1++ 등급 거세한우의 부위별 칼로리, 콜레스테롤, 콜라겐, 유리아미노산, 핵산관련물질 및 지방산조성 (Calorie, Cholesterol, Collagen, Free Amino Acids, Nucleotide-related Compounds and Fatty Acid Composition of Hanwoo Steer Beef with 1++ Quality Grade)

  • 본 연구는 국내에서 최고의 고급육으로 유통되는 한우고기의 부위별 성분조성 및 육질특성을 조사하고자 $1^{++}$ 육질등급을 받은 거세한우 10두에서 10부위(업진, 보섭, 채끝, 등심, 꾸리, 홍두깨, 목심, 설깃, 양지, 우둔)를 선정하여 성분조성 및 육질특성을 분석하였다. 10부위간에 칼로리 함량은 $2,205{\sim}3713.20\;cal/g$ 범위였으며 특히 등심, 채끝 및 업진부위가 다른 부위들과 비교하여 유의적으로 높은 것으로 나타났다(p<0.05). 콜레스테롤 함량도 $38.86{\sim}44.69\;mg/100g$ 수준으로 10부위 중에서 목심부위가 유의적으로 가장 높았고 홍두깨와 우둔부위가 다른 부위와 비교하여 유의적으로 가장 낮은 것으로 분석되었다(p<0.05). 가용성 및 불용성 콜라겐 함량에 있어서는 부위별로 유의적인 차이가 없었으나 총 콜라겐 함량은 꾸리, 목심 및 설깃부위가 가장 높았고 등심 및 채끝부위가 가장 낮은 것으로 나타났다(p<0.05). 맛과 관련이 있는 유리아미노산함량을 부위별로 비교한 결과 모든 부위에 glutamate와 alanine 함량이 각각 $275.99{\sim}536.04\;mg/100g$, $130.63{\sim}175.23\;mg/100g$으로 가장 높은 수준이었고 다음으로 arginine($7.00{\sim}10.49\;mg/100g$), phenylalanine($4.70{\sim}13.88\;mg/100g$), leucine ($5.98{\sim}10.26\;mg/100g$) 순으로 나타났다. 부위별 adenine diphosphate(ADP) 함량은 유의적인 차이가 없는 것으로 나타났으나(p>0.05). Adenine mono phosphate(AMP)는 보섭과 목심 부위에서 모두 1.09%로 다른 부위보다 유의적으로 가장 높게 나타났다(p<0.05). Inosine monophosphate(IMP)는 흥두깨 및 우둔부위가 각각 30.04, 33.44%로 다른 부위보다 유의적으로 가장 많았으며(p<0.05), inosine 함량은 보섭, 흥두깨 및 우둔부위가 각각 6.19, 6.25, 6.28%로 가장 많았고 반면에 hypoxanthine은 4.27%인 꾸리부위에서 가장 높게 나타났다(p<0.05). $1^{++}$ 등급 거세한우의 부위별 지방산 조성을 분석한 결과 C16:1n7, C20:3n6, C20:4n6을 제외한 부위별 지방산 조성은 유의적으로 차이가 없는 것으로 분석되었다(p>0.05).

숙성온도와 기간이 한우 거세우 배최장근의 육질 특성에 미치는 영향 (Effect of Ageing Temperature and Time on the Meat Quality of Longissimus Muscle from Hanwoo Steer)

