The Sterolic Properties of Heterotrophic Tetraselmis suecica

  • The heterotrophic production method for Tetraselmis suecica, a suggested alternative to photoautotrophic one in the economic sense, was studied in terms of cell growth and sterolic property. The alga in the 10 mM organic carbon (glucose) manifested cell growth. However, the alga produced by the heterotrophic method showed a unique property of sterol determined with an aid of GC and GC-MS. The photoautotrophic control T. suecica contained 6 detectable sterol species: $cholesta-5,\;22-dien-3\beta-o1$, $ergost-5-en-3\beta-o1$, cholest-5-en-3\beta-o1$, $24-methyl-cholesta-5,\;22-dien-3\beta-o1$, $24-methylcholesta-5,\;24-dien-3\beta-o1$, $24-ethylchlolesta-5,\;24-dien-3\beta­o1$, $24-methylcholesta-5-en-3\beta-o1$, and $24-ethylchlolesta-5en-3\beta-o1$. We discuss the sterolic properties of the alga along the heterotrophic progress, particularly focusing on the availability of the method in the aquaculture of bivalves which normally need sterols as a dietary source.

DISTRIBUTION CHARACTERISTICS AND AFFECTING FACTORS OF SPRING HETEROTROPHIC BACTERIA IN BOHAI SEA

  • Distribution characteristics, variation patterns and affecting factors of hetorotrophic bacteria were studied from April to May 1999 in Bohai Sea by standard Acridine Orange epifluorescence microscopy (AO method). The biomass in surface waters showed a small day-night variation, varying from 0.13-2.51$\mu\textrm{m}$$.$dm$\^$-3/ with an average of 0.84 $\mu\textrm{m}$$.$dm$\^$-3/. The biomass in bottom waters showed, however, a large variation, changing from 0.15-4.18 $\mu\textrm{m}$$.$dm$\^$-3/ with an average of 1.36 $\mu\textrm{m}$$.$dm$\^$-3/. The peak values were obtained at 5 and 11 am. The bottom water biomass showed a significant correlation with particulate organic carbon (r=0.639, p<0.05). Heterotrophic bacteria showed high biomass in nearshore waters and low values in offshore areas with a high biomass zone around Yellow Sea river mouth, which was consistent with the distribution of nutrients. The vertical distribution of heterotrophic bacteria showed biomass in bottom waters was higher than in surface water. The biomass of heterotrophic bacteria in Bohai Sea was similar with that in other marine waters.

Some Free-Living Heterotrophic Flagellates from Marine Sediments of Tropical Australia

  • The diversity of heterotrophic flagellates was examined at marine sediments around Cape Tribulation, Australia. The species described belong to the Alveolates, Apusomonadiae, Cercomonadida, Choanoflagellida, Cryptomonadida, Diplomonadida, Euglenozoa incertae sedis, Kathablepharidae, Kinetoplastida, Pedinellids, Stephanopogonidae, Stramenopiles, Stramenopiles incertae sedis, Thaumatomonadidae and Protista incertae sedis. Among the 51 species from 38 genera encountered in this study is one new taxon: Glissandra similis n. sp., and two new names are introduced: Goniomonas abrupta (Skvortzov 1924) nomen nodum and Cercomonas skvortzovi (Skvortzov 1977) nomen nodum. There was little evidence for endemism because most flagellates including one new taxon described here have been reported.

소양호 세균 군집의 계절적 분석 (Seasonal analysis of heterotrophic bacterial community in lake Soyang)

  • The numerical taxonomy of heterotrophic bacterial community in Lake Soyang was analysed. 95, 115, 88 and 75 strains which were isolated at each season from spring in 1987 to winter in 1988 were clustered by single matching coefficient. The diversity indices (H') were in the range of 0.511-1.684, and the community was most diverse in spring. THe seasonal variation of generic composition was significant. Of the domonant genera, Acinetobacter, Pseudomonas, and Flavobacterium were representative.

