Morphological Changes in the Skin of Hairless Mouse Fed Various Kimchi Diet (김치종류별 식이가 Hairless Mouse 피부의 형태학적 변화에 미치는 영향)

  • Skin is the most frequently exposed tissues to oxidative stress from exogenous and endogenous sources. Dietary antioxidants, which suppress oxidative stress including reactive oxygen metabolites, play an important role in protecting skin from deleterious reactive oxygen species. Kimchi containing lots of antioxidative compounds shows anti-aging effect on skin. Therefore the morphologic changes on the skin of hairless mice fed diets containing Korean cabbage, mustard leaf, and buchu kimchi for 16 weeks were determined. Although epidermal thickness was decreased with age, kimchi prevented this thinning of epidermis compared to control group. In kimchi groups, the staining area of cytokeratin was smaller and stratum corneum was thinner than control group. It suggests that various kinds of kimchi diets prevent the increase of keratinization in epidermis with aging. Type Ⅳ collagen, a major structural protein of basement membrane supporting matrices, existed greater amount in kimchi groups than control group, especially in mustard leaf kimchi group. Rough endoplasmic reticulum (RER) of fibroblast was well developed in dermis of Korean cabbage and mustard leaf kimchi groups, which means collagen synthesis at dermis increased in those kimchi groups. This morphological changes of skin suggest that kimchi consumption can retard skin aging due to the presence of antioxidant and anti-aging compounds, especially some components of mustard leaf kimchi may largely affect on the skin rejuvenescence.

UVB-INDUCED CHANGES OF BARRIERFUNCTION AND MORPHOLOGY OF THE HAIRLESS MOUSE SKIN (자외선-B에의한 Hairless mouse의 보호기능과 표면구조의 변화 영향)

  • Hairless mice (Skh:HR-1) exposed to single doses (0.5, 1.0 and 3. OMED) of UV-B radiation were displayed remarkable changes of barrier function and surface morphology. Trans- epidermal water loss (TEWL) as an index of barrier function was measured by evaporimeter, and wrinkle density (WD) as an index of morphological alteration was measured by image analyzer. Significant changes of TEWL were not observed in the control and 0. SMED group, but 1.OMED and 3. OMED groups noted significant difference. TEWL of 3. OMED group was rapidly increased to the 3rd day and decreased until the 14th day when it reached nearly to normal level, Time-courses of TBWL for 1. OMEB and 3. OMED groups displayed similar pattern, but different only in the magnitude. WD were significantly decreased during the 3rd-5th day in all of the irradiated groups and then increased during the last period to the 14th day, but did not recover the normal level at the 14th day. Time-courses of WD for all groups exhibited similarity, and were entirely dependent on the exposed doses. We also observed histological changes which included hyperplasia, sunburn cell (SBC) formation, accumulation of polymorphonuclear leukocyte (PMNs), and loss of collagen of UVB- exposed hairless mouse skin. Changes of TEWL and WD are helpful in understanding of epidermal and dermal damages by single exposure of UVB.

Effect of Insamyangyoung-tang on the Skin Barrier Function of Hairless Mice

  • Objective : To study the effect of the Insamyangyoung-tang(ISYT) extract on the skin barrier function, the skin pH, skin humidity and transepidermal water loss(TEWL) were measured and histological changes were observed in DNCB(2,4-dinitrochloro-benzen)-induced contact dermatitis(CD) hairless mice. Methods : The male hairless mice were divided into three groups. Each group consisted of 15 mice. The normal group which had acetone- olive oil applied. The control group which had intentionally induced CD by DNCB and it was fed normal saline orally. The ISYT group which had intentionally induced CD by DNCB and it was fed ISYT extract orally for 7 days. The three groups were checked 24h, 48h and 72h later after inducing CD, and the skin pH, skin humidity and TEWL were observed. Tissue samples were taken, and damage to the epithelial cell was observed. Statistical analysis was performed by using one way-ANOVA: significance was set at p values less than 5% (p<0.05). Results : ISYTextract efficiently maintained the pH balance, it kept the skin humidity at a normal level, and it inhibited TEWL of the DNCB-induced CD hairless mouse. The damage to the epithelium was decreased and the regeneration power of the skin was increased in the ISYT group. Conclusion : Insamyangyoung-tang has a good effect on the skin barrier function of DNCB induced contact dermatitis hairless mice.

The Killing Effect of Candida albicans on Hairless Mouse-2 Mouse Tissues by Non-Thermal Atmospheric Pressure Plasma (저온 상압플라즈마에 의한 Hairless Mouse-2 마우스 조직의 Candida albicans 사멸 효과)

  • The purpose of this study was to investigate the killing effect of Candida albicans on hairless mouse-2 (HRM-2) mouse tissues. We tested the effectiveness of a non-thermal atmospheric pressure plasma in killing C. albicans strains. The viability of C. albicans was determined by counting the colony forming units (CFU), after non-thermal atmospheric pressure plasma treatment. When non-thermal atmospheric pressure plasma was repeatedly treated on mouse skin which inoculated with C. albicans. The C. albicans cells were planted on skin tissue, and then the infected mouse tissue was exposed to non-thermal atmospheric pressure plasma for 0 sec, 60 sec, 180 sec and 300 sec. The death rate of C. albicans was increased in dependent with treatment times. The three times of non-thermal atmospheric pressure plasma at the interval of 10 minutes significantly showed the 6 log CFU/ml reduction of death rate on HRM-2 mouse tissues. Thus, non-thermal atmospheric pressure plasma could be used for the disinfection of C. albicans on oral surface.

