The inhibitory effect of IgY against Vibrio parahaemolyticus and Vibrio vulnificus responsible for seafood-borne diseases was investigated in this study. Water-soluble fractions (WSF) of protein containing IgYs were isolated from the egg yolk of hens initially immunized with formalin-inactivated V. parahaemolyticus or V. vulnificus. Protein, total and specific IgY contents of the WSF were determined. The inhibitory and protective effects of IgYs on the growth of V. parahaemolyticus and V. vulnificus were assayed in liquid medium and in mice. IgYs showed high affinity to their corresponding antigens with high titer from day 28 onwards. Protein contents and total IgY concentrations remained stable throughout the immunization period, whereas specific IgY concentrations increased steadily and reached a plateau at day 49. Specific IgY powder (150 mg/ml) significantly inhibited further multiplication of both V. parahaemolyticus and V. vulnificus in liquid medium as compared with the control IgY. The bacteria count in mice feces was lower in mice pretreated with specific IgYs than in those pretreated with PBS or control IgY. Higher survival of mice was observed in the experimental groups pretreated with either anti-V. parahaemolyticus (75% survival) or anti-V. vulnificus (87% survival) IgYs, compared with those in the control groups pretreated with PBS or nonspecific IgY. All mice in the control groups died within three days after bacteria inoculation; hence, the protective effect of specific IgYs against infection caused by V. parahaemolyticus and V. vulnificus was demonstrated.

Objective: Eggshell color is an important indicator of egg quality for consumers, especially for brown eggs. Various factors related to laying hens and their environment affect brown eggshell coloration. However, there have been no studies investigating hepatic functions of laying hens with variable intensity of brown eggshell color. Therefore, this study was aimed to identify potential factors affecting brown eggshell coloration in aged laying hens at the hepatic transcriptomic level. Methods: Five hundred 92-wk-old Hy-line Brown laying hens were screened to select laying hens with different intensity of brown eggshell color based on eggshell color fans. Based on eggshell color scores, hens with dark brown eggshells (DBE; eggshell color fan score = 14.8) and hens with light brown eggshells (LBE; eggshell color fan score = 9.7) were finally selected for the liver sampling. We performed RNA-seq analysis using the liver samples through the paired-end sequencing libraries. Differentially expressed genes (DEGs) profiling was carried out to identify their biological meaning by bioinformatics. Results: A total of 290 DEGs were identified with 196 being up-regulated and 94 being down-regulated in DBE groups as compared to LBE groups. The Kyoto encyclopedia of genes and genomes (KEGG) analysis revealed that these DEGs belong to several biological pathways including herpes simplex infection (toll-like receptor 3 [TLR3], cyclin-dependent kinase 1, etc.) and influenza A (TLR3, radical S-adenosyl methionine domain containing 2, myxovirus [influenza virus] resistance 1, etc.). Genes related to stress response (ceremide kinase like) and nutrient metabolism (phosphoenolpyruvate carboxy-kinase 1, methylmalonic aciduria [cobalamin deficiency] cblB type, glycine receptor alpha 2, solute carrier family 7 member 11, etc.) were also identified to be differentially expressed. Conclusion: The current results provide new insights regarding hepatic molecular functions related to different intensity of brown eggshell color in aged laying hens. These insights will contribute to future studies aiming to optimize brown eggshell coloration in aged laying hens.
A recent genetic study with Brucella abortus revealed the secretion activator gene A (SagA) as an autolysin component creating pores in the peptidoglycan (PGN) layer for the type IV secretion system (T4SS) and peptidoglycan hydrolase inhibitor A (PhiA) as an inhibitor of SagA. In this study, we determined the crystal structures of both SagA and PhiA. Notably, the SagA structure contained a PGN fragment in a space between the N- and C-terminal domains, showing the substrate-dependent hinge motion of the domains. The purified SagA fully hydrolyzed the meso-diaminopimelic acid (DAP)-type PGN, showing a higher activity than hen egg-white lysozyme. The PhiA protein exhibiting tetrameric assembly failed to inhibit SagA activity in our experiments. Our findings provide implications for the molecular basis of the SagA-PhiA system of B. abortus. The development of inhibitors of SagA would further contribute to controlling brucellosis by attenuating the function of T4SS, the major virulence factor of Brucella.
