Effects of Oenanthe javanica and Allium tuberosum on Lipid Content in Rats Fed a High-fat·High-cholesterol Diet (미나리 및 부추의 고지방․고콜레스테롤 식이 흰쥐의 지질 함량 개선에 미치는 영향)

  • This study was conducted to investigate the effects of Oenanthe javanica and Allium tuberosum powder on lipid metabolism in rats fed high fat·high cholesterol diet. Experimental rats were divided into five groups which were composed of normal diet group (N), high fat·high cholesterol diet group (HF), high fat·high cholesterol diet with 5% Oenanthe javanica powder diet group (OP), high fat·high cholesterol diet with 5% Allium tuberosum powder diet group (AP) and high fat·high cholesterol diet with 2.5% Oenanthe javanica and Allium tuberosum powder diet group (OAP). The serum TG content of the HF group was significantly increased compared to the N group, but that of the OAP group was significantly decreased. Serum HDL-cholesterol contents of the OAP group was significantly increased compared to the HF group. The serum total cholesterol, LDL-cholesterol and AI of the HF group were increased compared to the N group and especially the LDL-cholesterol of OP and OAP groups were significantly decreased compared to the HF group. The liver TG and total cholesterol contents of the HF group were significantly increased compared to the N group, while TG contents of the OAP group was significantly decreased compared to the HF group. Fecal total lipid and total cholesterol of OP, AP and OAP groups were significantly increased compared to the HF group. These results suggest that supplementation of Oenanthe javanica and Allium tuberosum may have a pronounced impact on markers of lipid metabolism in serum and liver of rats fed high fat·high cholesterol diets.

The Effect of Herbal-acupuncture with Moutan Cortex Radicis Extract (목단피 약침이 고지혈증과 동맥경화에 미치는 영향에 대한 실험적 연구)

  • Objective & Methods : The present study was to investigate the effect of Moutan Cortex Radicis Herbal-acupuncture (MCR-HA) at GB34 (Yangleungchean) on hyperlipidemia and arteriosclerosis induced by high fat diet in rats. Results : 1. Moutan Cortex Radicis herbal-acupuncture infusion solution (MCR-HAS) increased DPPH radical scavenging activity and HMG-CoA reductase inhibition rate in rat liver cells. 2. MCR-HA at GB34 decreased serum total cholesterol level, and increased serum phospholipid/total cholesterol in rats with high fat diet. 3. MCR-HA at GB34 decreased atherogenic index in rats with high fat diet. 4. MCR-HA at GB34 increased hepatic catalase activity in rats with high fat diet. Conclusion : These results suggested that Moutan Cortex Radicis herbal-acupuncture at GB34 has therapeutic effects on hyperlipidemia and related complications in rats with high fat diet.

Effects of gamgook(Chrysanthemum indicum L.) herbal-acupuncture on lipid lowering effect, anti-oxidative capacity and anti-inflammatory effect in rat fed high oxidized fat (감국약침이 과산화지질을 급여한 흰쥐의 지질대사, 항산화 및 면역계에 미치는 영향)

  • Objectives: To investigate the effects of Chrysanthemum indicum L. pharmacopuncture on lipids, antioxidant capacity and anti-inflammation in rats fed high-fat diet. Methods: Hyperlipidemic rats induced by high-fat diet were divided into 5 groups: no treatment control (normal, n=8), high-fat diet only control (control, n=8), high-fat diet and Chrysanthemum indicum L. pharmacopuncture at CV4 group (TI, n=8), high-fat diet and Chrysanthemum indicum L. pharmacopuncture at CV17 group (TII, n=8), and high-fat diet and Chrysanthemum indicum L. pharmacopuncture at EX-HN3 group (TIII, n=8). They were given pharmacopuncture accordingly every other day for two weeks followed by analyses of lowering lipids effects, oxidative capacity and anti-inflammatory effects. Results: Compared with the control, pharmacopuncture groups showed significantly decreased plasma total cholesterol (TC), liver thiobarbituric acid reactive substance (TBARS), catalase, glutathione peroxidase, neutrophils, monocytes, plasma and liver IL-$1{\beta}$, and plasma and liver IL-6. In other parameters including plasma and liver triglyceride, liver TC, LDL-cholesterol, HDL-cholesterol, liver TBARS, supraoxide dismutase, total protein, albumin, blood cell analysis, plasma and liver TNF-$\alpha$, and IL-10, there was no significant difference between control and pharmacopuncture groups. No clear acupoint-specificity was observed. Conclusions: Chrysanthemum indicum L. pharmacopuncture may improve control of hyperlipidemia.

