수용성 치자 색소의 추출에 관한 연구 (A Study on the Extraction of Soluble Colorants of the Gardenia jasminoides Ellis)

  • The study was performed to obtain the optimum extraction conditions for crocin from gardenia fructus. Generally crocin is unstable on heat, light, acid and base solution. The extraction efficiency of crocin from gardenia depended upon the extraction time, extraction temperature, pH in the extraction bath and the optimum conditions of crocin extraction were determined as 60 minutes of extraction time, 4$0^{\circ}C$ of extraction temperature, pH 7 of extraction bath. The molar extinction coefficient of crocin was 12,515 and the color yield of purified crocin was about six times higher than that of non-purified crocin. The heat-stability at extraction temperature and lightstability in irradiation with xenon lamp for one hour of the purified crocin were higher than those of non-purified crocin. Intensity of &{\lambda}_{max}&of crocin was decreased by irradiation for one hour but UV-Vis. spectra of crocin was not changed. The colors of purified and non-purified crocin dissolved wit methanol was evaluated by means of CIE L* a* b* system.

Monoglyceride의 지방산(脂肪酸) 조성(組成)이 Ice Cream 물성(物性)에 미치는 영향 (Effect of Fatty Acid Compositions by Monoglyceride on Rheological Properties of Ice Cream)

  • Ice cream용 유화제로 사용되는 monoglyceride의 지방산 조성에 따른 ice cream의 물성에 미치는 영향에 대해 검토하였다. 일반적으로 monolaurin부터 monostearin의 포화지방산 monoglyceride에는 각기 성질에 큰 차이가 없었으나 monocaprin및 불포화지방산인 monoolein에서는 ice cream의 점도가 현저히 상승되어 적합한 overrun과 보형성이 양호한 ice cream이 만들어지며 이의 원인은 지방구의 응집 때문으로 확인되었다. 한편 포화지방산 monoglyceride는 overrun이 freezing time의 초기에 발생되기 쉽지만 시간이 경과함에 따라 차츰 저하되는 반면 불포화지방산 monoglyceride는 액상 또는 저융점으로 인해 기름에 대한 융해성이 커짐으로써 overrun이 초기에는 어려우나 freezing time이 경과함에 따라 지방구가 입체적인 망상구조를 이룰뿐 아니라 지방구의 응집을 나타냄으로써 overrun이 개선되며 ice cream에 보형성을 증진시키는 결과를 얻었다. 또한 monostearin과 monoolein의 비율에 있어서는 30:70의 부근에서 지방구의 응집이 극대점을 나타내고 있음을 확인하였다.

Evolution of E. coli Phytase for Increased Thermostability Guided by Rational Parameters

  • Phytases are enzymes that can hydrolyze phytate and its salts into inositol and phosphoric acid, and have been utilized to increase the availability of nutrients in animal feed and mitigate environmental pollution. However, the enzymes' low thermostability has limited their application during the feed palletization process. In this study, a combination of B-value calculation and protein surface engineering was applied to rationally evolve the heat stability of Escherichia coli phytase. After systematic alignment and mining for homologs of the original phytase from the histidine acid phosphatase family, the two models 1DKL and 1DKQ were chosen and used to identify the B-values and spatial distribution of key amino acid residues. Consequently, thirteen potential amino acid mutation sites were obtained and categorized into six domains to construct mutant libraries. After five rounds of iterative mutation screening, the thermophilic phytase mutant P56214 was finally yielded. Compared with the wild-type, the residual enzyme activity of the mutant increased from 20% to 75% after incubation at $90^{\circ}C$ for 5 min. Compared with traditional methods, the rational engineering approach used in this study reduces the screening workload and provides a reference for future applications of phytases as green catalysts.

일방향응고시킨 Pivalic Acid-Ethanol 계에서의 Dendrite의 성장 (Growth of Dendrites in the Unidirectionally Solidified Pivalic Acid-Ethanol System)

  • Transparent organic materials have been frequently used as an analog of the solidifying metallic materials, because their transparency permits an in-situ observation of the microstructural development during solidification through optical microscopy. Pivalic acid (PVA)-ethanol system showing an anisotropic property in solid-liquid interfacial energy and interface kinetics was adopted in the present experiment, and the detailed experiments performed are as follows: (1) variation of dendrite tip temperature with growth velocity, (2) correlation between primary dendrite arm spacing (${\lambda}_1$) and the growth orientation away from the heat flow direction (tilt angle: ${\theta}$), (3) variation of dendrite tip radius (R) with growth velocity (V), (4) dendrite tip stability parameter (${\sigma}^*$) and its dependence on the concentration. Concerning the correlation between the dendrite tip temperature and growth velocity the present result is well suited to Hunt-Lu equation. As the tilt angle increases, the average primary dendrite spacing tends to increase.