The purpose of this survey was to collect basic data on breeding systems of animal welfare-approved laying hen farms in Korea. Questionnaires were mailed to 64 animal welfare-approved farms, and 20 questionnaires (31.3%) were returned. The housing systems were fabric coverlet (4 farms, representing 20%), naturally farmed (Yamagisi, 7 farms, 35%), and steel panel-framed housing (9 farms, 45%). The 20 farms had stocking densities of $2{\sim}3birds/m^2$ (2 farms; 10%), $4{\sim}5birds/m^2$ (10 farms; 50%), and $6{\sim}7birds/m^2$ (8 farms; 40%). Breeding methods were floor-housed (14 farms; 70%), free-range (3 farms; 15%), and floor plus free-range (3 farms; 15%). Stocking density was $4{\sim}6birds/m^2$ at most of the farms with fabric coverlet and naturally farmed housing and $6{\sim}7birds/m^2$ at seven farms (of 9 farms) with a steel panel-framed housing. The daily feed intake of 11 farms (55%) was between 120 and 130 g, which included 3 farms (15%) with fabric coverlet, 3 farms (15%) with naturally farmed housing, and 5 farms (25%) with steel panel-framed housing. The age of peak production was 24~28 weeks overall 20 farms. Over 80% of production on fabric coverlet, naturally farmed, and steel panel-framed house farms was on 3, 4 and 6 farms, respectively. Respiratory disease on the 20 farms represented 55% of total disease incidence, and of each housing type represented 75% (fabric coverlet), 70% (naturally farmed) and 33% (steel panel-framed). E. coli disease was only found in the steel panel-framed housing. Most of the animal welfare-approved eggs were sold at large markets or a real sale markets. Egg price was 200~250 won per egg. These results indicate the current situation of animal welfare-approved farms and could be caused that windowless poultry house was applied to animal welfare approved farms.

The specificity of the N- and C-terminal antigenic determinant($P_{17}$: sequence $Lys^1-{cys-}^6-Asn^{27},\;{Trp^{12}}_2-Cys^{127}-Leu^{129}$) of hen egg-white lysozyme(HL) was studied in more detail. In a Scatchard plot of the binding of $^{14}C$-acetyl HL with guinea pig purified anti-$P_{17}$ antibody experimental values bent sharply aear r=1. This suggests of two antibody populations with different affinities for HL or possible steric hindrance in the binding of a second HL molecule to the second binding site of the antibody molecule. The antigenic activities of various peptides were tested by measuring their inhibition of the binding of $^{14}C-acetyl-P_{17}$ with the antibody, Only $P_{17}$ and $P_{17}t$(sequence $Lys^1-cys^6-Homoser^{12},\;Trp^{123}-Cys^{127}-Leu^{128})$) were inhibitory, with $K_1$ values of $2.0{\times}10^4$ and $8.1{\times}10^3$, respectively. These results indicate that the direct binding site of $P_{17}$ to anti-$P_{17}$ antibody may be located in the terminal portion of $P_{17}$ (sequence $Lys^1-Cys^6-Homoser^{12},\;Trp^{123}-Cys^{127}-Leu^{129})$) while the rest of $P_{17}$ may be important in maintaining the conformation of this determinant. The single disulphide bond involved in this determinant is essential for manifestation of immunological activity.
Many of the protective functions of saliva can be attributed to the biological, physical, structural, and rheological characteristics of salivary glycoproteins. Therefore, the development of ideal saliva substitutes requires understanding of the rheological as well as biological properties of human saliva. In the present study, we investigated the changes of salivary enzymatic activities by saliva substitutes and compared viscosity of saliva substitutes with human saliva. Five kinds of saliva substitutes such as Moi-Stir, Stoppers4, MouthKote, Saliva Orthana, and SNU were used. Lysozyme activity was determined by the turbidimetric method. Peroxidase activity was determined with an NbsSCN assay. $\alpha$-Amylase activity was determined using a chromogenic substrate, 2-chloro-p-nitrophenol linked with maltotriose. The pH values of saliva substitutes were measured and their viscosity values were measured with a cone-and-plate digital viscometer at six different shear rates. Various types of saliva substitutes affected the activities of salivary enzymes in different ways. Stoppers4 enhanced the enzymatic activities of hen egg-white lysozyme, bovine lactoperoxidase (bLP), and $\alpha$-amylase. Saliva Orthana and SNU inhibited bLP activity and enhanced $\alpha$-amylase activity. MouthKote inhibited $\alpha$-amylase activity. Moi-Stir inhibited the enzymatic activities of bLP and $\alpha$-amylase. The pH values were very different according to the types of saliva substitutes. Stoppers4, MouthKote, and Saliva Orthana showed lower values of viscosity at low shear rates and higher values of viscosity at high shear rates compared with unstimulated and stimulated whole saliva. Moi-Stir and SNU displayed much higher values of viscosity than those of natural whole saliva. Collectively, our results indicate that each saliva substitute has its own biological and rheological characteristics. Each saliva substitute affects the enzymatic activity of salivary enzyme and finally oral health in different ways.

This study carried out to determine control factors for the improvement of productivity of laying hens suffering heat stress during hot weather. A total of 48,451 ISA Brown layers were housed in a farm located in Gyeongsangnam-do, Republic of Korea. Five thermo-hydrometer loggers were installed inside the house to collect data of dry-bulb temperature and relative humidity. The experiment continued for 81 days when the summer season begins from 19th June to 7th September, 2018. This study analyzed the correlations among layers' production index and daily average, highest, and lowest temperature; daily average, highest, and lowest relative humidity; and daily average, minimum, and maximum THI. The result indicated that feed consumption, hen-day egg production, egg weight, and FCR decreased as the daily average, highest and lowest dry-bulb temperature and THI rise (p<0.01). On the other hand, water intake increased as the daily average, highest and lowest dry-bulb temperature and THI rise (p<0.001). The relative humidity was not considered to have direct correlations to the layers' production index (p>0.05). However, it was noticeable that the mortality did not have significant relations with daily average and highest temperature; THI; or daily average, highest and lowest relative humidity while it was relevant to the daily lowest temperature and THI (p<0.05). In conclusion, to enhance the productivity of laying hens in a hot climate, it is recommended that daily average, highest, and lowest dry-bulb temperature and THI are maintained as low as possible. Especially, the daily lowest temperature is needed to lower to 20℃, which is the lowest critical temperature for layers.