  • 본 연구는 숙성온도와 숙성기간이 한우 거세우 배최장근의 육질 특성에 미치는 영향을 구명하고자 숙성온도를 0, $4^{\circ}C$로 하여 2, 7, 14, 21, 28, 49일 동안 숙성하면서 한우 거세우 배최장근의 pH, 전단력, MFI, 가열 및 육즙감량, 전자현미경 관찰, 관능검사 및 아미노산 조성을 분석하였다. pH는 숙성온도가 $0^{\circ}C$일 때 숙성 21일까지, 숙성온도가 $4^{\circ}C$일 때 숙성 14일까지 증가하다가 감소하였다(p<0.05). 전단력 값은 숙성 14일부터 28일까지 $4^{\circ}C$에서 숙성한 배최장근이 $4^{\circ}C$보다 유의적으로 낮았고, 숙성기간이 경과함에 따라 유의적으로 연도가 향상되었다(p<0.05). MFI의 경우 $4^{\circ}C$에서 숙성한 배최장근 보다 $0^{\circ}C$에서 높은 경향을 보였고, 숙성기간이 경과함에 따라 유의적으로 증가하였다(p<0.05). $0^{\circ}C$에서 숙성한 배최장근의 가열 및 육즙감량이 $4^{\circ}C$보다 적은 경향이었고, 숙성기간이 경과함에 따라 유의적으로 증가하였다(p<0.05). 관능검사에서 연도와 다즙성은 $0^{\circ}C$에서 숙성한 배최장근이 $4^{\circ}C$ 보다 좋은 평가를 보였고, 숙성기간이 경과하면서 유의적으로 높은 평가를 얻었다(p<0.05). 전자현미경에 의한 미세구조 변화는 $4^{\circ}C$에서 숙성한 배최장근이 $0^{\circ}C$보다 1주일 빠르게 Z-disk의 붕괴가 확실하게 관찰되었고, 숙성기간이 경과하면서 Z-disk의 붕괴정도가 많아졌다. 아미노산 함량 변화는 $4^{\circ}C$에서 숙성한 배최장근이 $0^{\circ}C$보다 높은 경향이었고, 숙성기간이 경과하면서 각 아미노산 조성의 함량이 유의적으로 증가하였다(p<0.05). 이상의 결과를 종합하면, 한우 거세우 등심의 최적 숙성 조건은 $0^{\circ}C$에서는 14일간, $4^{\circ}C$에서는 7일간 숙성하는 것이 바람직한 것으로 사료되며, 향후 숙성기술의 산업적 적용을 위하여 한우의 부위 및 근육별 적정 숙성기간 설정 연구가 필요할 것으로 생각된다.

등지방 두께가 한우 암소와 거세우의 도체 등급 요인 및 도체가격에 미치는 영향 (Effect of Backfat Thickness on the Carcass Grade Factors and Carcass Price in Hanwoo Cows and Steers)

