Studies on the Adrenergic Alpha-Receptor in the Guinea Pig Ileum (해명 회장 운동에 대한 아드레나린성 ${\alpha}$-수용체에 관한 연구)

  • Intestine is innervated by an interconnected plexus of both sympathetic and parasympathetic nerve fibers. Sympathetic influence causes inhibition of intestinal motility mediated by both ${\alpha}-\;and\;{\beta}-adrenergic$ receptors. The mechanism of intestinal relaxation by ${\beta}-receptors$ has been extensively studied, but the function of ${\alpha}-receptors$ in intestinal motility is still unclear. Although it is suggested that catecholamine reduces acetylcholine release and this may play an important role in ${\alpha}-receptor$ mediated intestinal relaxation, there is no definite evidences about the mechanism and site of action of ${\alpha}-receptor$ mediated relaxation. In this experiment, therefore, the effect and site of action of ${\alpha}-receptor$ agonists were investigated in the guinea pig ileum using electrical field stimulation. The results are summarized as follows : 1) Electrical field stimulation elicited tonic contraction in isolated guinea pig ileum ana this contraction was completely inhibited by the pretreatment of tetrodotoxin or atropine. 2) Norepinephrine, epinephrine and dopamine inhibited the contraction induced by electrical field stimulation but methoxamine and phenylephrine had little effects. 3) Inhibitory effects of norepinephrine and dopamine was partially blocked by yohimbine and phentolamine pretreatment. But haloperidol and propranolol pretreatment cause no effects on the electrical field stimulation induced contraction. Inhibitory effect of dopamine was completely blocked by both haloperidol and yohimbine pretreatment. 4) Inhibitory effects of norepinephrine and dopamine were little affected by the pretreatment with hexamethonium. It is suggested that electrical field stimulation causes tonic contraction of guinea pig ileum by releasing acetylcholine from postganglionic fiber, and this release is blocked by presynaptic ${\alpha}-receptor$ activation.

Characterization and Pharmacological Effect of Mung Bean Trypsin Inhibitor (녹두(Vigna radiata L.) Trypsin Inhibitor의 정제 및 약물학적 특성)

  • A kypsin inhibitor was isolated and purified from Mung bean (Vigna radiata L. wilczek) which has been used as a galenic and traditional food. In addition, we evaluated the pharmacological effect of the mung bean trypsin inhibitor (MBTI) using septic shock induced guinea pig model. Purification was carried out by Sephadex G-50 gel filtration, DEAE-cellulose ion exchange chromatography, and trypsin affinity column. The molecular weight of MBTI was estimated to be about 8,000 Da by 20% SDS-PACE under reducing condition. The chemically determined partial amino acid sequences of the purified MBTI perfectly coincide with those of previously reported MBTI which is BBI type trypsin inhibito. (Bowman-birk inhibitor type). These results suggest that the purified MBTI is authentic. Hypotension shock was prevented by the pretreatment of the MBTI (10 mg/kg of the body weight) on the septic shock guinea pig model caused by psedomonal elastase.

Effect of Panax Ginseng Alcohol Extract on the Contractility of Isolated Guinea Pig Tracheal Muscle (인삼(Panax Ginseng)주정추출액이 기관지 평활근의 수축력에 미치는 영향)

  • It has been reported that administration of Ginseng powder to the Guinea pig reduces anaphylactic shook induced by horse serum (Lee, 1939). However, Lee et al. (1960) and Paik et al. (1976) have demonstrated that Ginseng increases capillary permeabilites and histamine release from the mast cell. These facts suggest that Ginseng acts directly on the bronchial muscle causing it to dilate. Recently, a number of investigators(Kidakawa & Iwasiro 1963; Takagi et al. 1973) have reported that Ginseng reverses acetylcholine- or histamine- induced contraction in the isolated Guinea pig ileum. We, therefore, undertook the present study to examine if Ginseng relaxes the spasm of bronchial muscle induced by acetylcholine or histamine. We have also attempted to identify the mechanism of the Ginseng effect. Male Guinea Pig was sacrificed by a blow on the head, The trachea was removed and sectioned with scissors into about 12 rings. After the 'C' shaped ring of cartilage was sectioned the one end of ring was tied to the bottom of the incubation bath and the other end was connected to a force transducer (FTO 3C) to record tension on a Polygraph. When the antispasmodic action of Ginseng effect was first examined in the normal trachea which was not treated by the drug. And then the Ginseng effect was tested in the muscle treated by histamine hydrochloride, acetylcholine hydrochloride or barium chloride. The results indicate that Ginseng alcohol extract relaxes the contraction of isolated tracheal muscle induced by histamine $(1{\mu}g/ml{\sim}10{\mu}g/ml)$, acetylcholine $(1{\mu}g/ml{\sim}5{\mu}g/ml)$ and barium chloride (1.5 mg/ml). The mechanism of this action is in Pa.1 due to nonspecific antimuscarinic and antihistaminic effect and in part by predominant action in the adrenergic ${\beta}-receptor$ although the ${\alpha}-receptor$ is also involved. We, therefore, conclude that Ginseng can be act as a bronchodilator.