Two experiments were conducted to compare the effects of the natural and synthetic commercial pigments on the laying Performance, Pigmentation and fatty acid contents in e99 yolk of laying hens. The experimental diets were formulated to have isocalories and isonitrogen. In experiment I, the diet does not contain the com in which the xanthophylls are free, and in experiment II, the diet contained 54% of corn in which the level of xanthophylls are 19.34 ppm/kg (calculated levels). In the experiment 1, 480 ISA Brown laying hens were divided into eight groups. Each group has 60 birds fed the 0% of corn with 8 types of pigment levels for 4 weeks. in the experiment 2, 600 ISA Brown laying hens were divided into ten groups. Each group has 60 birds fed the 54% of com diet with 10 types of pigment levels for 4 weeks. Feed intake, e99 Production, egg weight and feed efficiency did not have significant difference in experiments I and II. Albumen height and haugh unit did not have significant difference in both experiments. In order to approach the yolk pigmentation to 12∼13 of Roche color fan, addition level of natural red pigment was 25∼30 ppm. In the case of synthetic red pigment, the level was 15∼20 ppm. In this experimental condition, the pigmenting effect of the synthetic pigment was better than that of the natural pigment. In the experiment 2, the pigmenting effect of mixing pigments were investigated between TM2 mixed with natural red pigment, and TM6 mixed with synthesis red pigment. The pigmenting effect of synthetic red pigment was greater than that of the natural red pigment. However, the pigmenting effect of natural pigment was greater than that of synthetic pigments when the levels of synthetic and natural red pigments in diet are higher in TM3 and TM8. The fatty acid content in yolk was not affected by pigment addition.

This study was conducted to evaluate the feeding value of the spent mushroom (Hypsizygus marmoreus) substrates (SMS) in laying hens (Hy-Line Brown). The fresh spent mushroom (Hypsizygus marmoreus) substrates collected from the Namkyung farm were fermented with Bacillus subtilis EJ3 for 2 weeks. A total of twenty-four laying hens were fed corn-soy based experimental diets containing 0% (T0), 5% (T1), 10% (T2) and 15%(T3) fermented SMS for 12 weeks. There were no significant differences among the treatments in egg production, egg weight, egg mass, feed conversion and viability during the experimental period. Feed intake was significantly lowered in T0 (118.3 g) than T1 (121.9 g), T2 (120.3 g) and T3 (122.4 g). There were no significant differences among the treatments eggshell breaking strength, thickness and haugh unit, whereas the yolk color of T1, T2 and T3 were significantly heavy than T0. The palatability of boiled meat was significantly better in the T3 laying hens than in the T0 laying hens. In conclusion, fermented SMS can be used as resource of feed in laying hen feed at 5.0-15% level without effect on performance and egg qualify.
Total 240 of ISA Brown layers were employed in this experiment to study the effects of single or mixed feeding of Lactobacillus and yeast on the performance and intestinal microflora of laying hens. They were randomly allocated to six dietary treatments; None(Control), Pichia farinosa(PF), Lactobacillus crispatus avihen1 (LCH), Lactobacillus vaginalis avihen1(LVH), LCH+PF, and LVH+PF. Viable microflora were added to meet 3${\times}$10$\^$6/ cfu PF and 10$\^$7/ cfu Lacrobacillus per g of feed. There were four replicates per treatment, and 10 birds per replicates. Laying performance was recorded for 10 weeks, followed by a metabolism trial during which nutrient utilization, pattern of intestinal microflora and fecal NH$\sub$3/ emission were examined. Egg production and daily egg mass of birds fed either single or mixed microorganisms were significantly higher than those of the control(P<0.05). Egg weight and feed intake were not statistically different among all treatments. However, feed conversion ratio tended to improve by the supplementation of microbes. Digestibility of crude protein, ether extract and crude ash tended to improve in Lactobacillus treatments, however, there were not statistically different. With regards to the number of intestinal microbes, number of anaerobes were increased in microbes feeding group. Eggshell quality of PF layers was significantly poorer than those of the other treatments. No consistent trend was found in Haugh Unit among all treatments. Fecal NH$\sub$3/ gas emission was significantly lower in LVH, LVH+PF and LCH+PF than the other treatments(P<0.05). From the result of this experiment, it could be concluded that single or mixed feeding of Lactobacillus and yeast improves the laying performance and decreases the fecal ammonia gas emission. No synergic effect was found when both microbes were mixed and fed to the layers.
