Growth and ingestion rates of heterotrophic dinoflagellates and a ciliate on the mixotrophic dinoflagellate Biecheleria cincta

  • To explore the interactions between the mixotrophic dinoflagellate Biecheleria cincta (previously Woloszynskia cincta) and heterotrophic protists, we investigated whether the common heterotrophic dinoflagellates Gyrodinium dominans, Gyrodinium moestrupii, Gyrodinium spirale, Oxyrrhis marina, and Polykrikos kofoidii, and the ciliate Strobilidium sp. were able to feed on B. cincta. We also measured growth and ingestion rates of O. marina and Strobilidium sp. on B. cincta as a function of prey concentration. In addition, these rates were measured for other predators at single prey concentrations at which the growth and ingestion rates of O. marina and Strobilidium sp. were saturated. All grazers tested in the present study were able to feed on B. cincta. B. cincta clearly supported positive growth of O. marina, G. dominans, and Strobilidium sp., but it did not support that of G. moestrupii, G. spirale, and P. kofoidii. The maximum growth rates of Strobilidium sp. and O. marina on B. cincta (0.91 and 0.49 $d^{-1}$, respectively) were much higher than that of G. dominans (0.07 $d^{-1}$). With increasing the mean prey concentration, the specific growth rates of O. marina and Strobilidium sp. on B. cincta increased, but either became saturated or slowly increased. The maximum ingestion rate of Strobilidium sp. (1.60 ng C $predator^{-1}\;d^{-1}$) was much higher than that of P. kofoidii and O. marina (0.55 and 0.34 ng C $predator^{-1}\;d^{-1}$) on B. cincta. The results of the present study suggest that O. marina and Strobilidium sp. are effective protistan grazers of B. cincta.

Interactions between the voracious heterotrophic nanoflagellate Katablepharis japonica and common heterotrophic protists

  • Recently, the heterotrophic nanoflagellate Katablepharis japonica has been reported to feed on diverse red-tide species and contribute to the decline of red tides. However, if there are effective predators feeding on K. japonica, its effect on red tide dynamics may be reduced. To investigate potential effective protist predators of K. japonica, feeding by the engulfment-feeding heterotrophic dinoflagellates (HTDs) Oxyrrhis marina, Gyrodinium dominans, Gyrodinium moestrupii, Polykrikos kofoidii, and Noctiluca scintillans, the peduncle-feeding HTDs Luciella masanensis and Pfiesteria piscicida, the pallium-feeding HTD Oblea rotunda, and the naked ciliates Strombidium sp. (approximately $20{\mu}m$ in cell length), Pelagostrobilidium sp., and Miamiensis sp. on K. japonica was explored. We found that none of these heterotrophic protists fed on actively swimming cells of K. japonica. However, O. marina, G. dominans, L. masanensis, and P. piscicida were able to feed on heat-killed K. japonica. Thus, actively swimming behavior of K. japonica may affect feeding by these heterotrophic protists on K. japonica. To the contrary, K. japonica was able to feed on O. marina, P. kofoidii, O. rotunda, Miamiensis sp., Pelagostrobilidium sp., and Strombidium sp. However, the specific growth rates of O. marina did not differ significantly among nine different K. japonica concentrations. Thus, K. japonica may not affect growth of O. marina. Our findings suggest that the effect of predation by heterotrophic protists on K. japonica might be negligible, and thus, the effect of grazing by K. japonica on populations of red-tide species may not be reduced by mortality due to predation by protists.

남해중앙부해역의 표층퇴적물중 유기물과 와편모조류 시스트 분포 (A Study of Organic Matter and Dinoflagellate Cyst on Surface Sediments in the Central Parts of South Sea, Korea)

