Streptozotocin 유도 당뇨쥐에서 C3G 및 D3G 안토시안이 풍부한 검정콩 CJ-3호 종피 추출물의 혈중지질 개선효과 (Effects of the C3G/D3G anthocyanins-rich black soybean testa extracts on improvement of lipid profiles in STZ-induced diabetic rats)

  • 기존 연구에서 주로 사용하는 기타 재래종 검정콩 종피추출물 (콩단백질 혹은 총 안토시안)의 효과를 연구하는 경우와 달리, 본 연구에서는 재래종 서리태보다 C3G와 D3G 안토시안이 2배 이상 풍부한 검정콩 CJ-3호 종피추출물을 이용하였다. STZ로 유도된 당뇨쥐에 4주간 검정콩 CJ-3호 종피추출물을 처치한 결과 비록 체중감소, 혈당강하, 인슐린저항성 완화 등의 효과는 미비하지만 근육량이 유의적으로 증가하였는데 이는 일반적으로 항비만 식이요법에 따라 나타나는 근육소모를 보완하는 중요한 효과이다. STZ유도 당뇨 쥐에서 억제된 글리코겐이용률이 검정콩 CJ-3호 종피추출물에 의하여 증가한 것은 포도당이 에너지원으로 사용되면서 지방조직의 지질이용률을 억제하여 혈중 TG를 개선시켰고 간에서 HMG-CoA reductase 활성을 조절함으로써 혈청 TC를 낮추는데 기여하였다. 4주간의 투여기간을 연장한다면 향후 당뇨에서 발생하는 인슐린저항성 및 당화적 산화성억제에 따른 지질의 과산화물의 생성을 억제할 수 있는 가능성도 기대할 수 있겠다. 또한 검정콩 CJ-3호는 병 저항성이 높고 종실특성이 우수한 녹자엽 검정콩으로 생산성이 높아서 기능성에 대한 반복 실험을 확인한다면 기능성식품시장의 산업화도 가능하다. 따라서 C3G와 D3G 안토시안이 풍부한 기능성 식품의 경우 당뇨, 비만 및 암 등 난치병질환에서 나타나는 혈중지질 개선을 위한 임상적 효과 또한 기대해 볼 수 있겠다.

Effect of corn gluten and its hydrolysate consumptions on weight reduction in rats fed a high-fat diet

  • This study examined the effects of com gluten (CG) and its hydrolysate consumptions on weight reduction in rats fed a high-fat diet. Eight-month-old male Sprague-Dawley rats (n=40) were fed a high-fat diet (40% calorie as fat) for 4 weeks. They were then randomly divided into four groups and fed the isocaloric diets with different protein sources for 8 weeks. The protein sources were casein (control group), intact CG (CG group), CG hydrolysate A (CGHA group, 30% of protein as peptides and 70% as free amino acids) and CG hydrolysate P (CGHP group, 93% of protein as peptides and 7% as free amino acids). Body weight gain, adipose tissue weights, nitrogen balance, absorptions of energy, protein and fat, lipid profiles in plasma, liver and feces and hepatic activities of camitine palmitoyl transferase (CPT), fatty acid synthase (FAS), malic enzyme (ME) and glucose-6-phosphate dehydrogenase (G6PDH) were assessed. The CGHA diet had the highest amount of BCAAs, especially leucine, and most of them existed as free amino acid forms. The CGHA group showed significant weight reduction and negative nitrogen balance. Protein absorption and apparent protein digestibility in the CGHA group were significantly lower than those in other groups. Adipose tissue weights were the lowest in the CGHA group. Activity of CPT tended to be higher in the CGHA group than in other groups and those of FAS, ME and G6PDH were significantly lower in the CGHA group than in other groups. In conclusion, the CGHA diet which had relatively high amounts of free amino acids and BCAAs, especially leucine, had a weight reduction effect by lowering adipose tissue weight and the activities of FAS, ME and G6PDH in experimental animals, but it seemed to be a negative result induced by lowering protein absorption, increasing urinary nitrogen excretion and protein catabolism.

산업장 남성 근로자의 지방간 유병율 및 위험요인 연구 (Prevalence and Risk Factors of Fatty Liver among Industrial Male Workers)

  • Purpose: This study was conducted to estimate the prevalence rate, and related factors,of fatty liver in male industrial workers. Method: Fatty liver was diagnosed using ultrasonography. The data for abdominal ultrasonography, BMI, smoking, alcohol drinking, exercise, liver enzymes, and lipid profiles were collected in 4,604 male who were examined with a health screening program in 2005. Prevalence rate and associated factors of fatty liver were analyzed using SPSS v. 12.0. Results: Overall prevalence of fatty liver was 34.1% and higher in the 30-39 year male group. Age, body mass index(BMI), hypertriglyceridemia, hypercholesterolemia, HDL cholesterol, r-GTP level were independently associated factors with presence of fatty liver in logistic regression analysis. Conclusion: The prevalence of fatty liver and tendency of associated factors were similar to those in the western world. To prevent development of this hepatic disorder, which may result in end-stage liver disease, risk factors such as obesity and dyslipidemia must be monitored and controlled within normal levels. The results of this study suggested maintenance of a healthy lifestyle, including diet, exercise, and behavioral change, as fundamental rehabilitation nursing implications.

