Effect of Chaihushugansan and Chaihuyigantang on the change of Monoamine Contents in the Hypothalamus of Mice induced by the Immobilization Stress (구속(拘束)스트레스로 유발된 생쥐 시상하부내(視床下部內) Monoamine의 변화에 대한 시호소간산(柴胡疎肝散)과 시호억간탕(柴胡抑肝湯)의 효능)

  • This study aimed to investigate the effects of Chaihushugansan and Chaihuyigantang on the change of monoamine-contents in the hypothalamus of mice induced by the immobilization stress. Each formula(1.0g/kg) was injected intragastrically to mice 1 hour prior to the immobilzation stress(15 minutes) before measuring the change of monoamine-contents level in mice hypothalamus. The results were as follows: 1. DA content at control group showed significant change. CHSGS group and CHYGT group showed significant decrease.(P<0.05) 2. NE content of the control group was increased than normal group. NE content of the CHYGT group was shown a significant decrease than control group.(P<0.05) 3. DOPAC contents didn't show significant changes at any groups. 4. 5-HT contents didn't show significant changes at any groups. 5. 5-HIAA contents at CHSGS group and CHYGT group showed signifiant changes.(P<0.05, 0.01) These data revealed that chaihushugansan and chaihuyigantang might have the anti-stress effect through the control of DA and NE contents in the hypothalamus.

  • Chelidonii Herba (Baekgulchae in Korean: CHE), a commonly used herb in Korea, Japan and China, is widely used in the treatment of stomach cancer, jaundice, gastric ulcer, edema and pain of stomach. In the present study, we demonstrated that CHE induces apoptosis in AGS cells, human stomach adenocarcinoma cell line. One of the most important recent advances in cancer research is the recognition that apoptosis plays a major role in both tumor formation and treatment response, In this study, CHE caused a decrease of viability in AGC cells. When AGS cells were treated with CHE, cells showed dose-dependent manner apoptotic cell death. Increased apoptotic cell death, exposured to CHE, resulted from induction of Bad translocation to mitochondria, cytochrome-c release from mitochondria to cytosol, activation of caspase-3, 8, 9, and PARP cleavage. These results suggest that CHE may be potential therapeutic approach in the clinical management of stomach adenocarcinoma.

  • Objectives : In this study the antidepressant effects of mixture of Lycii Radicis Cortex and Gardeniae Fructus on the change of HPA-Axis and Catecholamine system was investigated Methods : The forced swimming test was performed. The expression of CRF, c-Fos in the PVN, and TH in the VTA and LC were measured with immunohistochemical method. And the experimental groups were divided into the Extraction after Mix group(A) and Mix after Extraction group(B). The effects of both group were compared. Results : The duration of immobility in the FST was decreased significantly in three groups except B100(P<0.01). B100 was shown the significant increase(P<0.05). The expression of CRF in PVN was decreased significantly in all group(P<0.05~P<0.01). The expression of c-Fos in PVN was decreased in A100, A400 and was increased significantly in B100, B400(P<0.05). The expression of TH in VTA and LC were decreased significantly in all group(P<0.001). Conclusions : Effects of Extraction after Mix group(A) and Mix after Extraction group(B) on the HPA-Axis system and Catecholamine system were validated.

Effects of Taste and Quality of Drugs on the Plasma Corticosterone Level in Mice exposed to Heat and Cold Stress (한약(韓藥)의 기미(氣味)가 한열자극(寒熱刺戟)을 받은 생쥐의 혈중(血中) corticosterone에 미치는 효과(效果))