Protective effects of selenium on alcohol and/or paraquat-induced hepatotoxicity in guinea pigs (Guinea pig에서 alcohol과 paraquat에 의한 간독성에 미치는 selenium의 방어 효과)

  • Experiments were undertaken to examine the ability of selenium to protect against alcohol and/or paraquat-induced hepatotoxicity and to examine the additive effect between alcohol and paraquat. Protective effect against hepatotoxic functions was measured in serum from alcohol(15% v/v), paraquat(200ppm), alcohol and paraquat, and combination of sodium selenite(4ppm) in drinking water-fed guinea pigs ad libitum for 4 weeks. A total of 68 healthy 7-weeks-old male animals were assigned at random to 8 treatment groups(9~13 animals/group). Body and liver weight losses, and high serum concentrations in aspartate aminotransferase(AST), alanine aminotransferase(ALT, in only paraquat group), $\gamma$-glutamyltranspeptidase($\gamma$-GTP), cholesterol(Cho), creatinine, blood urea nitrogen(BUN), total bilirubin(TB), direct bilirubin(DB), total protein(TP), albumin and globulin as well as low values in alkaline phosphatase(ALP) and glucose were produced in a groups of alcohol or paraquat-fed. These values were not potentiated in a group given the combination of alcohol plus paraquat. Morphological changes in the liver were also observed in the alcohol or paraquat-fed group. Lipid droplet and cell swelling in the hepatocytes were observed in alcohol-fed guinea pig, especially Mallory's hyaline arounded hepatic vein. In the paraquat-fed guinea pig, lipid droplet, pyknosis and karyolysis were observed. When alcohol or paraquat was combined with selenium-fed, hyperplasia of Kupffer cell in liver were observed. However, the mean ALT, $\gamma$-GTP, Cho, BUN, TB, TP, albumin and globulin values were lower in groups given the combination of alcohol and/or paraquat plus selenium, compared with groups given alcohol and/or paraquat. Also, the ratio of liver weight to body weight and ALP values(exception of paraquat plus selenium group) were increased by selenium. These results suggest that an adequate selenium confers marked protection against alcohol and paraquat-induced hepatotoxicity.

ANTIGENICITY TEST OF COPTIS RHIZOME AND CROTON TIGLIUM L. EXTRACTS(CP-2) IN GUINEA PIG AND RABBITS

  • Antigenicity tests-ASA(Active Systemic Anaphylaxis), PSA(Passive Systemic Anaphylaxis), PCA(Passive Cutaneous Anaphylaxis)-of CP-2(Green cross Co). were performed according to the established regulations of National Institute of Safety Research. The results were as follows. 1. No specic clinical signs related anaphylaxis were observed, therefore, it was considered that CP-2 had not antigenicity in guinea pigs and rabbits. 2. No blue spots were observed on the back of guinea pig in the PCA test` CP-2 related IgE was not produced.

General Pharmacology of KI-60606 (KI-60606의 일반약리작용시험)

  • In this study general pharmacological profiles of KI-60606 on the central nervous system, the cardiovascular system and the other organs were investigated. The dosages given were 0,5, 10 and 25 mg/kg and drugs were administered intravenously. The animals used for this study were mice, rats, cats and guinea pigs. KI-60606 showed no effects on general behavior, motor coordination, spontaneous locomotor activity, hexobarbital-induced hypnosis time, body temperature, analgesic activity, anticonvulsant activity and contraction of nictitating membrane in cats. Furthermore KI-60606 showed no effects on blood pressure, heart rate, LVP (left ventricular peak systolic pressure), LVEDP (left ventricular end diastolic pressure), LVDP (left ventricular developing pressure), DP(double product), CFR(coronary flow rate), smooth muscle contraction using guinea pig ileum and gastric secretion at all dosage tested except the increase of gastrointestinal transport and urinary $K^+$ excretion.