  • 본 연구는 2009년도에 전국에서 출하된 한우 암소 및 거세우 69,159두를 대상으로 등지방두께가 도체등급요인 및 경락가격에 미치는 영향을 구명하였다. 평균 등지방두께는 거세우(12.57 mm)가 암소(11.93 mm) 보다 높았으며, 암소의 77.52%가 등지방두께 4-15 mm 구간에, 거세우의 85.91 %가 등지방두께 4-18 mm 구간에 속하였다. 암소 및 거세우의 도체중(평균, 315.5 kg과 413.2 kg)과 배최장근면적(평균, 76.73 $cm^2$와 86.23 $cm^2$)은 등지방두께가 높아짐에 따라 증가 하였으며(p<0.05), 육량등급은 C등급으로 낮아졌다(p<0.05). 평균 근내지방도는 거세우(5.29)가 암소(3.83)보다 우수하였으며 등지방두께 21 mm까지 증가하였고(p<0.05), 22 mm 이상부터 감소하였다. 암소 및 거세우의 평균 성숙도는 각각 6.11과 2.12로 암소의 골화 정도가 높았으며 평균 육색은 4.85 및 4.67 로써 비슷한 수준이었다. 암소의 평균 육질등급은 1등급과 2등급의 중간 수준(평균 3.55)이었고 거세우는 1등급과 $1^+$등급의 중간 수준(평균 2.58)으로 등지방두께가 21 mm까지 육질등급은 향상되었으나(p<0.05), 22 mm 이상부터 암소는 2등급 수준으로 거세우는 1등급 수준으로 낮아졌다. 암소도체 kg당 경락가격은 등지방두께 13-15 mm 구간에서 14,970원으로, 거세우는 등지방두께 10-12 mm 구간에서 17,318원으로 가장 높았다(p<0.05). 도체 kg당 경락가격은 암소는 등지방두께 18 mm이상, 거세우는 15 mm이상에서 유의적으로(p<0.05) 감소되었다. 특히, 거세우의 경우 도체 경락가격은 등지방 두께 15 mm 이상에서 유의적인 차이가 없었다. $1^{++}$등급 출현율은 암소는 등지방두께 19-21 mm 구간에서 7.33%, 거세우는 22-24 mm구간에서 25.04%로 가장 높았다. 육량 A등급은 등지방두께 4-6 mm구간에서 암소 및 거세우 각각 55.67%와 93.33%로 가장 높았다. 암소 및 거세우의 등지방 두께는 육량지수와 각각 $-0.97^{**}$$-0.96^{**}$로써 상관관계가 높았으며 근내지방도와 각각 $0.20^{**}$$0.14^{**}$ 상관관계가 있었고 육질 등급($1^{++}$등급 = 1, 3등급 = 5)과의 상관관계는 $-0.15^{**}$로 같았다. 본 연구에서 한우도체의 등지방두께는 도체등급 요인 및 도체가격에 영향을 미치는 것으로 분석되어 금후 한우도체의 등지방 감소를 위한 다양한 연구가 필요하다고 사료된다.

Effects of Italian ryegrass diet on the performance, carcass characteristics, and blood metabolites of Hanwoo steers

  • This study was conducted to develop a pasture feeding program for reducing the Hanwoo feeding cost. Italian ryegrass (IRG) is a major plant which is abundantly produced in the southern area of South Korea. Locally produced Italian ryegrass has a 30 - 50% reduced pasture cost compared to imported pasture grasses. We hypothesized that Italian ryegrass feed could reduce the feeding cost but not change the physiological characteristics of the Hanwoo steers in the feedlot. Four treatments (Control, short fed IRG silage, long fed IRG silage, and IRG hay) in a completely random design were used to feed 27 Hanwoo steers. Three or four steers were fed in the same pen, and 8 pens were used for the treatments. Blood was drawn from each steer on the first week of every month from 11 to 30 months. Overall, the average daily gain and feed efficiency were not different among the treatments (p < 0.05) except for the final fattening diets (p < 0.05). The long fed IRG silage group had a low ADG compared to the other groups (p < 0.05). The serum non-esterified fatty acid and glucose concentrations were reduced during the fattening periods (p < 0.05); however, the serum albumin, total proteins, triglycerides, and phosphorus were increased during the fattening periods (p < 0.05). The carcass weight, longissimus muscle area and back-fat thickness were low in the long fed IRG silage group (p < 0.05). The IRG diet induced saturated fatty acids in the longissimus dorsi (p < 0.05). These results show that the IRG diet increased the serum triglycerides but reduced the glucose level at the final fattening diet and decreased the meat yield score in Hanwoo steers.

Gender discrimination and multivariate analysis using deboning data

  • Recent favor on high quality food and concern on food safety have demonstrated the superiority of Hanwoo (Korean native cattle). In general, the price of cow is higher than those of steer and bull, causing cheating issues in the market. Hence, this study is to discriminate genders of Hanwoo with identification of factors which highly influence gender discrimination based on the big-size deboning data. Totally, there were 31 variables in the deboning data, and we divided into them two categories: data obtained before and after deboning. Discriminant function analysis was then applied into the data to determined the accuracy of gender discrimination in Hanwoo. The result showed that Hanwoo could be classified by gender with 99.2% of accuracy when using all 31 variables. In detail, it was possible to identify 93 of 94 bulls (98.9%), 96 of 96 cows (100%) and 74 of 75 steers (98.7%). The most significant variables was chuck, sirloin, armbone shin, plates, retail and cuts percentage, sequentially. With variables obtainable before deboning, accuracies of classification were 91.5% for bulls, 92.7% for cows, and 89.3% for steers. The most significant variables was water, cold carcass weight and back-fat thickness. The discrimination accuracy was higher with data obtainable after deboning: bulls (98.9%), cows (99.0%) and steers (98.7%). In this case, chuck, sirloin and armbone shin were the factors determined the classification ability. This study showed that Hanwoo can be classified based on deboning data with appropriate statistics, further suggesting weight of cut of beef might be the standard for gender classification.