  • 배소김의 열처리조건에 따른 성분의 변화와 저장중의 색소, 지방산, 유리아미노산, 색도, 갈변등의 변화를 측정하여 품질의 안정성을 검토한 결과를 요약하면 다음과 같다. 1. 열처리중 biliprotein 색소는 극히 불안정하였고, chlorophyll a 보다는 carotenoid가 비교적 안정하였다. 색소의 안정도는 열처리온도와 시간에 따라 다르나 chlorophyll과 carotenoid는 $60^{\circ}C$, 90분 ; $80^{\circ}C$, 60분 ; $100^{\circ}C$, 10분 ; $150^{\circ}C$, 5분 ; $200^{\circ}C$, 3분 정도의 열처리 하에서는 $90\%$ 이상의 잔존율을 보였다. Hunter color 값은 열처리 정도에 따라 L 및 a 값의 증가, b값의 감소가 뚜렷하였다. 지방산은 위에 적은 정도의 열처리조건에서는 크게 영향을 받지 않고 안정하였다. 2. 배소김의 저장중에는 열처리에 의한 안정화효과, 조미액의 항산화성, 살포한 기름의 보호역활동에 의하여 chlorophyll a, carotene지 등은 안정화 하였고 지방산은 조미배소김의 경우 $\alpha_w0.2$, 50일이상 저장한후에도 극히 안정함을 보였으나 기름살포 배소김의 장기 저장에서는 지질의 열화를 가져왔다. 3. 유리아미노산에 있어서는 열화처리중에 Thr, Gly 등은 $80\%$ 이상의 손실을 보였고 저장중에는 안정하여 기름살포김의 경우 $\alpha_w0.2$, 100일간 저장후 $80\%$ 이상이 잔존하였다. 그 중 Glu의 손실이 가장 심하였다. 기름의 살포는 유리아미노산의 안정화에 유효한 것으로 보였다.

세바식 산으로 중합된 폴리에스터 폴리올을 원료로 사용한 폴리우레탄 접착제에 관한 연구 (A Study on Polyurethane Adhesive Based Polyester Polyol Polymerized from Sebacic Acid)

  • 본 연구에서는 피마자유 유래의 sebacic acid를 isophthalic acid 및 glycol과 병용하여 폴리에스터 폴리올을 중합하고 이어서 폴리우레탄 접착제를 제조하였다. 접착력을 측정한 결과에서 sebacic acid를 병용한 폴리올이 adipic acid를 사용한 폴리올 보다 응집에너지가 커서 접착력이 우수하게 나타났고, glycol은 선형구조 보다 메틸기를 포함하는 가지구조가 더 좋은 접착력을 보였다. 쇄연장제는 1,4-butane diol(BD)를 사용한 것이 가장 접착력이 좋게 나타났다. 또한 TGA 측정결과 내열성은 BD가 가장 높고, 1,2-propylene glycol(PG)가 다음, ethylene glycol(EG)가 가장 낮았다. DMA 측정에서 유리전이온도는 PG, EG, BD의 순서로 높게 나타났다.

Stability of Antioxidant Properties and Essential Amino Acids in Squid Viscera Hydrolysate Produced using Subcritical Water

  • Subcritical water hydrolysis was carried out in a batch reactor to produce valuable materials, such as low-molecular weight (MW) peptides and essential amino acids with antioxidant properties, from heat-dried squid viscera. Hydrolysis of squid viscera was performed at 160 to $280^{\circ}C$ for 3 min. The yield was increased by increasing the temperature and pressure, while the protein content of squid viscera hydrolysate decreased with increasing temperature. Low-MW peptides were detected in all hydrolysates by sodium dodecyl sulfate polyacrylamide gel electrophoresis. The highest yields of free and structural amino acids in heat-dried squid viscera hydrolysate were at $160^{\circ}C$ and were $411.95{\pm}1.15$ and $346.62{\pm}1.25$ mg/100 g, respectively. All essential amino acids were detected in viscera hydrolysates; leucine was the most abundant. Antioxidant activities of hydrolysates were highest at $220^{\circ}C$. Greater than $98{\pm}0.26%$ of the ABTS antioxidant activity was retained in hydrolysates after long-term heat treatment.

