Effect of the Toluene Administration on the Liver Damage in Rats Fed Low or Standard Protein Diet (저 및 표준단백식이로 성장시킨 흰쥐에 Toluene 투여가 간 손상에 미치는 영향)

  • To study the effect of toluene administration on the liver damage, rats were previously fed a low (casein 7%, LP) or standard(casein 20%, SP) protein diet and for four days toluene(50% in olive oil) was given at 0.2ml/100g body weight/day to the male rats, and then the degree of liver damage in toluenetreated animals fed LP were compared with those fed SP. The increasing rate of liver weight/body weight and the serum levels of xanthine oxidase to the control group were higher in rats fed SP than those fed LP. The decreasing rate of protein contents in cytosol, mitochondria and glycogen, glutathione contents of liver to the control group were higher in rats fed SP than those fed LP. In histopathological findings, the swelling of hepatic cell around the central vein was demonstrated in all the two groups toluene-treated rats. But the degree of swelling severity in hepatocytes was somewhat higher in rats fed SP than those fed LP. Therefore it is assumed that the degree of liver damage severity in toluenetreated animals was higher in rats fed SP than those fed LP.

Dietary Nigella sativa and Peganum harmala Oils Reverses Hyperglycaemia, Hepatotoxicity, and Metabolism in Rats

  • This study aims to evaluate the therapeutic action of administration of Nigella sativa (NS) and Peganum harmala (PH) oils in diabetes and hepatic toxicity. Results show that treatment of diabetic rats with NS oil or PH oil ameliorate hyperglycaemia induced stress oxidative and hepatic dysfunction in diabetic rats. Administration of NS or PH oil to diabetic rats caused an anti-diabetic and antioxidant activities by the decrease in plasmatic glucose level and increase in hepatic superoxide dismutase (SOD), catalase (CAT), and glutathione peroxidase (GPX) activities, reduced glutathione (GSH) and glycogen contents compared to untreated diabetic rats. Besides, NS and PH oils protect the hepatic function observed by decrease of triglyceride (TG), total cholesterol (TCh), and increase of high density lipoprotein-cholesterol (HDL-Ch) levels in serum and hepatic tissues. Moreover, a diminution in the bilirubin, transaminase glutanic pyruvic (TGP), and transaminase pyruvic oxaloacetic (TPO) contents in serum and the thiobarbituric acid-reactive substances levels (TBARs) in hepatic tissues are also detected.

Antidiabetic and Antioxidative Effects of Corni fructus In Streptozotocin-Induced Diabetic Rats (산수유 추출물이 Streptozotocin으로 유발된 흰쥐의 항당뇨 및 항산화 작용에 미치는 효과)

  • The extraction yield of Corni fructus was about 47.5% by extract apparatus. This study was done to investigate the antidiabetic and antioxidative effects of Corni fructus in Streptozotocin (STZ)-induced diabetic rats. The contents of serum glucose, total cholesterol and hepatic lipid peroxide, glutathione were significantly decreaed (p<0.05) in Corni fructus treated group compared to the those of STZ-control group, also content of triglyceride (TG), atherogenic index (AI) and activity of catalase were decreased, but not statistically significant. High density lipoprotein (HDL)-cholesterol and HDL-cholesterol/total cholesterol ratio (HTR) were increased in Corni fructus treated group compared to the those of STZ-control group. The activities of glutathione-S-transferase (GST) and superoxide dismutase (SOD) were significantly decreaed (p<0.05) in Corni fructus treated group compared to the those of STZ-control group. The content of hepatic glycogen and activities of glucose-phosphate dehydrogenase (G-6 PDH), glucokin-ase were significantly increased, but activity of glucose-Grphosphatase (G-6 Pase)was decreased in Corni fructus treated group compared to the those of STZ-control group. Therefore, these results indicated that ethanol extract of Corni fructus would have antidiabetic and antioxidative effects in STZ-induced diabetic rats.

