Development of Host-Vector Systems for Lactic Acid Bacteria (유산균의 Host-Vector System 개발)

  • Lactic acid bacteria (LAB) are widely used for various food fermentation. With the recent advances in modern biotechnology, a variety of bio-products with the high economic values have been produced using microorganisms. For molecular cloning and expression studies on the gene of interest, E. coli has been widely used mainly because vector systems are fully developed. Most plasmid vectors currently used for E, coli carry antibiotic-resistant markers. As it is generally believed that the antibiotic resistance markers are potentially transferred to other bacteria, application of the plasmid vectors carrying antibiotic resistance genes as selection markers should be avoided, especially for human consump-tion. By contrast, as LAB have some desirable traits such that the they are GRAS(generally recognized as safe), able to secrete gene products out of cell, and their low protease activities, they are regarded as an ideal organism for the genetic manipulation, including cloning and expression of homologous and heterologous genes. However, the vec-tor systems established for LAB are stil insufficient to over-produce gene products, stably, limiting the use of these organisms for industrial applications. For a past decade, the two popular plasmid vectors, pAM$\beta$1 of Streptococcus faecalis and pGK12 theB. subtilis-E. coli shuttle vector derived from pWV01 of Lactococcus lactis ssp. cremoris wg 2, were most widely used to construct efficient chimeric vectors to be stably maintained in many industrial strains of LAB. Currently, non-antibiotic markers such as nisin resistance($Nis^{r}$ ) are explored for selecting recombi-nant clone. In addition, a gene encoding S-layer protein, slp/A, on bacterial cell wall was successfully recombined with the proper LAB vectors LAB vectors for excretion of the heterologous gene product from LAB Many food-grade host vec-tor systems were successfully developed, which allowed stable integration of multiple plasmid copies in the vec-mosome of LAB. More recently, an integration vector system based on the site-specific integration apparatus of temperate lactococcal bacteriophage, containing the integrase gene(int) and phage attachment site(attP), was pub-lished. In conclusion, when various vector system, which are maintain stably and expressed strongly in LAB, are developed, lost of such food products as enzymes, pharmaceuticals, bioactive food ingredients for human consump-tion would be produced at a full scale in LAB.

Ameliorating Effects of Sulfonylurea Drugs on Insulin Resistance in Otsuka Long-Evans Tokushima Fatty Rats

  • OLETF (Otsuka Long-Evans Tokushima Fatty) rats are characterized by obesity-related insulin resistance, which is a phenotype of type 2 diabetes. Sulfonylurea drugs or benzoic acid derivatives as inhibitors of the ATP-sensitive potassium $(K_{ATP})$ channel are commercially available to treat diabetes. The present study compared sulfonylurea drugs (glimepiride and gliclazide) with one of benzoic acid derivatives (repaglinide) in regard to their long-term effect on ameliorating insulin sensitivity in OLETF rats. Each drug was dissolved and fed with drinking water from 29 weeks of age. On high glucose loading at 45 weeks of age, response of blood glucose recovery was the greatest in the group treated with glimepiride. On immunohistochemistry analysis for the Kir6.2 subunit of $K_{ATP}$ channels, insulin receptor ${\beta}$-subunits, and glucose transporters (GLUT) type 2 and 4 in liver, fat and skeletal muscle tissues, the sulfonylurea drugs (glimepiride and gliclazide) were more effective than repaglinide in recovery from their decreased expressions in OLETF rats. From these results, it seems to be plausible that $K_{ATP}$-channel inhibitors containing sulfonylurea moiety may be much more effective in reducing insulin resistance than those with benzoic acid moiety. In contrast to gliclazide, non-tissue selectivity of glimepiride on $K_{ATP}$ channel inhibition may further strengthen an amelioration of insulin sensitivity unless considering other side effects.

Galactinol is Involved in Induced Systemic Resistance against Bacterial Infection and Environmental Stresses

  • We previously demonstrated that root colonization of the rhizobacterium, Pseudomonas chlororaphis O6, induced expression of a galactinol synthase gene (CsGolS1), and resulting galactinol conferred induced systemic resistance (ISR) against fungal and bacterial pathogens in cucumber leaves. To examine the role of galactinol on ISR, drought or high salt stress, we obtained T-DNA insertion Arabidopsis mutants at the AtGolS1 gene, an ortholog of the CsGolS1 gene. The T-DNA insertion mutant compromised resistance induced by the O6 colonization against Erwinia carotovora. Pharmaceutical application of 0.5 - 5 mM galactinol on roots was sufficient to elicit ISR in wild-type Arabidopsis against infection with E. carotovora. The involvement of jasmonic acid (JA) signaling on the ISR was validated to detect increased expression of the indicator gene PDF1.2. The T-DNA insertion mutant also compromised tolerance by increasing galactinol content in the O6-colonized plant against drought or high salt stresses. Taken together, our results indicate that primed expression of the galactinol synthase gene AtGolS1in the O6-colonized plants can play a critical role in the ISR against infection with E. carotovora, and in the tolerance to drought or high salt stresses.