  • 남해 중앙부해역의 해양환경특성을 파악하기 위해 표층 퇴적물중의 와편모. 조류 시스트와 유기물 분포에 대한 조사를 2002년 4월부터 2003년 1월가지 총 6회 실시하였다. 분석 항목 중 함수율, 강열감량, 화학적 산소요구량, 입자성 유기탄소, 입자성 유기질소, 식물색소량은 각각 $38.7\~68.9\%\;3.9\~\;12.5\%,\;9.60\~44.05\;m9O_2\;gdry^{-1},\;3.12\~13.14mgC\; gdry^{-1},\;0.49\~2.01mgN\;gdry^{-1}$$1.61\~29.51{\mu}g\;gdry^{-1}$ 범위로 나타났다. 시스트는 18속, 31종 2미동정으로 총 33종이 동정되었으며, 우점종은 주로 독립영양종인 Spiniferites bulloideus와 Scrippsiella trocho-idea가 나타났으며 현존량은 $42\~2,880\;cysts\;gdry{-1}$ 범위로 출현하였다. 유기물 농도와 시스트 현존량은 연안해역보다 육상에서 멀리 떨어진 외해역에서 높게 나타났으며, 유기물 기원은 육상기원보다는 해역자체 생산에 의해 지배되고 있는 것으로 나타났다. 주성분분석결과 4월과 7월 모두 제1주성분은 "외해 유기물 집적"을, 제2주성분은 "시스트 출현량"에 의해 집약되는 특성으로 판단 할 수 있었으며, 득점분포도에 의해 남해 중앙부 해역은 3개의 해역으로 나누어졌다.

Development of an automatic system for cultivating the bioluminescent heterotrophic dinoflagellate Noctiluca scintillans on a 100-liter scale

  • Noctiluca scintillans is a heterotrophic dinoflagellate that causes red-colored oceans during the day (red tides) and glowing oceans at night (bioluminescence). This species feeds on diverse prey, including phytoplankton, heterotrophic protists, and eggs of metazoans. Thus, many scientists have conducted studies on the ecophysiology of this species. It is easy to cultivate N. scintillans at a scale of <1 L, but it is difficult to cultivate them at a scale of >100 L because N. scintillans cells usually stay near the surface, while prey cells stay below the surface in large water tanks. To obtain mass-cultured N. scintillans cells, we developed an automatic system for cultivating N. scintillans on a scale of 100 L. The system consisted of four tanks containing fresh nutrients, the chlorophyte Dunaliella salina as prey, N. scintillans for growth, and N. scintillans for storage, respectively. The light intensities supporting the high growth rates of D. salina and N. scintillans were 300 and 20 µmol photons m-2 s-1, respectively. Twenty liters of D. salina culture from the prey culture tank were transferred to the predator culture tank, and subsequently 20 L of nutrients from the nutrient tank were transferred to the prey culture tank every 2 d. When the volume of N. scintillans in the predator culture tank reached 90 L 6 d later, 70 L of the culture were transferred to the predator storage tank. To prevent N. scintillans cells from being separated from D. salina cells in the predator culture tank, the culture was mixed using an air pump, a sparger, and a stirrer. The highest abundance of N. scintillans in the predator culture tank was 45 cells mL-1, which was more than twice the highest abundance when this dinoflagellate was cultivated manually. This automatic system supplies 100 L of N. scintillans pure culture with a high density every 10 d for diverse experiments on N. scintillans.

Ichthyotoxic Cochlodinium polykrikoides red tides offshore in the South Sea, Korea in 2014: II. Heterotrophic protists and their grazing impacts on red-tide organisms

  • Occurrence of Cochlodinium polykrikoides red tides have resulted in considerable economic losses in the aquaculture industry in many countries, and thus predicting the process of C. polykrikoides red tides is a critical step toward minimizing those losses. Models predicting red tide dynamics define mortality due to predation as one of the most important parameters. To investigate the roles of heterotrophic protists in red tide dynamics in the South Sea of Korea, the abundances of heterotrophic dinoflagellates (HTDs), tintinnid ciliates (TCs), and naked ciliates (NCs) were measured over one- or two-week intervals from May to Nov 2014. In addition, the grazing impacts of dominant heterotrophic protists on each red tide species were estimated by combining field data on red tide species abundances and dominant heterotrophic protist grazers with data obtained from the literature concerning ingestion rates of the grazers on red tide species. The abundances of HTDs, TCs, and NCs over the course of this study were high during or after red tides, with maximum abundances of 82, 49, and $35cells\;mL^{-1}$, respectively. In general, the dominant heterotrophic protists differed when different species caused red tides. The HTDs Polykrikos spp. and NCs were abundant during or after C. polykrikoides red tides. The mean and maximum calculated grazing coefficients of Polykrikos spp. and NCs on populations of co-occurring C. polykrikoides were $1.63d^{-1}$ and $12.92d^{-1}$, respectively. Moreover, during or after red tides dominated by the phototrophic dinoflagellates Prorocentrum donghaiense, Ceratium furca, and Alexandrium fraterculus, which formed serial red tides prior to the occurrence of C. polykrikoides red tides, the HTDs Gyrodinium spp., Polykrikos spp., and Gyrodinium spp., respectively were abundant. The maximum calculated grazing coefficients attributable to dominant heterotrophic protists on co-occurring P. donghaiense, C. furca, and A. fraterculus were 13.12, 4.13, and $2.00d^{-1}$, respectively. Thus, heterotrophic protists may sometimes have considerable potential grazing impacts on populations of these four red tide species in the study area.

