식물성 항균소재 처리가 Botrytis cinerea의 생리기능에 미치는 저해효과 (Inhibitory Effect of Grapefruit Seed Extract Mixture on the Physiological Function of Botrytis cinerea)

  • BAAG의 첨가배지상에서 미생물세포의 생육이 크게 억제된다는 실험결과를 토대로, 미생물에 대한 항균작용의 방식을 알아 보고자, Botrytis cinerea를 배양하여 분리한 여러 가지 효소 활성에 미치는 영향을 조사하였다. 특히 에너지 대사와 관련된 효소들에 대한 영향을 살펴보기 위하여 여러가지 대사경로에서 주요한 역할을 하는 효소인, hexokinase,glucose 6-phosphate dehydrogenase, malate dehydrogenase와succinate dehydrogenase에 대한 영향을 살펴 본 결과, BAAG가 외ucose-6-phosphate dehydrogenase 및 malate dehydrogenase의활성에는 별다른 효과를 나타내지 않았으나 succinate dehydrogenase 및 hexokinase 효소활성에서는 약간의 효소활성 억제작용을 관찰할 수 있었다. 또한, 세포막에 미치는 영향을 알아 보기 위하여 $\beta$-galactosidase의 인위적 기질인 ONPG의 분해 여부를 조사한 결과, BAAG는 세포막의 기능에 영향을 끼침을 관찰하였다. BAAG의 항균 성분을 분리확인 하기 위해서 silica gel column chromatography를 하여 ethyl ether분획물에서 3개, ethyl acetate분회물에서 4개의peak를 얻었으며, 각 용매분획물에서 1개씩의 분획에서 강한항균 활성을 검출하였다. 이 분획을 NMR 및 fast atomicbombardment측정기에 의해서 분석한 결과, 화학구조를limonin과 naringin으로 동정할 수 있었다. 이상의 연구결과로 미루어, 본 연구에서 그 기능과 작용을 구명한 천연항균소재 BAAG는 수확한 과채류의 직접 처리제재 및 항균 포장 소재로 과채류의 선도유지를 목적으로 활용하여 그 효과를 기대할 수 있어, 과채류의 생산기반 보호와 농가소득에큰 도움이 될 것으로 생각된다.

Stress-Governed Expression and Purification of Human Type II Hexokinase in Escherichia coli

  • The full encoding sequence for human type II hexokinase (HXK II) was cloned into the E. coli expression vector pET 21b and expressed as a C-terminally hexahistidine-tagged protein in the BL2l (DE3) strain. The IPTG-induced HXK II approximately accounted for 17% of the total E. coli proteins, and 81% of HXK $II_{6{\times}His}$ existed in inclusion bodies. To improve the production of soluble recombinant HXK II protein, in the functionally active form, we used low temperature, and the osmotic stress expression method. When expressed at $18^{\circ}C$, about 83% of HXK $II_{6{\times}His}$ existed in the soluble fraction, which amounted to a 4.1-fold yield over that expressed at $37^{\circ}C$. The soluble form of HXK $II_{6{\times}His}$ was also highly produced in the presence of 1M sorbitol under the standard condition $(37^{\circ}C)$, which indicated that temperature downshift and low water potentials were required to improve the yield of active recombinant HXK II protein. The expressed protein was purified by metal chelate affinity chromatography performed in an IDA Excellose column charged with $Ni^{2+}$ ions, resulting in about 40mg recombinant HXK II protein obtained with purity over 89% from 51 of E. coli culture. The identity of HXK $II_{6{\times}His}$ was confirmed by Western blotting analysis. Taken together, using the stress-governed expression described in this study, human active HXK II can be purified in sufficient amounts for biochemical and biomedical studies.