인삼양영탕(人蔘養榮湯)이 PTU로 유발된 Rat의 갑상선기능저하증에 미치는 영향 (Effects of Insamyangyoung-tang Aqueous Extracts on the Hypothyroidism Induced by Propylthiouracil in Rats)

  • Objectives : The object of this study was to evaluate the effect of Insamyangyoung-tang aqueous extracts (ISYYT) on the propylthiouracil (PTU) induced rat hypothyroidism. Methods : The rats were divided into 6 groups : intact control, PTU control, Levothyroxine (LT 4 ), 500, 250 and 125 mg/kg ISYYT treated groups. In ISYYT treated groups, PTU and ISYYT were administered for 4 weeks after 500, 250 and 125 mg/kg ISYYT were administered for 2 weeks. In LT 4 group, PTU and LT 4 were administered for 4 weeks. The changes were observed : the weigh, serum thyroid hormone levels, serum sex hormone levels, serum lipid profiles, serum liver enzyme levels, liver and testis antioxidant defense system, histopathology of thyroid gland, liver, epididymis, prostate and testis. Results were compared with PTU control group in this experiment. Results : In comparison with PTU control group, 500 and 250 mg/kg ISYYT treated groups showed significant increase of body, thyroid, liver, testis, epididymis and prostate weights, decrease of serum TSH levels with increase of serum T3 and T4 level, increase of serum testosterone and DHT levels with decrease of serum FSH levels, decrease of serum HDL with increase of TG and increase of serum AST levels. Histopathological inspections of hepatic and male reproductive organ damage induced by PTU were improved. And the changes of antioxidant defense system of Liver and testis induced by PTU were improve. There was no significant difference between 125 mg/kg ISYYT treated group and PTU control group in this experiment. Conclusions : The results obtained in this study considered that Insamyangyoung-tang may be effective in hypothyroidism and related organ damages.

수유기에 식이와 함께 섭취한 DHA가 산모의 혈액과 모유의 지질조성에 미치는 영향 (The Influence of DHA Supplementation in Maternal Diets on Fatty Acid Compositions of Plasma Lipids and Human Milk)

  • The most abundant long-chain polyunsaturated fatty acid in brain lipids is docosahexaenoic acid(C22 : 6 N-3, DHA). It is incorporated into nerve tissues mostly in utero and during the first year of life. DHA in brain is derived from either pre-formed DHA in human milk or by infant hepatic synthesis from linolenic acid in milk. This study was designed to investigate the effects of DHA supplementation on fatty acid profiles in maternal plasma lipid and breast milk. Twenty lactating women participated in the study. Seven women took 3g of fish oil per day and vitamin E for 28 days starting from the day of giving birth. Five women consumed 1.5g of fish oil as well as tivamin E, and the rest took vitamin E supplements for the same period of time. Dietary questionnaires and 3 consecutive 24-h recalls were collected to evaluate theri nutritional status and food habits. Finding that DHA intake from fish was not significantly different among three experimental groups, the partcipants were instructed to continue eating their usual home diets. Milk samples were taken on the day of giving birth, as well as the 7th, 14th and 28th day being the supplement phase, and finally 2 weeks after the cessating of DHA supplements. The amounts of the fish oil supplements produced significant dose-dependent increased in the DHA content of milk and plasma, but to a lesser degree. Base-line for 28 days raised the level to 2.05$\pm$0.43% and 1.5g/day supplement produced DHA levels of 1.02$\pm$0.19%. The results of this study indicated that relatively small amount of dietary DHA supplementation significantly elevats DHA content in milk. This would clearly elevate the infant's DHA intake which in turn may have implications for the infant's brain development.