Red beet(Beta vulgaris L.) leaf supplementation improves antioxidant status in C57BL/6J mice fed high fat high cholesterol diet

  • The effect of diet supplemented with red beet (Beta vulgaris L.) leaf on antioxidant status of plasma and tissue was investigated in C57BL/6J mice. The mice were randomly divided into two groups after one-week acclimation, and fed a high fat (20%) and high cholesterol (1%) diet without (control group) or with 8% freeze-dried red beet leaf (RBL group) for 4 weeks. In RBL mice, lipid peroxidation determined as 2-thiobarbituric acid-reactive substances (TBARS value) was significantly reduced in the plasma and selected organs (liver, heart, and kidney). Levels of antioxidants (glutathione and $\beta$-carotene) and the activities of antioxidant enzyme (glutathione peroxidase) in plasma and liver were considerably increased, suggesting that antioxidant defenses were improved by RBL diet. Comet parameters such as tail DNA (%), tail extent moment, olive tail moment and tail length were significantly reduced by 25.1%, 49.4%, 35.4%, and 23.7%, respectively, in plasma lymphocyte DNA of RBL mice compared with control mice, and indicated the increased resistance of lymphocyte DNA to oxidative damage. In addition, the RBL diet controlled body weight together with a significant reduction of fat pad (retroperitoneal, epididymal, inguinal fat, and total fat). Therefore, the present study suggested that the supplementation of 8% red beet leaf in high fat high cholesterol diet could prevent lipid peroxidation and improve antioxidant defense system in the plasma and tissue of C57BL/6J mice.

Effects of Eleutherococcus senticosus and Several Oriental Medicinal Herbs Extracts on Serum Lipid Concentrations (가시오가피와 수종 한약자원 혼합 조성물이 고지방 식이에 있어 혈청 지질농도에 미치는 효과)

  • This study evaluated the hypolipidemic effects of Eleutherococcus senticosus combined with several oriental medicinal herbs. In addition, it addressed whether consuming a diet of healthy food along with a daily supplement of this composite could affect the health status of individuals suffering from diet-related disease such as obesity and hyperlipidemia. The effects of the combined medicinal herbal extracts(MHE) extracted with hot water on reducing serum lipids and improving blood parameters were examined in rats fed a high-fat diet for 5 weeks. Sprague-Dawley rats were randomly assigned to 4 groups: basal diet only(BDG), high fat diet without MHE(FCG), high fat diet and 10% MHE(F10E) and high fat diet and 20% MHE(F20E). Serum lipid contents and blood variables were examined after the experimental period. The results of were as follows. The hematological data for the 4 groups were similar indicating no significant differences. There were moderate level of serum GOT activity in the F10E and F20E groups as compared to the FCG group. Total cholesterol, LDL-cholesterol, triglyceride in serum and the atherogenic index were remarkably reduced in the MHE-supplemented groups as compared to the control group. However, F10E and F20E groups had significantly higher HDL-cholesterol levels than the control group. These results imply that combination of several medicinal herbal extracts could be used to reduce of serum lipid concentrations. The data from this study will be used as basic information with the field of functional food research to address how we can apply oriental medicinal resources to foods. Such research currently drawing considerable attention world-wide.

