제주도 한우 집단에서 Galactose mutarotase (GALM) 유전자의 삽입/결실 다형성과 도체형질의 연관성 분석 (Associations between Insertion/deletion Polymorphisms in Galactose Mutarotaseand Carcass Traits in the Hanwoo Population of Jeju Island)

  • 이 연구는 제주도 한우집단에서 galactose mutarotase (GALM) 유전자형과 도체형질의 연관성을 시험하였다. GALM 유전자형은 3'-비해독부위(3'-UTR)의 14-bp (5'-GGTCTAATGACCAG-3') 삽입/결실 다형성을 이용하였다. 한우 비육우 집단에서 GALM 유전자의 세 가지 유전자형(LL, LS, SS)이 모두 관찰되었다. 연관성 분석결과는 근내지방의 함량과 밀접한 상관을 보이는 육질등급과 근내지방도의 수준과, 등지방두께의 수준이 유전자형에 따른 유의적인 차이를 나타내었다(p<0.05). 동형접합인 SS 유전자형을 보유한 도체에서 LL 또는 LS 유전자형인 도체에 비해 근내지방 함량 수준은 더 높고, 등지방두께도 더 얇은 수준을 보였다. 반면, 도체중, 등심단면적, 육색, 지방색 등은 GALM 유전자형에 따른 유의적인 차이는 없었다(p>0.05). 3'-UTR에서 14-bp 절편의 결실은 RNA의 2차 구조의 변형과 RNA-결합 단백질, microRNA와의 결합능력에 대한 방해를 통해 유전자 발현에 영향을 줄 수 있는 것으로 예측되었다. GALM 유전자의 3'-UTR 영역에서 14-bp 삽입/결실 다형성에 대한 이번 연구결과는 소에서 근육과 등지방 조직에서 galactose 대사에 의한 지방 축적을 통해 성장형질, 도체형질에 영향을 주는 것으로 판단된다.

식품영양 관련 전공 남녀 대학생의 육우고기에 대한 인식조사 (Perception of Youku (Domestically Produced Holstein Steer) Meat among College Students Majoring in Food and Nutrition Studies)

  • A survey was conducted to determine the perception of youku meat among college students majoring in food and/or nutrition. The survey participants were located nationwide, and the responses from the 2,454 students were analyzed. More male and higher grade students answered that they had heard about youku while only 20.0% had learned about Youku from class. Approximately 37.8% of the subjects recognized youku as 'dairy cattle which are too old to produce milk', 54.0% as 'all cattle grown for the purpose of meat', and 23.1% as 'all cattle except for Hanwoo'. Only 37.4% recognized youku correctly. Compared with the same quality grade, 25.3% recognized youku meat as being cheaper than imported beef, and only 25.6% of them recognized that youku meat has less fat than imported beef. As much as 83.3% of subjects did not know whether or not they were served youku meat, and 23.7% of subjects wanted increased availability of youku meat. As much as 22.9% of subjects opposed the increased use of youku meat, and the reasons were "it does not taste good" (18.1%), "it is not Hanwoo" (15.1%), "it is not sanitary" (13.1%), and "it is imported" (6.0%). The findings provide basic information on barriers regarding youku meat promotion among subjects who will be dieticians in food service or managers in purchase departments of catering companies in the future.

Proteome analysis of the m. longissimus dorsi between fattening stages in Hanwoo steer

  • The objective of this study was to identify proteins in the m. longissimus dorsi between early (12 months of age) and late (27 months of age) fattening stages of Hanwoo (Korean cattle) steers. Using two-dimensional electrophoresis and mass spectrometry, 8 proteins of 11 differentially expressed spots between the 12 and 27 month age groups were identified in the loin muscle. Among those that were differentially expressed, zinc finger 323 and myosin light chain were highly expressed in late-fattening stage, and two catabolic enzymes, triosephosphate isomerase (TPI) and succinate dehydrogenase (SDH) were expressed more in the early versus the late-fattening stage. In particular, the quantification of TPI and SDH by immunoblotting correlated well with fat content. Our data suggested that TPI and SDH are potential candidates as markers and their identification provides new insight into the molecular mechanisms and pathways associated with intramuscular fat contents of bovine skeletal muscle.