A Whole Genome Association Study on Meat Palatability in Hanwoo

  • A whole genome association (WGA) study was carried out to find quantitative trait loci (QTL) for sensory evaluation traits in Hanwoo. Carcass samples of 250 Hanwoo steers were collected from National Agricultural Cooperative Livestock Research Institute, Ansung, Gyeonggi province, Korea, between 2011 and 2012 and genotyped with the Affymetrix Bovine Axiom Array 640K single nucleotide polymorphism (SNP) chip. Among the SNPs in the chip, a total of 322,160 SNPs were chosen after quality control tests. After adjusting for the effects of age, slaughter-year-season, and polygenic effects using genome relationship matrix, the corrected phenotypes for the sensory evaluation measurements were regressed on each SNP using a simple linear regression additive based model. A total of 1,631 SNPs were detected for color, aroma, tenderness, juiciness and palatability at 0.1% comparison-wise level. Among the significant SNPs, the best set of 52 SNP markers were chosen using a forward regression procedure at 0.05 level, among which the sets of 8, 14, 11, 10, and 9 SNPs were determined for the respectively sensory evaluation traits. The sets of significant SNPs explained 18% to 31% of phenotypic variance. Three SNPs were pleiotropic, i.e. AX-26703353 and AX-26742891 that were located at 101 and 110 Mb of BTA6, respectively, influencing tenderness, juiciness and palatability, while AX-18624743 at 3 Mb of BTA10 affected tenderness and palatability. Our results suggest that some QTL for sensory measures are segregating in a Hanwoo steer population. Additional WGA studies on fatty acid and nutritional components as well as the sensory panels are in process to characterize genetic architecture of meat quality and palatability in Hanwoo.

Effect of Total Digestible Nutrients Level of Concentrates on Growth Performance, Carcass Characteristics, and Meat Composition of Korean Hanwoo Steers

  • This study was conducted to investigate the effect of the total digestible nutrients (TDN) level of commercial concentrates on growth performance, carcass characteristics, and meat composition of late fattening Hanwoo steers. A total of 28 steers were randomly assigned to one of four dietary groups; T1 (73.30% TDN), T2 (74.50% TDN), T3 (76.40% TDN), and T4 (77.10% TDN). Average daily gain (ADG) was slightly but not significantly higher in the T2 than in the other treatments. Dry matter intake (DMI) and feed conversion ratio (FCR) were higher in the T2 than in the other treatments; however, the differences were not statistically significant. Carcass back fat thickness was thicker in the T4 and marbling score was higher in the T2 than in the other treatments; however, the differences were not statistically significant. The TDN level of concentrates had no effect on the physicochemical characteristics and fatty acid composition of the longissimus muscle. The finding of this study indicate that less than 74% or greater than 75% TDN in the commercial concentrate did not contribute to improve ADG, FCR, marbling score; therefore, in the present study, the recommendable TDN level in the commercial concentrate for late fattening period was 74% to 75% in terms of growth performance and marbling score of Hanwoo steer.