益氣補血湯 물 추출물이 PTU로 유발된 Rat의 갑상선기능저하증에 미치는 영향 (Effects of Yikgeebohyul-tang Aqueous Extracts on the Rat Hypothyroidism Induced by Propylthiouracil)

  • Objectives The object of this study was to evaluate the effect of Yikgeebohyul-tang aqueous extracts (YKBHT) on the propylthiouracil (PTU)-induced rat hypothyroidism. Methods The rats were divided into 6 groups : intact vehicle control, PTU control, LT4, YKBHT 500, 250 and 125 mg/kg treated groups. Hypothyroidism was induced by daily subcutaneous treatment of PTU 10 mg/kg for 28 days. YKBHT aqueous extracts were administered once a day as an oral dose of 500, 250 and 125 mg/kg for 42 days. The changes were observed : weight of body, thyroid gland, liver, testis, epididymis and prostate, serum thyroid hormone levels, serum male sex hormone levels, serum lipid profiles, liver and testis antioxidant system. These results were compared with LT4 0.5 mg/kg intraperitoneally treated rats. Results These PTU induced hypothyroidism and related hepatic and male reproductive organ damages were favorably and dose-dependently inhibited by treatment of YKBHT 500, 250 and 125 mg/kg, and YKBHT also effectively regulated the PTU-induced abnormal antioxidant defense factor changes in the both liver and testis. Although, LT4 also inhibited PTU-induced hypothyroidism and relative liver damages. But it deteriorated the hypothyroidism related testis, epididymis and prostate damages through testicular oxidative damages. Conclusions : The result of this study suggests that YKBHT has favorable effects on the hypothyroidism and related liver and reproductive organ damages with augmentation of antioxidant defense factor in the testis and liver. YKBHT 500, 250 and 125 mg/kg dose-dependently inhibited PTU-induced hypothyroidism and related liver and male reproductive organ damages in rats.

비만과 이상지질혈증을 동반한 비알코올 지방간질환 환자 치험 1례 (A Case Report of Nonalcoholic Fatty Liver Disease with Obesity and Dyslipidemia)

  • Objectives: This study aims to report the clinical efficacy of Korean medical treatment of a patient with nonalcoholic fatty liver disease accompanied by obesity and dyslipidemia. Methods: A 52-year-old man with nonalcoholic fatty liver disease, obesity, and dyslipidemia was treated with Saenggangunbi-tang extract from November 3, 2021 to January 8, 2022. During the treatment period, the patient also undertook exercise and maintained a hypocaloric diet to reduce body weight. Changes were observed in the patient's symptoms, laboratory findings, such as liver enzymes and lipid profiles, and bioelectrical impedance analyzer results. Results: After taking Saenggangunbi-tang extract for approximately two months, with concomitant adjustments to exercise and diet, the serum levels of liver enzymes and triglyceride were decreased and the patient's body weight, body mass index, hepatic steatosis index, and visceral fat area were also reduced. In addition, symptoms including fatigue and dyspepsia improved. Conclusion: This study suggests that Saenggangunbi-tang extract with exercise and a regulated diet could be a helpful treatment strategy for managing nonalcoholic fatty liver disease with metabolic disorders.

대사이상 관련 지방간질환의 생간건비탕 치험 1례 (Case Report of Metabolic Dysfunction-Associated Fatty Liver Disease Treated with Saenggangunbi-tang)

  • Objective: This study identified the effects of Korean medicine treatment on a patient with metabolic dysfunction-associated fatty liver disease (MAFLD). Methods: A 43-year-old man with MAFLD was treated with Saenggangunbi-tang with regular exercise from August 13, 2022, to December 24, 2022, to reduce fatigue and dyspepsia and to improve laboratory findings, such as liver enzymes and lipid profiles. We observed changes in symptoms and laboratory findings during the approximately four-month treatment. Results: Treatment with Saenggangunbi-tang resulted in decreased serum levels of liver enzymes, triglycerides, hepatic steatosis index scores, and clinical symptoms. During the treatment, the patient performed regular exercise; however, there was no significant change in body weight until the end of the study. Conclusion: This study suggests the availability of Saenggangunbi-tang as a therapeutic option for managing MAFLD patients.

갈대순분말이 고지방을 급여한 마우스의 지질대사 및 적혈구 항산화방어계에 미치는 영향 (Effect of Young Phragmites communis Leaves Powder on Lipid Metabolism and Erythrocyte Antioxidant Enzyme Activities in High-Fat Diet Fed Mice)