A Study on Chromium Electroplating of Piston Ring Groove's Surface (엔진피스톤링 홈의 크롬도금에 관한 연구)

  • When the low heavy oil is using as fuel oil to the diesel engine, some problems such as corrosion resistance, wear resistance and heat resistance are happened in diesel engine's internal material, especially the adhesive wear of piston ring groove was occurred as a important problem. Therefore to prevent adhesive wear of its groove, the surface of its groove used to be electroplated with Chrominum and for its Chromium electroplating, Fe anode is being used until nowadays because of its Special shape. However in case of using Fe anode, there were some problems such as deterioation of solution, property of Chromium film, and condition of coation. In this paper Pb anode electroplated withPb to the steel plate was investigated for its Chromium electroplating for Pb's high corrosion resistance in acid solution, and Pb anode is not dissolved compared with Fe anode and deterioration degree of solution in case of Pb anode is smaller than that of Fe anode and also property of Chromium film was better than that of Fe anode. Moreover it was known that the optimum cathodic current density for Pb electroplating to steel plate as insoluable anode for Chromium coating of piston ring groove is 30mA/$cm^2$ by experimental results obtained.

Prevalence, species, and antimicrobial resistance of Acinetobacter in surgical practice and laboratory dog husbandry room environments

  • Acinetobacter is a bacteria found in the environment and clinical specimens, causing nosocomial infection and antimicrobial resistance (AMR) threats. This study examined the prevalence, species, and AMR characteristics of Acinetobacter isolated from surgical practice and the laboratory dog husbandry room environments (n = 235) at Rajamangala University of Technology Tawan-ok veterinary hospital during 2018-2019. The prevalence of Acinetobacter in the laboratory dog husbandry room and veterinary belongings were 2.55% and 0.43%, respectively. Species determination was Acinetobacter hemolyticus (2.13%) and Acinetobacter baumannii (0.43%) from environments in the laboratory dog husbandry room, and Acinetobacter junii (0.43%) from the shoes used in the surgical practice room. AMR was observed in both study environments and the specimens sent to the Veterinary Diagnostic Center. These isolates had a high resistant percentage to amoxicillin-clavulanic acid (84.62%), sulfamethoxazole-trimethoprim (61.54%), and cephalexin (53.85%) but were susceptible to imipenem. Compared to the isolates recovered from the clinical specimens, most isolates derived from environments exhibited multidrug resistance and shared correlated resistance patterns. These results highlight the need for sanitization in the dog husbandry room. Furthermore, the AMR results can be used as a preliminary baseline for studying AMR Acinetobacter contamination in animals and their environments.

  • First of all, the properties imparted to PET fabrics are resistance to and recovery from creasing or wrinkling when wet or dry; high resistance to stretch in the filament yarns but not in the staple; high abrasion resistance; good texture and appearance; resistance to heat ageing; good chemical resistance and good resistance, behind glass, to sunlight. But, the low moisture regain of PET fabric conduces to static troubles in textile processing. Furthermore, garments made from PET may, during wear, develop electric charges which attract to the fabric particles of soil(dirt, swarf, dust) flying in the air, so that the cuffs of shirts, for example, become soiled quickly and are not easily laundered clean. The sericin constitutes 25$\sim$30% of silk protein and surrounds the fibroin fiber with sticky layer that supports the formation of a cocoon. The useful biochemical properties of sericin protein are oxidation resistant, antibacterial, UV resistant, hydrophilic property, and good affinity with hydrophobic material. These properties can be used as an improving reagent or a coating agent for natural and synthetic fibers, fabrics, and other intermediate products. The sericin is also applied to cross-link, and can be blended with other materials. In this study, we modified the surface of PET fabric by mixture of sericin finishing agent; sericin, polyuretane binder and 1,2,3,4-butanetetracarboxylic acid (BTCA) cross-link agent. Also, we investigated the finshing effect; moisture regain, stiffness, handle, drape and electrostatic. The moisture regain of PET fabric treated with sericin finishing agent was higher than that of untreated PET fabric. As a result of evaluating influence about handle of PET fabrics treated with sericin finishing agent, it was confirmed that the sericin finishing agent could be use as a linen like finishing agent.

