Enzymatic Preparation and Antioxidant Activities of Protein Hydrolysates from Tenebrio molitor Larvae (Mealworm) (갈색거저리 유충 단백가수분해물의 제조 및 항산화 활성)

  • The present study was carried out to evaluate the applicability of Tenebrio molitor larvae (mealworm) as a health functional food material in order to contribute to the development of the domestic insect industry and health functional food industry. Protein hydrolysates were prepared from mealworm powder by enzymatic hydrolysis using five different proteases (alcalase, bromelain, flavourzyme, neutrase, and papain), and the hydrolysates were then tested for their antioxidant activities. Based on available amino group contents and sodium dodecyl sulphate-polyacrylamide gel electrophoresis analyses, mealworms treated with alcalase ($4,781.39{\mu}g/mL$), flavourzyme ($5,429.35{\mu}g/mL$), or neutrase ($3,155.55{\mu}g/mL$) for 24 h showed high degree of hydrolysis (HD) value, whereas HD values of bromelain ($1,800{\mu}g/mL$) and papain-treated ($1,782.61{\mu}g/mL$) mealworms were much lower. Protein hydrolysates showing high HD values were further separated into > 3 kDa and ${\leq}3kDa$ fractions by a centrifugal filter system and then lyophilized, and the production yields of the low molecular weight protein hydrolysates (${\leq}3kDa$) by alcalase, flavourzyme, and neutrase were 42.05%, 26.27%, and 30.01%, respectively. According to the RC_{50} values of the protein hydrolysates (${\leq}3kDa$) obtained from three different antioxidant analyses, all three hydrolysates showed similar antioxidant activities. Thus, alcalase hydrolysates showing the highest production yield of low molecular weight protein hydrolysates were further tested for their inhibitory effects on peroxidation of linoleic acid by measuring thiobarbituric acid values, and the results show that peroxidation of untreated linoleic acid increased dramatically during 6 days of incubation. However, pretreatment with the hydrolysates ($100{\sim}800{\mu}g/mL$) significantly inhibited linoleic acid peroxidation in a dose-dependent manner over 6 days.

  • Background : In 1993, American Thoracic Society (ATS) recommended a guideline for the initial management of adults with community-acquired pneumonia(CAP). However, etiologic organisms and medical system in Korea seem to be different from those in Western countries. Retrospective analysis was done to evaluate the efficacy of antibiotics chosen by ATS guideline in the treatment of Korean patients with CAP admitted to a tertiary university medical center. Methods : Hospitalized patients with CAP at Samsung Medical Center from April 1997 through March 1998 were retrospectively reviewed. Patients who fulfilled all of the following criteria were included in this study : (1) fever ${\geq}38^{\circ}C$ (2) purulent sputum (3) pulmonary infiltrates on chest X-ray. Patients were classified as : 1) ATS group ; patients whose initial antibiotics were chosen by ATS guideline 2) Non-ATS overuse group ; additional antibiotics administered more than those of ATS guideline, and 3) Non-ATS underuse group ; initial antibiotics were insufficient to ATS guideline. Response of empirical antibiotics and etiologic organisms of 3 groups were identified. Results : Sixty-four patients were enrolled. Thirty-six patients were classified into ATS group, 10 patients Non-ATS overuse group, and 18 patients Non-ATS underuse group. Thirty-three patients of 36 ATS group, 9 patients of 10 Non-ATS overuse group, and 14 patients of 18 Non-ATS underuse group showed improvement by initial empirical antibiotics. There was no statistical difference in antibiotic response between 3 groups (p>0.05). S. pneumoniae (12.5%), K. pneumoniae (9.4%), and P. aeruginosa (4.7%), Mycoplasma (3.1%) were the most commonly isolated organisms. In 18 patients with severe CAP, P. aeruginosa was isolated only in 1 patient and Legionella organism not isolated. Conclusion : Initial empirical antibiotics chosen by ATS guideline were effective in the management of Korean patients with CAP admitted to a tertiary hospital. However, well-designed large-scale prospective study is needed to identify etiologic organisms and choose an adequate initial empirical antibiotics in Korean adults patient with severe CAP.