  • The aerobic heterotrophic bacteria in the histosphere associated with grasses (Gramineae, Caryphyllaceae, Crucifereae) and rice cultivars in saline and reclaimed saline paddy soils were varied with species and rice cultivars. The fraction of aerobic heterotrophic $N_2$-fixing bacteria to the total aerobic heterotrophic bacteria were averaged to eighteen percent in the histosphere of grasses and rice. Acetylene reducing activity of these bacteria were ranged from 1 to 24 n mole/tube/hr. Most of the bacteria strains were predominated of hydrogen utilizing bacteria. The majority of these bacteria were closed to Pseudomonas, Azospirillum, Klebsiella and Agrobacter.

금강 하구 퇴적토에서의 종속영양 세균의 분포에 미치는 환경요인과 계절적 우점속에 관하여 (Environmental factors affecting seasonal destribution of heterotrophic bacteria in Kum river estuarine sediments)

  • Dominant genera of heterotrophic bacteria were investigated seasonally in Kum River estuarine sediments. Numerical taxonomy on the isolated heterotrophic bacteria were carried out according to their morphological, physiological, nutritional and biochemical characters. Heterotrophic bacteria in the depths of 0 to 10 cm of sampled sediments were almost facultative anaerobes, and 77.9% to 86.9% of them were Gram negative rods. Kominant genera among the isolates were Pseudomonas, Flavobacterium, Micrococcus, Cnterobacteria and Vibrio during sampling periods. According to their character analyses by similarity index(S), the isolates were clustered in fourteen groups at clay type sediments (Site 1) and four groups at sand type sediments (Site 2).

요인실험계획을 이용한 수도관 생물막 형성 영향 인자의 효과 분석 (Analysis of Effects of Factors Influencing Biofilm Formation in Drinking Water Distribution Pipe Using Factorial Experimental Design)

  • This study evaluated the effect of factors influencing the initial biofilm formation in drinking water distribution pipe by running experiments using a $2^{4-1}$ fractional factorial experimental design with a replicate. Important variables used for assessing biofilm formation included BDOC(biodegradable dissolved organic carbon), viable heterotrophic bacteria present in drinking water, water temperature, and shear stress at two levels each. Based on the statistical analysis of biofilm levels measured as attached HPC(heterotrophic plate count) and community-level assay, the main factors that have significant effects on biofilm formation were found to be viable heterotrophic bacteria and BDOC. Water temperature only exhibited significant effect on the levels of attached HPC, while shear stress was not a significant factor under given conditions. Moreover, the statistical analysis revealed that interactions between the important variables were not statistically significant at a 0.05 significance level.

Effects of an Artificial Breakwater on the Distributions of Planktonic Microbial Communities

  • The summer distributions of planktonic microbial communities (heterotrophic and phtosynthetic bacteria, phtosynthetic and heterotrophic nanoflagellates, ciliate plankton, and microphytoplankton) were compared between inner and outer areas of Lake Sihwa, divided by an artificial breakwater, located on the western coast of Korea, in September 2003. The semi-enclosed, inner area was characterized by hyposaline surface water (<17 psu), and by low concentrations of dissolved oxygen (avg. $0.4\;mg\;L^{-1}$) and high concentrations of inorganic nutrients (nitrogenous nutrients $>36\;{\mu}M$, phosphate $>4\;{\mu}M$) in the bottom layer. Higher densities of heterotrophic bacteria and nanoflagellates also occurred in the inner area than did in the outer area, while microphytoplankton (mainly diatoms) occurred abundantly in the outer area. A tiny tintinnid ciliate, Tintinnopsis nana, bloomed into more than $10^6\;cells\;L^{-1}$ at the surface layer of the inner area, while its abundance was much lower ($10^3-10^4\;cells\;L^{-1}$) in the outer area of the breakwater. Ciliate abundance was highly correlated with heterotrophic bacteria (r = 0.886, p < 0.001) and heterotrophic flagellates (r = 0.962, p < 0.001), indicating that rich food availability may have led to the T. nana bloom. These results suggest that the breakwater causes the eutrophic environment in artificial lakes with limited flushing of enriched water and develops into abundant bacteria, nanoflagellates, and ciliates.