SEM Observation for the Damage of Inner Hair Cell Stereocilia of Guinea Pig Cochlea after Loud Tone Exposure (격음노출 후 기니픽 달팽이관 내유모세포 부동섬모에 관한 SEM(전자투사식현미경) 관측)

  • The inner hair cell stereocilia of the guinea pig cochlea was examined under a scanning electron microscope (SEM) after loud tone exposure onto the ear drum of the animal. Before and after guinea pigs were exposed to intensive and continuous tone such as 106 dB SPL in intensity, the functioning of the cochlea was monitored by N1-N2 audiograms. The structural damage of the stereocilia of inner hair cells (IHCs) and outer hair cells (OHCs) was examined using the SEM in x 1500 magnification. The comparison between the functional change of the cochlea and the structural damage of the IHC stereocilia is done by means of photographic observation. It can be shown that the functional change might be related to the structural damage of the IHC stereocilia after intensive acoustic trauma.

Pharmacological Effects of Mungbean Trypsin Inhibitor (MBTI) and MBTI-polymer Conjugate (Mungbean Trypsin Inhibitor(MBTI) 및 MBTI-polymer 포합체의 약물학적 특성)

  • Mungbean trypsin inhibitor (MBTI) was isolated and purified from Mung bean which has been used as a galenic and traditional food. MBTI and poly(ethylene glycol) were conjugated by using water soluble carbodiimide. We evaluated the therapeutic value of the MBTI and MBTI-PEG conjugate using animal models, sublethal septic shock model in guinea pig caused by pseudomonal elastase, shock model in rat caused by lipopolysaccharide, and the vascular permeability test by using pseudomonal elastase. In two shock model in guinea p Is and in rat, hypotesion shock was inhibited by pretreatment of MBTI. And also the vascular permeability caused by pseudomonal elastase reduced by pretreatment of MBTI. Also, therapeutic value of the MBTI-PEG conjugate was evaluated by using the sublethal septic shock model caused by pseudomonal elastase. The MBTI-PEG conjugate was more effective than native MBTI against pseudomonal elastase induced septic shock in guinea pig model.

  • Cyclical changes in the fine structures of endometrial stroma of guinea pig during the estrous cycle were studied by electron microscopy. Cytochemical studies were made in order to investigate the ultrastructural localization of the acid phosphatase,alkaline phosphatase and ATPase in these cells. The results obtained are as follows: 1. During estrus collagen fibers were most abundant in the stroma. The stromal cells showed increases in the number of several cytoplasmic organelles, especially the rough endoplasmic reticulum was significantly increased and the structures were greatly differentiated. 2. Many cytoplasmic processes and cell debris have been distributed in the stroma of metestrus. The distributions were increased and degenerated mitochondria were observed during diestrus. 3. Cytochemical studies indicated that during metestrus and diestrus acid phosphatase activities were localized in the degenerating collagen fibers. Alkaline phosphatase activities were weak in the collegen fibers during proestrus and estrus which intense activities were localized around the cell membrane during metestrus and diestrus. ATPase activities were present on the cell membrane and intercellular space of stromal cell during proestrus and estrus.

Effects of Shinsu(BL23) and Chishil(BL52) Herbal-Acupuncture with Palmijihwangtang on Blood Picture in Rabbits, Guineapigs and Rats (신유(腎兪), 지실(志室)의 팔미지황탕약침처리(八味地黃湯藥鍼處理)가 토끼, 기니어 픽 및 Rat의 혈액상(血液象)에 미치는 영향(影響))

  • Counts of RBC and proportion of packed cell volume(PCV) showdc no difference among treatment groups. the values of hemoglobin in rat showed a tendeuy to increase in herbal-acupuncture group, however the values of hemoglobin in guineapig and rabbit showed no significant difference in the treatnent groups. the count of WBC showed high in the chishiI(BL52) herbal-acupuncture group. The proprtion of neutrophils showed no significant difference in rabbit, however the values in guinea-pig and rat showed a tendency to increased in herbal-acupuncture groups. The proportion of lymphocytes in rabbit and guinea-pig showed a tendency to decreast in herbal-acupuncture groups, however the values in rat showed no significant difference. The proportion of Eosinophils and monocytes showed no significant difference in all the treated groups. Results from this study indicated that shinsu(BL23) and chishil(BL52) herbal-acupuncture with palmijihwangtang elivate the biglogical function in Rabbit Guinea-pig and Rat.
