Possible existence of tetrodotoxin-like toxins in cultured river puffer fish, Takifugu obscurus (양식산 황복에서 tetrodotoxin 유사 독소의 미량 존재 가능성 제시)

  • It was examined whether the common belief that "cultured puffer fishes do not contain tetrodotoxin (TTX)", the major lethal substance that accidently causes death in consumers of those fishes, is true in river puffer fish Takifugu obscurus. In mouse bioassay, lethal levels of toxins were detected in the ranks: gonad>liver>intestine>muscle>skin in wild puffer fish. In contrast, no mortality occurred in the mouse bioassay on cultured fish. However, there were sleepiness, sluggish behavior, and hind limb paralysis with the tissue extracts of cultured fish suggesting the presence of TTX or other similarly acting toxins. An attempt to confirm the presence of TTX in cultured fish with high performance liquid chromatography (HPLC) was not very successful. The results suggest possible existence of TTX toxins or similarly acting toxins.

The Effect of UV-B Irradiation and Hot-Air Drying on the Vitamin D$_2$Content of Shiitake Mushroom (Lentinus edodes) (자외선 B파 조사와 열풍건조가 표고버섯의 비타민 D$_2$함량에 미치는 영향)

  • Fresh Shiitake mushrooms were irradiated by UV-B or dried by using hot air to increase the vitamin D$_2$content. Vitamin D$_2$content was determined by high-performance liquid chromatography Irradiating the inside of the mushroom cap was more effective in increasing vitamin D$_2$content than irradiating the outside of the mushroom cap. Irradiation of UV-B at 0, 2.5, 5.0, 7.5 and 10 J/㎠ increased the content of vitamin D$_2$in the mushroom to 21.51, 120.78, 144.38, 168.08, and 187.69 $\mu\textrm{g}$/g, respectively. Vitamin D$_2$content of Shiitake mushrooms with irradiation of 10 J/㎠ decreased from 187.69 to 98.78 $\mu\textrm{g}$/g by 6 hours of hot-air drying. When three kinds of drying method (UV-B irradiation followed by hot-air drying, hot-air drying, and sunlight drying) were compared, the combination of UV-B irradiation and hot-air drying was the most effective in terms of increasine the vitamin D$_2$content in the mushroom.

Analysis of Gliotransmitters in ADHD Mice (ADHD (주의력결핍 과잉행동장애) 생쥐 모델에서의 별아교세포 유래 신경전달물질 분석)

  • Although the core mechanisms of Attention Deficit/Hyperactivity Disorder (ADHD) are unknown, several ADHD-associated proteins have been studied. G-protein - coupled receptor kinase interacting protein-1 (GIT1) is a multifunctional adapter protein that affects neuron growth and dendrite formation. GIT1-deficient mice have shown ADHD-like behavior and also recovered through amphetamine treatment. In this study, gliotransmitters were investigated in both intracellular and extracellular space from GIT1-deficient mice. To measure the amount of gliotransmitters, primary astrocyte cultures were taken from the cerebral and cerebellar cortices of wild (WT), hetero (HE), and knock-out (KO) mice. Major gliotransmitters were analyzed using high-performance liquid chromatography. It was observed that the amount of excitatory and inhibitory gliotransmitters were dependent on genotype and showed a change in excitation/inhibition ratios. Interestingly, the major excitatory gliotransmitter, glutamate, existed at the lowest level in WT mice, but the amount of inhibitory gliotransmitters, gamma-aminobutyric acid (GABA) and glycine, varied depending on brain region. Remarkably, an increased amount of GABA was measured at the intracellular cerebrum in WT mice compared with KO mice. It was presumed that KO mice would secrete more inhibitory gliotransmitters to compensate for GIT1 depletion or else acquire a defect to reuptake-secreted GABA. This may be a possible mechanism for ADHD pathology.