  • 본 연구는 고지방을 급여한 마우스의 체내 지질개선 및 항산화 방어계에 미치는 갈대순의 영향을 구명하고자 하였다. 4주령의 ICR마우스(n=24)를 1주일간 적응시킨 후 정상식이를 급여한 정상군, 고지방을 급여한 고지방군, 고지방-갈대순분말 급여군으로 나누어 8주간 사육하였다. 갈대순분말은 사람이 하루에 3잔의 차를 마시는 양을 고려하여 식이에 1% 수준이 되도록 첨가 조제하여 급여하였다. 갈대순분말군의 체중, 일일 식이섭취량과 에너지섭취량은 고지방군에 비하여 감소하는 경향이었다. 갈대순분말 급여 시 혈장의 총 콜레스테롤 함량은 고지방군에 비하여 19% 감소되었고 중성지질 함량은 36% 유의적인 감소를 보였다. 갈대순분말은 고지방식이로 축적된 간 조직의 중성지질 함량 변화에는 영향을 미치지 않았으나 콜레스테롤 함량은 고지방군에 비하여 유의적으로 낮추었다. 심장조직의 중성지질과 콜레스테롤 함량 역시 정상군에 비하여 고지방군이 각각 145%, 112%의 유의적인 증가를 나타내었으나 갈대순분말 급여로 중성지질 함량은 감소경향을 보였으며 콜레스테롤 함량은 정상군과 유사한 수준으로 개선되었다. 갈대순분말은 고지방군에 비하여 분변으로의 중성지질과 콜레스테롤 배설을 각각 27%와 28% 유의적으로 증가시켰다. 갈대순분말은 간 조직의 지방산 산화에는 영향을 미치지 않았고 지방산 합성효소 활성을 유의적으로 억제하였다. 간 조직 중의 HMG-CoA reductase와 ACAT 활성도 역시 갈대순급여군이 고지방군에 비하여 유의적으로 낮았다. 적혈구 중의 SOD와 CAT 활성은 정상군에 비하여 고지방식이 급여 시 유의적인 증가를 보였으나 GSH-Px 활성은 고지방식이로 인한 변화가 관찰되지 않았다. 반면, 갈대순분말 급여는 적혈구의 SOD와 CAT 활성과 지질과산화물 함량을 고지방군에 비하여 유의적으로 낮추었다. 이와 같이 갈대순분말은 고지방식 이를 급여한 마우스의 간 조직에서의 지방산과 콜레스테롤 합성을 저해하고 분변으로의 지질배설을 증가시킴으로써 혈장 지질개선에 효과적이었으며 적혈구의 항산화 효소활성도와 지질과산화물 함량도 개선하는 것으로 나타났다.

Monascus Pilosus로 발효시킨 검정콩 첨가 식이가 고지방식이 마우스의 체중과 혈청 지방함량 및 간 조직 항산화계 효소활성에 미치는 영향 (Dietary effects of black bean fermented by Monascus pilosus on body weight, serum lipid profiles and activities of hepatic antioxidative enzymes in mice fed high fat diets)

  • The anti-obesity effects of fermented black bean were tested with mice fed a high fat diet for seven weeks. Body weight gain and feed efficiency ratio (FER) in the high fat diet control (HC) group were markedly higher, compared with those of the normal control (NC) group, but were significantly lower in the 2% black bean powder supplemented high fat diet (BB) group and 2% black bean powder fermented by M. pilosus supplemented high fat diet (BBM) group, compared with those of the HC group. Food intake in the HC and BB groups was significantly lower than that of the NC and BBM groups. Water intake in the HC group was significantly lower than that of the NC group, but was higher in the BB and BBM groups, compared with that of the HC group. On the other hand, relative liver and kidney weight in the HC group was lower than that of the NC group, but was higher in the BB and BBM groups, compared with that of the HC group. In addition, whereas epididymal fat weight in the HC group was markedly higher than that of the NC group, it was significantly lower in the BB and BBM groups, compared with that of the HC group. Meanwhile, hepatic GSH in the HC group was significantly lower than that of the NC group, but was slightly higher in the BB and BBM groups, compared with that of the HC group. Although hepatic LPO in the HC group was dramatically higher than that of the NC group, it was significantly lower in the BB and BBM groups, compared with that of the HC group. In addition, serum TG, total cholesterol, and LDL-cholesterol in the HC group was significantly higher than that of the NC group, but was significantly lower in the BB and BBM groups, compared with that of the HC group. On the contrary, HDL-cholesterol in the HC group was significantly lower than that of the NC group, but was higher in the BB and BBM groups, compared with that of the HC group. In addition, activity of XOR D type in the HC group was lower than that of the NC group, but was slightly higher in the BB and BBM groups, compared with that of the NC group. Activities of ROS scavenging enzymes, such as SOD, GPX, and GST in the HC group were significantly lower than those of the NC group, but were significantly higher in the BB and BBM groups, compared with those of the HC group. In addition, serum ALT activity in the HC and BB groups was higher than that of the NC group, but was significantly lower in the BB and BBM groups, compared with that of the HC group. In histopathological findings, hepatic fat accumulation in the HC group was higher than that of the NC group, but was lower in the BBM group, compared with that of the HC and BB groups. In particular, antiobese, hypolipidemic, and antifatty liver effect of black bean powder fermented by M. pilosus was specifically higher than that of non-fermented steamed black bean. In conclusion, the constituents of black bean fermented by Monascus pilosus have been proven to not only inhibit obesity and hyperlipidemia but also decrease hepatic fat accumulation in high fat diet-induced obese mice.
