  • We previously reported the antimutagenicity, anti-lipid peroxidation and antiinflammtory properties of the three Chinese herb medicine, Kalopanax pictus, Pueraria thunbergiana and Rhus verniciflua, which have been used as therapeutics for diabetes mellitus and inflammation in Korea. Since these drugs have been used for the prescriptions consisted of several crude drugs, we investigated whether the extract of the three drugs could increase the individual drugs or not. The three extracts were independently prepared from K. pictus, P. thunbergiana, and R. verniciflua, which extracts were named the K-1, P-1 and R-1 in this order and the extract (KPR-1) was also prepared from the combined plant materials with each same weight of the three drugs. • The three crude drugs of the Kalopanax pictus (Araliaceae) roots (K), Pueraria thunbergiana (Leguminosae) flowers (P), and the Rhus verniciflua (Anacardiaceae) heartwood(R) used for anti-thirst drugs in Oriental herb medicine were extracted with MeOH, respectively, and the successive fractionation of the extract gave EtOAc extract. Certain amount ratios of the three extracts were also prepared to compare the antimutagenicity in Ames test. In N-methyl-N(-nitro-N-nitrosoguanidine (MNNG; 0.4 (g/plate)-induced test, the activities of complex mixture were observed between the highest antimutagenic activity of K extract and the lowest P extract. In aflatoxin (AFB$_1$)-induced test, the EtOAc complex (K : P : R=l : 1 : 3) labeled E-113 decreased the revertants of Salmonella typhimurium TA100 by 95%, which activity were highest among other extracts or complexes mixture used. Fractionation of organic solvent mostly increased the antimutagenicity. These trends were also observed in the antimutagenicity test of the mixture of each active component of kalopanaxsaponin A, tectorigenin and sulfuretin. These results supported that many kinds of anti-thirst herb medicine in the prescription could effectively prevent cancer disease.

  • An oncology-specific database called OncoRx (http://bit.ly/cancerRx) was previously set up in cyberspace to aid clinicians in identifying interactions of anticancer drugs (ACDs) and chemotherapy regimens with traditional Chinese medicines (TCMs) and complementary and alternative medicines (CAMs). Since then, users have requested the drug-CAM interactions (DCIs) of 5 specific CAMs (cranberry, melatonin, co-enzyme Q10, huachansu, reishi mushroom) to be updated in the database. Pharmacokinetic properties (metabolism, enzyme induction/inhibition, elimination), TCM properties and DCIs of each CAM were collated with 117 ACDs using 9 hardcopy compendia and online databases as resources. Additionally, individual ACDs and CAMs were used as keywords for PubMed searches in combination with the terms 'anticancer drugs', 'drug interactions', 'herb-drug/drug-herb interactions', 'pharmacokinetic interactions' and 'pharmacodynamic interactions'. DCI parameters consisted of interaction effects, evidence summaries, proposed management plans and alternative non-interacting CAMs, together with relevant citations and update dates of the DCIs. OncoRx is also used as a case to introduce the "Four Pharmaco-cybernetic Maxims" of quality, quantity, relationship and manner to developers of digital healthcare tools. Its role in Hayne's "5S" hierarchy of research evidence is also presented. OncoRx is meant to complement existing DCI resources for clinicians and alternative medicine practitioners as an additional drug information resource that provides evidence-based DCI information for ACD-CAM interactions.

  • A literature study on cancer therapy of warm-hot oriental medicine was done, and the results were as follows. 1. In oriental medicine, oncogens are six exopathogens, seven modes of emotion, overwork, pathogenic factors, and especially related with pathologic cold situation. 2. There are many capillaries in tuomr, and because temperature of inner space of tumor is higher than normal organization. Tumor cell has a character which is weak for high temperature. 3. Warm-hot herb drugs have effects of dissipating mass, warming kidney to reinforce yang and dispering, so it has a function of suppressing tumor as well as improving immunity in cancer therapy. 4. In traditional medical books, main prescriptions of cancer therapy are xinzhiyinyanggongjiwan(新製陰陽攻積丸), qianjinxiaoshiwan(千金硝石丸), feiqiwan(肥氣丸), xibenwan(息賁丸), fuliangwan(伏梁丸), beiqiwan, bentunwan(賁豚丸), zengsunwujiwan(增損五積丸), and these are composed of warm-hot herb drugs. 5. In current, the study of warm-hot drugs is progressed in immunological capacity, anti-tumor activity, stimulating bone marrow and regulating hormone secretion. It will be expected that advanced study of these must be accomplished in cancer patients.

  • Near infrared reflectance spectroscopy (NIRS) was applied for discrimination according to geographical origin (Korea, China) of herb drugs. Herbal medicine has an important role in clinical therapy in Asian countries such as Korea and China. The objective of this study is to provide a convenient and accurate method to determinate geographical origin (Korea, China) of herbal medicine for quality control whose quality is generally different according to geographical origin. A rapid, nondestructive and accurate discrimination was achieved by NIRS. Second derivative spectra of herb drugs were subjected to discriminant analysis. Herbal medicine drugs such as Cassia Semen, Ganoderma and Smilacis Rhizoma was discriminated accurately according to geographical origin using PLS regression method.

  • In Oriental medicine, the prescriptions composed of several herb medicines have been used. It is still unclear how the sum of several extracts of anti-thirst drugs represents the anti-lipid peroxidative action. Three anti-thirst herb medicines, Kalopanax pictus (K), Pueraria thunbergiana (P) and Rhus verniciflua (R), were extracted with MeOH and $H_2O$, respectively, and the former one was fractionated into the resultant EtOAc extract. Each extract was reconstituted to give KPR311, KPR131 and KPR113 where, for example, KPR311 represents the complex of K-P-R {3:1:1 (w/w/w)} of the three extracts. The order of the inhibitory effect in bromobenzene-induced lipid peroxidation in rats was as follows: EtOAc extract>$H_2O$ extract>MeOH extract. Extract complexes were found to be more potent than the extracts of individual crude drugs. The KPR131 of EtOAc extract was found to be the most potent among the tested samples. These anti-lipid peroxidative effects were also supported by the decrease of aniline hydroxylase activity and aminopyrine N-demethylase activity, on the other hand by the increase of epoxide hydrolase activity. All the tested samples were assayed in vitro antioxidative effects such as DPPH assay, ADP/NADPH/Fe$^{3+}$ assay and ascorbic acid/Fe$^{2+}$ assay. The EtOAc extracts also showed the most significant antioxidative effects. These results suggest that the sum of anti-thirst drugs could reflect the effects of respective crude drugs.s.s.