A STUDY OF MONOMER RELEASE FROM PIT AND FISSURE SEALANTS ACCORDING TO VARIOUS LIGHT SOURCES (광원에 따른 수종의 치면열구전색제로부터 용리되는 모노머에 관한 연구)

  • The aim of this study was to identify and quantify the major or detectable monomers released from any of five commercially-available, light-cured pit and fissure sealants with three different light sources : conventional halogen light curing unit, plasma arc light curing unit and LED curing unit. After curing, specimens were immediately immersed in distilled water for different time intervals. The time related release of monomers were analyzed by high performance liquid chromatography(HPLC). Identification and quantitative analysis of monomers were performed by the comparison of the elution time and the absorption peak height of the eluates with those of the authentic sample. The result of this study can be summarized as follows. 1. Standard solution peaks with retention times of 2.3, 3.2, 5.6, 6.5, 10.4 minutes were identified as BPA, TEGDMA, UDMA, Bis-GMA, Bis-DMA, respectively. 2. None of the chromatograms of the tested sealants displayed peaks with the same retention time as that of the standard solution, except for TEGDMA. 3. The highest release rate of TEGDMA was observed during the 12hr period for all samples and declined thereafter. 4. The elution of TEGDMA from curing with Halogen curing unit for 20 second and LED for 10 second was less than that resulting from curing with Plasma arc for 3 second. 5. TEGDMA was detected at much lower levels in eluates from the Pit & Fissure $Sealant^{TM}$ than other sealants. The elution of TEGDMA from the $Helioseal^{(R)}$ F cured with Halogen light curing unit, the $Concise^{TM}$ cured with Plasma arc curing unit and the $Teethmate^{(R)}$ F-1 cured with LED curing unit were higher than other sealants.

  • Objectives : This study was performed to investigate the discriminative criteria of 6 kinds of Achyranthis Radix by HPLC/DAD. Methods : 20-hydroxyecdysone is isolated by silica gel column chromatography ($CHCl_3$:MeOH, 7:1-1:1 v/v) and identified by nuclear magnetic resonance, A high-performance liquid chromatographic method with diode array detection was used to identify 20-hydroxyecdysone in A. japonica. The analysis was performed using $C_{18}$ column with isocratic elution consisted of 18% acetonitrile and 82% water and the detection was carried out by DAD at 254 nm. 6 kinds of Achyranthis Radix from different locations were extracted in MeOH. Each extracts was analyzed by HPLC in same condition as used in analysis of 20-hydroxyecdysone. The identities of each extracts were determined by comparing the retention time and UV spectrum with that of reference compound. Results : 1. A. japonica and A. bidentata showed the similar patterns of HPLC chromatogram and 20-hydroxycedysone was present in both of them because the peaks having the same retention time and UV spectrum as 20-hydroxyecdysone were shown in the HPLC chromatograms of A. japonica and A. bidentata 2. Cyathula officinalis and C. capitata showed the similar patterns of HPLC chromatogram. The peak having the same retention time and UV spectrum as 20-hydroxyecdysone was shown in the HPLC chromatogram of C. capitata but not shown in the HPLC chromatogram of C. officinalis. 3. Two species of medicinal drugs from Sacheon province showed similar patterns of HPLC chromatogram. Achyranthis Radix from Sacheon(wild) did not have 20-hydroxycedysone but Achyranthis Radix from Sacheon(cultivated) showed the peak having the same retention time as 20-hydroxyecdysone but UV spectrum of the peak was different from that of 20-hydroxyecdysone. Conclusions : These results suggested that 20-hydroxyecdysone could be the discriminative criteria for Achyranthis Radix contain 20-hydroxyecdysone though they belong to different genus and species. And the patterns of HPLC chromatogram also could be the discriminative criteria as the different species of Achyranthis Radix belonging to the same genus showed similar patterns of HPLC chromatogram.