Effects of ethanolic extract of Ulmus davidiana Root on Lipid Metabolism in High-Fat Diet Fed Mice (느릅나무 뿌리 에탄올 추출물이 고지방 식이를 섭취한 마우스의 지질대사에 미치는 영향)

  • This study was performed to investigate the effects of ethanolic extract of Ulmus davidiana root (UE) on lipid metabolism in mice fed a high-fat diet (HF) for 7 weeks. Forty male ICR mice were randomly divided into four groups; normal diet group (N), high-fat diet group (HF), HF with 0.5% UE (HF-L) and 1% UE (HF-H) group. Body weight, body weight gain, and liver weight in the HF group was significantly higher than in the N group, while those of the HF-L and HF-H group were unchanged. UE improved HF-induced dyslipidemia by reducing serum triglyceride, total cholesterol, and the atherogenic index. There was no difference in serum HDL-cholesterol among experimental groups. However, the HDL-cholesterol/total cholesterol ratio was significantly increased in the HF-L and HF-H group. Histological analysis showed that HF-fed mice developed hepatocellular microvesicular vacuolation as a result of fat accumulation. These changes were attenuated by 1% UE supplementation. In addition, hepatic triglyceride and cholesterol levels in the HF-H group significantly reduced. Taken together, these results demonstrated that lipid levels in the blood and liver were reduced by UE, suggesting that it might be beneficial for the prevention and treatment of hyperlipidemia and fatty liver.

Inhibitory Effect of Daesiho-tang(Dachaihu-tang) Extracts on High-fat Diet-induced Obesity (고지방 식이 유도 비만에 대한 대시호탕 추출물의 억제 효과)

  • Objectives : The aim of this study is to investigate Daesiho-tang(Dachaihu-tang) water extracts (DSTE) have potent anti-obesity activities in a high-fat diet-induced obesity mouse model. Methods : In this study, we designed three groups (normal diet group, high-fat diet group, high-fat diet plus DSTE group for 7-weeks oral administration). Results : Increases in body weight were inhibited by 7-weeks oral administration of DSTE at a 500 mg/kg concentration in this animal model. Results from blood lipid analysis showed that the levels of triglyceride, total cholesterol and LDL-cholesterol were significantly lowered by DSTE administration, also HDL-cholesterol was increased more than high-fat diet-induced obese mouse. To understand the underlying mechanism at the molecular level, the effects of DSTE were examined on the expression of the genes involved in lipogenesis by real-time PCR. In epididymal fat and liver of DSTE-treated mice, the mRNA level of lipogenic genes such as sterol regulatory element binding protein 1 and fatty acid synthase were decreased, which was well correlated with the reduction of the tissues weight. Conclusions : These results suggest that DSTE may have great potential as a novel anti-obesity agent.

Effects of Onion Peel Water Extract on the Blood Lipid Profiles in Mice Fed a High-Fat Diet (고지방식이를 섭취한 마우스에서 양파껍질 열수 추출물이 혈중지질에 미치는 영향)

  • Onion (Allium cepa L.) is one of the richest sources of flavonoids in human diet. Onion peel contains over 20 times more quercetin than onion flesh. In this study, we studied the effects of onion peel water extract (OPE) on the blood lipid profiles in mice. The onion peel extracts was extracted with hot water. The experimental groups were divided with 3 groups (n = 6) of ICR male mice: normal diet + distilled water (NC), high-fat diet + distilled water (HF), high-fat diet + onion peel water extract 20 mg/kg (OPE-20). The oral administration was conducted daily. The experimental period was 7 weeks. Onion peel water extract showed higher concentration of polyphenol gallic acid and anti-oxidant trolox equivalent than the ethanol extract. The body weight gain and food efficiency ratio was significantly lower in the OPE-20 group as compared with HF group (p < 0.05). The epididymal fat and retroperitoneal fat showed significantly lower weights and sizes in the OPE-20 group as compared with HF group (p < 0.05). The serum levels of total cholesterol, LDL cholesterol and triglyceride were significantly lower in the OPE-20 group as compared with HF group (p < 0.05). The OPE-20 group showed higher HDL cholesterol concentration than HF group (p < 0.05). Atherogenic index was ignificantly lower in as compared with HF group (p < 0.05). The serum levels of glucose, GOT and GPT were significantly lower in the OPE-20 group as compared with HF group (p < 0.05). In these results, we suggests that onion peel water extracts supplementation can reduces the serum lipid components and improves the lipid metabolism in hyperlipidemic mice induced with a high-fat diet.

