A DNA Strand-Nicking Principle of a Higher Plant, Caesalpinia sappan

  • To find anticancer agents from higher plants, DNA strand-scission assay method was employed for bioassay-guided fractionation as well as for screening the crude extracts. During the screening, an ethyl acetate extracts of the heartwood of Caesalpinia sappan L. (Leguminosae) exhibited potent DNA strand-scission activity. Therefore, the ethyl acetate extracts of the dried heartwood of C. sappan was subjected to the bioassay-guided fractionation, which led to the isolation of a known compound, brazilin (1) as the active constituent. In addition, caesalpine J (2) was also isolated as an inactive constituent.

Radial Penetration of Safranine in Populus tomentiglandulosa T. Lee

  • An experiment was conducted to observe the safranine penetration depth in radial directions of Populus tomentiglandulosa. Radial penetration was considered from bark to pith. In radial direction, ray parenchyma and intercellular space were considered for the measurement of safranine penetration depth. It was found that sapwood conducted safranine 24.23% higher in radial direction compared with heartwood. Intercellular space conducted safranine 39.27% higher depth compared with ray parenchyma and the penetration depth was 39.41% higher in sapwood compared to heartwood. During safranine penetration, it formed a curvature in the lumen of ray parenchyma. Initially safranine penetration was found high and decreased gradually.

  • Liquid permeabilities of red oak and several softwoods were measured by the pressure bomb method and a modified liquid permeability method in order to investigate their efficacy. The effect of preboiling and prefreezing on wood permeability were also examined for both green and resaturated specimens. Regardless of some disadvantages these two methods were revealed as a handy tool for quick evaluation of the permeability of an unknown species. The permeabilities of the resaturated specimens increased when preboiled. but decreased when prefrozen. For green specimens, however, pre freezing increased permeability. The discrepancy of the pre freezing effect on two specimens partially attributes to their difference of initial permeabilities. For all species except radiata pine heartwood, the radii of the effective capillary pores, derived from the water potential equation, distribute from $0.42{\mu}m$ to $7.2{\mu}m$. Those of radiata pine heartwood are below $0.46{\mu}m$.

  • From the $CHCl_3$ and BuOH soluble fractions of the heartwood of Hemiptelea davidii, eleven compounds have been isolated. On the basis of spectral data, they were identified as ${\beta}-sitosterol$ (1), scopoletin (2), kaempferol (3), 4-hydroxybenzoic acid (4), 2-(4-hydroxyphenyl) ethanol (5), aromadendrin (6), scopolin (7), kaempferol 6-C-glucoside (8), aromadendrin 6-C-glucoside (9), taxifolin 6-C-glucoside (10) and quercetin 6-C-glucoside (11), respectively. Among these compounds, compounds 3, 8, 10, and 11 showed potent DPPH radical scavenging activity with $IC_{50}$ values of 11.9, 14.7, 10.3 and $6.2\;{\mu}g/ml$, respectively.

Free Radical Scavengers from the Heartwood of Juniperus chinensis

  • The antioxidant activity of Juniperus chinensis (Cupressaceae) was determined by measuring the radical scavenging effect on DPPH (1,1-diphenyl-2-picrylhydrazyl). The methanolic extract of J. chinensis heartwood showed the strong antioxidant activity. The antioxidant activity of n-BuOH soluble fraction was stronger than that of the others, and the fraction was subjected to purification by repeated silica gel and Sephadex LH-20 column chromatography. Quercetin, naringenin, taxifolin, aromadendrin and isoquercitrin were isolated from the n-BuOH fraction. Their structures were elucidated by physico-chemical and spectroscopic studies.

Relationship between Anatomical Properties and Modulus of Rupture (MOR) of Larix kaempferi Carr

  • Larix kaempferi is a tree with a major economic impact and is processed in large quantity every year in Korea. This study was carried out to collect basic data for the reasonable use of Larix kaempferi and to investigate the relation between anatomical properties and modulus of rupture (MOR) for heartwood and sapwood. As the length of earlywood tracheid and the radial wall thickness of earlywood tracheid and latewood tracheid increased, the modulus of rupture (MOR) increased, but decreased with increasing microfibril angle. Statistical analysis by the stepwise regression technique shows that the main factors affecting the modulus of rupture (MOR) of heartwood are the microfibril angle and the radial wall thickness of latewood tracheid, while those affecting MOR of sapwood are the length of earlywood tracheid and the microfibril angle.