Antidiabetic Effect of Glechoma hederacea LINNAEUS in Streptozotocin-Induced Diabetic Rats (Streptozotocin 유발 당뇨 흰쥐에 대한 금전초 추출물의 혈당 강하 효과)

  • This study was done to investigate the antidiabetic effect of Glechoma hederacea LINNAEUS in Streptozotocin (STZ)-induced diabetic rats. Diabetes was induced by intravenous injection of STZ at a dose of 45 mg/kg dissolved in citrate buffer. The methanol extract of Glechoma hederacea was orally administrated once a day for 6 days. The contents of serum glucose, triglyceride (TG), total cholesterol and Atherogenic Index (Al) were significantly decreased, but high density lipoprotein (HDL)-cholesterol and HDL-cholesterol/total cholesterol ratio (HTR) were significantly increased in Glechoma hederacea treated STZ-sample group compared to the those of STZ-control group. The content of hepatic lipid peroxide and activity of catalase were decreased, but content of glutathione as well as activities of glutathione-S-transferase and superoxide dismutase were increased in Glechoma hederacea treated STZ-sample group compared to the those of STZ-control group. The content of hepatic glycogen and activities of glucose-6-phosphate dehydrogenase, glucokinase were significantly increased, but activity of glucose-6-phosphatase was decreased in Glechoma hederacea treated STZ-sample group compared to the those of STZ-control group. These results indicated that methanol extract of Glechoma hederacea would have antidiabetic effect in STZ-induced diabetic rats.

Diabetes was induced by intravenous injection of STZ at a dose 45mg/kg.b.w. dissolved in citrate buffer(pH4.5). The ethanol extract of Cibotium barometz was orally administrated once a day for 7 days. The contents of serum glucose, triglyceride(TG) and total cholesterol were significantly decreased(p<0.05) in Cibotium barometz treated group compared to the those of STZ-control group, The content of glutathione(GSH) and activity of gluthathione-s-transferase(GST) were significantly increased (P<0.05) in Cibotium barometz treated group compared to the those of STZ-control group. and activityes of catalase(CAT) and glutathione peroxidae(GSH-Px) were signiicantly decreased (P<0.05) in Cibotium barometz treated group compared to the those of STZ-control group. Also the content of hepatic glycogen and activities of glucose-phosphate dehydrogenase(G-6-PDH)and glucokinase(GK) were significamtly increased(p<0.05), but activity of glucose-6-phosphatase (G-6-Pase) was significamtly decreased (p<0.05) in Cibotium barometz treated group compared to the those of STZ-control group. These results indicated that ethanol extract of Cibotium barometz would have antidiabetic and antioxidant effects in STZ-induced diabetic rats.

But not statistically significant In Lycii fructus treated group compared to the those of STZ-control group These results indicated that ethanol extract of Lycii fructus would have antidiabetic and antioxidant effects in STZ-induced diabetic rats.

Ethanol Effect of Nelumbo nucifera Root on Carbohydrate Methabolism Related Enzyme Activities and Antioxidative Effect in Streptozotocin-Induced Diabetic Rats (Streptozotocin으로 유발된 당뇨 흰쥐에 대한 연근 에탄올 추출물의 당대사 효소활성과 항산화 작용에 미치는 영향)

  • This study was done to investigate the carbohydrate metabolism related enzyme activities and antioxidative effects of Nelumbo nucifera(N.N) Root in streptozotocin (STZ)-induced diabetic rats. The contents of serum glucose, triglyceride (TG) and Total cholesterol were significantly decreaed in N.n treated group compared to the those of STZ-control group. The activity of glucose-6-pase(G-6-Pase) was significantly decreased in N.N treated group. Also the activity of glucokinase(Gk) was significantly increaed in N.N treated group. The content of hepatic glycogen was significantly increaed in N.N treated group, in addition, content of malondialdehyde(MDA) was significanly decreased in N.N treated group. Also, content of glutathione(GSH) was significanly increased in N.N treated froup. whereas, activity of catalase(CAT) was significantly decreaed in N.N treated group compared to the those of STZ-control group. activity of glutathione peroxidase(GSH-Px) was inecreaed. In conclusion, these results indicated that ethanol extract of N.N would have carbohydrate metabolism antioxidative effects in STZ-induced diabetic rats.