  • This study was performed to investigate what effect the taste and quality of drugs would have on the cold and heat mechanisms of human body. We administered Hwangryeunhaedok-tang composed of bitter-tasted, cold-qualified drugs and Gungangbuza-tang composed of spice-tasted. hot-qualified drugs, respectively to the mice 1 hr before exposure to heat stress or cold stress. Plasma corticosterone level of mice was measured. The results were as follows: 1. The elevated corticosterone level in the mice exposed to heat stress was significantly decreased after administration of Gungangbuza-tang but there was no decrease after administration of Hwangryeunhaedok-tang. 2. The elevated corticosterone level in the mice exposed to cold stress was significantly decreased after administration of Hwangryeunhaedok-tang but there was mild decrease after administration of Gungangbuza-tang. 3. When the doses, 3g/kg and 1g/kg were administerd to mice exposed to heat stress. both dose showed significant decrease of corticosterone level and the dose. 3g/kg was more effective. However, in the mice exposed to cold stress, the dose, 3g/kg showed mild decrease and 1g/kg showed significant decrease. These data suggested that HW decreased the plasma corticosterone level in the mice exposed to cold stress and GB also decreased the plasma corticosterone level in the mice exposed to heat stress. In conclusion, our study revealed that the taste and quality of drugs controled the cold and heat mechanism of human body.

  • Object : The effects of Rehmanniae Radix(RR) on HPA Axis system and Catecholaminergic system was investigated. Methods : we performed the Forced Swimming Test(FST). Also the expression of Corticotropin -Releasing Factor(CRF), c-Fos and Tyrosine Hydorxylase(TH) was measured by immunohistochemical method at Paraventricular Nucleus(PVN), Locus Coeruleus(LC) and Ventral Tegmental Area(VTA). Results : 1. The duration of immobility in the Forced Swimming Test was significantly decreased in the Rehmanniae Radix 400 mg/kg groups in comparison with the control group(p<0.05). 2. Comparing to the control group, CRF expression was significantly decreased in the Rehmanniae Radix 400 mg/kg treated group in comparison with the control group(p<0.01). 3. c-Fos expression was significantly decreased at PVN in the Rehmanniae Radix 100 mg/kg treated group in comparison with the control group(p<0.01). 4. TH expression was significantly increased at LC in the Rehmanniae Radix 100 mg/kg and 400 mg/kg treated group in comparison with the control group(p<0.001). 5. TH expression was significantly decreased at VTA in the Rehmanniae Radix 100 mg/kg and 400 mg/kg treated group in comparison with the control group(p<0.001). Conclusion : According to the results, it can be considered that Rehmanniae Radix has antidepressant effect by showing the reduction of immobility in FST through the decreased expression of CRF, c-Fos in PVN and TH in VTA.

Effect of Banggibongnyeongtang on the immunohistological change in LPS-induced depression rats (방기복령탕(防己茯苓湯)이 백서에서 LPS로 유도된 우울증에서 면역 조직학적 변화에 미치는 효과)

  • Objective : This study is accomplished in order to investigate the effect of banggibongnyeongtang on the immunohistological change in LPS-induced depression rats to confirm the histological result of the previous behavioral and biochemical effect. Methods : LPS 5 ㎍ was injected to lateral ventricle and experimental groups were administered BBT intraperitoneally. The concentration of 5-HT in the Medial Prefrontal Cortex, Striatum, Hippocampus, Amygdala was measured by ELISA. IL-1β, TNF-α mRNA and BDNF mRNA expression in the hippocampus was examined by RT-PCR. Result : BBT enhanced 5-HT concentration at all part of brain but no significantly difference at medial prefrontal cortex and striatum. LPS+BBT400 group increased 5-HT concentration significantly than LPS group at hippocampus and amygdala (p<0.05). BBT decreased IL-1β mRNA expression dose dependently but only with significantly decrease in LPS+BBT400 group than LPS group's in Hippocampus (p<0.05). But BBT did not decrease TNF-α mRNA expression significantly in Hippocampus. BBT increased the expression of BDNF mRNA at hippocampus and LPS+BBT400 group significantly increased comparing with LPS group does (p<0.05). Conclusion : It is postulated that the anti-depressant effect of BBT can be validated through the anti-inflammatory effect, 5-HT concentration increase, and the neuro-protective effect mediated by BDNF by combining the results of the previous report about the behavioral and biochemical effect.