Cryptoperidiniopsis brodyi (Dinophyceae)의 생활사 (Life Cycle of Heterotrophic Dinoflagellate Cryptoperidiniopsis brodyi (Dinophyceae))

  • Pfiesteriaand Pfiesteria-like organisms were reported to be linked to major fish kills(involving well over a billion fish) in North Carolina and Maryland estuaries on the U.S. east coast during the 1990s. Occurrences of these species have been recently reported from Korean waters including Chinhae Bay and the coast of Yeosu. In this study, the life cycle of Cryptoperidiniopsis brodyi and Pfiesteria piscicida were examined using DAPI staining. Their excystment and growth were stimulated directly by the addition of prey cells such as Rhodiminas salina. Amoeboid stages in C. brodyi and P. piscicida were never observed in culture, even after addition of filter-sterile fish mucus and tissue. The dominant life cycle stages consisted of motile flagellated zoospores and cysts. A typical dinoflagellate life cycle was demonstrated by direct observation and DAPI staining.

Abundance of Heterotrophic-and Photosynthetic Dinoflagellates and Factors Controlling Their Abundance and Distribution in Korean Coastal Waters During Summer, 1994

  • We investigated the abundance and biomass of dinoflagellates and factors controlling their abundance in marine planktonic ecosystems in Korean coastal waters. The abundance of photosynthetic (PDNF) and heterotrophic dinoflagellates (HDNF) was in the range of 0.7${\times}$10$^2$ cells/1-14.0${\times}$10$^6$ cells/1 and in the range of 3.0${\times}$10$^2$ cells/1-6.47${\times}$10$^5$ cells/I, respectively. Their biomass was 0.5${\times}$10$^{-1}$-2.56${\times}$10$^4$ ${\mu}gC/I$ and 2.0${\times}$10$^{-1}$-1.5${\times}$10$^{2}$ ${\mu}gC/I$, respectively. In order to find factors controlling their abundance, stepwise regression and best subsets regression analyses were used. We found that during the summer the most important factors controlling PDNF abundance are DO, P, N and S (abiotic factors), and for HDNF, the abundance of zooplankton, ciliates and HF (biotic factors), and that high turbidity may effect the distribution of dinoflagellate species.

제주 해안주변해역 표층퇴적물 중 와편모조류 시스트 군집의 분포특성 (Distribution of Dinoflagellate Cysts in Surface Sediments of the Coastal Areas around Jeju Island, Korea)

