Effect of Black Garlic and Herb Formulas on Lipid Profiles and Antioxidant Status in Rats by Interval Running Training (간헐적 운동부하에서 흑마늘과 생약재의 복합처방이 흰쥐의 체내 지질 성분 및 항산화에 미치는 영향)

  • To improve the functionality of black garlic drinks, black garlic extract (5%) and five herb extracts (1%) were mixed in 70:30 (v/v) ratios as BHF1, and BHF2 was prepared using a 3X concentration of BHF1. After the black garlic and herb formulas (BHFs) were administered over the course of five weeks in rats by interval running training, the lipid profiles and the antioxidant enzyme activities were tested. The total phenolic content of the BHFs were significantly higher in BHF2 than they were in BHF1, and their antioxidant activities were dependent upon the total phenolic content. No significant difference was found in the total serum protein levels among the rats in the Ex-con group by interval running training and the rats in the BHFs-fed groups. However, the albumin level was significantly higher in the Ex-BHF2 to Ex-con group. AST and ALT activities significantly decreased in the BHFs-fed groups compared to the Ex-con group. In terms of changes in the serum lipid profiles, no significant difference was found between the specimens that underwent interval running training and those that did not undergo interval running training. Triglyceride levels, total cholesterol, LDL-C, and HTR levels in the serum were significantly decreased in the Ex-BHF2 to Ex-con group. No significant difference was found in the total lipid levels in the livers of the BHFs-fed groups and the Ex-con group. The triglyceride levels and total cholesterol levels in the Ex-BHF2 group were significantly lower compared to another group. Hepatic catalase activity was significantly increased in the Ex-BHF2 group, but SOD and GSH-px activities were significantly increased as the concentration of the BHF. The antioxidant enzyme activities by supplementation of BHFs increased; thus, three intakes of BHF each day could improve antioxidant status against different types of oxidative stress.

Effects of Uncooked Powdered Food on Antioxidative System and Serum Mineral Concentrations in Rats Fed Unbalanced Diet (생식제품 급여가 영양불균형식이를 섭취하는 흰쥐의 항산화체계 및 혈청 무기질 농도에 미치는 영향)

  • Antioxidative function of uncooked powdered food (Sangsik) was evaluated in rats consuming nutritionally unbalanced diet including 1% cholesterol, high proportion of animal lipids (lard : soybean oil : 8 . 2) , sub-optimal levels of vitamin and mineral mixture along with 0.5% ethanol in drinking water. The uncooked powdered food tested in the present study was a mixture composed of 42 kinds of plant foods (cereals, legumes, seaweeds, vegetables, and fruits) supplemented with vitamins and minerals, and dietary fiber. Control rats were fed the semi-purified diet based on the AIN-93G composition, and nutritionally unbalanced rats were divided into 3 groups, and fed one of the following diets with 0.5% ethanol in drinking water for 5 weeks : unbalanced control diet (UC) ,20% Sangsik powder supplemented diet (S20), and 40% Sangsik powder supplemented diet (S40) . Food efficiency ratio was significantly higher in rats fed S40 compared to the value for rats fed UC (p < 0.05). Hepatic level of thiobarbituric acid reactive substances (TBARS) was significantly lower in rats fed UC compared to that for control rats (p < 0.05) , and was not influenced by dietary supplementation of the Sangsik powder. Hepatic superoxide dismutase (SOD) activity was significantly higher in rats fed UC compared to that for control rats (p < 0.05) , and significantly reduced in rats fed S20 or S40 compared to the value for unbalanced control rats. Feeding unbalanced control diet significantly reduced the ratio of hepatic GSH-Px + catalase/SOD activities compared to the value for control rats, and this decrease in the ratio of antioxidant enzyme activities was reversed by adding the Sangsik powder to the diet at 20% (p <0.05) . Based on the results of antioxidant enzyme activities, feeding uncooked powdered diet appears to provide a favorable environment for body's antioxidative defense mechanism. Serum levels of Fe and Cu were significantly lower in rats fed the Sangsik powder supplemented diets compared to the value for unbalanced control rats (p < 0.05) , and levels of Se, Mn, and Zn were also tended to be decreased by dietary supplementation of the Sangsik powder. These results postulate the possibility that ingredients used in the uncooked powdered food may decrease the bioavailability of trace elements in rats.

