This survey was undertaken to evaluate the status of clonorchiasis in Chang-gok Myun, Hong-Seong Gun adjacent to Yedang reservoir from Sept., 1980 to Oct., 1981. A total of 550 subjects (male 318, and female 232) were examined by cellophane-thick smear technique and formalin-ether method. In order to identify the intensity of the Clonorchis sinensis infection, Stoll's egg dilution counting method was also applied to the C. sinensis positive cases. The following results were obtained in this survey; 1. In the examination of 550 subjects, Clonorchis sinensis eggs were found in 167 cases (30.4%). Male subjects showed 43.4% positive rates and female represented 12.5%. 2. The highest prevalence rate of Clonorchis sinensis was found in 40~49 year group in male (64.2%) and 30~39 year group in female (22.0%), although 14.3%~16.7% positive rates were found in young aged group. 3. Mean E.P.G. of Clonorchis sinensis infected cases were 6,148, showing 7,533 in male and 5,193 in female. 4. The degree of intensity of Clonorchis sinensis infection by E.P.G. count was distributed as 30.4% in light infection (1~999), 45.7% in moderate infection (1,000~29,999) and 5.8% in very heavy infection (over 30,000) in male. Female showed 48.3% in light, 37.9% in moderate, 10.3%. in heavy and 3.5% in very heavy infection. 5. Other helminthic infection rate was 15.6% in Ascaris lumbricoides infection, 15.1% in Trichuris trichiura infection and 4.4% of male subjects showed eggs of Taenia species.
Human schistosomiasis caused by Schistosoma japonicum and Schistosoma mekongi is a chronic and debilitating helminthic disease still prevalent in several countries of Asia. Due to morphological similarities of cercariae and eggs of these 2 species, microscopic differentiation is difficult. High resolution melting (HRM) real-time PCR is developed as an alternative tool for the detection and differentiation of these 2 species. A primer pair was designed for targeting the 18S ribosomal RNA gene to generate PCR products of 156 base pairs for both species. The melting points of S. japonicum and S. mekongi PCR products were $84.5{\pm}0.07^{\circ}C$ and $85.7{\pm}0.07^{\circ}C$, respectively. The method permits amplification from a single cercaria or an egg. The HRM real-time PCR is a rapid and simple tool for differentiation of S. japonicum and S. mekongi in the intermediate and final hosts.

Eosinophilia occurs commonly in many diseases including allergic diseases and helminthic infections. Toxocariasis has been suggested as one cause of eosinophilia. The present study was undertaken to examine the prevalence of toxocariasis in patients with eosinophilia and to identify the risk factors for toxocariasis. This prospective cohort study recruited a total of 81 patients with eosinophilia (34 males and 47 females) who visited the outpatient clinic at Seoul National University Hospital from January 2017 to February 2018 and agreed to participate in this study. The prevalence of toxocariasis was examined by T. canis-specific ELISA, and the various risk factors for toxocariasis were evaluated by a questionnaire survey. Among 81 patients with eosinophilia, 18 were positive for anti-T. canis antibodies (22.2%); 88.9% were male (16/18) and 11.1% were female (2/18). Multivariate statistical analysis revealed that males (OR 21.876, 95% CI: 1.667-287.144) with a history of consuming the raw meat or livers of animals (OR 5.899, 95% CI: 1.004-34.669) and a heavy alcohol-drinking habit (OR 8.767, 95% CI: 1.018-75.497) were at higher risk of toxocariasis in patients with eosinophilia. Toxocariasis should be considered a potential cause of eosinophilia when the patient has a history of eating the raw meat or livers of animals in Korea. A single course of albendazole is recommended to reduce the migration of Toxocara larvae in serologically positive cases with eosinophilia.

A serological survey was performed using ELISA to estimate the prevalence of tissue and luminal helminthic infections among hospital patients in Hanoi region, Vietnam. An overall seroprevalence of tissue and luminal helminthiases was 64.0% (95% CI 61.2-66.8) among 1,120 patients who visited Hanoi Medical University Hospital, Vietnam in 2018. The highest seroprevalence was observed against Toxocara spp. (59.0%), followed by Strongyloides stercoralis (46.3%), Gnathostoma spp. (25.5%), cysticercus (12.8%), Angiostrongylus cantonensis (10.5%), Fasciola spp. (11.1%), and Clonorchis sinensis (8.7%). Mono-infection by one species (11.1%) was lower than multiple infections (53.0%) (P<0.05). The seroprevalence in males (59.3%) was lower than in females (66.2%) but not statistically significant (P>0.05). Children (<15 years) revealed lower seroprevalence (34.0%) than adults (68.4%), and the age group 51-70 years revealed the highest seroprevalence (76.0%). Among the seropositive patients, eosinophilia (≥8.0%) was noted in 80.2%. The present results suggested active transmission of various tissue and luminal helminths among people in Hanoi, Vietnam.