  • 제주 해안선 주변 항만 표층퇴적물의 와편모조류 시스트 군집구조를 파악하기 위하여, 제주 해안을 따라 위치하는 22곳의 항만을 대상으로 2012년에서 2016년까지 격년단위로 3회 조사하였다. 조사는 소형 에크만 채니기를 이용하여 0~2 cm 두께의 표층퇴적물 표본을 채집하였다. 분석결과 출현이 확인된 와편모조류 시스트는 6그룹 9속 29종으로 Protoperidinioid Group이 13종으로 44.8%의 점유율을, Gonyaulacoid Group이 9종으로 31.0%, Calciodineloid Group이 3종으로 10.3%, Gymnodinioid Group이 2종으로 6.9%, Tuberculodinoid Group과 Diplosalid Group이 각각 1종으로 3.5%의 점유율을 보였다. 또한 정점에 따른 변화는 1~6 종으로 매우 낮았다. 시스트의 세포밀도는 13~220 cysts/g dry의 범위로 낮지만, 퇴적물의 함수율과 유의적인 회귀식을 나타내었다. 또한 일부 세립퇴적물을 나타내는 항만 및 개발에 의해 인위적으로 형성된 조수웅덩이에서는 종속영양종 출현비가 높아, 오랜 시간 유기물이 퇴적되고 있음을 시사하였다. 우점종은 cyst of Gymnodinium sp 및 Protoperidinium 속 시스트가 최우점하였고, 이외 cyst of Pyrodinium bahamense, cyst of Scrippsiella trochoidea 등이 우점 출현하였다. 특히 열대 및 아열대성 유독와편모조인 P. bahamense 시스트는 제주연안에서 처음 기록되는 종으로 제주 해안주변해역의 인위적 조수웅덩이 등의 해역 수질환경 및 열대나 아열대에서 유입되는 새로운 유독와편모조류에 대한 모니터링 및 관리방안 수립이 시급한 것으로 판단되었다.

소양호에서 Peridinium bipes에 의한 적조가 수층 미생물 순환고리에 미치는 영향 (Effects of Freshwater Red Tide by Peridinium bipes on Microbial Loop in the Water Column of Soyang Reservoir)

  • Effects of freshwater red tide of Peridinium bipes (dinoflagellate) in August of 1991 on the carbon flux through the microbial loop were studied in Soyang Reservoir. Chlorophyll a and primary production, bacterial production in red tide area were 22, 18 and 400 times higher, respectively, than other sites. Phytoplandton biomass comprised 97% and 20% of food source of zooplankton grazing within and without red tide, respectively. The percent bacterial production supported by phytoplankton exudate was 14% within red tide and >100% without red tide. In laboratory experiments, more than 85% organic carbon of Peridinium biomass was released or degraded by heterotrophic bacteria within 14 days. As results of red tide of Peridinium with sudden influx of organic carbon in water column, the main food source of zooplankton and dependency of bacteria on phytoplankton exudate were changed. Therefore, the relative importance of microbial loop to grazing food web was changed.

와편모조류 시스트 분포에 의한 한국 남서해역의 해양환경 특성 2. 가막만 주상퇴적물중 유기물 및 와편모조류 시스트의 분포특성에 의한 해양환경변화 추정 (Estimation on the Variation of Marine Environment by the Distribution of Organic Matter and Dinoflagellate Cyst in the Vertical Sediments in Gmmak Bay, Korea)

  • 가막만 주상 퇴적물중의 유기물농도와 와편모조류 시스트의 분포특성을 파악하기 위해 만내 6개 정점을 대상으로 2000년 6월부터 2001년 4월까지 격월간으로 총 6회 현장조사를 실시하였다. 유기물 농도중 강열감량은 3.21∼11.18%, 화학적산소요구량은 12.25∼99.26 mgO₂/g dry, 황화물량은 nd∼10.29 mgS/g dry, 입자성 유기탄소가 2.91∼116.18 mgC/g dry, 입자성 유기질소가 0.53∼2.82 mgN/g dry, 식물색소량이 2.49∼116.18 ㎍/g dry로 나타났다. 와편모조류 시스트는 21속, 36종, 2미동정 종 등 총 38종이 동정되었으며,출현세포수는 18∼3,836 cysts/g dry의 범위를 보였다. 만내 주상퇴적물중의 유기불과 와편모조류 시스트의 연직적인 분포특성은 대부분의 정점에서 표층퇴적층에서 아래로 갈수록 농도가 감소하였다. 와편모조류 시스트중 종속영양종의 시스트 분포가 유기물량과 밀접한 관련성을 나타내었으며, 퇴적물중 유기물 농도와 와편모조류 시스트의 분포특성 및 기타 해양환경변화 자료로부터 가막만 북서부 해역은 약 1980년대 이후부터 부영양화가 급속히 진행되고 있는 것으로 판단되었다.