Anti-atherogenic Effect of Isoflavone through Hypolipidemic, Anti-oxidative and Anti-inflammatory Actions in C57BL/6 Mice (C57BL/6 Mice에서 이소플라본의 지질강하, 항산화, 항염증효과를 통한 항동맥경화 효과)

  • This study was carried out to investigate the effect of isoflavone on the atherogenic effect in C57BL/6 mice. C57BL/6 female mice, 5 weeks of age, were fed on chow diets for 2 weeks during adjustment period. Mice weighing approximately $17.9{\pm}0.9\;g$ were divided into 4 groups and were fed on the experimental diets containing isoflavone for 8 weeks. Experimental groups were control (atherogenic diet), IF-10 (atherogenic diet with isoflavone 10 mg/100 g diet), IF-40 (atherogenic diet with isoflavone 40 mg/100 g diet) and IF-100 (atherogenic diet with isoflavone 100 mg/100 g diet). Food efficiency ratio was not different among the experimental groups. Plasma triglyceride (TG) concentrations were lower after 4 weeks in isoflavone supplementation groups than in control group, whereas monocyte chemoattractant protein-1 and thiobarbituric acid reactive substances (TBARS) levels of plasma were significantly (p<0.05) decreased in the isoflavone supplementation groups in a dose dependent manner. Both hepatic TG and cholesterol levels were significantly lowered in IF-100 than control. Hepatic glutathione concentrations were higher in the IF-100 group than in the other groups. Hepatic antioxidant enzyme activities including glutathione-reductase, glutathione-peroxidase, catalase, and Mn-superoxide dismutase were significantly higher in the isoflavone supplemen-tation groups in a dose dependent manner. From the above results, it is concluded that isoflavone may reduce the risk of atherosclerosis via hypolipidemic, anti-oxidative and anti-inflammatory effects.

Effect of Crude Saponins from Soybean Cake on Body Weight and Glucose Tolerance in High-Fat Diet Induced Obese Mice (대두박 사포닌 보충이 식이성 유도 비만마우스의 체중과 내당능에 미치는 영향)

  • This study investigated the beneficial effects of crude saponins from soybean cake on body weight and glucose tolerance in high-fat (37% calories from fat) diet fed C57BL/6 mice. The mice were supplemented with three doses of saponins (0.5%, 1.0%, and 1.5%, wt/wt) and 1.0% Garcinia cambogia (wt/wt), positive control for 9 weeks. The body weight, visceral fat weight and epididymal adipocyte area were significantly reduced in the saponin supplemented groups in a dose dependent manner compared to the high-fat group. Saponins did not significantly affect food intake; however, cambogia significantly lowered food intake compared to the high-fat fed control group. The crude saponins from soybean cake supplement significantly lowered plasma leptin, triglyceride and total cholesterol levels, whereas they significantly elevated the fecal excretion of triglyceide in a dose dependent manner compared to the high-fat group. Cambogia did not affect the fecal excretion of lipid in the diet-induced obese mice. Supplementation of 1.5% saponin reduced the hepatic triglyceride content compared to the high-fat group. High-fat induced glucose intolerance with the elevation of blood glucose levels compared to the normal group; however, the saponins supplement significantly improved postprandial glucose levels. After 9 weeks of being fed a high-fat diet, the mice presented with significantly increased activities of hepatic fatty acid synthase and fatty acid ${\beta}$-oxidation; however, saponins and cambogia normalized these activities. These results indicate that saponins from soybean cake exhibit a potential anti-obesity effect and may prevent glucose intolerance by reducing body weight and plasma lipids, increasing fecal lipid excretion and regulating hepatic lipid metabolism in high-fat fed mice.

Effect of Black Garlic and Gaeddongssuk (Artemisia annua L.) Extracts on the Lipid Profile and Hepatic Antioxidant Enzyme Activities of Exercised Rats (흑마늘 및 개똥쑥 추출물의 급이가 강제운동 시 흰쥐의 체내 지질 성분 및 간조직의 항산화효소 활성에 미치는 영향)

  • To develop functional products based on black garlic, a black garlic extract (BG) of 7 brix, a gaeddongssuk extract (GS) of 0.7 brix and two types of mixtures (MBS-I, black garlic 7 brix : gaeddongssuk 0.7 brix; MBS-II, black garlic 14 brix : gaeddongssuk 1.4 brix, 93:7, v/v) were supplemented to rats training on a treadmill for 4 weeks. Body weight from the training did not decrease during the experimental period. Serum albumin content significantly increased in the groups fed an experimental diet compared to the control. The BUN content significantly decreased in BG and MBS-II groups compared to the control. AST and ALP activities significantly decreased in the groups fed an experimental diet compared to the control. Serum triglyceride and total cholesterol levels in MBS-I and MBS-II groups significantly decreased compared to the control. Lipid levels in the serum and liver tissue were not significantly different between the MBS-I and MBS-II groups. The lipid peroxide content in the serum and liver tissue was significantly reduced in the groups fed all extracts compared to the control; the serum and liver lipid contents was lowest in the MBS-I and MBS-II groups, respectively. Hepatic catalase activity in the GS and MBS groups increased by 1.8~2.3 times compared to the control. SOD and GSH-px activities significantly increased from treatment with the extracts by 1.3~1.5 times and 1.2~1.7 times, respectively. These results indicate that a mixture of BG and GS extracts has higher biological activity than a single supplementation of BG or GS extract. Therefore, the addition of gaeddongssuk to black garlic (MBS-I and MBS-II) is effective as a defense material against oxidative stress. MBS-I may be especially effective for its biological activities.

