Reuse Information based Thrashing Resistant Cache Management Scheme

  • In recent computing systems, LRU replacement policy has been widely used because it can be simply implemented and applicable to most programs. However, if the working set size of the program is bigger than the actual cache size, LRU replacement policy may occur thrashing problem. Thrashing problem means that cache blocks are consistently replaced without re-referencing in the cache. This paper proposes a new cache management scheme to solve the thrashing problem in the second-level cache. The proposed scheme measures per set reuse frequency using EAF structure to find thrashing sets. When the cache miss occurs, it tests whether the address of the missed block is stored or not. If the address of the missed block is stored, it means that the recently evicted block is re-requested, so the reuse frequency is predicted high. In this case, the corresponding counter of the set is increased. When the counter value is bigger than the threshold value, we assume that the corresponding set shows high reuse frequency. The proposed scheme assigns the set with high reuse frequency to the additional small size cache to keep the blocks in the cache for a long time. Our experimental results show that the proposed scheme improves the IPC by 3.81% on average.

20(S)-ginsenoside Rh2 ameliorates ATRA resistance in APL by modulating lactylation-driven METTL3

  • Background: 20(S)-ginsenoside Rh2(GRh2), an effective natural histone deacetylase inhibitor, can inhibit acute myeloid leukemia (AML) cell proliferation. Lactate regulated histone lactylation, which has different temporal dynamics from acetylation. However, whether the high level of lactylation modification that we first detected in acute promyelocytic leukemia (APL) is associated with all-trans retinoic acid (ATRA) resistance has not been reported. Furthermore, Whether GRh2 can regulate lactylation modification in ATRA-resistant APL remains unknown. Methods: Lactylation and METTL3 expression levels in ATRA-sensitive and ATRA-resistant APL cells were detected by Western blot analysis, qRT-PCR and CO-IP. Flow cytometry (FCM) and APL xenograft mouse models were used to determine the effect of METTL3 and GRh2 on ATRA-resistance. Results: Histone lactylation and METTL3 expression levels were considerably upregulated in ATRA-resistant APL cells. METTL3 was regulated by histone lactylation and direct lactylation modification. Overexpression of METTL3 promoted ATRA-resistance. GRh2 ameliorated ATRA-resistance by downregulated lactylation level and directly inhibiting METTL3. Conclusions: This study suggests that lactylation-modified METTL3 could provide a promising strategy for ameliorating ATRA-resistance in APL, and GRh2 could act as a potential lactylation-modified METTL3 inhibitor to ameliorate ATRA-resistance in APL.

Changing Patterns of Serum and Bile Antibodies in Re-infected Rats with Clonorchis sinensis

  • Rats develop strong resistance to re-infection and super-infection by Clonorchis sinensis. The present study investigated the antibodies present in the sera and bile juice of rats that were primary infected and re-infected with C. sinensis. The serum level of specific IgG antibodies, which were elevated 2 wk of the primary infection, peaked at 4 wk and subsequently remained unchanged even during re-infection. The total IgE level in serum increased slowly from 388 ng / ml to 3,426 ng / ml beginning 2 wk after the primary infection, and remained high up to 8 wk but dropped to a normal level (259 ng / ml) after treatment. In resistant re-infected rats, the serum IgE level increased rapidly and peaked within 1 wk, whereas no increase was observed in immunosuppressed rats. The serum level of specific IgA antibodies was elevated beginning 1 wk after infection, and decreased 4 wk after treatment. The total bile IgA level unchanged during the primary infection but increased in treated and re-infected rats. The elevated levels of serum IgE and bile IgA indicate that these immunoglobulins may be correlated with the development of resistance to re-infection by C. sinensis in rats.