Enterobiasis is common helminthic infections found in man. But control of this disease is still troublesome because of its difficulty in the d diagnosis and prevention of infection. Considering the difficulty of accurate diagnosis of pinworm infection, which is very common and somewhat pathogenic, reevaluation of cellotape anal swab method for the diagnosis of enterobiasis was performed. A total of 147 children ranging the ages of 1-12 years in 3 orphanages in the suburbs of Seoul, Korea was subjected for this study. Repeated cellotape anal swabs were carried out against 70 children, 7 times for 3 days interval, in the morning 6am. Finally 10 mg/kg body weight pyrantel pamoates were given to all children including egg negative cases and whole stools of 3 following successive days were collected for the confirmation of residual worm ourden at the time of treatment. Cellotape anal swabs were also performed to another 77 children at 6am, 3pm, 9pm, twice for 3 days interval. The resultant findings were summarized as follows; 1) While the each time average detection rate of Enterobius egg was 28.8% in this study group, the accumulative detection rate up to 7th examination was 62.8%. The accumulative detection rate rose continuously up to 6th examination. 2) After administration of pyrantel pamoate 10 mg/kg body weight, final infection rate was increased to 72.9% by adding worm positive cases who didn't show any evidence of infection in the cellotape anal swabs. 3) Although pinworms were detected in 35 among 70 children treated, 7 cases (20%) of them were egg negative cases in 7 consecutive cellotape anal swabs. 4) Pinworms were expelleded in 14 (87.5%) among 16 children whose swab result was positive in the last examination which was done concurrently with drug administration. 5) Estimated infection rate calculated by best asymptotically normal estimate of Neuman from Moriya's modification revealed 71.5% similar to 72.9% of present results. 6) The result of anal swab performed at 6am was higher than that of 3pm or 8pm. In conclusion, cellotape anal swab method for Enterobius vermicularis infection was considered as method having relatively high positive accuracy. However to gain the reliable infection rate, at least 6 examination is required in the group with 25-30% infection rate by single swab. Moriya's modification of Neuman could be used efficiently in the mass control of this diease for the estimation of true infection rate of E. vermicularis in the sampled population.

Serodiagnosis of Clonorchis sinensis infections will probably be a first choice tool for screening of clonorchiasis in a future because of increasing difficulties in collection and examination of stools. The sensitive test such as ELISA can he used effectively. However there are some limitations in serological diagnosis for the detection of serum antibody. One of the major problems is the non-specificity of the antigens which produce cross reaction with other helminthic infection sera. To solve this problem. many investigators have tried to purify the antigens used. In this study, we determined the antigenic profile of the crude saline extract antigen of C. sinensis at early developmental stage based on SDS-PAGE and immunoblotting techniques for the purpose of understanding the nature of C. sinensis worm antigen The following results were obtained : 1) The SDS-PAGE showed many protein hands ranging from 10Kd to 91Kd relative molecular weight. Among them, 66, 46, 40, 33, 27, 24, 16, 14 and 10Kd bands were observed as a principle bands. The protein components of C. sinensis changed chronologically during their early developmental period. 44Kd band was stained unclearly in antigen of 2 weeks worm, but changed to concentrated state in antigen of 5 weeks worm. 35Kd band was found in antigen of 2 weeks worm, however this band was disappeared in antigen of 5 weeks worm. 22Kd band also lost its staining property gradually. 2) In spite of differences in antigenic profile, there was no differences in the data obtained by microplate ELISA using each antigen preparation. Absorbance value began to rise in between 2 to 3 weeks after infection. 3) By EITB. serum antibody recognized major protein bands with molecular weight of 91, 85, 63, 46, 40, 33, 24, 14 and 10Kd hand respectively. Among them 66, 33, 17 and 14Kd bands were observed as non-specific band because they reacted even in normal control sera. Generally, gradual increase of positive reactions were observed as the infection period of C. sinensis was prolonged. In other hand, the reaction of 10Kd hand did not occurred when 26th week sera was tested. 4) The positive reactions using antigens of 2 weeks worm, especially on 40 and 24Kd bands, were most strong and sharply demarcated compared to those of 3~5 weeks worm antigen.
Enzyme-linked immunoelectrotransfer blot (EITB) using crude worm antigen of adult Paragonimus westermani was performed for human patients sera to identify the species-specific components. Crude antigen was obtained by homogenizing and centrifuging 24-week old adult worms at 10,000 rpm for 60 minutes in phosphate buffered saline (PBS, PH 7.2) containing: Phenyl methyl sulfonyl auoride (PMSF). Gradient sodium dodecyl sulfate polyacrylamide gel electrophoresis(SDS-PAGE) was performed and blotted electrophoretically onto a sheet of nitrocellulose paper. The sheet was cut into strips and exposed to sera diluted 1 : 200 with PBS. SDS-PAGE showed 26 protein bands ranging 229 to 10 kDa. Of them 229, 91, 60, 50, 35∼31, 27, 25, 21, 17, 11 and 10 kDa components showed positive reaction with serum antibody of patients with p. westermani. Sera of patients infected with Clcnorchis sinensis reacted with 35∼31, 19, and 11 kDa bands. Human sera from cysticercosis and diphyllobothriasis cases showed non-specific cross reactions with 229, 35∼31, 27, 25 and 17kDa bands. Protein bands of 91, 60, 21 and 10kDa showed strong positive reaction without cross reactions with sera from other helminthic infections.