Effect of Chronic Alcohol Feeding and 2-Acetylaminofluorene Treatment on Hepatic Mitochondrial ATPase Activity and Membrane Lipid Composition in Rats (만성 알코올 섭취시 2-Acetylaminofluorene 투여가 흰쥐 간 미토콘드리아 ATPase 활성도와 막지질 조성에 미치는 영향)

  • This study was done ot investigate the effect of chronic alcohol feeding and acetylaminofluorene(2-AAF) treatment on hepatic mitochondrial ATPase activity andmembrane lipid composition. Male Sprague-Dawley rats, weighing 120~125g, were fed for 6 weeks on a liquid diet containing 35% of calories as ethanol. After 4 weeks of experiment diet feeding, 2-AAF(100mg/kg body weight) was injected twice a week intraperitoneally. Body weight and percent liver weight per body weight were significantly changed by ethanol feeding. Hepatic mitochondrial ATPase activity significantly decreased by ethanol feedings but not by 2-AAF treatment. In comparison to control, the ATPase activity of ethanol-AAF group decreased 29.3%. Since phospholipid(PL) content of mitochondria has an interaction effect between ethanol and 2-AAF treatment, 2-AAF treatment significantly increased phospholipid content in only ethanol fed group. Total cholesterol(C) level of mitochondria significantly increased by ethanol feeding. Consequently C/PL ratio of ethanol group was significantly higher than that of control group. The analysis of mitochondrial PL composition showed that cardiolipin(CL) significantly increased by 2-AFF treatment in control group. Phosphatidyl choline(PC) significantly increased by ethanol feeding, whereas PC significanlty decreased and phosphatidyl ethanolamine(PE) significantly increased by 2-AAF treatment. 2-AAF treatment also showed a significant increase in PE/PC ratio. Fatty acid patterns of mitochondria were also changed by either ethanol or 2-AAF although the severity of the changes was not great. These data suggest that the reduced mitochondrial ATPase activity in ethanol-AAF group may be a consequence of a changes in mitochondrial membrane lipid composition such as PE/PC ratio, C/PL ration and fatty acid patterns.

Effect of Dietary Supplementation with Bitter Melon on Lipids and Hepatic Enzyme Levels in Streptozotocin Induced Diabetic Rats (여주열매 첨가식이가 당뇨 흰쥐의 지질과 항산화효소 수준에 미치는 영향)

  • This study examined the effect of Momordica charantia L. (bitter melon: BM) on lipid and hepatic antioxidative enzyme levels in diabetic rats. Diabetes mellitus was induced in male Sprague-Dawley rats by injection of streptozotocin (STZ), and rats were fed for 4 weeks with experimental groups divided into four groups: a normal control group, STZ-control and STZ-BM 5% & STZ-BM 10% treated groups. Levels of free fatty acids (FFA), high-density lipoprotein cholesterol (HDL-chol), triglycerides (TG) in plasma and malondialdehyde (MDA) & protein in liver, catalase (CAT), superoxide dismutase (SOD), glutathione-S-transferase (GST), and xanthine oxidase (XOD) were measured in liver cytosol. Level of HDL-chol significantly increased in the STZ-BM 5% diabetic group. TG & FFA levels were significantly higher in all diabetic groups compared to the control group. MDA and protein levels were significantly higher in the STZ-BM 5% group compared to all other experimental group. CAT level was higher in the supplementary group with BM compared to the STZ-control group, although the difference was not significantly different. SOD level was not significant in any experimental groups. GST level was significantly higher in the BM-treated groups compared to the STZ-control group. XOD level was significantly lower in the BM 5% group and significantly decreased in all experimental groups. These results show that supplementation of BM fruit powder may have beneficial effects on diabetic complications and damage caused by oxidative stress.