Drug Resistance and R-Plasmid Profile of Shigella Isolated in 1986-1988 (이질균의 약제 내성과 R-Plasmid)

  • These studies were made to assess the present stage of resistance of Shigella species to antibiotics and to find characteristics of R-plasmid of these bacteria. From 1986 to 1988, 125 strains of Shigella species were isolated from patients specimens collected in Chung Cheong-do Hospital, Kyunghee Medical Center, city an provincial health & environmental institutes. These specimens were tested for resistance to 12 kinds of antimicrobial agents by agar dilution method. Using Muller-Hinton agar for the assay of drug resistance and Trypticane Soy Broth as propagating medium for conjugation. All the strains (100%) were resistant to one or more antibiotics. Drug resistance patterns of isolated strains were found as the highest resistance to ampicillin (98%) in 1986, to tetracycline (98%) in 1987, to tetracycline (100%) in 1988, all strains were sensitive to gentamicin, amikacin, tobramycin. Chronologically, resistance decreased gradually as it was shown in relation to kanamycin, rifampicin in 1986, 1987 and 1988, (4%, 2%) (4%, 2%) (0%, 0%) respectively. But, resistance was increased year by year as it was shown in relation to tetracycline, nalidixic acid, streptomycin in 1986, 1987, 1988 (89%, 19%, 45%) (98%, 46%, 71%) and (100%, 58%, 88%). The resistance in correlation to more than 5 drugs, which was 13 strains among 47 strains in 1986, 38 strains among 87 strains in 1987, 23 strains among 26 strains in 1988, was increased gradually. In the transfer test of drug resistance by conjugation methods, the rate which was 3 strains (50%) in 1986, 8 strains (62%) in 1987, 3 strains (100%) in 1988, was increased gradually. When the donor strains were conjugated with the recipient strains, the conjugation rate was high in the multiple resistant strains. The relationships of transferring patterns of drug resistance and molecular weight of R-plasmid were variable. However, only a plasmid which has more than 35 Mgd was transferred.

Genomic Insights of Weissella jogaejeotgali FOL01 Reveals Its Food Fermentation Ability and Human Gut Adaptive Potential for Probiotic Applications in Food Industries

  • Although the genus Leuconostoc, generally found in various fermented foods, has often been suggested to be a novel probiotic for food fermentation and health promotion, the strains in this genus showed low acid tolerance and low osmotic stress resistance activities, which are required for survival during food fermentation events. Recently, a novel species of Weissella, W. jogaejeotgali $FOL01^T$ (= KCCM 43128 = JCM 30580), was isolated from Korean fermented clams. To determine the genomic features of this new species, its genome was completely sequenced and analyzed. The genome consists of a circular chromosome of 2,114,163 bp of DNA with a G+C content of 38.8%, and the plasmid pFOL01 consists of 35,382 bp of DNA with a G+C content of 39.1%. The genome analysis showed its potential for use in food fermentation and osmotic stress resistance abilities for processing in food industries. In addition, this strain was predicted to have acid tolerance and adhesion to the mucosal layer for survival and colonization in the gut. Subsequent experiments substantiated these abilities, suggesting that W. jogaejeotgali may have probiotic potential and a high survival rate during food fermentation. Therefore, it may be suitable as a novel probiotic strain for various applications in food industries.

Physiological Characteristics and Immunomodulation Activity of Lactobacillus fermentum 450 isolated from Raw Milk (원유에서 분리한 Lactobacillus fermentum 450의 생리적 특성과 면역활성)

  • The aim of this study was to investigate the physiological characteristics and immunomodulatory activity of Lactobacillus fermentum 450 isolated from raw milk. L. fermentum 450 showed optimum growth at $40^{\circ}C$ and exhibited immunomodulatory effects on $interleukin-1{\alpha}$, tumor necrosis $factor-{\alpha}$, and nitrous oxide at concentrations of >2,500 pg/mL, >2,000 pg/mL, and $11.55{\pm}2.95{\mu}M$, respectively. Of the 16 antibiotics tested, L. fermentum 450 exhibited the highest sensitivity to rifampicin, followed by penicillin-G, and the highest resistance to kanamycin, followed by neomycin and polymyxin B. The strain showed high acid phosphatase activity and was comparatively tolerant to bile juice and acid. Moreover, the strain displayed high resistance to Salmonella Typhimurium (63.86%). These results demonstrate that L. fermentum 450 has potential for use as a probiotic with immunomodulatory activity.

Effect of Solution Temperature for Al Alloy Anodizing on Cavitation Characteristics (캐비테이션 특성에 미치는 알루미늄 합금의 양극 산화 용액 온도의 영향)

  • The commercialization of aluminum had been delayed than other metals because of its high oxygen affinity. Anodizing is a process in which oxide film is formed on the surface of a valve metal in an electrolyte solution by anodic oxidation reaction. Aluminum has thin oxide film on surface but the oxide film is inhomogeneous having a thickness only in the range of several nanometers. Anodizing process increases the thickness of the oxide film significantly. In this study, porous type oxide film was produced on the surface of aluminum in sulfuric acid as a function of electrolyte temperature, and the optimum condition were determined for anodizing film to exhibit excellent cavitation resistance in seawater environment. The result revealed that the oxide film formed at $10^{\circ}C$ represented the highest cavitation resistance, while the oxide film formed at $15^{\circ}C$ showed the lowest resistance to cavitation in spite of its high hardness.
