Effect of Puffing in the Extraction of Active Ingredients from the Roots of Paeonia lactiflora and Astragalus membranaceus

  • In Asia, the roots of Paeonia lactiflora and Astragalus membranaceus have been used as therapeutic agents for thousands of years. Once the medicinal plants are harvested, they are dried and their ingredients are extracted by heat-mediated reflux extraction. However, the condensed structure of organic products (especially roots) limits the extraction of bioactive components. In this study, we assessed the effect of the puffing method (using high temperature and pressure) before the extraction process in relation to the profile and antioxidant capacity of active ingredients. We demonstrated that the additional puffing process before extraction methods improves the yield of polyphenol concentrations and antioxidant activities from the roots of P. lactiflora and A. membranaceus.

  • Feedwater heaters of many nuclear power plants have recently experienced severe wall thinning damange, which will increase as operating time progresses. Several nuclear power plants in Korea have experienced wall thinning damage in the area around the impingement baffle inside feed-water heater installed downstream of the turbine extraction stream line. At that point, the extract steam from the turbine is two phase fluid at high temperature, high pressure, and high speed. Since it flows to reverse direction after impinging the impingement baffle, the shell wall of feedwater heaters may be affected by flow-accelerated corrosion. In this paper, to compare wall thinning range according to change entrance nozzle diameter and position with reference numerical analysis model's wall thinning range, various numerical analysis models applied. In case of changing diameter, four different diameter is applied. And a side of nozzle position, two different position-vertical type and parallel type-is applied. And then this paper describes operation of numerical analysis which is composed similar condition with real feed water heater. In conclusion, this study shows effective design for shall wall thinning by changing nozzle diameter and position.

Stability of Proteasomes Extracted from Pressurized, Aged Skeletal Muscles

  • The present paper describes the effects of pressure and post-mortem aging treatments on in situ proteasome activity in rabbit and bovine skeletal muscles. Synthetic peptide hydrolyzing activity of rabbit proteasomes remained in the muscle after exposure to pressures up to 100 MPa. However, when a pressure of 400 MPa or more was applied, proteasomes were markedly inactivated. The extraction of proteasomes from excessively pressurized muscle appeared to be difficult. Proteasomes in aged muscle remained relatively stable throughout the aging process, with activity after 168 h (7 days) being 35%, 48%, 53% and 31% of the 0 h post-mortem LLVY, LSTR, AAF and LLE total hydrolyzing activities, respectively. The synthetic peptide hydrolyzing activities of bovine muscle proteasomes were similar to those of rabbit skeletal muscle proteasomes. The results suggest that synthetic peptide hydrolyzing activity remains in muscle exposed to relatively low pressures. Furthermore, it is known that high-pressure treatment induces fragmentation of myofibrils, modification of actin-myosin interaction and activation of intramuscular proteinases, cathepsins and calpains. Thus, proteasomes are probably involved in the tenderization process in combination with other intramuscular proteinases under high-pressure conditions. Our findings confirmed that proteasomes play a role in meat tenderization induced by high-pressure treatment or aging.

  • Subcutaneous cervicofacial, mediastinal emphysemas are complications associated with head and neck surgery, trauma, infectious processes, tooth extraction. Drill cooling stream and dental syringe air ject are the sources of high pressure air that may enter exposed soft tissue. Since the introduction of the high-speed air turbine drill in the 1960s, The incidence of iatrogenic subcutaneous emphysema has increased. Most cases begin to resolve after 2 to 3 days and residual swelling is usually minimal at the end of 7 to 10 days. Surgical approach is not advised because it is likely to be ineffective. The differential diagnosis of neck swelling after dental procedure includes hematoma, cellulitis, angioedema, allergic reaction, subcutaneous emphysema. We report a rare case of patient with subcutaneous cervicofacial emphysema and mediastinal emphysema secondary to third molar extraction.

Quality Characteristics of Fish, Crab and Red-Crab Stock Prepared by High Pressure Extract Method (고압 가열 추출 방식을 이용한 생선 육수, 붉은 대게 육수, 대게 육수의 품질 특성)

  • Using the high-pressure heating extraction process suitable for mass-production, crab stock(CS), red crab stock(RCS) and fish stock(FS) were manufactured and their quality and organic features were compared. The results are as follows. In mineral content, the content of essential amino acids, the content of delicious amino acids and preference, CS and RCS were rated higher than FS, and especially RCS had the most abundant essential amino acid content and had the highest rating in organic evaluation. Also, in terms of cost, RCS was more economical than regular CS, which is relatively expensive. Therefore, it is believed that replacing fish bones with RCS in making stock will be more cost-saving and have higher preference.