Possibility of N-Nitrosamine Formation during Fermentation of Kimchi (김치 숙성중(熟成中) N-Nitrosamine의 생성요인(生成要因)에 관한 연구(硏究))

  • The possibility of formation of carcinogenic N-nitrosamines such as nitrosodimethylamine(NDMA), nitrosodiethylamine (NDEA) and nitrosopyrolidine (NPYR) during the fermentation of Kimchi was investigated. Three different types of Kimchi, formulated with chinese cabbage, red pepper powder and garlic, with or without one of both fermented shrimp and anchovy juice, were cured for 75 days at $5^{\circ}C$. The changes in contents of nitrates, nitrites, pH, ascorbic acid, secondary amines, trimethyl-aminoxide (TMAO), trimethylamine (TMA) and NDMA were analyzed periodically during the fermentation. TMAO, TMA. DMA, nitrate, nitrite and ascorbic acid were analyzed by colorimetric methods, and NDMA, NPYR and NDEA were determined by the method of GLC-TEA. Although the total secondary amines markedly increased, no significant changes in the levels of TMAO and TMA were observed during the fermentation Kimchi added with fermented shrimp or anchovy juice. The predominating component of secondary amines was confirmed to be dimethylamine by means of nitrosating technique coupled with gas chromatography. No appreciable increase in the level of nitrites was appeared although nitrate level in the Kimchi apparently decreased. Non detectable or trace level of nitrosamine formation was detected whereas the nitrates fairly decreased during the fermentation of Kimchi. This could be explained by the fact that the lack of nitrites was resulted in the system due to rapid consumption of nitrites formed from nitrates by the reactions with ascorbic acid and amino acids which have been known as inhibitors of nitrosation reaction.

  • The home health care industry has grown rapidly and can be expected to continue to grow in the foreseeable future. Home health care refers to the practice of nursing applied to clients with a health condition in the clients place of residence. clients and their designated care givers are the focus at home health nursing practice. The goal of care is to initiate. manage and evaluate the resources needed to promote the clients optimal level of well-being and function. Nursing activities necessary to achieve this goal may warrant preventive maintenance and restorative emphases to prevent potential problems from developing. Many project program were suggested home health care model for Korea's health care system and policy direction for expansion and establishment of home health care .But the aim of this paper is to provide on overview for theoretical frame work in home health care. Theories and conceptual frameworks or models are important nursing because they define and guide the boundaries of professional practice and identify key nurse-patient-caregiver relationships that emerge with caring. Following is the research with an investigation of the literature review in the University of Arizona international medline database, In conclusion, are as followers: First, many nursing theorists have had a tremendous impact on nursing practice. the following highlights those nursing theorists that are particularly helpful in understanding home health care. 1. Florence Nightingale : Our earliest theoretical legacy. Nightingale's believes are reflected in basic infection control practice such as hand washing and infectious waste disposal and are key nursing interventions in home care. 2. Martha Roger's :Science of unitary human beings theory. Rorger's believed that the focus of shared. non invasive healing modelities is the human environmental field rather than direct physical care. These modelities continue to evolve as our awareness (reflecting greater diversity, faster rhythms, motions, and ways of knowing) transcends time and space, allowing individuals to get in touch with their integral nature of unbroken wholeness. On people as ever changing energy fields have special relevance in home care especially with hospice and palliative care applications. 3. Madeline Leininger's; Transcultural nursing theory. Home care nurses move through a variety of communities and often care for patients from different cultural back grounds. Therefore Leininger's work has a good that with home care because home care nursing practice is very culturally focused. 4. Dorothea Orem's : Self care deficit theory. Orem's theory views care as something to be performed by both nurses and patients. The role of the nurse is to provide education and support that help patients acquire the necessary activities to perform self-care. Orem's theory is foundational to have care because it begins to truly acknowledge the role of the patient in managing his or her own health. which is referred to as self-care. 5. Margaret Neuman's; Health as expending consciousness theory. Neuman believes that health compasses disease and reflects an underlying pattern of person-environment interaction. A key application of 'Neuman's work to home care is for nurses to understand that health and illness do not necessarily exist at opposite ends of a continuum. 6. Jean Watson's: Theory of human caring. Watson's theory of human caring in nursing proposes human caring as the moral ideal of nursing. Nurses participate human caring to protect, enhance and preserve humanity by assisting individuals to fing meaning in illness. pain and existence and to help others gain self knowledge. self control. and self healing such thinking lends richness to theory development. as well as clinical practice in home care. Second, Robin Rice : Dynamic self determination for self care. (A theoretical framework for home care) Dynamical self determination for self care can be useful to home care nurses in a variety of ways. As research tool it can be reflected in the interview process when the home visit. The home care nurse's role is that of facilitator of patient self-determination for self care through numerous strategies. including patient education and case management.