  • 지구 환경문제의 하나인 오존충의 파괴는 지구상에 분포되어있는 자외선량을 증가시키는 외에 지금까지 지상에 도달하지 않았던 단파장역의 자외선량의 증가를 초래하여, 이것에 피부암, 백내장등의 발병율 증가등의 건강 피해가 염려된다. 이들 발병기전은 아직 확실치 않으나 에너지가 큰 단파영역의 자외선에 폭로되면 세포내의 물분자의 이온화에 기인되어 발생하는 활성산소종이 막지질, 핵산. 단백질등에 산화적 손상을 가져와 이것에 돌연변이, 세포사를 초래하는 것이 그 원인의 하나라고 생각된다. 본 연구는 자외선 중 UVB의 조사로 인한 장해와 유도단백질을 찾아내어, 자외선의 유해성을 밝히는데 목적을 두었다. UVB를 농도별로 1회 hairless mouse에게 조사하여, 경시적으로 피부를 채취하여, UVB 조사로 인해 유도되는 단백질을 2차 전기영동법을 이용해 관찰하고. 유도단백질이 HSP인지를 면역염색을 통해 밝히고, 유도된 단백질을 protein sequence를 하여, 어떤 단백질인지 밝혔다.

Effect of Ethanolamine Salts and Enhancers on the Percutaneous Absorption of Meloxicam from a Pressure Sensitive Adhesive Matrix

  • The purpose of this study was to investigate the effect of salt formation on the percutaneous absorption of meloxicam through hairless mouse skin from a pressure sensitive adhesive (PSA) matrix. In addition, the influences of enhancers on the permeation of meloxicam or meloxicam-ethanolamine (MX-EA) salts across the hairless mouse skin were evaluated using a flow-through diffusion cell system. The salt formation of meloxicam resulted in lower permeation rate than the parent drug. $Span^{(R)}$ 80 provided the highest enhancing effect for meloxicam and meloxicam monoethanolamine salt. The maximum amount of the drug that can be loaded without retarding permeation rate was different depending on the compound. No relationship was found between the fluxes of meloxicam or MX-EA salts from saturated solutions and those from PSA matrices containing the same enhancer.

Penetration Enhancement of β2-Selective Agonist, Tulobuterol, Across Hairless Mouse Skin

  • The effects of various pressure sensitive adhesives (PSA) and enhancers on the percutaneous absorption of tulobuterol were investigated. The permeation rate of tulobuterol through hairless mouse skin from various adhesives was evaluated using a flow-through diffusion cell system at $37^{\circ}C$. The permeability of tulobuterol was variable depending on the physicochemical property of the PSA. The permeation rate of tulobuterol from polyethylene oxide grafted acrylic adhesive matrix was higher than that from other PSA matrices. The flux of tulobuterol was $4.37{\pm}0.34\;{\mu}g/hr/cm^2$ from polyethylene oxide grafted acrylic adhesive matrix. When the effects of various enhancers on the percutaneous absorption of tulobuterol from grafted acrylic adhesive were evaluated, Plurol $oleique^{\circledR}$ showed higher flux than all other enhancers tested.

Determination of Water Content in Skin by using a FT Near Infrared Spectrometer

  • The water content of skin was determined using a FT near infrared (NIR) spectrometer. NIR diffuse reflectance spectra were collected from hairless mouse, in vitro, and from human inner arm, in vivo. It was found that the variation of NIR absorbance band 1450 nm from OH vibration of water and 1940 nm from the combination involving OH stretching and OH deformation, depending on the absolute water content of separated hairless mouse skin, in vitro, using the FT NIR spectrometer. Partial least squares regression (PLSR) was applied to develop a calibration model. The PLS model showed good correlation. For practical use of the evaluation of human skin moisture, the PLS model for human skin moisture was developed in vivo on the basis of the relative water content of stratum corneum from the conventional capacitance method. The PLS model predicted human skin moisture with a standard errors of prediction (SEP) of 3.98 at 1130-1830 nm range. These studies showed the possibility of a rapid and nondestructive skin moisture measurement using FT NIR spectrometer.

  • A new capsaicin analog modified with 4-hydroxyl and alkyl chain of capsaicin was a very potent antiinflammatory analgesic drug and may be clinically useful for those who have rheumatoid arthritis, diabetic neuropathy and cancer. The purpose of this study was to investigate histopathology after short and long term application of poloxamer-based gels, and percutaneous absorption of various topical formulations. Poloxamer-based gel was prepared by cold method using poloxamer 407. The poloxamer gels was applied to dorsal sites of hairless mouse skin during one week or one month for the evaluation of skin irritation. The applied site was then sectioned for histopathologic examination. The topical formulations were also prepared using CMC, HPMC, MC, carbopol and glycerylmono stearate. Skin variation of poloxamer gels was studied using excised hairless mouse, rat, hamster and human penis skin. Franz-type diffusion cells were used far skin penetration of drug against receptor phase filled with about 10$m\ell$ of 0.9% saline solution kept at 32$^{\circ}C$. The concentration of drug was determined by the reverse phased C18, Symmetry HPLC with fluorometeric detector. No skin erythema was observed after dorsal application of poloxamer-based gels for one week or one month. No histopathologic changes was also examined, suggesting no skin toxicity of poloxamer-based gels. The order of flux rate was HPMC > MC ( CMC > poloxamer >> glycerylmono stearate ( carbopol. There was a skin variation of poloxamer gels. The flux rate of poloxamer gels was highest in case of hairless mouse followed by rat, human and hamster skin.