Screening of Skin Whitening Activity of Codonopsis lanceolata Extract by Complex Steaming Process (복합 고온 전처리 더덕 추출물의 미백 활성 탐색)

  • According to previous reports, antioxidant activities of Codonopsis lanceolata could be increased by a steaming process. This study was performed to improve its antioxidant activity and skin whitening activities of C. lanceolata by high pressure and stepwise steaming complex process. The complex processed C. lanceolata showed highest free radical scavenging acitivity as 45.21%, and for phenol and flavonoid contents, complex processed C. lanceolata contained higher than those from conventional extraction process or steaming process alone. The Cytotoxicity of all C. lanceolata extracts also showed low cytotoxicity against human fibroblast cell (CCD-986sk) as 4.49 ~ 10.40%. In whitening activity, high inhibition of tyrosinase activity was estimated as 25.08% by adding the extracts from complex process. We found that whitening and antioxidant activity of complex processed C. lancolata extract was higher than those obtained from conventional extraction and a steaming process because various kinds of antioxidant compounds could be easily released by combined process, compared to one of each process.

Optimal Extraction Conditions of Active Components from the Adventitious Roots of Noni (Morinda citrifolia) (노니 부정근으로부터 유효성분의 최적 추출조건)

  • To determine the optimum condition for the extraction of active components, noni adventitious roots were extracted under various conditions employing various solvents including methanolic water at different ratio (20, 40, 60, 80, and 100% water), extraction times and extraction methods. Anthraquinones, phenolic compounds, and flavonoids extraction using different solvents showed that 1 h of ultrasonic extraction was effective in 60-80% methanol, and 2 h of reflux extraction was effective in 80% methanol. To compare the extraction efficiency of active components according to different extraction methods and extraction times for noni adventitious roots, the active components were extracted by ultrasonic extraction, shaking extraction, reflux extraction, homogenizer extraction, high-pressure extraction, and soaking extraction. The highest phenolic contents were found in the extracted from ultrasonic extraction and anthraquinones and flavonoids contents were highest in the reflux extraction.

  • Thermoelastoplastic analysis of a typical device consisting of fixed tube and movable case having an inital clearance in between, which is subjected to pressure and thermal load, has been carried out to examine the cause of malfunction mainly at high temperature condition, and to improve the design. Stresses, deformed shape, interface state and their effects on normal function of case are discussed by using finite element method. The extraction energy can be remarkably reduced by changing the configuration of tube from the present design (Parallel type) to the improved design (Tapered type). This effect has been proved by sustained cyclic function test.

Process conditions of valuable compound for benign prostatic hyperplasia(BPH) from saw palmitto by supercritical CO2 (초임계 CO2를 이용한 Saw palmitto로부터 전립선 비대 유효 성분의 추출 조건)

  • Supercritical $CO_2$ extraction was applied to extraction of the valuable compound for benign prostatic hyperplasia from saw palmitto. Three levels of parameters, temperature $(35^{\circ}C,\; 60^<\circ}C)$, pressure 22∼30 Mpa and ethanol modified solvent concentration, were used as the independent variables of central composite to rotate the design. The extracting pressure and temperature interaction was significantly affected the extraction of valuable compounds. The ethanol was an efficient modifier to extract valuable compound by supercritical carbon dioxide. The optimal processing conditions were extracting pressure, 30 Mpa, extracting temperature, $60^<\circ}C$, ethanol modifier, 15% for extracts of the valuable compounds from saw palmitto. In conclusion, it was found that supercritical $CO_2$ extraction can successfully be applied to extract high quality extracts from saw palmitto.

Ultrasonic-assisted Micellar Extraction and Cloud-point Pre-concentration of Major Saikosaponins in Radix Bupleuri using High Performance Liquid Chromatography with Evaporative Light Scattering Detection

  • A new ultrasonic-assisted micellar extraction and cloud-point pre-concentration method was developed for the determination of major saikosaponins, namely saikosaponins -A, -C and -D, in Radix Bupleuri by high performance liquid chromatography with evaporative light scattering detection (HPLC-ELSD). The non-ionic surfactant Genapol X-080 (oligoethylene glycol monoalkyl ether) was chosen as the extraction additive and parameters affecting the extraction efficiency were optimized. The highest yield was obtained with 10% (w/v) Genapol X-080, a liquid/solid ratio of 200:1 (mL/g) and ultrasonic-assisted extraction for 40 min. In addition, the optimum cloud-point pre-concentration was reached with 10% sodium sulfate and equilibration at $60^{\circ}C$ for 30 min. Separation was achieved on an Ascentis Express C18 column (100 ${\times}$ 4.6 mm i.d., 2.7 ${\mu}M$) using a binary mobile phase composed of 0.1% acetic acid and acetonitrile. Saikosaponins were detected by ELSD, which was operated at a $50^{\circ}C$ drift tube temperature and 3.0 bar nebulizer gas ($N_2$) pressure. The water-based solvent modified with Genapol X-080 showed better extraction efficiency compared to that of the conventional solvent methanol. Recovery of saikosaponins ranged from 93.1 to 101.9%. An environmentally-friendly extraction method was successfully applied to extract and enrich major saikosaponins in Radix Bupleuri.