  • Five flavonoids were isolated from the heartwoods of Maackia fauriei. On the basis of spectroscopic evidences, the structure of these compounds were established as (-)maa ckiain(3-hydroxy-8,9-methylenedioxypterocarpan), ononin(4'-methoxyisoflavone- 7-O-${\beta}$-D-glucoside), wistin(4',6-dimethoxyisoflavone-7-O-${\beta}$-D-glucoside), daidzin(4'-hydroxyisoflavone-7-O-${\beta}$-D-glucoside) and 2'-methoxy-4'- hydroxyisoflavone-6-O-${\beta}$-D-glucoside. In these compounds, 2'-methoxy-4'-hydroxyisoflavone-6-O-${\beta}$-D-glucoside was for the first time isolated from the plant source.

Characteristics and Dyeability of Juniperus Chinensis Extracts (향나무 추출 색소의 특성 및 염색성)

  • The purpose of this study was to investigate the characteristics of the pigment and the dyeability of juniperus chinensis needles, berry, bark and heartwood extracts using distilled water, methanol, normal butanol and ethyl acetate as a solvent. Ultraviolet-visible absorption spectrum bands of the extracts were measured at around 280nm to 320nm in all the solvent extracts. The maximum absorption wavelength was able to determine tannin. All the solvent extracts except for distilled water extracts were able to confirm the presence of chlorophyll. Infrared absorption spectra (FT-IR) of all parts of the extracts showed broad absorption bands of OH due to phenolic-OH, benzene CH peak of phenol chemical structure, ether-based stretching vibration peak and the peak of flavonoid compounds that appeared in all the solvent extracts. The yield of juniper needles and heartwood in distilled water and methanol extraction were effective. Extraction of berry yields in distilled water was also effective. The yield of ethanol extraction from the bark showed better efficiency. As a result of using distilled water for the dye solution, the color of the fabrics dyed with all extracts of the needles and Y series berries generally showed light Y progression with a strong red tinge. By using a mixture of 20% ethanol and 80% distilled water for the dye solution, the color of the fabrics dyed with needles and berry extracts showed Y series dominantly. The color of the fabrics dyed with Bark and heartwood extracts were dominantly R series.

Physical Characteristics of Korean Red Pines According to Provinces (Goseong, Hongcheon and Bonghwa-gun) (한국산 소나무의 지역(고성, 홍천 및 봉화군)에 따른 물리적 특성)

  • Physical characteristics of Korean red pine (Pinus densiflora) were investigated with different cultivation locations in Taebaek Mountains as Goseong-gun, Hongcheon-gun and Bonghwa-gun as experimental sites. Moisture content based on air-dried or green (artificial) wood was no significant differences with different cultivation places. Specific gravities of both sapwood and heartwood of red pine from Bonghwa-gun were higher than those from other two sites (Hongcheon-gun or Goseong-gun). Specific gravity of heartwood of red pine from Goseong-gun was higher than it from Hongcheon-gun, but this trend was opposite in case of sapwood. Higher specific gravity of red pine heartwood from Goseong-gun maybe resulted in higher strength than those of other sites. In shrinkage ratio, there was no significant difference among different cultivation places in radial directions, but red pine from Bonghwa-gun was higher than those of other sites in tangential directions. Hygroscopicity was no significant differences with different cultivation places. These results will be helpful information for efficient use of pinewood and good quality of pinewood production for genetical breeding improved.

Studies on Wood Quality of Pinus koraiensis Sieb. et Zucc. (II) -On the Gross Structural Features of Stem - (잣나무의 재질(材質)에 관(關)한 연구(硏究) (제(第)2보(報)) -수간(樹幹)의 구조적(構造的) 형질(形質)-)

  • A study on the wood quality from planted Pinus koraiensis Sieb. et Zucc. has been undertaken by the University Forest in order to gain results of the gross structural features of stem. For this study some groups of trees were felled and several characteristics such as the degree of stem, slenderness, eccentric growth, heartwood diameter, height of butt sweep and butt-swell were analyzed. The results of the study are as follows. 1) The relation between diameter of log and its height in sample trees are nearly parallel to each other. But their degrees of slenderness in logs cut from intermediate and codominant trees are some what gentle as compared with that of logs from dominant tree. 2) The eccentric growth at transverse section of sample trees is generally decreased with the increase of tree height on all sample trees. 3) On the eccentric growth at transverse section of tree, a rather distinct tendency was noticed that all of sample trees showed the direction of eccentric growth in $R_2$ side. 4) Heartwood diameter appeares to become larger as tree growth decrease under same diameter of logs and log diameter in which heartwood appeares is larger in rapid growth rate trees. 5) The maximum height of butt-sweep generally shows that the larger the breast height diameter of tree is, the smaller its height becomes. On the contrary it was noticed that the larger the breast height diameter of trees, the higher the maximum height of butt-swell became. The direction of butt-sweep and butt-swell were shown in $R_1$ side in all sample trees.