삼천포만 인근해역과 저질토에서 종속영양세균과 분변성 기원 세균의 분포 (Distribution of Population Densities of Heterotrophic Bacteria and Fecal-Origin Bacterial Group in Seawater and Sediments near Samcheonpo Bay)

  • 본 연구는 경상남도 사천시 삼천포 항을 중심으로 인근의 40개의 조사정점을 선정하여 해수와 저질토에서 해양종속영양세균, E. coli, 그리고 Salmonella와 Shigella의 분포를 2003년 7월부터 12월가지 3차에 걸쳐 실시하였다. 본 연구의 조사기간(2003년 $7{\sim}12$월)중 삼천포만 인근해역의 조사정점에서 해양 종속영양세균의 분포는 $1.7{\pm}0.9{\times}10^3{\sim}2.4{\pm}0.9{\times}10^5CFUmL^{-1}$의 범주에서 변화하였으며, 정점 34에서 9월에 가장 높은 값을 나타냈다. 전반적으로 9월이 7월과 12월에 비해 모든 정점에서 다소 높은 값을 보였다. 저질토에서 혐기성 세균의 분포는 $2.2{\pm}0.2{\times}10^3{\sim}2.0{\pm}0.2{\times}10^5CFUg^{-1}dry$ wt.의 범주에서 변화하였다. 혐기성 세균의 경우 외해 정점에서는 내만에 위치한 정점들보다 년 중 낮은 균체수를 유지하고 있어 물리, 화학적인 환경요인들에 의해 별 영향인 없음을 알 수 있다. E. coli의 분포는 해수의 경우, 7월중에만 사천만 인근(St. 1, 2, 3, 4),진주만과 창선도 인근 (St. 15, 23, 31), 삼천포항 인근(St. 14)에서 검출되고 9월과 12월에는 전 정점에서 전혀 검출되지 않았다. 저질토의 경우, 7월에 사천만 인근(St. 1, 2)과 창선도 인근(St. 16, 31)에서만 검출 되었다. Salmonella와 Shigella의 분포는 해수와 퇴적토에서 7월과 9월에 일부 해역에서만 검출되었다.

Arthrospira (Spirulina) platensis의 종속영양배양과 r-Linolenic Acid 생산 (Growth and r-Linolenic Acid Production of Arthrospira (Spirulina) platensis in Heterotrophic Culture.)

  • Arthrospira에 대한 종속영양배양 결과, A. platensis M9108(A. platensis PCC 9108의 고농도 glucose 내성 돌연변이)은 glucose 5.0 g/L 첨가된 SOT 배지에서 8일 동안 배양하여 0.54 g/L의 생물량을 얻었으며, 일일생산량은 0.068 g/L/day을 보였고, specific growth $rate(\mu)$는 0.014/h를 보였다. A. platensis M9108의 일일생산량과 $\mu$는 모균주인 A. platensis PCC 9108에 비하여 각각 1.3배와 1.2배 향상된 결과를 보였으며, 이전에 보고된 결과에 비하여 각각 1.3배와 1.8배 증가하였다. Arthrospira의 건체량 중 지방산과 r-linolenic acid의 함량은 각각 $1.94\sim4.97%$$0.13\sim1.33%$로 균주에 따라 상당히 다양하였다. Arthrospira는 유기탄소원으로 glucose를 첨가한 조건에서 성장이 가장 좋았으며, A. platensis M9108의 경우 maltose의 이용능도 glucose 이용 능과 비슷하였다. A. platensis M9108의 성장은 glucose의 5.0 g/L까지는 증가하였지만, 10.0 g/L에서는 저해되는 양상을 보였다. 또한 고농도의 탄소원을 이용한 종속영양배양 중 발생하는 세포의 뭉침 현상이 A. platensis M9108에서는 관찰되지 않아 안정적으로 고농도 생산에 유용할 것으로 판단된다.
