Fluorescent Method for Observing Intravascular Bonghan Duct (형광염색을 이용한 혈관내봉한관의 관찰)

  • Observation of intra-vascular threadlike structures in the blood vessels of rats is reported with the images by differential interference contrast microscope, and fluorescence inverted microscope of the acridine-orange stained samples. The confocal microscope image and the hematoxylin-eosin staining revealed the distinctive pattern of nuclei distribution that clearly discerned the threadlike structure from fibrin, capillary, small venule, arteriole, or lymph vessel. Physiological function of the intra-vascular thread in connection with acupuncture is discussed. Especially, this threadlike duct can be a circulation path for herb-liquid flow, which may provide the scientific mechanism for therapeutic effect of herbal acupuncture.

HISTOCHEMICAL STUDY ON THE CALCIFIED TISSUE INDUCED BY POTASSIUM PERMANGANATE IN THE RABBIT DENTAL PULP (가토 치수에 있어서 KMnO4투여후 형성된 경조직의 성상에 관한 연구)

  • The author observed histochemically the nature of the calcified tissue in the rabbit dental pulp, induced by pulpal injection of potassium permanganate. The pulp of rabbit mandibular incisors were exposed and enlarged by a dental hand reamer. The exposed pulps were injected with 0.05ml of 20mM solution of potassium permanganate dissolved in Ringer's solution in experimental tooth. Also the control tooth received a pulpal injection of 0.05ml of Ringer's solution. After pulpal injection, the tooth was plugged with a gutta-percha root canal point. The staining techniques were hematoxylin-eosin stain, van Gieson stain, PAS reaction, toluidine blue stain, alcian blue-hematoxylin stain and colloidal iron-picric acid stain. The results were as follows: 1. The pulp on experimental tooth showed osteodentin-like calcified tissue. Also, in some areas, false denticle-like substance were observed. 2. The central portion of the calcified matrix showed metachromasia in toluidine blue stain had strong staining capacity in alcian blue stain. 3. The peripheral portion of the calcified tissue revealed marked van Gieson positive reaction for collagen. But their staining ability in alcian blue was slight and metachromasia was not appeared.

  • DTBP (dimethyl 3,3`-dithiobispropionimidate) was applied to collagen and gelatin coating on BCP granules and a crosslinking agent. The DTBP crosslinking was done for decreasing the solubility of the coating and hence increasing the stability. The nanostructure of collagen and gelatin coating surfaces were observed by SEM technique. Based on the DSC thermograms and FT-IR spectrums, the crosslinkings were confirmed between collagen molecules and gelatin molecules. The compressive strength was measured before crosslinking and after that. In-vitro study was carried out by measuring cell viability and observing cell morphology after DTBP crosslinking. Moreover, the proliferation ability of MG-63 osteoblast-like cells on the crosslinked BCP granules was evaluated by Western blot assay. The BCP granules were implanted into rabbit femur for 4 weeks and 12 weeks. The bone tissue formation was analyzed with micro-computed tomography (micro-CT) and histological analysis was also carried out by hematoxylin and eosin (H&E) staining for visualization of cells.

Effect of Yam Yogurt on Colon Mucosal Tissue of Rats with Loperamide-induced Constipation

  • The effects of lactic acid fermented yam yogurt (Yam/YG) on colon mucosal tissue were investigated in a loperamide-induced constipation rat models. Sprague-Dawley rats were fed for 6 weeks with 3 types of diets (normal, supplemented with lactic acid bacteria, and supplemented with Yam/YG), and were then administered loperamide intraperitoneally twice daily for 5 days. Administration of loperamide decreased fecal excretion and the moisture content of feces with increasing of numbers of pellets in the colon. On the histopathologic findings from hematoxylin and eosin (H& E) and alcian blue stainings, supplementation with Yam/YG resulted in the recovery of depleted goblet cells and mucin, and increased the numbers of Ki-67 positive cells, indicating restoration of colonic mucosa through cell proliferation and crypt regeneration against damages observed in crypt epithelial cells of loperamide-induced rats. These results indicate that Yam/YG improves evacuation and mucus production in the gastrointestinal tracts of constipated-induced rats.

  • Leiomyoma is a type of benign smooth muscle neoplasm that is a common neoplasm of the uterus and gastrointestinal tract but rarely affects the head and neck region and is especially unlikely to affect the oral cavity. The diagnosis of leiomyoma is mainly determined by histopathological studies due to variation in its clinical appearance and symptoms. In the present paper we report two rare cases of gingival angioleiomyoma in the posterior maxilla and mandible. After total excision, hematoxylin-eosin and smooth muscle actin staining confirmed the diagnosis of angioleimyoma.

