Global Proteomic Analysis of Mesenchymal Stem Cells Derived from Human Embryonic Stem Cells via Connective Tissue Growth Factor Treatment under Chemically Defined Feeder-Free Culture Conditions

  • Stem cells can be applied usefully in basic research and clinical field due to their differentiation and self-renewal capacity. The aim of this study was to establish an effective novel therapeutic cellular source and create its molecular expression profile map to elucidate the possible therapeutic mechanism and signaling pathway. We successfully obtained a mesenchymal stem cell population from human embryonic stem cells (hESCs) cultured on chemically defined feeder-free conditions and treated with connective tissue growth factor (CTGF) and performed the expressive proteomic approach to elucidate the molecular basis. We further selected 12 differentially expressed proteins in CTGF-induced hESC-derived mesenchymal stem cells (C-hESC-MSCs), which were found to be involved in the metabolic process, immune response, cell signaling, and cell proliferation, as compared to bone marrow derived-MSCs(BM-MSCs). Moreover, these up-regulated proteins were potentially related to the Wnt/β-catenin pathway. These results suggest that C-hESC-MSCs are a highly proliferative cell population, which can interact with the Wnt/β-catenin signaling pathway; thus, due to the upregulated cell survival ability or downregulated apoptosis effects of C-hESC-MSCs, these can be used as an unlimited cellular source in the cell therapy field for a higher therapeutic potential. Overall, the study provided valuable insights into the molecular functioning of hESC derivatives as a valuable cellular source.

Extracts from the Red Algae Gracilaria vermiculophylla have Antioxidant Effects in Human Bone Marrow Mesenchymal Stem Cells

  • The red algae Gracilaria vermiculophylla is widely spread around seaside areas across the globe, and has been used as a food resource in Southeast Asian countries. Previous studies have shown that Gracilaria red algae extracts have beneficial antihypercholesterolemic, antioxidant, anti-inflammatory, and antimicrobial effects. In this study, we investigated the antioxidant effects of Gracilaria vermiculophylla extracts (GV-Ex) on human bone marrow mesenchymal stem cells (hBM-MSCs). The acetone and DMSO/ethanol solvents of the tested GV contain higher total flavonoid and polyphenolic contents that can strongly scavenge reactive oxygen species (ROS). Pre-treatment with GV-Ex protected hBM-MSCs against oxidative stress induced by hydrogen peroxide treatment. The protective effects of GV-Ex treatment were confirmed by MTT assay. The elevated levels of ROS in hBM-MSCs caused by hydrogen peroxide induced oxidative stress were significantly decreased by GV extract treatment. The levels of the antioxidant proteins superoxide dismutase 1 (SOD1), superoxide dismutase 2 (SOD2), and catalase (CAT) were also restored or protected by GV-Ex treatment, suggesting that GV extracts moderate excess ROS levels and prevent cells from oxidative damage.

Protective Effects of Changbudodam-tang on Cell Death Signals on the Bone Marrow-Derived Human Mesenchymal Stem Cells via Regulation of MKK7/JNK/c-Jun Signaling Pathway

  • Objectives: Polycystic ovary syndrome (PCOS) is one of the most common disorders and it shows up to 20% prevalence in reproductive-aged women populations, but no cures are available to date. We aimed to investigate the protective effects of Changbudodam-tang (CBD) on cell death signaling pathways, inflammation, and oxidative stress observed in Bone-Marrow derived human mesenchymal stem cell (BM-hMSC) by means of PCOS therapeutics in the future. Methods: BM-hMSCs were applied with cell deaths and injuries. Apoptosis and pyroptosis signals were quenched with their related signaling pathways using quantitative PCR, Western blot, and fluorescence image analysis. Results: Our data clearly displayed hydrogen peroxide- and nigericin-treated cell death signaling pathways via regulations of mitochondrial integrity and interleukin (IL)-1β at the cellular levels (p < 0.01 or 0.001). We further observed that pre-treatment with CBD showed protective effects against oxidative stress by enhancement of antioxidant components at the cellular level, with respect to both protein and mRNA expression levels (p < 0.05, 0.01 or 0.001). The mechanisms of CBD were examined by Western blot analysis, and it showed anti-cell death, anti-inflammatory, and antioxidant effects via normalizations of the Jun N-terminal kinase/mitogen-activated protein kinase kinase 7/c-Jun signaling pathways. Conclusion: This study confirmed the pharmacological properties of CBD by regulation of cellular oxidation and the inflammation-provoked cell death condition of BM-hMSCs, which is mediated by the MKK7/JNK/c-Jun signaling pathway.

