Growth Inhibitory Activities of Kalopanaxsaponins A and I against Human Pathogenic Fungi

  • Antifungal activities of the compounds isolated from Kalopanax pictus against representative fungi of dermatomycosis were investigated using paper disc diffusion method. It was found that kalopanaxsaponins A and I were effective in inhibiting the growth of Candida albicans KCTC 1940 and Cryptococcus neoformans KCTC 7224 with minimum inhibitory concentration(MIC) of 25${\mu}g$/ml. It showed that antifungal activity of both compounds have strong selectivity against the fungi of dermatomycosis.

세균에 의한 남조 Anabaena cylindrica의 분해에 대한 연구 (A Study on the Lysis of a Bluegreen Alga Anabaena cylindrica by a Bacterium)

  • A Gram (-), rod-shaped bacterium in size of 1.6-2.8 $\times$ 0.4 μm was isolated from a eutrophic reservoir, which exhibited growth-inhibiting effect against a bluegreen alga (Anabaena cylindrica). This isolate showed positive reactions for catalase and oxidase, and optimal conditions of 35-40°C and pH 9.0. This isolate was designated AC-1 in this manuscript. In a mixed-culture of A. cylindrica and AC-1, their growth patterns were inversely correlated and the bluegreen algal vegetative cells completely disappeared within 24-36 hours. AC-1 showed similar lytic activity in natural water as in an artificial medium. The lytic activity of AC-1 was dependent on the photosynthetic activity of A. cylindrica. When observed under phase contrast microscope, the isolate lysed vegetative cells of A. cylindrica in scattered state in a liquid medium, whereas heterocysts have not been lysed.

Molecular Analysis of Freeze-Tolerance Enhanced by Treatment of Trinexapac-Ethyl in Kentucky Bluegrass

  • Trinexapac-ethyl[ 4-(cyclopropyl- $\alpha$ -hydroxy-methylene)-3,5-dioxocyclohexane carboxylic acid ethylester] is a growth-retardant for plants by inhibiting a key step in biosynthesis of GA. A treatment of trinexapacethyl generally induces a reduction in vegetative growth and also inhibits heading. In addition, the trinexapacethyl was known to enhance the freeze-tolerance in annual bluegrass, however, the mechanism is not known yet. One possible reason for the enhanced freeze-tolerance may be the antifreeze protein known to be accumulated in intercellular space of the leaf during cold acclimation. In order to see the possible in-duction of the synthesis of antifreeze proteins by trinexacpacethyl, the apoplastic proteins extracted from Kentucky bluegrass treated with trinexapacethyl were analyzed by SDS-PAGE and the presence of the antifreeze protein was observed. In addition, western analysis showed the identity of the protein induced by both a cold acclimation and a trinexapacethyl treatment. It appears that an enhanced freeze-tolerance of the turf grass by trinexapacethyl is due to the synthesis and/or accumulation of the antifreeze protein similar to the enhanced freeze tolerance induced by cold acclimation.

Relationship between Chemical Structure and Antimicrobial Activities of Isothiocyanates from Cruciferous Vegetables against Oral Pathogens

  • We evaluated the potentials of 10 isothiocyanates (ITCs) from cruciferous vegetables and radish root hydrolysate for inhibiting the growth of oral pathogens, with an emphasis on assessing any structure-function relationship. Structural differences in ITCs impacted their antimicrobial activities against oral pathogens differently. The indolyl ITC (indol-3-carbinol) was the most potent inhibitor of the growth of oral pathogens, followed by aromatic ITCs (benzyl ITC (BITC) and phenylethyl ITC (PEITC)) and aliphatic ITCs (erucin, iberin, and sulforaphene). Sulforaphene, which is similar in structure, but has one double bond, showed higher antimicrobial activity than sulforaphane. Erucin, which has a thiol group, showed higher antimicrobial activity than sulforaphane, which has a sulfinyl group. BITC and iberin with a short chain exhibited higher antimicrobial potential than PEITC and sulforaphane with a longer chain, respectively. ITCs have strong antimicrobial activities and may be useful in the prevention and management of dental caries.