Phenolic Compounds in Sweet Potatoes and Their Antioxidative Activity (고구마 페놀화합물의 항산화 활성)

  • The phenolic compounds of Korean sweet potatoes, Mokpo 18 and Yulmi, were extracted by using 70%-methanol and the extracts(ME) were fractionated and obtained three fractions such as free phenolic acid(FPAF), soluble phenolic acid ester(SPAF) and insoluble bound phenolic acid(BPAF) fractions. The antioxidative activities(AA) was represented as the peroxide values(POVs). The POVs were calculated by measuring the oxidation of linoleic acid and lard emulsions at $60^{\circ}C$. AA of FPAF has shown the most effective. AA of FPAF were more effective than those of ME in both Yulmi and Mokpo 18. AA of the ME of Mokpo 18 were more effective than those of Yulmi, however, those of FPAF in Ulmi were more effective than in Mokpo 18. The POVs of ME and FPAF of the peel part in both sweet potatoes were more effective than those of peeled part. The qualitative and quantitative analysis of the phenolic compounds in both sweet-potatoes were performed by using high performance liquid chromatography(HPLC) and the major phenolic compounds were identified as chlorogenic acid and caffeic acid. The contents of caffeic acid were 0.684mg/g in the peel part and 0.028mg/g in the peeled part of Yulmi and 0.472mg/g in the peel part and 0.046mg/g in the peeled part of Mokpo 18 and those of chlorogenic acid was 0.674mg/g, 0.926mg/g, and 0.012mg/g, respectively. In comparative test of antioxidative activities between a standard chlorogenic acid and caffeic acid, AA of caffeic acid were more effective than those of chlorogenic acid.

Korean Red Ginseng attenuates anxiety-like behavior during ethanol withdrawal in rats

  • Background: Korean Red Ginseng (KRG) is known to have antianxiety properties. This study was conducted to investigate the anxiolytic effects of KRG extract (KRGE) during ethanol withdrawal (EW) and the involvement of the mesoamygdaloid dopamine (DA) system in it. Methods: Rats were treated with 3 g/kg/d of ethanol for 28 d, and subjected to 3 d of withdrawal. During EW, KRGE (20 mg/kg/d or 60 mg/kg/d, p.o.) was given to rats once/d for 3 d. Thirty min after the final dose of KRGE, anxiety-like behavior was evaluated in an elevated plus maze (EPM), and plasma corticosterone (CORT) levels were determined by a radioimmunoassay (RIA). In addition, concentrations of DA and 3,4-dihydroxyphenylacetic acid (DOPAC) in the central nucleus of the amygdala (CeA) were also measured by high performance liquid chromatography (HPLC). Results: The EPM test and RIA revealed KRGE inhibited anxiety-like behavior and the over secretion of plasma CORT during EW. Furthermore, the behavioral effect was blocked by a selective DA D2 receptor (D2R) antagonist (eticlopride) but not by a selective DA D1 receptor (D1R) antagonist (SCH23390). HPLC analyses showed KRGE reversed EW-induced decreases of DA and DOPAC in a dose-dependent way. Additionally, Western blotting and real-time polymerase chain reaction (PCR) assays showed that KRGE prevented the EW-induced reductions in tyrosine hydroxylase (TH) protein expression in the CeA and TH mRNA expression in the ventral tegmental area (VTA). Conclusion: These results suggest that KRGE has anxiolytic effects during EW by improving the mesoamygdaloid DA system.

Investigation of ginsenosides in different tissues after elicitor treatment in Panax ginseng