대나무섬유/PLA 바이오복합재료의 기계적 특성, 충격강도 및 열변형온도에 미치는 대나무섬유 수처리의 영향 (Water Treatment Effect of Bamboo Fiber on the Mechanical Properties, Impact Strength, and Heat Deflection Temperature of Bamboo Fiber/PLA Biocomposites)

  • 본 연구에서는 셀룰로스계 천연섬유인 대나무와 poly(lactic acid) (PLA)로 구성된 펠렛을 압출공정으로 제조하고, 여러 가지 함량의 대나무섬유/PLA 바이오복합재료를 사출공정을 통해 성형하였다. 바이오복합재료의 굴곡, 인장, 충격 특성 및 열변형온도에 미치는 대나무섬유의 수처리 영향을 조사하였다. 천연섬유의 열안정성, 바이오복합재료의 굴곡특성, 인장탄성률 및 충격특성은 섬유 함량은 물론 수처리 유 무에 의존한 반면, 열변형온도는 주로 수처리에 의해 영향을 받았다. 바이오복합재료의 기계적 특성과 충격특성의 증가는 이를 구성하고 있는 대나무섬유와 PLA 매트릭스 사이의 계면결합력이 대나무섬유의 수처리에 의해 향상되었기 때문이다. 연구결과는 친환경적이고 작업친화적인 물을 이용한 천연섬유의 전처리가 바이오복합재료의 성능을 향상시키는데 기여할 수 있다는 것을 제시하여 준다.

Identification and Antimicrobial Activity Detection of Lactic Acid Bacteria Isolated from Corn Stover Silage

  • A total of 59 lactic acid bacteria (LAB) strains were isolated from corn stover silage. According to phenotypic and chemotaxonomic characteristics, 16S ribosomal DNA (rDNA) sequences and recA gene polymerase chain reaction amplification, these LAB isolates were identified as five species: Lactobacillus (L.) plantarum subsp. plantarum, Pediococcus pentosaceus, Enterococcus mundtii, Weissella cibaria and Leuconostoc pseudomesenteroides, respectively. Those strains were also screened for antimicrobial activity using a dual-culture agar plate assay. Based on excluding the effects of organic acids and hydrogen peroxide, two L. plantarum subsp. plantarum strains ZZU 203 and 204, which strongly inhibited Salmonella enterica ATCC $43971^T$, Micrococcus luteus ATCC $4698^T$ and Escherichia coli ATCC $11775^T$ were selected for further research on sensitivity of the antimicrobial substance to heat, pH and protease. Cell-free culture supernatants of the two strains exhibited strong heat stability (60 min at $100^{\circ}C$), but the antimicrobial activity was eliminated after treatment at $121^{\circ}C$ for 15 min. The antimicrobial substance remained active under acidic condition (pH 2.0 to 6.0), but became inactive under neutral and alkaline condition (pH 7.0 to 9.0). In addition, the antimicrobial activities of these two strains decreased remarkably after digestion by protease K. These results preliminarily suggest that the desirable antimicrobial activity of strains ZZU 203 and 204 is the result of the production of a bacteriocin-like substance, and these two strains with antimicrobial activity could be used as silage additives to inhibit proliferation of unwanted microorganism during ensiling and preserve nutrients of silage. The nature of the antimicrobial substances is being investigated in our laboratory.

Thermal Decomposition Behavior and Durability Evaluation of Thermotropic Liquid Crystalline Polymers

  • The thermal decomposition behavior and degradation characteristics off our different thermotropic liquid crystalline polymers (TLCPs) were studied. The thermal decomposition behavior was determined by means of thermogravimetric analysis (TGA) at different heating rates in nitrogen and air. The order of the thermal stability was as follows: multi-aromatic polyester > hydroxybenzoic acid (HBA)/hydroxynaphthoic acid (HNA) copolyester > HNA/hydroxyl acetaniline (HAA)/terephthalic acid (TA) copolyester > HBA/Poly(ethylene terephthalate) (PET) copolyester. The activation energies of the thermal degradation were calculated by four multiple heating rate methods: Flynn-Wall, Friedman, Kissinger, and Kim-Park. The Flynn-Wall and Kim-Park methods were the most suitable methods to calculate the activation energy. Samples were exposed to an accelerated degradation test (ADT), under fixed conditions of heat ($63{\pm}3^{\circ}C$), humidity ($30{\pm}4%$) and Xenon arc radiation ($1.10\;W/m^2$), and the changes in surface morphology and color difference with time were determined. The TLCPs decomposed, discolored and cracked upon exposure to ultraviolet radiation.