Yookmijihwang-Tang on the Plasma Corticosterone level in Mice exposed to Heat, Cold and Immobilization Stress (한(寒), 열(熱), 구속(拘束) 스트레스에 대한 사군자탕(四君子湯), 사물탕(四物湯), 육미지황탕(六味地黃湯)의 효능(效能))

  • In order to investigate the effect of Sagoonja-Tang, Samool-Tang and Yookmijihwang-Tang against the cold, heat and immobilization stress, each formula was injected intragastically to mice exposed to forced cold water swimming($4^{\circ}C$), forced hot water swimming($42^{\circ}C$) and immobilization stress before measuring the change of plasma corticosterone level of mice. The results were as follows: 1. One hour after each Sagoonja-Tang, Samool-Tang, Yookmijihwang-Tang with the dose of 1g/kg, 3g/kg each were administered to mice, we put cold stress($4^{\circ}C$) on mice for 3min. Then those prescriptions didn't show any significant effect on plasma corticosterone level. however, Yookmijihwang-Tang(1g/kg) showed a significant increase(p〈0.05), and Samool-Tang(3g/kg) showed a small increase. 2. One hour after Samool-Tang, Sagoonja-Tang and Yookmijihwang-Tang with the dose of 1g/kg, 3g/kg each were administerd, mice exposed to heat stress($42^{\circ}C$) for 3 min. Samool-Tang(1g/kg) and Yookmijihwang-Tang(lg/kg) showed significant decrease of corticosterone level(p〈0.01, p〈0.05) but Sagoonja-Tang didn't change it. In the case of the dose of 3g/kg, only Samool-Tang decreased corticosterone level(P〈0.01), but Sagoonja-Tang, Yookmijihwang-Tang didn't make significant effect on plasma corticosterone level of the mice exposed to heat stress($42^{\circ}C$ ). 3. Those mice which exposed to immobilization stress one hour after Sagoonja-Tang, Samool-Tang, Yookmijihwang-Tang with the dose of 1g/kg and 3g/kg didn't show effective decrease of corticosterone level. However, Sagoonja-Tang(3g/kg), Samool-Tang(lg/kg, 3g/kg) showed the effect of mild increase of corticosterone level rather than decrease. These data revealed that the effect-variation of prescriptions depended on the kinds of stress and the dose of prescriptions had a different effect on therapy.

Effect of Sihogayonggolmoryeotang on SPS-induced PTSD in Rats (시호가룡골모려탕(柴胡加龍骨牡蠣湯)이 흰쥐에서 SPS로 유도된 PTSD에 미치는 효과)

  • Objective : To investigate the effect of sihogayonggolmoryeotang (SY) on Single Prolonged Stress(SPS)-induced Post Traumatic Stress Disorder(PTSD). Method : To confirm the effects of SY on SPS-induced PTSD, Changes in body weight, sucrose intake open field test(OFT) and forced swimming test(FST)were observed. After behavioral tests, the plasma corticosterone(CORT) from the abdominal aorta, serotonin(5-HT) from prefrontal cortex, hippocampus, amygdala and striatum, norepinephrine(NE) and dopamine(DA) from hippocampus was measured by ELISA. mRNA expression of brain-derived neurotrophic factor(BDNF) and cAMP response element-binding protein(CREB) in hippocampus was measured by RT-PCR. Result : Weight change and sucrose intakes of rats in 14th day after the administration of SY were significantly increased in the SPS + SY450 group compared to the SPS group (p<0.05). Numbers of crossing in the central zone in the OFT were significantly increased in the SPS + SY450 group (p<0.05) compared with the SPS group. The immobility time of FST was significantly decreased in SPS + SY450 group compared with SPS group (p<0.05). The change of plasma CORT concentration was significantly decreased in SPS + SY450 group compared with that in SPS group (p<0.05). The change of 5-HT concentration was significantly increased in the SPS + SY450 group at hippocampus and amygdala compared with the SPS group (p<0.05). The concentration of DA was significantly increased in the SPS + SY450 group compared with the SPS group (p<0.05). The expression of BDNF and CREB were significantly increased in SPS + SY450 group compared with the SPS group (p<0.05). Conclusion : SY administration lowered the increase of CORT caused by PTSD and increases the 5-HT concentration and reversed the decreased expression of NE and DA and BDNF and CREB by PTSD. It is postulated that SY is effective in treating PTSD by restoring cognitive function, memory impairment, unstable emotional disturbances.