Effects of Planting Soil on the Soil Moisture and the Growth of Vitex rotundifolia for Green Roof (옥상녹화 식재지반이 토양수분과 순비기나무의 생육에 미치는 영향)

  • This study focuses on the appropriate planting soil for Vitex rotundifolia by planting soil. Different soil depth levels were achieved at 15cm and 25cm in the green roof module system that was created with woody materials for a $500{\times}500{\times}300mm$ area. The soil mixture ratio was $S_{10}$, $L_{10}$, $S_7L_3$, $S_5L_5$, $P_7P_1L_2$, $P_6P_2L_2$, $P_5P_3L_2$ and $P_4P_4L_2$. This study was carried out over five months between April and September, 2006. The amount of soil moisture tends to decrease according to the planting soil. For the experimental items $S_{10}$, $S_7L_3$ and $S_5L_5$, the amount of soil moisture tends to decrease rapidly. However, for the experimental items $P_7P_1L_2$, $P_6P_2L_2$, $P_5P_3L_2$ and $P_4P_4L_2$, conditions containing perlite and peat moss, the amount of moisture tends to decrease more gradually. As a result, the use of soil-improving amending for the afforestation planting of roofs with a low level of management is need. After experimenting with the ratio of soil mixture for Vitex rotundifolia, the planting soil for experimental item $P_6P_2L_2$, $P_5P_3L_2$ and $P_4P_4L_2$ appeared excellent. For experimental item $S_{10}$, the growth of Vitex rotundifolia seemed to be weaker than that of others, because of the low levels of moisture and organic matter in the soil. For experimental item $L_{10}$, there appeared to be a low level of growth, even when the levels of moisture and organic matter were high. This may have occurred because of the low level of soil pH and the excessive amount of exchangeable cation. At the depth of 25cm, the growth of Vitex rotundifolia is vigorous overall. For experimental item at 15cm, Vitex rotundifolia was able to survive for 14 days without any rainfall and Vitex rotundifolia was better in amended soil, $P_6P_2L_2$, $P_5P_3L_2$ and $P_4P_4L_2$, than natural soil, SL.

Evaluation of Image Quality in Micro-CT System Using Constrained Total Variation (TV) Minimization (Micro-CT 시스템에서 제한된 조건의 Total Variation (TV) Minimization을 이용한 영상화질 평가)

  • The reduction of radiation dose from x-ray is a main concern in computed tomography (CT) imaging due to the side-effect of the dose on human body. Recently, the various methods for dose reduction have been studied in CT and one of the method is a iterative reconstruction based on total variation (TV) minimization at few-views data. In this paper, we evaluated the image quality between total variation (TV) minimization algorithm and Feldkam-Davis-kress (FDK) algorithm in micro computed tomography (CT). To evaluate the effect of TV minimization algorithm, we produced a cylindrical phantom including contrast media, water, air inserts. We can acquire maximum 400 projection views per rotation of the x-ray tube and detector. 20, 50, 90, 180 projection data were chosen for evaluating the level of image restoration by TV minimization. The phantom and mouse image reconstructed with FDK algorithm at 400 projection data used as a reference image for comparing with TV minimization and FDK algorithm at few-views. Contrast-to-noise ratio (CNR), Universal quality index (UQI) were used as a image evaluation metric. When projection data are not insufficient, our results show that the image quality of reconstructed with TV minimization is similar to reconstructed image with FDK at 400 view. In the cylindrical phantom study, the CNR of TV image was 5.86, FDK image was 5.65 and FDK-reference was 5.98 at 90-views. The CNR of TV image 0.21 higher than FDK image CNR at 90-views. UQI of TV image was 0.99 and FDK image was 0.81 at 90-views. where, the number of projection is 90, the UQI of TV image 0.18 higher than FDK image at 90-views. In the mouse study UQI of TV image was 0.91, FDK was 0.83 at 90-views. the UQI of TV image 0.08 higher than FDK image at 90-views. In cylindrical phantom image and mouse image study, TV minimization algorithm shows the best performance in artifact reduction and preserving edges at few view data. Therefore, TV minimization can potentially be expected to reduce patient dose in clinics.