Thermal Characteristic Evaluation of Functionally Graded Composites for PSZ/Metal

  • The functionally graded material (FGM) is the new concept for a heat resisting material. FGM consists of ceramics on one side and metal on the other. A composition and microstructure of an intermediate layer change continuously from ceramics to metal at the micron level. This study is carried out to analyze the thermal shock characteristics of functionally graded PSZ/ metal composites. Heat-resistant property was evaluated by gas burner heating test using $C_2H_2/O_2$ combustion flame. The ceramic surface was heated with burner flame and the bottom surface cooled with water flow. Also, the composition profile and the thickness of the graded layer were varied to study the thermo mechanical response. Furthermore, this study carried out the thermal stress analysis to investigate the thermal characteristics by the finite element method. Acoustic emission (AE) monitoring was performed to detect the microfracture process in a thermal shock test.

  • The alkaline elastase is an extracellular serine protease of the alkalophilic Bacillus strain Ya-B. To increase the gene copy number and the production level of the alkaline elastase Ya-B, we designed, on the B. subtilis chromosome, a gene amplification of the 10.6 kb repeating unit containing amyE, aleE (alkaline elastase Ya-B gene) and tmrB. The aleE was inserted between amyE and tmrB, and B. subtilis APT119 strain was transformed with this amyE-aleE-tmrB-junction region fragment. As a result, we succeeded in obtaining tunicamycin-resistant (Tm$^{r}$) transformants (Tf-1, Tf-2) in which the designed gene amplification of 10.6 kb occurred in chromosome. The transformants showed high productivity of $\alpha $-amylase and alkaline elastase Ya-B. The copy number of the repeating unit (amyE-aleE-tmrB) was estimated to be 25, but plasmid vector (pUC19) was not integrated. The amplified aleE of chromosome was more stable than that of plasmid in absence of antibiotics.

  • The earthquake-resistant structural systems have to ensure the sufficient stiffness and ductility for the stability. For those purposes, recently, the performance design concept to increase the degree of absorbed energy level of structures has been proposed. One practical way of the performance design in the spatial structures is to apply the isolation system to boundary parts of roof system and sub-structure to obtain the target performance. So, it is necessary to examine the characteristics of dynamic behavior of spatial structures governed by higher modes rather than lower modes different from the cases of high rise buildings. The objectives of this paper are to develop the equivalent lumped mass model to simplify the analytical processes and to investigate the dynamic behavior of roof system according to the mass and the stiffness of sub-structures as a fundamental study of performance design for the spatial structures.

  • In this paper, polymer nanocomposites were fabricated by mixing fire-resistant and high pseudoplastic Nylon 6,6 with MWNT(Multiwalled Nanotube), which has mechanical, electrical, and heat stable properties. The experiments were performed in order to investigate their mechanical and electrical properties depending on the level of MWNT and the presence of acid treatment on Nylon 6.6. Morphology of polymer nanocomposites was observed using Scanning Electron Microscopy technique. The results indicate that the polymer nanocomposites have the best mechanical and electrical properties in the optimal conditions of Nylon 6.6 and MWNT(10wt%).

  • In both untreated and conventionally stabilized swine manures antibiotic resistant (AR) microorganisms, Staphylococcus-like and Salmonella-like microorganisms were detected. Also pathogens with MAR phynotype were detected. Presence of such microorganisms suggest high level of pathogen-related health risk to farmers who may be in direct contact with the manure and its conventionally stabilized product In contrast the autothermal thermophilic aerobic digestion (ATAD) treatment have efficiently reduced AR and pathogenicity from the swine manure. When soil was fertilized using swine manure and its stabilized products, despite no detection of MAR-exhibiting pathogen-like microorganisms in fertilized soil, potential pathogen-related health risk could not be ruled out from the fertilized soil since the organic fertilization led to increase in AR and pathogenicity in the soil microbial communities. As conclusion, this microbiological study demonstrated that an ATAD process is applicable in control of pathogen-related health risk in livestock manure.

  • The effectiveness of three antifungal antibiotics amphotericin B, clotrimazole and 5-fluorocytosine was tested against 120 clinical isolated Candida species. The minimum inhibitory concentrations of amphotericin B was considerably lower than those of clotrimazole and 5-fluorocytosine. High-level resistance to 5-fluorocytosine was present in 50% of the isolates. The combination of clotrimazole and 5-fluorocytosine produced synergistic inhibition against all 20 strains of Candida albicans tested that were relatively resistant to both antifungal agents.