Effects of Green Tea on Hepatic Antioxidative Defense System and Muscle Fatigue Recovery in Rat after Aerobic Exercise (녹차가 유산소 운동 후 흰쥐 간조직의 항산화 작용 및 근피로 회복에 미치는 영향)

  • The purpose of this study was to investigate the effects of green tea on hepatic antioxidative defense system and recovery of muscle fatigue in rat after aerobic exercise. Male Sprague-Dawley rats weighing 150$\pm$ 10 g were randomly assigned to one normal (N) group and aerobic exercise training groups. Exercise training groups were classified into two groups: training (T) group and green tea (TG) group which were supplemented the distilled water and green tea extracts by dringking water during experimental periods, respectively. The experimental rats in exercise training groups (T and TG) ran on a treadmill 30 min/day at a speed of 28 m/min (7% incline) 5 days/week or were cage confined (Normal group) for 4 weeks. And rats were sacrificed with an overdose of pentobarbital injection just after running. Hepatic xanthine oxidase (XOD) activities were not significantly different among three groups. The activity of superoxide dismutase (SOD) in T group was no significant difference from N group, but those of TG groups were significantly increased, compared with that of T group. Hepatic glutathione peroxidase (GSHpx) activites of TG groups showed a similar tendency to that of normal group, but it was increased to 20% in TG group, compared with normal group. The reduced glutathione (GSH) contents in liver was not significantly different from that of any three group. The oxidized glutathione (GSSG) contents in T group was increased to 69%, compared with the normal group, but TG group significantly decreased, compared with the T group. The ratio of GSH/GSSG in liver of T group was lower than that of normal group, but those of TG group was a similar tendency to that of normal group. Contents of thiobarbituric acid reactive substance(TBARS) in T group was increased to 52%, compared with that of normal group but those of TG group were recovered the normal level. Contents of hepatic glycogen in T group were decreased to 23% compared with those of normal group, while that of TG group was the same as normal levels. The contents of serum lactic acid in T group were increased to 261%, compared with normal group, but those of TG group maintained the normal level by green tea supplementations. In conclusion, the effects of green tea in exercise training rats would appear to reduce peroxidation of tissue as an antioxidative defense mechanism and promote recovery of muscle fatigue.

(-) Epigallocatechin gallate restores ethanol-induced alterations in hepatic detoxification system and prevents apoptosis

  • The present study was designed to estimate the protective effect of (-) epigallocatechin gallate (EGCG) on ethanol-induced liver injury in rats. Chronic ethanol administration (6 g/kg/day ${\times}$ 60 days) caused liver damage that was manifested by the elevation of markers of liver dysfunction - aspartate aminotransferase, alanine aminotransferase, alkaline phosphatase, lactate dehydrogenase, bilirubin and ${\gamma}$-glutamyl transferase in plasma and reduction in liver glycogen. The activities of alcohol metabolizing enzymes such as alcohol dehydrogenase and aldehyde dehydrogenase were found to be altered in alcohol-treated group. Ethanol administration resulted in the induction of cytochrome p450 and cytochrome-$b_{5}$ activities and reduction of cytochrome-c reductase and glutathione-S-transferase, a phase II drug metabolizing enzyme. Further, ethanol reduced the viability of isolated hepatocytes (ex vivo) as assessed by trypan blue exclusion test and induced hepatocyte apoptosis as assessed by propidium iodide staining. Treatment of alcoholic rats with EGCG restored the levels of markers of liver injury and mitigated the alterations in alcohol metabolizing and drug metabolizing enzymes and cyt-c-reductase. Increased hepatocyte viability and reduced apoptotic nuclei were observed in alcohol + EGCG-treated rats. These findings suggest that EGCG acts as a hepatoprotective agent against alcoholic liver injury.

The Effect of Meliae toosendan fructus Ethanol Extract on Blood Glucose, Lipid metabolism, Carbohydrate Methabolism Related Enzyme Activities and Antioxidative Effect in Streptozotocin-Induced Diabetic Rats (천련자 에탄올 추출물이 Streptozotocin으로 유발된 당뇨 흰쥐에 대한 혈당, 지질대사, 당대사 효소 활성과 항산화 작용에 미치는 영향)
