse environment is increasing the risk of food security. Wheat cultivars have been actively released by public side since 1960s in Korea. Each variety has been developed for superior regional adaptation, pest resistance and mostly high yield. Heat stress tolerance is one of the major parameters that threaten wheat production in Korea. Heat stress during grain filling period has been conceived as critical level and directly influences on wheat production. We evaluated 11 common wheat cultivars ("Baegjoong", "Dajung", "Goso", "Hanbaek", "Jokyoung", "Joeun", "Jopum", "Keumgang", "Olgeuru", "Sinmichal", "Uri") that were exposed to abnormally high temperature during the grain filling period. Each plant was grown well in a pot containing "Sunshine #4" soil in controlled phytotron facility set on $20^{\circ}C$ and 16 h photoperiod. At 9 day-after-anthesis (DAA9), plants were subjected to a gradual increase in temperature from $20^{\circ}C$ to $33^{\circ}C$ and maintained constantly at $33^{\circ}C$ for 5 days. After the treatment, plants were subjected to gradual decrease to normal temperature ($20^{\circ}C$) and continue to grow till harvest. Seeds were harvested from each tiller/plant. Total chlorophyll contents decrease level as well as grain parameters were measured to evaluate varietal tolerance to heat stress. We also divide each spike into five regions and evaluate grain characteristics among the regions in each spike. The obtained results allow us to classify cultivars for heat stress tolerance. The pedigree information showed that typical wheat lines provide either tolerance or susceptible trait to their off-springs, which enable breeders to develop heat stress tolerance wheat by appropriate parental choice.

Fabrication and Analysis of Characteristics of PRT using High-fine Laser Trimming Technology (고정밀 레이저 가공 기술을 이용한 PRT 제작 및 특성 분석)

  • In this paper, we fabricated PRT(platinum resistance thermometers) with the trimming technology using high fine laser system. U. V.(wavelength: 355nm) laser was mainly used for adjusting Pt thin films resistors to 100Ω at 0$^{\circ}C$. Internationally, the accepted A-class tolerance of temperature sensor is ${\pm}$0.06Ω at 0$^{\circ}C$. according to DIN EN 60751. The width of trimmed lines was about 10$\mu\textrm{m}$ and the best trimming conditions of Pt thin films were power : 37mW, frequency : 200Hz and bite size:1.5$\mu\textrm{m}$. And 96 resistors, fabricated by photolithography and etching process, have 79∼90Ω and 91∼102Ω as the proportion of 45.7% and 57.3%, respectively. As result of sitting Pt thin films resistors to the target value(109.73Ω at 25$^{\circ}C$), 82.3% of all resistors had the tolerance within ${\pm}$0.03Ω and the others(17.7%) were within ${\pm}$0.06Ω of A-class tolerance. The PRTs which wore fabricated in this research had excellent characteristics as follows; high accuracy, international standard TCR(temperature coefficient of resistance) value, long-term stability, wide temperature range, good linearity and repeatability, rapid response time, etc.

Growth and Environmental Tolerances (Water Temperature and Low Salinity) of Hybrid Female Red Sea Bream Pagrus major × Male Black Sea Bream Acanthopagrus schregeli (교잡어, 참돔 (Pagrus major, ♀) × 감성동 (Acanthopagrus schregeli, ♂)의 성장 및 수온변화와 저염분 환경에서의 내성)

  • The growth and tolerance for water temperature and salinity were compared among red sea bream Pagrus major (RSB) black sea bream Acanthopagrus schregeli (BSB) and their hybrid ($F_1$), female RSB ${\times}$ male BSB. The growth of the $F_1$ fish did not differ until 27 days after hatching (dAH), after which the most rapid growth was observed until approximately 300 dAH, followed by RSB and BSB. However, the RSB had out grown the $F_1$ fish by approximately 303 dAH. By 480 dAH, the RSB were largest, followed by the $F_1$ and BSB groups. The tolerances for high and low water temperature were significantly different for each species and growth stage. The largest tolerance spectrum was observed in the BSB group, and the tolerance spectrums gradually decreased with increasing final body weight. During the salinity tolerance trials, all of the species started to die following transfer into freshwater (0 psu). BSB showed the highest survival rate when kept in fresh water for an average of 29.9 hours, while the $F_1$ fish were more tolerant than the RSB fish. The fish were increasingly tolerant to fresh water and changes in water temperature as they grew, while the size of the temperature spectrum remained unchanged but shifted to a lower temperature range with growth. Our results demonstrate that $F_1$ performed well in terms of growth compared to parental fish, with higher temperature and salinity tolerances than RSB, and is thus suggested to be a suitable aquaculture species for Korea and northeast China.