Mean Field Analysis of Exchange Coupling in Amorphous RE$Fe_2$-B (RE=Dy, Sm) Alloy Ribbons (비정질 RE$Fe_2$-B (RE=Dy, Sm) 합금 리본에서 평균장 이론에 의한 교환상호작용 계산)

  • Experimental magnetization-temperature curves for melt-spun ribbons of amorphous alloys (Dy$\_$0.33/Fe$\_$0.67/)$\_$1-x/B$\_$x/(x=0 ,0.05, 0.1, and 0.15) and (Sm$\_$0.33/Fe$\_$0.67/)$\_$1-x/B$\_$x/(x=0, 0.01, 0.02, and 0.03) (in atomic fraction) are fitted with theoretical equations based on the mean field theory in order to calculate exchange couplings between constituent elements as a function of the B content. In the case of the DyFe$_2$-B system, the sign of the exchange coupling between Dy and Fe is negative, indicating that the magnetization direction of Dy is antiparallel to that of Fe. The sign of the other two couplings are positive indicating a parallel alignment. The exchange coupling between Fe ions are greatest, while that between Dy ions is negligible. In the case of the SmFe$_2$B alloys, the sign of all the couplings are positive, indicating ferromagnetic coupling between the spins. The exchange couplings between Fe ions, and Fe and Sm are comparable to each other, but they are much greater than that between Sm ions. The high exchange coupling between Fe and Sm, which is considered to occur indirectly, is rather unexpected, but it is considered to be unique characteristics of amorphous Sm-Fe alloys. In both alloy systems, the exchange coupling between Fe ions increases with increasing B content. and this may be explained by the increase of the Fe-Fe separation with increasing B content. The exchange coupling between Fe and RE also increases with increasing B content. As the B content increases, the magnetization decreases over the whole temperature range, and the Curie temperature also decreases.

Expression and Purification of Recombinant Human Interferon-gamma Produced by Escherichia coli (대장균이 생산한 재조합 인체 감마인터페론의 발현과 정제)

  • For the production of the recombinant human interferon-gamma(rhIFN-${\gamma}$) in Escherichia coli, human glucagon and ferritin heavy chain were used as fusion partners. Even though rhIFN-${\gamma}$ is expressed as an inclusion body form in E. coli because of strong hydrophobicity of itself, over 50% of fused rhIFN-${\gamma}$ was expressed as soluble form in E. coli $Origami^{TM}$(DE3) harboring pT7FH(HE)-IFN-${\gamma}$ which encodes ferritin heavy chain-fused rhIFN-${\gamma}$. In the case of using glucagon-ferritin heavy chain hybrid mutant as a fusion partner, 6X His-tag was additionally introduced to N-terminus of GFHM(HE)-IFN-${\gamma}$ for enhancing purification yields of rhIFN-${\gamma}$. Fusion protein HGFHM(HE)-IFN-${\gamma}$ with two 6X His-tag was more effectively bound to Ni-NTA agarose bead than GFHM(HE)-IFN-${\gamma}$ with a 6X His-tag. rhIFN-${\gamma}$ was completely purified from enterokinase-treated HGFHM(HE)-IFN-${\gamma}$ by Ni-NTA affinity column. For high-level production of rhIFN-${\gamma}$, glucose was used as the sole carbon source with simple exponential feeding rate($2.4{\sim}7.2g/h$) in fed-batch process. The effective lactose concentration for the expression of the rhIFN-${\gamma}$ was $10{\sim}20mM$. Under the fed-batch culture conditions, rhIFN-${\gamma}$ production yield reached 11 g DCW/L for 6 hours after lactose induction.

