Regulatory Effects of Exercise and Dietary Intervention in Mitogen Activated Protein Kinase Signaling Pathways in Rats

  • As a central component of a novel protein kinase cascade, the activation of the mitogen-activated protein (MAP) kinase cascade has attracted considerable attention. We sought to determine the effect of exercise and diet on the activation of the extracellular-signal regulated protein kinase (ERK) 1/2 and the p38 MAP kinase pathways in rat soleus muscle. Forty-eight Sprague-Dawley rats were assigned to one of two dietary conditions: high-carbohydrate (CHO) or high-fat (FAT). Animals having each dietary condition were further divided into one of three subgroups: a sedentary control group that did not exercise (NT), a group that performed 8 weeks of treadmill running and was sacrificed 48 h after their final treadmill run (CE), and a group that was sacrificed immediately after their final routine exercise training (AE). A high-fat diet did not have any significant effect on phosphorylated and total forms of ERK 1/2 or p38 MAP kinase. In chronically trained muscle that was taken 48 h after the last training, phosphorylated ERK 1/2 significantly increased only in the FAT but not in the CHO groups. In the case of total ERK 1/2, it increased significantly for both groups. In contrast, both phosphorylated and total forms of p38 MAP kinase decreased markedly compared to sedentary muscle. In muscle that was taken immediately after a last bout of exercise, phosphorylated ERK 1/2 increased in both groups but statistical significance was seen only in the CHO group. Total ERK 1/2 in acutely stimulated muscle increased only in the CHO-AE group even though the degree was much lower than the phosphorylated status. Muscle that was taken immediately after the routine training increased in phosphorylation status of p38 MAP kinase for both dietary conditions. However, statistical significance was seen only in the CHO group owing to a large variation with FAT. In conclusion, a high-fat diet per se did not have any notable effect versus a high-carbohydrate diet on MAP kinase pathways. However, when diet (either CHO or FAT) was combined with exercise and/or training, there was differentiated protein expression in MAP kinase pathways. This indicates MAP kinase pathways have diverse control mechanisms in slow-twitch fibers.

Capsaicin Increases Swimming Endurance Capapcity in High-Fat-Fed Mice

  • Increase in fat mobilization by capsaicin(CAP) was investigated in high-fat-fed mice using an adjustable current water pool. Male ICR 7-wk-old mice were fed a high fat diet [50% total energy content in the diet(E%) fat, 20 E% protein, 30E% carbohydrate] for 2 wk and one group (HCAP) was orally administered CAP at 2 h before swimming. After being accustomed to swimming , the mice were subjected to forced swimming every 2d in the current water pol and the total swimming period until exhaustion was measured . The total swimming period was used as index of swimming capacity. Swimming time to exhaustion of treated mice was significantly longer than that of the high-fat-fed control group (100.2$\pm$10.6 vs. 58.0$\pm$8.5min, P<0.01) after 2wk of training. The concentration of serum-free fatty scids gradually increased up to 2 h in CAP -administered mice. The perirenal adipose tissue weight of CAP -administered mice (HCAP) before swimming was lower than that of the high-fat-fed mice adminstered placebo solution (HP) which had not ingested CAP during the 2 wk. These results suggest that the increase of swimming capacity of CAP-administered high-fat-fed mice was due to an increase of fat mobilization that was induced by CAP.

  • Objective : Obesity is an important cause of diabetes, and lipotoxicity causes insulin resistance. Recently a lot of research is being done on PPAR-${\alpha}$. TNF-${\alpha}$. adiponectin, and leptin, which are important obesity related factors. In this study, we investigated the effects of Supungsunki-hwan on high fat. high carbohydrate diet-induced obese type 2 diabetic mouse models. Methods: Diabetes was induced in ICR male mouse (30${\pm}$5g) with Surwit's high fat, high sucrose diet. Mice were divided into 4 groups(n=10) of Normal. Control. Supungsunkj-hwan group. and acarbose group. The Supungsunki-hwsn group was given 10% Supungsunkj-hwan in their diet. and the acarbose group was given 0.5% acarbose in their diet. After 6 weeks. body weight. food intake, FBS and OGTT. lipid profile and liver enzymes, epididymal fat weight, and gene expression of leptin, adiponectin, TNF-${\alpha}$ and PPAR-${\alpha}$ were measured. Leptin. adiponectin. tumor necrosis factor(TNF)-${\alpha}$ and peroxisome proliferator-activated receptor (PPAR)-${\alpha}$ were evaluated by reverse transcription-polymerase chain reaction. Results : Supungsunkj-hwan increased the gene expression of PPAR-${\alpha}$, which reduces lipotoxicity and insulin resistance. Supungsunkj-hwan also significantly reduced triglyceride. AST. ALT serum levels. and 1 hour oral glucose tolerance levels. Conclusion : These results show that Supungsunkj-hwan improves insulin resistance in the liver and muscles, by reducing triglyceride levels and lipotoxicity through increased PPAR-${\alpha}$ gene expression. This is supported by the fact that Supungsunkj-hwan significantly reduces 1 hour oral glucose tolerance levels. Therefore we suggest that Supungsunkj-hwan would be an effective treatment for obese type 2 diabetic patients.