새송이버섯 수확후배지의 사료적 가치 (Feeding Value of Spent Mushroom (Pleurotus eryngii) Substrate)

  • 버섯수확후배지는 버섯을 수확한 후 남겨진 배지를 말하며 버섯배지의 주원료는 가축 사료원료인 콘코브, 미강, 밀기울, 면실피, 비트펄프 등이고 버섯재배과정에서 배지영양원의 약 15-25% 정도만 버섯에 의해 이용되고 나머지 75-85% 정도는 버섯수확후배지에 남아있기 때문에 버섯수확후 배지는 사료자원으로써 활용가치가 매우 높은 농산부산물이다. 버섯수확후배지 중 사료자원으로 이용할 수 있는 것은 새송이, 팽이, 느타리버섯 수확후배지이며 새송이버섯 수확후배지는 난분해성 물질인 톱밥이 함유되어 있기 때문에 섬유소 함량은 높고 가소화양분 함량은 낮다는 단점이 있지만 cellulase와 xylanase 활성이 높은 미생물을 가축사료 첨가용 생균제로 이용하여 버섯수확후배지 발효사료를 제조하면 버섯수확후배지의 섬유소 함량은 낮아질 수 있다. 그러나 새송이버섯 수확후배지는 가소화양분 함량이 낮기 때문에 새송이버섯 수확후배지 발효사료를 단독으로 급여하기보다는 버섯수확후배지 발효사료를 일정량 첨가한 배합사료를 제조하여 급여하는 것이 영양학적인 측면에서 바람직하며 발효과정에 의해 기호성과 저장성이 개선된 새송이버섯 수확후배지 발효사료의 급여는 축우의 사료비 절감과 고급육 생산 효과를 기대할 수 있다.

Quality grading of Hanwoo (Korean native cattle breed) sub-images using convolutional neural network

  • The aim of this study was to develop a marbling classification and prediction model using small parts of sirloin images based on a deep learning algorithm, namely, a convolutional neural network (CNN). Samples were purchased from a commercial slaughterhouse in Korea, images for each grade were acquired, and the total images (n = 500) were assigned according to their grade number: 1++, 1+, 1, and both 2 & 3. The image acquisition system consists of a DSLR camera with a polarization filter to remove diffusive reflectance and two light sources (55 W). To correct the distorted original images, a radial correction algorithm was implemented. Color images of sirloins of Hanwoo (mixed with feeder cattle, steer, and calf) were divided and sub-images with image sizes of 161 × 161 were made to train the marbling prediction model. In this study, the convolutional neural network (CNN) has four convolution layers and yields prediction results in accordance with marbling grades (1++, 1+, 1, and 2&3). Every single layer uses a rectified linear unit (ReLU) function as an activation function and max-pooling is used for extracting the edge between fat and muscle and reducing the variance of the data. Prediction accuracy was measured using an accuracy and kappa coefficient from a confusion matrix. We summed the prediction of sub-images and determined the total average prediction accuracy. Training accuracy was 100% and the test accuracy was 86%, indicating comparably good performance using the CNN. This study provides classification potential for predicting the marbling grade using color images and a convolutional neural network algorithm.

The difference of castration timing of Korean Hanwoo bulls does not significantly affect the carcass characteristics