Effect of Banggibongnyeongtang on LPS-induced Depression in rats (방기복령탕(防己茯苓湯)이 흰쥐에서 LPS로 유도된 우울증에 미치는 효과)

  • Objective : This study is conducted in order to investigate the effect of Banggibongnyeongtang(BBT) on Lipopolysaccharide (LPS)-induced depression. Method : LPS $5{\mu}g$ was injected to lateral ventricle. Experimental groups were administered BBT intraperitoneally. Depressive behavior was confirmed by weight change, sucrose preference, open field test(OFT), and forced swimming test(FST). The plasma concentration of $IL-1{\beta}$ and $TNF-{\alpha}$, Corticotropin-Releasing Factor(CRF), Adrenocorticotropin Hormone(ACTH) and Corticosterone(CORT) were measured by ELISA. Result : BBT did not change the body weight significantly than LPS group, but on sucrose preference, BBT increased significantly in LPS+BBT400 group compared to LPS group (P<0.05). In the OFT, BBT increased spending time in the central zone and decreased grooming number. LPS+BBT400 group increased central zone-spending time, and decreased grooming number than LPS group significantly (P<0.05). In the FST, LPS+BBT400 group decreased immobility time than LPS group significantly (P<0.05). BBT decreased $IL-1{\beta}$ concentration does-dependently, but only with significant decrease in LPS+BBT400 group than LPS group in plasma (P<0.05). But BBT did not decrease $TNF-{\alpha}$ concentration significantly in plasma. BBT decreased plasma CRF, ACTH, and CORT. And CRH and CORT of LPS+BBT400 group were shown significant decrease comparing with LPS group (P<0.05). Conclusion : It is postulated that the anti-depressant effect of BBT can be validated through inhibition of HPA axis abnormal activity by the anti-inflammatory effect.

An Overview of the Herbal Remedies for Severe Acute Respiratory Syndrome (SARS) in WHO Official Report (2004) (중증급성호흡기증후군(SARS) 치료에 사용된 한약 처방에 대한 고찰 - WHO 보고서(2004)에 사용된 처방을 중심으로)

  • Background : Early in November 2002, severe acute respiratory syndrome (SARS) began to spread throughout thirty-two countries around the world. A total of 5,327 cases were reported in China, including 1,755 cases in Hong Kong and 655 in Taiwan. The total number of cases reported from Canada and Singapore was more than 200. The total number of SARS cases world-wide reached 8,437 with incidences. Mortality from SARS is estimated at around 11%. Under the guidance of the World Health Organization (WHO), clinical studies on the use of integrated herbal medicine and western medicine for treating SARS were carried out in China and Hong Kong. The official report suggested that integrated treatment was more effective than western medicine alone in clinical symptoms, lung inflammation, blood oxygen saturation, immunological functions and others. Objective : To overview the treatment principle and the prescriptions according to the regimens and the stages for SARS of the reports, and further to broaden our knowledge of treatment of acute infectious diseases using natural herbal medicine. Methods : We reviewed nine of WHO SARS reports that comprehensively described the principles and methods of treatment and summarized them into eight treatment methods. We analyzed the herbal formulae on the basis of their treatment principles, evaluated them in accordance with warm disease study, listed frequently used herbs, and assessed patent prescriptions and herbal injections that were mentioned in the reports. Results and Conclusion : The reports divided the course of SARS into 3 to 6 stages such as high fever stage, fastigium stage, and convalescent stage. Frequently used herbs were Radix Scutellariae, Radix Paeoniae, Radix Astragali, Semen Armeniacae, Radix Pseudostellariae, Radix Ophiopogonis, and Fructus Schizandrae. Herbal decoctions were the primary method of treatment, while extracts or injections were secondary. Prescriptions were mainly based on the warm disease study. SARS is a viral disease caused by corona virus, and herbal medicine is proven to be effective against it. We believe that evidences and experiences from SARS cases can be a good reference to further researches on acute infectious diseases.
