Chromosome Number in Several Species of the Genus Fusarium (Fusarium 속 균종들의 염색체수)

  • The chromosome of Fusarium species during the vegetatve nuclear divisions in hyphae were observed by use of HCl-Giemsa technique on light microscope. The haploid chromosome number of Fusarium anthophilum 7472 was n=7, n=6 in F. anthophilum 7481 and n=6 in F. oxysporum 7500. The haploid chromosome number was 7 in F. napiforme 6129 and F. napiforme 6144. Those of F. caucasicum F. caucasicum ATCC 18791 and F. aquaeductuum ATCC 15612 were n=5. F. coeruleum ATCC 20088 was n=6, n=8 in F. camptoceras ATCC 16065 and n=7 in F. sambucinum NRRL 13451. From these results and previous papers, it may be concluded that the basic haploid chromosome number of the genus Fusarium is n=4.

Chromosomal Studies on the genus of Rhizopus I (Rhizopus속의 염색체에 관한 연구 1)

  • This experiment was designed to elucidate the life cycle of 7 species (Rh.nigricans, Rh. delemar, Rh.oryzae, Rh.acidus, Rh.tritici, Rh. formosaensis and Rh. japonicus) in genus Rhizopus isolated from Korean soil, so as to seize the appropriate stage for detecting their chromosomal number and nuclear size under the method of thin layer slide culture using modified Rogers(1965a) medium. The results are summarized as the folowings ; 1. The haploid chromosome number are found 16 in Rh. japonicus are 8, respectively. 2. Comparing the 7 species of Rhizopus with each other, it may be concluded that the basic haploid chromosome number of genus Rhizopus distributed in Korean soil are 8 and that Rh. nigricans is double of the basic hapolid chromosome number (n = 16). Besides them, the other two species (Rh. tritici and Rh. formosaensis) are believed aneuploids.

  • This study was conducted to establish the maternal haploid method for the practical breeding of M tabacum using the interspeciflc hybridization between M tabacum and N. aflicana. The frequency of surviving seedling per seed capsule of interspecific hybridization was 4.15. Among them, the frequencies of maternal haploid and hybrid were 1.20 and 2.95, respectively. The chromosome numbers of n=24 for maternal haploid and 2n=47 for hybrid were identified in surviving seedling from interspecific hybridiztion. Except the chromosome number, distinguishable morphological differences of material haploid from hybrid were observed at seedling stage.

Studies on the Culture of Haploid Tobacco Leaf (담배 반수성의 유엽배양에 관한 연구)

  • Young haploid leaf derived from the anthers of tobacco plant was cultuerd and plantlets of various ploidies were obtained. When the leaf was put on the medium supplemented with kinetin as growth regulator, plantlets developed directly from the leaf, and the plants coming out in early stage of culture were all haploid. Plants developing in later stage were mostly haploids with some exception of diploid and aneuploid. Leaves were also cultured on the callus-inducing media supplemented with 2,4-D and kinetiion, and the calluses were sub-cultured for six months. Plants developed from these calluses were mostly aneuploids of various chromosome numbers. In view of the fact that the plants directly developed from the leaf were all haploid, the tissue of the original leaf explant was assumed to be uniform as far as chromosome number was concerned. On the other hand, it seemed that the occurrence of various ploidies in the plants derived from the calluses of same origin was the result of the influence of in vitro culture. Apical meristem tissues and various multicellular bodies were formed in the epidermal and inner mesophyll tissues as well as in the sub-epidermal cells.

  • As a series of systematic classification of paramphistomes, the worms in the rumen and reticulum were collected on 214 Korean cattle slaughtered at Jeonju abattoir from January, 1986 to April, 1987 and were classified by means of morphology. Afterwards, the karyotype of Paramphistomum cervi(Zeder, 1790) was detected by means of modified air.drying method from germ cells of the worms. The results were summarized as follows: 1. In the chromosome number of 254 p. cervi, the haploid cell was n:9 and the diploid 2n=18. The meiotic divisions were observed frequently; 1,924 haploid and 32 diploid cells were reliable. Nine pairs of mitotic chromosomes were homologous in the metaphase stage, and the chromosomes were composed of five medium-sized metacentrics (m) , subtelocentrics(st) or submetacentrics(sm) and four small-siRed subtelocentrics(st) or submetacentrics(sm). Meiotic metaphase was composed of five medium and four small chromosomes in size. 2. As a series of C-banding method, C-band was showed in centromeric region from all of the haploid germ cells. Whereas chromosome No. 3 and 5 included heterochromatin on the tip region, chromosome No. 4 on the distal region and No. 6 proximal region. And chromosomes No. 2 and 8 showed a remarkable C-band distinguished from other chromosomes.