Effects of Green Tea Powder on the Disorders of Lipid Metabolism and Hepatic Functions in Rats treated by 2,3,7,8-Tetrachlorodibenzo-p-dioxin (TCDD) (녹차가 다이옥신계 TCDD(2,3,7,8-tetrachlorodibenzo-p-dioxin)에 노출된 흰쥐의 지질대사 및 간 독성물질대사에 미치는 영향)

  • This study investigated the effects of green tea on the disorders of lipid metabolism, oxidative system and hepatic functions induced by 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD), using adult male rats (SD) for 3 weeks. These 36 animals were divided into four groups. TCDD ($50{\mu}g/kg$ BW) was intraperitoneally injected at the beginning of experiment. Green tea powder was added 1% or 3% levels in basal diets respectively. Relative weights of thymus were decreased about one-third of control group, but those of liver, brain and testis were significantly increased in rats treated TCDD. Neutrophill% and lymphocyte% by TCDD treatment was improved by green tea diets. In liver functional enzyme, elevation of glutamic oxaloacetic transaminase (GOT) and alkaline phosphatase (ALP) activities due to TCDD treatment was lowered by green tea diets. The concentrations of serum and liver lipids were significantly increased by TCDD treatment, however, those of serum and liver triglyceride tended to decrease by green tea diets. Fecal lipid excretion was increased in rats fed green tea diets. Especially, fecal total cholesterol level was significantly elevated by 3% green tea diets. The activities of superoxide dismutase (SOD) were increased in rats fed 3% green tea diets. Increment of benzphetamine N-demethylase (BPND) activity by TCDD treatment was declined by 1% green tea diets. These results indicate that green tea can exert improving effects on liver lipid accumulation and unfavorable hepatic functions, and elevate antioxidation.

Protective Effects of Extracts of Hovenia dulcis Thunb on Hepatotoxicity in Carbon Tetrachloride Intoxicated Rats (지구자나무 추출물이 사염화탄소로 유발된 흰쥐의 간손상에 미치는 영향)

  • This study was performed to investigate the protective effects of Hovenia dulcis Thunb on hepatotoxicity in carbon tetrachloride-intoxicated rats. Male Sprague-Dawley rats (220~240 g) were used as experimental groups, which were divided into 7 groups; Control group, $CCl_4$-treated group, hexane fraction pretreated and $CCl_4$-treated group, chloroform fraction pretreated and $CCl_4$-treated group, ethylacetate fraction pretreated and $CCl_4$-treated group, butanol fraction pretreated and $CCl_4$-treated group, $H_2O$ fraction pretreated and $CCl_4$-treated group. After 6 days, the activities of aminotransferase, contents of cholesterol, TG and hepatic lipid peroxide content in chloroform fraction pretreated and $CCl_4$-treated group were significantly decreased (p<0.05) compared to the only $CCl_4$-treated group. The content of glutathione and activities of GST in chloroform fraction pretreated and $CCl_4$-treated group were also significantly increased (p<0.05) compared to the only $CCl_4$-treated group. In addition, activities of SOD, catalase and GSH-Px in chloroform fraction pretreated and $CCl_4$-treated group were significantly decreased (p<0.05) compared to the only $CCl_4$-treated group. These results indicated that the chloroform fraction of Hovenia dulcis Thunb methanol extract showed hepatoprotective effect in carbon tetrachloride-intoxicated rats.

Lipase-Inhibitory and Anti-Oxidative Activity of the Methanol Extract and the Powder of Phellinus linteus (상황버섯 자실체 메탄올 추출물과 분말의 지방소화효소 억제 및 항산화 활성)

  • Phellinus linteus (PL) has been known to exhibit potent biological activity. The present study was designed to investigate lipase-inhibitory and anti-oxidative activity of the methanol extract and the powder of PL fruiting body. The methanol extract of PL appeared to have the inhibitory activity against pancreatic lipase with an $IC_{50}$ value of $36.3\;{\mu}g/mL$, and the scavenging activity of DPPH radical with an $IC_{50}$ value of $20.1\;{\mu}g/mL$, which was similar to that of vitamin C ($IC_{50}\;18.3\;{\mu}g/mL$). To investigate the lipase-inhibitory and anti-oxidative effect of PL on animal, Sprague-Dawley rats were fed with high-fat diet supplemented with either 2% or 5% PL powder for 8 weeks. Total food intake was significantly increased, but body weight was not changed by PL powder supplementation. However, fecal fat excretion of the experimental groups fed with the PL powder were higher than that of the control group. PL powder showed a decrease in the plasma total cholesterol, LDL-cholesterol, and the hepatic total cholesterol levels. The anti-oxidative enzyme activities were also affected by PL supplementation. Glutathione peroxidase (GSH-Px) in the plasma and liver were significantly increased by 98% and 46% in the 2% PL group, and 99% and 32% in the 5% PL group, respectively. Total superoxide dismutase (T-SOD) activity was not affected by PL supplementation. DNA damage was measured by the comet assay in the lymphocytes collected after 2 weeks, 4 weeks, and 8 weeks of feeding PL supplemented diet. Lymphocyte DNA damage was decreased in the PL supplemented group. Furthermore, PL feeding enhanced the resistance to lymphocyte DNA damage caused by an oxidant challenge with $H_2O_2$.