Inhibitory Effects of Ethanolic Extracts from Aster glehni on Xanthine Oxidase and Content Determination of Bioactive Components Using HPLC-UV (섬쑥부쟁이 에탄올 추출물의 잔틴산화효소 저해 효능 및 HPLC-UV를 이용한 유효성분의 함량 분석)

  • This study aimed to establish an optimal extraction process and high performance liquid chromatography-ultraviolet (HPLC-UV) analytical method for determination of 3,5-dicaffeoylquinic acid (3,5-DCQA) as a part of materials standardization for the development of a xanthine oxidase inhibitor as a health functional food. The quantitative determination method of 3,5-DCQA as a marker compound was optimized by HPLC analysis using a Luna RP-18 column, and the correlation coefficient for the calibration curve showed good linearity of more than 0.9999 using a gradient eluent of water (1% acetic acid) and methanol as the mobile phase at a flow rate of 1.0 mL/min and a detection wavelength of 320 nm. The HPLC-UV method was applied successfully to quantification of the marker compound (3,5-DCQA) in Aster glehni extracts after validation of the method with linearity, accuracy, and precision. Ethanolic extracts of A. glehni (AGEs) were evaluated by reflux extraction at 70 and $80^{\circ}C$ with 30, 50, 70, and 80% ethanol for 3, 4, 5, and 6 h, respectively. Among AGEs, 70% AGE at $70^{\circ}C$ showed the highest content of 3,5-DCQA of $52.59{\pm}3.45mg/100g$ A. glehni. Furthermore, AGEs were analyzed for their inhibitory activities on uric acid production by the xanthine/xanthine oxidase system. The 70% AGE at $70^{\circ}C$ showed the most potent inhibitory activity with $IC_{50}$ values of $77.01{\pm}3.13{\sim}89.96{\pm}3.08{\mu}g/mL$. The results suggest that standardization of 3,5-DCQA in AGEs using HPLC-UV analysis would be an acceptable method for the development of health functional foods.

Growth Response of Lettuce to Various Levels of EC and Light Intensity in Plant Factory (배양액 농도와 광도가 식물공장에서 재배되는 적축면 상추의 생장에 미치는 영향)

  • To investigate the influence electrical conductivity (EC) of nutrient solution and light intensity on growth of red leafy lettuce, fresh and dry weights, number of leave, chlorophyll concentration and production efficiency were evaluated through nutrient film technique system. The levels of EC were 0.5, 1.0, 1.5, 2.0, 3.0, and $6.0dS{\cdot}m^{-1}$, and those of light intensity were 120, 150, and $180{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$. Under photoperiod of 16 h/day, the temperature was maintained in the range of $20{\sim}25^{\circ}C$. Planting density was $10{\times}10cm$ (100 plants/$m^2$). When red leafy lettuce were grown in the EC range of $0.5{\sim}1.5dS{\cdot}m^{-1}$, the fresh and dry weights decreased as the EC levels and light intensity were lowered, however, Hunter's a value showed no significant differences among the treatments of EC and light intensity levels (Ex. 1). The fresh and dry weights and production efficiency ($g{\cdot}FW/kw$) were the highest in the treatment of $3.0dS{\cdot}m^{-1}$ and $180{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$ when crops were grown under the EC range of EC $1.5{\sim}6.0dS{\cdot}m^{-1}$ (Ex. 2). But the fresh and dry weights, number of leaves, and production efficiency of $2.0dS{\cdot}m^{-1}$ were the highest when the light intensity was $180{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$ (Ex. 3). The SPAD value increased gradually as EC levels were elevated. From the above results, we concluded that optimum levels of EC and light intensity were $2.0dS{\cdot}m^{-1}$ and $180{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$, respectively, for production as well as production efficiency of red leaf lettuce in plant factory.