Cucurbitacin B Suppresses Hyperglycemia Associated with a High Sugar Diet and Promotes Sleep in Drosophila melanogaster

  • Secondary metabolites enable plants to protect themselves from herbivorous insects. Among these, cucurbitacin B (cuc-B) is a bitter-tasting compound with promising pharmacological potential. Dietary exposure to cuc-B lowered the hemolymph glucose levels of Drosophila melanogaster fed with a high carbohydrate diet, which is homologous to high blood glucose in humans, and its effect was comparable to that of metformin, a well-known glucose-lowering drug. Furthermore, cuc-B reduced tissue sugar levels and glycogen levels, as well as triacylglycerol levels. Our results thus highlight the potential applicability of this compound to treat chronic metabolic diseases such as diabetes and obesity. Additionally, we analyzed sleep quality and taste-associative memory enhancement after cuc-B and metformin treatment. Both supplements increased nighttime bout length and metformin increased memory consolidation. Therefore, discarded shell of Cucurbitaceae could be processed into health supplements.

  • This study was designed to compare by the body composition in 4 groups of Sprague-Dawley rats during starvation previously fed with standard, high Carbohydrate, high protein or high fat diet. A) In different internal organs and skeletal muscles, the weights were differently reduced during starvation. Especially the weight reduction of liver was the highest level at $50{\sim}70%$, and adrenal glands did not change significantly. After death due to starvation, body weight decreased morethan 40% comparing to the initial body weight and epididmal fat pads disap peared completely. B) In rats starved to death, the changes of body composition (%) were following; lipid content was markedly reduced, while protein was similar to that of zero day starvation, ash increased slightly and water content increased. C) Five days starvation did not change body composition and body weight significantly. D) Effects of succeeding starvation after feeding 4 different diets were not significant, and it seemed that 4 different diets did not affect life span due to starvation.

Effect of long-term high-fat diet and fasting on energy metabolic substrates utilization in resting rats

  • The effects of a high-fat diet and fasting on resting energy expenditure and energy substrate utilization were examined using the method of measuring whole body energy metabolism and oxygen uptake. Eight 4-week old male Sprague-Dawley rats were used for the high-fat diet experiment. Energy metabolism was measured using acrylic metabolic chambers over 24 hours. After 1-week of preliminary feeding, 4 rats were fed a chow diet, whereas the remaining 4 rats were fed a high-fat diet (HF) ad libitum, which contained 40% (w/w, calorie base 60%) more fat than that in the chow diet. The flow rate to measure energy metabolism inside the chamber was controlled at a mean of 3.5 L/min, and five chambers were subjected to measurement. One of the five chambers was used to correct errors by measuring the atmosphere. As a result of 5 weeks of control diet and high-fat diet feeding, body weight of the high-fat diet group tended to increase more than that in the control diet fed group, but the difference was not significant. Oxygen uptake and carbon dioxide production changed as time went on over the 24 hr. The respiratory exchange ratio also changed during the 24 hr, and the difference between the groups was significant. The control group showed significantly more carbohydrate oxidation than that of the high-fat diet fed group. A fasting experiment was conducted using six 7-week old Sprague-Dawley male rats. Energy metabolism measurements were performed using the same method as that used in the high-fat diet experiment; resting metabolism was measured prior to fasting, and a fasting condition began from 9:00 am the next day for 3 days to calculate energy metabolism. Both body weight and 24-hour oxygen uptake decreased significantly as a result of 3-day fasting. Total oxygen uptake in the first day decreased, and declined significantly on day 3 of fasting. Total 24-hour carbon dioxide production decreased significantly over the 3 days. The mean 24-hour respiratory exchange ratio decreased significantly. Additionally, energy expenditure during the dark period (20:00-08:00), which is the active period for rats, decreased significantly with fasting, whereas energy expenditure during the light period (08:00-20:00) did not increase by fasting.