  • By spectrophotometric assay method, the following enzymes could be detected in Dunaliella tertiolecta and Chlorella pyrenoidosa cell-free extracts: Hexokinase; Glucose-6-phosphate, 6-Phosphogluconate and Triosephosphate dehydrogenase; Transketolase; Phosphogluco and Ribosephosphate isomerase; Phosphoglucomutase; Phosphofructokinase; Fructosediphosphate aldorase and Ribulosephosphate 3-epimerase. Such enzymes are in accordance with the proposed pathway of glucose catabolism by D. tertiolecta as well as C. pyrenoidosa. Also, it could be estimated, under the presence of NADP, that pentose phosphate pathway were more active than glycolytic pathway in D. tertiolecta cell-free systems.

  • Transgluaminase반응을 이용하여 ATP analog들$(C^8-ATP와 N^6-ATP)$을 casein에 고정화하였다. 고정화 ATP는 hexokinase에 의해 탈인산화되어 약55%가 ATP형으로 전환되었으며, 이는 acetate kinase에 의해 역으로 인산화되어 약80%가 ATP형으로 전환 되었다. $\beta$-Casein은 $$\alpha$_s_1-casein$에 비하여 glutamine 잔기의 수가 많으며,$N^6-ATP$의 경우 이의 alkyl carbamyl기와 casein의 carboxamide기 간의 정전기적 반발에 원인이 있는 것으로 보인다. ATP는 고정화하므로써 안정성이 증대되었으며, 고정화 ATP의 km치는 유리상의 ATP및 ATP analog의 그것과 비슷하였으나, 최대속도는 감소되었다. 고정하 ATP는 반응액에서 calcium의 첨가로 쉽게 침전되어 용액으로부터 거의 완전히 회수 되므로써, 반응액으로부터의 회수분리가 가능한 이점을 가졌다.

식이 내 곡류 종류와 지방수준이 성장기 이후 비만유도 흰쥐의 당대사와 항혈전능에 미치는 영향 (The Impact of Kinds of Dietary Grain and Dietary Lipid Level on the Glucose Metabolism and Antithrombogenic Capacity of Full Grown Obesity Induced Rats)

  • This study was designed to evaluate impact of kinds of dietary grain and dietary lipid level on the glucose metabolism and antithrombogenic capacity in obesity induced rats. Total of 80 Sprague-Dawley male rats were raised for one month with control diet containing $50\%$ (w/w) well-milled rice powder and $20\%$(w/w) of dietary lipids. The rats were blocked into 8 groups and raised for two months with diets containing well-milled rice, brown rice, black rice, or glutinous barley powder and 8 or $20\%$(w/w) of dietary lipids. The contents of total dietary fiber in experimental grains were in following order; glutinous barley > black rice > brown rice > well-milled rice. Weekly food intake were lower in glutinous barley group among all experimental groups. Body weight gain was high in high level of fat groups ($50\%$w/w) than medium level of fat groups ($8\%$ w/w). Plasma glucose concentration was not different significantly in each groups. But brown rice group was a little lower than others. Plasma insulin concentration was lower in black rice and glutinous barley group than rice group. Plasma glucagon concentration did not differ significantly among all experimental groups. Hexokinase activities in skeletal muscle are different significantly according to level of dietary fat and grain variety factors. Brown rice group was significantly highest among all experimental groups in hexokinase activity. Plasma $TXB_2$ concentrations in black rice and glutinous barley groups were lower as compared to rice and brown rice groups. Plasma 6-keto-$PGF_{1\alpha}$ concentrations in glutinous barley group was higher as compared to others. In conclusion brown rice has a little lowering effect glucose concentration. Black rice and glutinous barley intakes enhance antithromboenic capacity. It is suggested that the intakes of mixed gains are recommend.