In order to observe the infestation rate and intensity of C. sinensis in Yeoju Eup, Kyung-gi Do, 9,512 stool specimens were collected from the inhabitants through student's helps. The specimens were examined by cellophane thick smear technique and Stoll's egg dilution technique. The epidemiological status was analysed statistically by the regession equation and catalytic curve with the results obtained from this area. The results are as follows: 1) Collection rate oft he stool samples was 66.5%, 9.512 out of 14,300 inhabintants complied with our survey. 2) The infestation rate of C.sinensis was 5.0%, and for male 7.8%, for female 2.3%. 3) Average E.P.G. by Stoll's egg dilution technique in this area was 1,572, and for male 1,853, for female 676. 4) The degree of the intensity of C. sinensis infection by E.P.G. count was distributed as 65.2% in light infection, 32.0 % in moderate infection, 2.8% in heavy infection and none in very heavy infection. 5) The intensity of endemicity in this area was represented with the regression equation calculated with cumulative percentages of E.P.G. counts. Regression equation was y=3.887+1.695 log x and Cs. $D_{50}$ was 4.54. 6) The two-stage catalytic model was applied and the calculation lead to the equation $y=0.267(e^{-0.004t}-e^{-0.019t})$; a=0.004 < b=0.019. 7) Other helminthic infection rate in this area was 1.4% in A.lumbricoides 2.7% in T. trichiura, 1.6% in M. yokogawai md 0.2% in Taenia sp. respectively.

This study was undertaken to evaluate the present status of parasitic infection and swine pen human latrine system in Cheju Do, from July to September 1982. The 663 stool specimens (male 323 and female 340) and 579 scotch tape anal swabs collected from 161 households of 2 areas in Cheju Do were examined. The methods employed were formalin-ether technique for the prevalence rate of various helminthic and protozoan infection, and scotch tape anal swab technique for the prevalence rates of Enterobius vermicularis. In addition to these, questionaire was used to evaluate the present status of swine pen human latrine system and prevalence rates of taeniasis in these areas. The results are as follows ; 1) Prevalence rates of parasitic infections of any kind was 33.9%. It was 35.7% in Cheju City in contrast to 32.1% in North Cheju Gun. The infection rates of Trichuris trichiura was 10.0% and it was the highest prevalence rate in this survey. The prevalence rates of the other parasites were as follows ; Ascaris lumbricoides 2,3%, Hookworm 0.2%, Clonorchis sinensis 0.5%, Hymenolepis nana 1.5%, Entamoeba coli 3.2%, and Giardia lamblia 0.5%. 2) The infection rates of Enterobius vermicularis in 579 peoples (male 285, female 294) by applying scotch tape anal swab technique was 13.1% through the survey. It was 16.9 in Cheju City and 8.5% in North Cheju Gun. 3) The Infection rate of Taenia species by applying the stool examination and making up a question was 19.2%(21.4% in Cheju City and 16.7% in North Cheju Gun). 4) Sexual distribution of the parasitic infections showed slightly higher rate in female than that of male. 5) The positive rates of parasitic infection by the stool examination and questionaire positive cases of taeniasis were higher in 0-9 and over 60 year old than any other age group. 7) The swine pen human latrine systems were used in 46 households (28.6%). 7) Relationship between swine pen human latrine system and taeniasis was not noted.
Cystic echinococcosis (CE) caused by E. granulosus is a serious helminthic zoonosis in humans, livestock and wildlife. Xinjiang is one of high endemic province for CE in China. A total of 55 sheep and cattle livers containing echinococcal cysts were collected from slaughterhouses in Changji and Yining City, northern region of Xinjiang. PCR was employed for cloning 2 gene fragments, 12S rRNA and CO1 for analysis of phylogenetic diversity of E. granulosus. The results showed that all the samples collected were identified as G1 genotype of E. granulosus. Interestingly, YL5 and CJ75 strains were the older branches compared to those strains from France, Argentina, Australia. CO1 gene fragment showed 20 new genotype haploids and 5 new genotype haplogroups (H1-H5) by the analysis of Network 5.0 software, and the YLY17 strain was identified as the most ancestral haplotype. The major haplotypes, such as CJ75 and YL5 strains, showed identical to the isolates from Middle East. The international and domestic trade of livestock might contribute to the dispersal of different haplotypes for E. granulosus evolution.