  • It is already well known that castration improves marbling quality but exact timing of castration is still highly debated in beef cattle production industry. After castration, blood hormonal changes occur in steer and objective of this study was to investigate the effects of growth hormone (GH) levels on adipocyte differentiation in stromal vascular cells (SVCs) and transdifferentiation into adipocytes in C2C12 myoblasts. Total GH concentrations were measured via enzyme-linked immunosorbent assay (ELISA) in 24 male calves and 4 female calves. Cell proliferation, cellular triglyceride (TG) accumulation, and the cell's lipolytic capability were measured in C2C12 myoblasts and SVCs. Myogenic, adipogenic, and brown adipocyte-specific gene expression was measured via real-time polymerase chain reaction (PCR) using SYBR green. Serum GH levels were the highest in late-castrated calves. Treatment with 5 ng/mL GH resulted in greater TG accumulation as well as increased CCAAT-enhancer-binding protein (C/EBP)α and peroxisome proliferator-activated receptor (PPAR)γ expression compared to that after treatment with 15 ng/mL GH. Treatment with 5 ng/mL GH also resulted in lower myogenin (myo)G and myoD expression compared to that after treatment with 15 ng/mL GH. The expression of bone morphogenetic protein (BMP) 7 after treatment with 5 ng/mL GH was higher than that after treatment with 15 ng/mL GH. But carcass characteristics data showed no significant difference between early and late castrated steers. Therefore, our results indicate that castration timing does not seem to be inevitable determinate of carcass qualities, particularly carcass weight and marbling score in Hanwoo beef cattle.

천연물질을 이용한 반추동물 항생제 대체용 미량광물질 및 천연유화제의 이용 (Supplmentation of the Mineral and Surfactant for Replacement of Antibiotics for Ruminant Aninmal)

  • The present study investigated the effect of inclusion of chromium propionate (Cr-P) and lysophopholipid (LPL) in diet on blood parameters and meat quality of Hanwoo steer. Feeding trial was performed from late fattening period to slaughter and blood parameters (insulin, blood glucose and non-esterified fatty acid (NEFA) concentration) and meat quality were examined. Total 4 experimental groups including control (no addition), T1 (Cr-P 0.2%), T2 (LPL 0.2%) and T3 (Cr-P 0.1% + LPL 0.1%) were employed. For blood parameters, insulin concentration in T1 and T3 showed an elevating patterns from $3.13{\mu}U/mL$ to $3.35{\mu}U/mL$ (T1) and from $4.38{\mu}U/mL$ to $5.23{\mu}U/mL$ (T3). The changes of NEFA in all groups were detected as a decreasing patterns according to days of feeding. However, significant difference was not found. In growth performance, T2 showed greater daily gain and T1 showed greater carcass yield compared to others. However, there were no significance in difference. In meat quality, T1 showed greater yield and intra-muscular fat levels and lower sharing force compared to others. However, significant differences were not detected.

거세한우의 혈청 및 간장내 비타민 A 농도와 도체형질간의 상관관계 (Correlated Relationships Between Vitamin A Concentration in Serum and Liver and Carcass Characteristics of Hanwoo Steers)

  • 본 연구는 사료 중 비타민 A 농도 조절을 통한 고급육 생산 기술 개발을 위한 기초실험의 일환으로, 비타민 A가 거세 한우의 육질에 미치는 영향을 구명하고자 실시되었다. 본 연구에서는 (사)한국종축개량협회에서 주관하는 제 4회 전국한우능력평가대회(1999)에 출품된 거세한우 210두와 일반농가 거세한우 118두에 대하여 혈청 및 간장내 비타민 A 농도를 분석하고, 비타민 A 농도와 육질 및 육량 판정 항목들간 상관관계를 조사하였다. 비육전기 거세한우의 혈중 비타민 A 농도는 평균 212.0$\pm$32.7 IU/$d\ell$ 이었으며, 비육말기 거세한우의 혈중 비타민 A 농도는 117.56$\pm$43.15 IU/$d\ell$를 나타내었으며, 비육말기 거세한우의 간장내 비타민 A 농도는 평균 143.62$\pm$110 IU/g로서 개체별, 지역별, 농가별로 큰 변이를 나타내었다. 또한, 거세한우의 혈청내 비타민 A 농도는 근내지방도(r=-0.24, P<0.01), 등심내 조지방 함량(r=-0.21, P<0.01) 및 육질등급(r=-0.20, P<0.01)과 유의한 부(負)의 상관을 나타내었으며, 간장내 비타민 A plamitate는 등심내 조지방 함량(r=-0.18, P<0.01) 및 육질등급(r=-0.16, P<0.05)과 유의한 부(負)의 상관을 나타내었다. 이상의 결과를 종합해 보면, 거세한우의 혈청 및 간장내 비타민 A 농도와 근내지방도 및 육질등급간에는 유의한 부(負)의 상관이 있는 것으로 나타났다. 이러한 결과들은 한우 고급육 생산시 사료내 비타민 A 함량을 조절함으로서 근내지방도를 향상시킬 수 있다는 가능성을 시사한다. 그러나 한우 고급육 생산을 위한 거세한우의 사료내 비타민 A의 함량과 급여량 및 급여시기 등에 대한 보다 구체적인 연구가 계속해서 수행되어야 할 것으로 생각된다.