  • Necrotizing sialometaplasia (NS) which mimics malignancy both clinically and histopathologically is an uncommon benign, self-limiting inflammatory disease of the mucus-secreting minor salivary glands. The lesion is believed to be the result of vascular ischemia that may be initiated by trauma. Till date, the diagnosis of NS remains a challenge. This report demonstrates a case of NS in a 73-year-old male patient who presented with an ulcerative lesion in his palate. He had a history of local trauma and was long-term user of salbutamol inhaler. An incisional biopsy was carried out and the diagnosis was established through history, clinical examination, histopathology using Hematoxylin and Eosin stain. The patient was given symptomatic treatment and the lesion healed in about 7 weeks.

  • The author has observed the tissue reaction of the absolute alcohol infection of rat oral mucosa. 0.5ml absolute alcohol was injected subcutaneously on the mucobuccal fold of rat. And the rats were sacrifieced at intervals of one day, 3rd, 1 week, 2 week and 4 week after alcohol injection. The microscopic tissue sections were made and stained with hematoxylin and eosin. The results were are as follows; 1. Degeneration and shrinkage of fibroblasts and coagulative necrosis were observed one day to and three day after alcohol injection. 2. Although coagulative necrosis and tissue degeneration occurred, the inflammatory infiltration was not prominent especially there were scarcely any polymorphonuclear leukocytes in that field. 3. Granulation tissue with moderate small round cell infiltration were replaced the necrotic area at one week after injection and the fibroblast proliferate into the granulation tissue at two week group. 4. User Interface Application for Cancer Classification using Histopathology Images

  • User interface for cancer classification system is a software application with clinician's friendly tools and functions to diagnose cancer from pathology images. Pathology evolved from manual diagnosis to computer-aided diagnosis with the help of Artificial Intelligence tools and algorithms. In this paper, we explained each block of the project life cycle for the implementation of automated breast cancer classification software using AI and machine learning algorithms to classify normal and invasive breast histology images. The system was designed to help the pathologists in an automatic and efficient diagnosis of breast cancer. To design the classification model, Hematoxylin and Eosin (H&E) stained breast histology images were obtained from the ICIAR Breast Cancer challenge. These images are stain normalized to minimize the error that can occur during model training due to pathological stains. The normalized dataset was fed into the ResNet-34 for the classification of normal and invasive breast cancer images. ResNet-34 gave 94% accuracy, 93% F Score, 95% of model Recall, and 91% precision.

Studies on the Fish Kills by Histopathological Characteristics in Gills and Caudal Fins (아가미 혈종과 지느러미 표피탈락 현상을 이용한 어류 폐사원인 연구)

  • Histopathological changes of gills and caudal fins isolated from fishes, Cyprinus carpio, Carassius auratus, and Hernibarbus labeo, which were killed by oxygen deficiency or toxic chemicals, were studied. The toxic chemicals were HCl, NaOH, chloroform, benzene, heavy metals(Cu, Cr, Zn, Pb, Hg), and o-dichlorobenzene. The exposure level was enough to kill the fishes within 30 minutes. Oxygen deficient water was prepared by aeration of nitrogen gas and the oxygen concentration was less than l ppm. Cryocutting was used for the rapid preparation of tissue slides and the tissues were stained by hematoxylin/eosin. In the fishes killed by hazardous chemicals, congestion and/or hyperplasia of secondary lamella and erosion of fin were found as the major histopathological changes. Whereas, these characteristics were not observed in gills or caudal fins of fishes killed by oxygen deficiency. These different bioindications appeared in the fishes killed by toxic substances or natural causes, can be used for the rapid identification of the causes of fish kills.

PULP RFSPONSE TO SELF-CURING RESIN (즉시중합(卽時重合) 레진에 대(對)한 치수반응(齒髓反應))

  • This study was performed to reveal the effect of self-curing acrylic resin to dental pulp when the resin was used directly on the freshly cut dentin. Class V cavity preparations were made on the teeth of dogs with No. 701 carbide bur at a speed of 200,000 rpm and with an air water spray. Petrolatum jelly was coated on the inner surfaces of the cavities. After that, doughs of self-curing acrylic resin were filled in these cavities and before completely set these were removed and cemented with zinc oxide and eugenol cement. The dogs were sacrified between 1 day and 3 weeks at the intervals of one week and routinely processed with hematoxylin and eosin stain for histopathologic interpretation. The results were as followings; 1. The inflammatory changes of the pulp can be seen after the use of self-curing acrylic resin on freshly cut dentin. 2. The inflammatory changes of the pulp were not completely subsided until the end of third experimental week.