경직상태의 근육막에 MgADP를 첨가시킨 결과에 대한 연구 (A Study on The Effect of Added MgADP to The Rigor Muscle Membrane)

  • To study the relationship between elementary biochemical states and structural states of the actomyosin crossbridges in muscle, the effects of binding of MgADP to myosin heads in the rigor muscle were examined by X-ray diffraction using synchrotron radiation. X-ray diffraction studies have been made to investigate the effects of binding of ADP on the structure of glycerinated rabbit skeletal muscle in the rigor state. The intensity increase was accompanied by a slight but distinct decrease in the 5.9 am layer-line intensity close to the meridian. These results strongly suggest that myosin heads altered their attached conformation in the proximal end toward the plane perpendicular to the fiber axis when MgADP was bound to them. We found that the intensity of the 14.5 nm-based meridional reflections increase by 20-50% when MgADP was added to the rigor muscle in the presence of hexokinase and myokinase inhibitor.

Effect of Dissolved Oxygen Concentration on the Metabolism of Glucose in Pseudomonas putida BM014

  • The effect of dissolved oxygen concentration on the metabolism of glucose in Pseudomonas putida BM014 was investigated. Glucose was completely converted to 2-ketogluconate via extracellular oxidative pathway and then taken up for cell growth under the condition of sufficient dissolved oxygen concentration. On the other hand, oxygen limitation below dissolved oxygen tension (DOT) value of 20% of air saturation caused the shift of glucose metabolism from the extracellular oxidative pathway to the intracellular phosphorylative pathway. Specific activities of hexokinase and gluconate kinase in intracellular phosphorylation pathway decreased as the DOT increased, while 2-ketogluconokinase activity in extracellular oxidative pathway increased under the same condition. This result can be usefully applied to microbial transformation of glucose to 2-ketogluconate, the synthetic precursor for iso-vitamine C, with almost 100% yield via extracellular oxidation by simple DOT control.

오미자, 매실 및 당밀을 함유하는 한방스포츠음료가 실험동물의 지구력과 에너지 대사 성분에 미치는 영향 (Effects of Herbal Sports Drinks with Omija, Maesil and Molasses on the Endurance and Energy Metabolites of Experimental Animals)

  • 한방재료 즉 오미자 및 매실 추출물과 당밀이 혼합된 미네랄 음료의 항피로효과 및 지구력 증진효과를 연구하기 위하여 ICR 생쥐의 강제수영능력 및 생화학적 요인을 측정하였다. 6% 당과 전해질용액에 오미자추출물, 매실추출물 및 당밀을 혼합한 음료들을 실험군들에게 4주 동안 자유로이 섭취시켰다. 음료들을 섭취한 10 group의 실험동물들은 28일 후 강제수영하였으며 한방음료를 섭취한 실험군(6번 음료; 190.8${\pm}$25.6 sec, 10번 음료; 173.6${\pm}$21.8 sec; p<0.05)의 강제수영시간은 대조군(93.2${\pm}$10.4 sec)에 비하여 1.5${\sim}$2배 증가하였다. 또한 6번 음료 및 10번 음료의 hexokinase 및 citrate synthase 활성 역시 대조군에 비하여 유의적으로 높았다 (hexokinase 활성 - 대조군:5.23${\pm}$0.38 ${\mu}mol$/g tissue; 6번 음료: 5.99${\pm}$0.18 ${\mu}mol$l/g tissue, 10번 음료: 6.13${\pm}$0.25 ${\mu}mol$/g tissue, p<0.05, citrate synthase 활성 - (대조군: 42.9${\pm}$1.87 ${\mu}mol$/g tissue; 6번 음료: 56.8${\pm}$3.98 ${\mu}mol$/g tissue, 10번 음료; 59.5${\pm}$3.09 ${\mu}mol$/g tissue, p<0.05). 한방음료를 섭취한 실험군 이 대조군에 비하여 훨씬 오랜 시간 강제수영을 하였으나 비복근 및 간의 glycogen 함량은 실험동물군들 사이에서 유의적인 차이가 나타나지 않았다. 이것은 지구력이 증진되었음을 보여주는 한 예이다. 이러한 실험결과는 실험에 사용한 한방음료가 피로예방, 지구력 증진 및 운동수행능력을 증진하는데 매우 효과가 있음을 말해준다.