  • Background: The effect of methyl jasmonate (MJ) on ginsenoside production in different organs of ginseng (Panax ginseng Meyer) was evaluated after the whole plant was dipped in an MJ-containing solution. MJ can induce the production of antioxidant defense genes and secondary metabolites in plants. In ginseng, MJ treatment in adventitious root resulted in the increase of dammarenediol synthase expression but a decrease of cycloartenol synthase expression, thereby enhancing ginsenoside biosynthesis. Although a previous study focused on the application of MJ to affect ginsenoside production in adventitious roots, we conducted our research on entire plants by evaluating the effect of exogenous MJ on ginsenoside production with the aim of obtaining new approaches to study ginsenoside biosynthesis response to MJ in vivo. Methods: Different parts of MJ-treated ginseng plants were analyzed for ginsenoside contents (fine root, root body, epidermis, rhizome, stem, and leaf) by high-performance liquid chromatography. Results: The total ginsenoside content of the ginseng root significantly increased after 2 d of MJ treatment compared with the control not subjected to MJ. Our results revealed that MJ treatment enhances ginsenoside production not in the epidermis but in the stele of the ginseng root, implying transportation of ginsenosides from the root vasculature to the epidermis. Application of MJ enhanced protopanaxadiol (PPD)-type ginsenosides, whereas chilling treatment induced protopanaxatriol (PPT)-type ginsenosides. Conclusion: These findings indicate that the production of PPD-type and PPT-type ginsenosides is differently affected by abiotic and biotic stresses in the ginseng plant, and they might play different defense mechanism roles.

In vitro and in vivo anti-inflammatory activities of Korean Red Ginseng-derived components

  • Background: Although Korean Red Ginseng (KRG) has been traditionally used for a long time, its anti-inflammatory role and underlying molecular and cellular mechanisms have been poorly understood. In this study, the anti-inflammatory roles of KRG-derived components, namely, water extract (KRG-WE), saponin fraction (KRG-SF), and nonsaponin fraction (KRG-NSF), were investigated. Methods: To check saponin levels in the test fractions, KRG-WE, KRG-NSF, and KRG-SF were analyzed using high-performance liquid chromatography. The anti-inflammatory roles and underlying cellular and molecular mechanisms of these components were investigated using a macrophage-like cell line (RAW264.7 cells) and an acute gastritis model in mice. Results: Of the tested fractions, KGR-SF (but not KRG-NSF and KRG-WE) markedly inhibited the viability of RAW264.7 cells, and splenocytes at more than 500 mg/mL significantly suppressed NO production at $100{\mu}g/mL$, diminished mRNA expression of inflammatory genes such as inducible nitric oxide synthase, cyclooxygenase-2, tumor necrosis factor-${\alpha}$, and interferon-${\beta}$ at $200{\mu}g/mL$, and completely blocked phagocytic uptake by RAW264.7 cells. All three fractions suppressed luciferase activity triggered by interferon regulatory factor 3 (IRF3), but not that triggered by activator protein-1 and nuclear factor-kappa B. Phospho-IRF3 and phospho-TBK1 were simultaneously decreased in KRG-SF. Interestingly, all these fractions, when orally administered, clearly ameliorated the symptoms of gastric ulcer in HCl/ethanol-induced gastritis mice. Conclusion: These results suggest that KRG-WE, KRG-NSF, and KRG-SF might have anti-inflammatory properties, mostly because of the suppression of the IRF3 pathway.

Chemical Composition and Biological Activities of Immunostimulants Purified from Alkali Extract of Poria cocos Sclerotium (복령 균핵의 알칼리추출물에서 정제한 면역활성 증강물질의 작용과 화학구성)

  • An one percent sodium carbonate extract prepared from sclerotia of Poria cocos activated the proliferation of the T lymphocytes as measured by mixed lymphocyte responses(MLR). The active fraction, PCSC22, was isolated from an one percent sodium carbonate extract by a combination of fractionation procedures, including ethanol precipitation and chromatographies on column of DEAE-cellulose and Sephadex G50. Carbohydrate and peptide contained in PCSC22 were 78 : 22% in ratio. On employing gel filtration high performance liquid chromatography, PCSC22 exhitited a homogeneous peak with an average molecular weight of 8 kDa. The sugar moiety of PCSC22 was composed with mannose (92%), galactose (6.2%) and arabinose (1.3%), which might be indicated as heteromannan. Fifteen amino acids were found in peptide moiety of the polysaccharide and aspartic acid, serine, and valine were major components. PCSC22 activated the primary proliferation of T lymphocytes measured by mixed lymphocyte responses, the antibody production of the B lymphocytes and the secretion of nitric oxide from macrophage cell line, RAW264.7.