한우 후대검정우 체중, 체척 및 도체형질간 상관분석 (Correlation Analyses on Growth Traits, Body Size Traits and Carcass Traits in Hanwoo Steers)

  • 본 연구는 한우 성장형질간의 상관분석을 통하여 6, 12, 18, 24개월 체중, 일당증체량, 도체형질과 18개월 체형형질간의 성장단계별 형질간의 상관관계를 파악하고자 실시하였다. 분석에 이용한 자료는 농협중앙회 한우개량사업소에서 후대 검정을 실시한 2004년에서 2008년까지의 거세우 1,838두의 체중과 체형형질을 이용하였고 도체형질은 24개월령 도축성적을 이용하였다. 본 논문에서 상관분석은 관측치 상관과 고정효과를 고려한 잔차상관으로 하였다. 도축 전 생체중과 도체중은 0.92의 높은 상관관계를 가지고 있었다. 도체형질에서는 육량지수와 등지방두께 간에 -0.92의 높은 부의 상관을 보였다. 체형형질에서 십자부고와 체고는 0.89로서 높은 상관관계가 있었으며 흉폭과 흉심이 0.73의 상관관계를 나타냈다. 체위 형질간 상관에서 요각폭에 대해 흉심과 흉폭이 각각 0.75, 0.74의 상관을 보였고 곤폭은 요각폭과 0.74의 상관을 보였다. 좌골폭에 대해서는 흉심, 흉폭, 요각폭, 곤폭이 각각 0.73, 0.70, 0.75, 0.75로 정의 높은 상관을 나타냈다. 체장과 도체중간의 관측치상관은 0.48이였고, 흉위는 도체중과 잔차상관이 0.51로 다른 도체형질보다 상관정도가 높게 나타났다. 따라서 본 연구결과는 한우 후대검정우 유전평가시 기초 자료로 활용 가능할 것으로 예상된다.

Multiple Regression Analysis to Estimate the Unit Price of Hanwoo (Bos taurus coreanae) Beef

  • This study were estimated the contribution of carcass traits to unit price, to analyze the marbling score as a categorical variable rather than a numerical variable, and to develop an optimal model that also includes the holiday effect and the raising period. The data for this study were acquired from the Quality Evaluation of the Korea Institute for Animal Products, and consisted of the trading records of 1,613,699 heads at 12 wholesale markets from 2010 to 2014. The unit price of a cow was estimated from the following parameters: -52.50 Won/mm, $8.93Won/cm^2$, 7.20 Won/kg, and -1.04 Won/day for backfat thickness, eye muscle area, carcass weight, and raising period, respectively. Parameters for the dummy variables of marbling scores varied from 0 to 8328.74 Won/kg, which means that each marbling score grade had a different price value. The unit price of a steer was estimated from the following parameters: - 92.12 Won/mm, $20.22Won/cm^2$, 1.30 Won/kg, and -1.72 Won/day for backfat thickness, eye muscle area, carcass weights, and raising period, respectively. Parameters for dummy variables of marbling scores varied from 0 to 7338.80 Won/kg, which means that the grades of each marbling score had different price values. The unit price of sales during traditional holidays was significantly higher (827.71 Won/kg for cows, and 645.15 Won/kg for steers) than during non-holidays.We conclude that the use of categorical values for marbling scores would be needed to evaluate the price of Hanwoo beef using multiple regression analysis based on carcass traits and environmental factors.