미강(米糠)과 야채첨가식이(野菜添加食餌)가 흰쥐의 체내대사(體內代謝)에 미치는 영향(影響)( II ) -열량소의 소화흡수율에 미치는 영향을 중심으로- (The Metabolic Effects of Rice Bran and Vegetable Cellulose Supplemented Diets on Albino Rats -Especially on Apparent Digestibility of Intake Nutrients-)

  • The effects of dietary cellulose to utilize the intake nutrients in the albino rats were investigated by the two consecutive experiments in this study. The author wanted to observe the effects of the age of animals and different level of dietary cellulose, high and low in the first experiment and the effects of dietary cellulose from three different sources, rice bran, vegetable and pure-cellulose at high level in the second experiment. The results were summarized as follows. (1) The obvious effect of dietary cellulose caused to increase feces weight. Consequently apparent digestibilities of intake nutrients in the diet decreased. It was noteworthy that fecal glucose excretion was significantly affected by cellulose intake. Since three nutrients, protein, fat and carbohydrate serve as energy sources in the body, this result can be interpreted to be lowered in the efficiency of energy utilization out of intake from the diet. (2) The nutrients loss due to dietary cellulose resulted in reduction of skeletal muscle and bone weights which lead eventually to affect in reduction of body weight. (3) The effects of dietary cellulose showed differently according to the amount of cellulose ingested, dietary nutrients composition, the kinds of dietary cellulose supplemented and the age of experimental animals. When the intake diet was proportionately imbalanced in three different energy sources, carbohydrate, fat and protein, the cellulose intake was adversely affected in the utilization of the nutrients in the body. Cellulose from rice bran decreased body weight gain, cellulose from vegetable maintained medium level in body weight and pure-cellulose increased body weight. Growing rats showed more sensitive effects of cellulose tolerance in the body than grown rats in every concerns of this experiment.

경기 일부 중학생의 당류 간식 섭취가 정서지능에 미치는 영향 (The Effect of Carbohydrate Processed Snacks on Middle School Students' Emotional Intelligence)

  • Adequate nutrients intake and regular eating habit are so important for the students to develop physical and mental health, and they contribute significantly for the development of intellectual capacity. The intake of simple sugar contained in the processed foods is known to have been related not only with such physical problems as obesity and diabetes but also with hyperactivity during growing period. This study analysed the effects of carbohydrate processed snacks on the middle school students' emotional intelligence and intended to demonstrate the correlation and the influence of the foods. The subjects of this study were 476 the 1st grade middle school students sampled from two public middle schools of Gyeonggi Province, Korea. Questionnaire contained 47 questions including the kinds of sugar containing processed foods, the frequency of intake, the frequency of normal diet, and the emotional test. All data obtained in this study were processed by SAS 8.2 program. 77.9% of the students had snacks more than once a day and the favored foods were in the order of bread, cookies, fruit, milk and dairy products, flour foods, ramen and ice cream. Less of the sugar contained processed snacks consumed high emotional intelligence showed(p<0.001). And low intake of carbohydrate processed snacks linked with having more regular meals and the higher levels of emotional intelligence. There was no gender difference in consuming simple sugar containing processed foods, however, out of these foods, carbonated drinks, ramen and pizza affected significantly the emotional intelligence of the male students. It was found that sugar contained processed snacks affect the eating habits like meal skipping and the emotional intelligence. It is determined that the effects of carbohydrate processed snacks on emotional intelligence can not be denied.

Activities of Enzymes Involved in Fatty Acid Metabolism in the Colon Epithelium of Piglets Fed with Different Fiber Contents Diets

  • The present study was conducted to evaluate the influence of dietary fiber on the activities of malic enzyme and citrate lyase involved in fatty acid metabolism in the colon epithelium of pigs. Thirty-six weaned 5 weeks old crossbred (Yorkshire${\times}$Swedish Landrace) piglets originating from twelve litters were randomly assigned to either a low fiber diet containing 10% non-starch polysaccharides (NSP), a control diet containing 14.7% NSP or a high fiber diet containing 20% NSP. The activity of malic enzyme in the colonic epithelium of pigs significantly (p<0.05) increased with age during the suckling-weaning transition. There was a tendency (p<0.10) of decreased malic enzyme activity in the colonic epithelium of pigs fed on the high fiber diet. At week 6, a lowered (p<0.01) activity of malic enzyme in pigs fed on the low fiber diet compared with that in pigs fed on the high fiber and the control diets. Nevertheless, there were no significant differences in the activity of citrate lyase observed either between pigs with different ages or between pigs fed with various diets. The current data suggest that piglets during the suckling-weaning transition have a limited capacity to synthesize fatty acids from carbohydrate derivatives in the coloncytes. In addition, lipogenesis in coloncytes was enhanced with age during the suckling-weaning transition. A tendency (p<0.10) to an increased capacity to utilize acetyl-CoA in coloncytes of pigs has been observed for the high fiber diet. Moreover, the present work indicated that dietary fiber resulted in a lowered rate of lipogenesis and a reduced activity of malic enzyme.