Effects of Rhizoma Alismatis on Lipid Composition and TBARS Concentration in Rat Fed High Fat Diet (택사농축액이 고지방식이를 급여한 흰쥐의 지질구성 및 TBARS량에 미치는 영향)

  • Objectives : Effects of Rhizoma Alismatis extract on lipid composition and TBARS (thiobarbituric acid reactive substances) formation were investigated in rat fed high fat diet. Methods : Seventy male Sprague-Dawley rats were used in these experiments and housed with food and water ad libitum and Twenty rats were selected healthy over 400g body weight and fed high fat diet for 8 weeks. Rhizoma Alismatis extract was administerd at dose 100mg/kg/day, 200mg/kg/day and 300mg/kg/day p.o. for 4 weeks. Then Triglyceride, Total cholesterol, HDL-cholesterol, LDL-cholesterol, TBARS concentration and ALT(Alanine Aminotransferase), AST(aspatate transaminase) activities were determined. Results : Plasma total LDL-cholesterol showed a tendancy to decrease in all treated group(P<0.05), however plasma HDL-cholesterol, total cholesterol, triglyceride showed no significantly different. Rhizoma Alismatis extract feedings resulted in a decrease in plasma TBARS(p<0.05) concentration and AST, ALT activites but contents of TBARS in liver showed no significantly different among all the treatments. Conclusions : Reviewing these experimental results, it appears that Rhizoma Alismatis extract was effective for inhibiting lipid peroxidation and decreasing LDL-cholesterol concentration.

  • Objective : In order to investigate the effects of Soongijeseub-bang(here in after referred to SJB) on the obese gene and obese inhibitory, C57BL/6 mice were induced by high fat diet Methods : C57BL/6 mice were divided into three groups(normal, high fat diet with control, high fat diet with SJB extract) and fed for 15 weeks. And measured body weight change, the weight change of the adipocytes in body, the levels of ALT, AST, creatinine, total cholesterol, LDL-cholesterol, HDL-cholesterol, triglyceride, NEFA(non-esterified fat acid), glucose in serum and the expression of ${\beta}3AR$, leptin, and TNF- ${\alpha}$ gene in primary adipocytes and adipocytes tissue. Results : The following results were obtained in this study. 1. SJB 500 mg/kg extract group showed significant decrease in final weight. 2. SJB 500 mg/kg extract group showed significant decrease in the amount of adipocyte in weight. 3. All experimental group showed significant decrease in ALT and AST levels. 4. All experimetal group did not show significant decrease in glucose and creatinine levels. 5. All experimental group showed significant decrease in total cholesterol, LDL-cholesterol, triglyceride and NEFA levels. HDL-cholesterol were increase significantly in SJB 500 mg/kg extract group. 6. SJB 500 mg/kg extract group showed significant decrease in leptin levels. 7. All experimental group showed significantly increased expressions of fJ 3AR in primary adipose cell and 3T3-Ll cell, and those of leptin in primary adipose cell and 3T3-Ll cell were decreased significantly. SJB $100{\mu}l/ml$ extract group showed significant decrease in the expression of TNF-${\alpha}$. 8. SJB 500 mg/kg extract group showed decrease in the size of adipocyte in adipocytes tissue. 9. All experimental group showed significant decrease in adipose vacuoles in liver tissue.