Engraftment of Human Mesenchymal Stem Cells in a Rat Photothrombotic Cerebral Infarction Model : Comparison of Intra-Arterial and Intravenous Infusion Using MRI and Histological Analysis

  • Objective : This study aimed to evaluate the hypotheses that administration routes [intra-arterial (IA) vs. intravenous (IV)] affect the early stage migration of transplanted human bone marrow-derived mesenchymal stem cells (hBM-MSCs) in acute brain infarction. Methods : Male Sprague-Dawley rats (n=40) were subjected to photothrombotic infarction. Three days after photothrombotic infarction, rats were randomly allocated to one of four experimental groups [IA group : n=12, IV group : n=12, superparamagnetic iron oxide (SPIO) group : n=8, control group : n=8]. All groups were subdivided into 1, 6, 24, and 48 hours groups according to time point of sacrifice. Magnetic resonance imaging (MRI) consisting of T2 weighted image (T2WI), $T2^*$ weighted image ($T2^*WI$), susceptibility weighted image (SWI), and diffusion weighted image of rat brain were obtained prior to and at 1, 6, 24, and 48 hours post-implantation. After final MRI, rats were sacrificed and grafted cells were analyzed in brain and lung specimen using Prussian blue and immunohistochemical staining. Results : Grafted cells appeared as dark signal intensity regions at the peri-lesional zone. In IA group, dark signals in peri-lesional zone were more prominent compared with IV group. SWI showed largest dark signal followed by $T2^*WI$ and T2WI in both IA and IV groups. On Prussian blue staining, IA administration showed substantially increased migration and a large number of transplanted hBM-MSCs in the target brain than IV administration. The Prussian blue-positive cells were not detected in SPIO and control groups. Conclusion : In a rat photothrombotic model of ischemic stroke, selective IA administration of human mesenchymal stem cells is more effective than IV administration. MRI and histological analyses revealed the time course of cell migration, and the numbers and distribution of hBM-MSCs delivered into the brain.

인간 중간엽 줄기세포로부터 골아세포로의 분화시 관찰되는 유전자 발현 분석 (Gene Expression Profile Associated with the Differentiation of Osteoblasts from Human Mesenchymal Stem Cells)

  • 인체의 골수내에 존재하는 중간엽 줄기세포는 성장인자나 환경적 요인에 의해 지방세포, 골아세포, 연골모세포 및 연골세포 등으로 분화됨이 알려져 있다. 본 연구에서는 정상 인체의 골수으로 얻어진 중간엽 줄기세포로부터 골아세포의 분화 가능성을 알아보고 이에 관여하는 유전자의 발현을 조사하였다. 정상 골수의 중간엽 줄기세포를 골유도성 자극보조제로서 $\beta$-glycerol phosphate, L-ascorbic acid 및 dexamethasone을 첨가하여 골아세포로의 분화를 유도한 세포와 골유도성 자극보조제를 첨가하지 않은 세포를 배양하여 일정 간격으로 cDNA microarray를 이용하여 각각의 단계에서 발현되는 유전자를 검사하고 이로 인해 얻어진 유전자의 발현량을 분석하기 위해 real time quantitative RT-PCR 을 시행하였다. 골유도성 자극보조제를 첨가한 군에서 첨가하지 않은 군에 비하여 정상적인 골아세포로의 성장이 유도되었고, 분화과정에서 36개의 유전자의 발현이 증가되었고, 59개의 유전자의 발현이 억제되었다. 주로 골 생성 과정과 연관이 있다고 알려진 osteoprotegerin, LRP5 및 metallothionein 2A 등의 유전자들이 분화과정에서 발현 증가되어 나타났고, 줄기세포로부터 분화될 수 있는 조직들 중 근육, 지방, 연골, 혈관 및 신경 조직과 연관된 유전자들은 분화 후기에 감소하거나 혹은 전분화 과정 동안 발현이 억제되었다. 본 연구에서는 골아세포의 분화와 연관된 유전자 발현을 확인함으로써 특정 조건하에서 중간엽 줄기세포로부터 골아세포로의 분화가 가능함을 확인할 수 있었고, 이 과정에 관련된 특정 유전자의 발현 양상을 밝힐 수 있었다.

Diallyl Biphenyl-Type Neolignans Have a Pharmacophore of PPARα/γ Dual Modulators

  • Adiponectin secretion-promoting compounds have therapeutic potentials in human metabolic diseases. Diallyl biphenyl-type neolignan compounds, magnolol, honokiol, and 4-O-methylhonokiol, from a Magnolia officinalis extract were screened as adiponectin-secretion promoting compounds in the adipogenic differentiation model of human bone marrow mesenchymal stem cells (hBM-MSCs). In a target identification study, magnolol, honokiol, and 4-O-methylhonokiol were elucidated as PPARα and PPARγ dual modulators. Diallyl biphenyl-type neolignans affected the transcription of lipid metabolism-associated genes in a different way compared to those of specific PPAR ligands. The diallyl biphenyl-type neolignan structure provides a novel pharmacophore of PPARα/γ dual modulators, which may have unique therapeutic potentials in diverse metabolic diseases.