  • by use of HCl-Giemsa technique and light microscope, dividing vegetative nuclei in hyphae of Fusarium species were observed and the results are summerized. The chromosome number of these fungi was ranged 4 to 8. Of the 20 strains, the highest haploid chromosome number is 8 in F. solani S Hongchun K4, F. moniliforme (from banana) and F. raphani (from radish). The lowest is 4 in F. sporotrichioides NRRL 3510 and F. equiseti KFCC 11843 IFO 30198. F. solani 7468 (from Sydney), F. solani 7475 (from Sydney), F. oxysporum(from tomato). F. roseum (from rice), F. sporotrichioides C Jngsun 1, F. equiseti C Kosung 1 and F. avenaceum 46039 are n=7. F. moniliforme (from rice) F. graminearum, F. proliferatum 6787 (from Syndey), F. proliferatum 7459 (from Synder) and F. anguioides ATCC 20351 are n=6. F. moniliforme NRRL 2284, F. poae NRRL 3287 and F. trincinctum NRRL 3299 are n=5. From these results, it may be concluded that the basic haploid chromosome number of the genus Fusarium is 4 and mat have been evolutionary variation of chromosome number through aneuploidy and polyploidy.

Effect of Genotype of Donor Plants on the Success of Anther Culture in Sweet Pepper (Capsicum annuum L.)

  • To study the effect of genotype of donor plants on anther culture, anthers of nine hybrid cultivars (Derby, Special, Bossanova, Minipaprika, Fiesta, Boogie, Phenlene, Kufrah, and Clarity) of sweet pepper (Capsicum annuum L.) were cultured in a petridish containing C medium (Sibi, Dumas De Vaulx medium) supplemented with 0.1 mg/L 2,4-D and 0.1 mg/L kinetin, 3% sucrose and 0.32% Phytagel. The cultures were incubated in the dark at $35^{\circ}C$ for seven days, and then cultured at $25^{\circ}C$ with a photoperiod of 16 hr. daylength for 40 days. Frequency of callus formation and plant regeneration was varied among cultivars. Callus formation was ranged from 6% in Phenlene to 69.8% in Kufrah. The highest percentage of regenerated plantlets was obtained in cv. Phenlene (2.67%) followed by Bossanova (2.41%). Result of ploidy analysis; chromosome number observation and flowcytometry analysis, showed that haploid plants could be developed from all of these hybrid cultivars except cv. Fiesta, where highest percentage of haploid plants were obtained in Minipaprika (40%) followed by cv. Bossanova (36.1%). Haploid plants derived from these hybrid cultivars contained single set of chromosome (12 in numbers), higher stomata density (numbers), and smaller sized stomata as compare to diploid plants. The mean length of stomata was 26.9 ${\mu}m$ in haploid plants and 35.7 ${\mu}m$ in diploids.

  • After the previous paper, this chromosomal studies on the fungi were dealt with 17 species in genus of Rhizopus. The results are sumarized as the followings; The haploid chromosome number of 17 species were confirmed as of 6(Rh. oligosporus), 8(Rh. homothallicus, Rh. liquefaciens, Rh. shanghaiensis, Rh, acetorinus), 12(Rh. microsporus, Rh. pseudochinensis, Rh, rhizopodiformis, Rh, thermosus, and Rh. kazanensis), 14(Rh. stolonifer), and 16(Rh. suinus), respectively. Referring to the above fact and the previous paper, it is strongly presumed that the basic chromosome number of Rhizopus are 4.

  • Meiotic metaphase chromosomes of the testis of two storage insects, tobacco moth (Ephestia elutella H.) and cigarette beetle (Lasioderma serricorne F.) were examined to study their karyotypes. The number of haploid chromosomes of the tobacco moth was 31 and the karyotype was characterized by 29 metacentric and 2 submetacentric chromosomes. The metaphase chromosome length of tobacco moth ranged ca. 1.44-2.11${\mu}{\textrm}{m}$ and the average length was 1, 78${\mu}{\textrm}{m}$. The number of haploid chromosomes of the cigarette beetle at metaphase was 10 and the karyotype was characterized by 8 metacentric and 2 submetacentric chromosomes. The range of metaphase chromosome length of the cigarette beetle was ca. 1.79- 2.39${\mu}{\textrm}{m}$ and the average length 2.09${\mu}{\textrm}{m}$.

  • The life cycle of 17 species of Trichoderma was elucidated to seize the proper stage for observing the nuclear behavior and chromosome count. The most convenient stage for the purpose in their life cycle was the stage just before producing the asexual spore. Of the 17 species in the genus Trichoderma the haploid chromosome numbers were counted 5,6,7 and 10. Six chromosomes were most frequently observed. It is believed that the basic chromosome number is placed between 4 and 10, and that the number might be 6, referring to the related papers. It appears necessary to reclassify the single genus of Trichoderma into at least two or three genera.