Determination of in situ degradation parameters and feeding level of pineapple (Ananas comosus L.) cannery by-product to Hanwoo steers

  • Objective: The objectives of this study were to determine the in situ degradation parameters and appropriate feeding level of pineapple cannery by-products (PCB) based on the growth performance and blood parameters of growing Hanwoo (Bos taurus coreanae) steers fed various levels of PCB. Methods: Two ruminally cannulated Holstein cows were used for in situ disappearance rate measurements. Nylon bags (5×10 cm, 45 ㎛ pore size) filled with 5 g of PCB in triplicate were inserted into the ventral sac of the two cannulated Holsteins cows and incubated for 0, 2, 4, 8, 16, 24, and 48 h. A total of 16 castrated growing Hanwoo steers (12.5±0.5 months old, 302.9±25.7 kg of initial body weight [BW]) were used for the experiment. Animals were stratified by initial BW and then randomly assigned to one of four experimental diets (0%, 1.5%, 3.0%, or 6.0% of PCB, on the dry matter [DM] basis) fed for 91-d, including 30-d of adaptation. Results: Soluble fraction a of DM and crude protein (CP) was 61.9% and 86.0%, fraction b of DM and CP was 32.7% and 11.2%, and indigestible fraction c of DM and CP was 5.4% and 2.8%. The 6.0% PCB feeding group showed lower productivity compared to animals in the other treatments. Increasing the dietary level of PCB did not alter DM intake, but it was numerically lowest in the 6.0% feeding group. The gain to feed ratio was linearly decreased by increasing of PCB. The quadratic broken-line test estimated that 2.5% (DM basis) was the maximum feeding level of PCB in growing Hanwoo steers (y = 0.103 - 0.001×[1.245-x]2, R2 = 0.18). Conclusion: Diets containing up to 2.5% PCB can be fed to growing Hanwoo steers without adverse effects on growth performance.

Diagnosis of Laryngeal Cyst using Respiratory Endoscopy in Hanwoo Cattle with Chronic Bronchopneumonia

  • A seven-month-old Hanwoo steer was presented immediately after transport with respiratory symptoms including a nasal discharge, depression, and anorexia. Though repeated treatments, bronchopneumonia had not been improved and had persisted for 10 months. Then, obstructive breath sound was heard. A cyst adjacent to the epiglottis could be observed with respiratory endoscopy. Consequently, chronic bronchopneumonia induced laryngeal cyst formation, resulting in obstructive dyspnea. And respiratory endoscopy may be useful for differentiating the causes of dyspnea in bovine clinical practice.

Change of performance, serum metabolite, and carcass characteristics on high energy diet of Hanwoo steers

  • The aim of this experiment was to determine the effect of a high-energy diet on the level of serum metabolites and on carcass characteristics of Hanwoo steers. High energy diets have been used for enhancing intramuscular adipose tissue in high quality beef cattle. However, there is not much information about the physiological reactions to this diet. We hypothesized that a high energy diet would increase blood metabolites and the meat quality of Hanwoo steers during the early and final fattening periods. A $2{\times}2$ factorial arrangement (High, Control, and Early, Final) in a completely randomized design was used to feed 24 Hanwoo steers. Two steers were kept in the same pen and 12 pens were used for the experiment. Blood was drawn from each steer on the first week of every other month from 11 to 28 months. Overall Average Daily Gain (ADG) and feed efficiency were not different between high energy and control diets (p > 0.05). However, Dry Matter Intake (DMI greater with the control diet than DMI with the high energy diet during the final fattening period (p < 0.05). Serum non-esterified fatty acid (NEFA) concentrations were greater in the control diet group than in the high energy diet group during the final fattening period (p < 0.05). However, serum albumin, glucose, total protein, triglyceride, and phosphorus were greater in the high energy group than those of the control group (p < 0.05). Carcass traits or physico-chemical characteristics were not different between high energy diet treatment and the control. These data indicated that a high energy diet (+ 3% TDN) increased serum triglyceride during early fattening periods and decreased non-esterified fatty acids during final fattening periods in Hanwoo steers.