Micromonospora속 균주들의 protoplast생성, 균사체로의 환원 및 융합에 대한 연구 (Formation, Regeneration, and Fusion of Protoplast of Micromonospora spp.)

  • Conditions for effcient formation and regeneration of protoplasts of Micromonospora rosaria and Micromonospora purpurea were investigated. The state of inoculm, culture stage and growth in a medium containing partially growth-inhibiting concentration of glycing have significant effects on portoplasting. A high frequency of regeneration (up to 30%) was accomplished with a hypertonic regeneration agar medium defined by Okanishi for Strptomyces. Using the optimal conditions for protroplasting and regeneration, protoplast fusion of auxotrophic M.rosaria was carried out. Polyethylene glycol 1,000 was chosen for fusogenic agent. When signgle auxotrophs were used, the recombinant frequency of auxortrophic markers varied from 1.3 to 3.2%. Using two double auxotrophs, the recombinant frequencies of 0.7-4.3% were obtained. Much lower frequencies(three or more orders of magnitude) were observed by the conventional matings.

Chromatographically Purified Porphyran from Porphyra yezoensis Effectively Inhibits Proliferation of Human Cancer Cells

  • In this study, we isolated porphyran was isolated from the red seaweed Porphyra yezoensis and assessed in terms of in vitro anti-proliferative activity. Sequential anion-exchange and gel-filtration chromatography led to purification of 3 porphyrans of different molecular masses, which contained <$50\;{\mu}g/mL$ protein and >$10\;{\mu}g/mL$ porphyran. Crude porphyran inhibited cell growth in a dose-dependent manner (0-5 mg/mL). When HT-29 colon cancer cells and AGS gastric cancer cells were cultured with various concentrations of the purified porphyran, cancer cell growth was inhibited by 50% at a low concentration (5 or $10\;{\mu}g/mL$). Furthermore, the polysaccharide portion of the porphyran preparation, rather than the protein portion, is the most effective at inhibiting cancer cell proliferation via apoptosis, as indicated by increased caspase-3 activity. Our results indicate that purified porphyran has significant in vitro anti-proliferative activity (p<0.05).

  • The heat transfer performance improvement in closed cooling water system of an electric power generation can be achieved by a corrosion control using corrosion inhibitors. The effect of trisodium phosphate and sodium nitrite upon carbon steel at various $Cl^{-1}$ ion containing water concentrations was examined by an integrated corrosion monitoring system. Nitrite was found to be the most effective inhibitor among tested inhibitors for carbon steel. The inhibiting process is considered as adsorption of nitrite ions in oxide layer which form a passive film on the carbon steel surface.

Role of GALNT14 in lung metastasis of breast cancer

  • Aberrant expression of the polypeptide N-acetyl-galactosaminyltransferase (GALNTs) has been associated with cancer, but their function(s) in metastasis remains elusive. We have recently identified GALNT14, one of the O-GalNAc glycosylation-initiating enzymes, as a prognostic marker for pulmonary relapse in breast cancer patients. Furthermore, we showed that GALNT14 promotes lung metastasis by the following novel mechanisms: 1) enhancing metastasis initiation by inhibiting the anti-metastatic effect of BMP produced from the lung stroma, 2) exploiting growth signals (e.g. FGF) supplied by macrophages, for their growth into macrometastases in the lung environment. These multi-faceted roles of GALNT14 in lung metastasis are achieved by GALNT14-mediated inhibition and activation of the BMP and FGF signaling pathways, respectively. The link among GALNT14, its downstream pathways and lung metastasis, provides us with an opportunity to develop effective therapeutic intervention for breast cancer.

Pleiotropic Effects of Caffeine Leading to Chromosome Instability and Cytotoxicity in Eukaryotic Microorganisms

  • Caffeine, a methylxanthine analog of purine bases, is a compound that is largely consumed in beverages and medications for psychoactive and diuretic effects and plays many beneficial roles in neuronal stimulation and enhancement of anti-tumor immune responses by blocking adenosine receptors in higher organisms. In single-cell eukaryotes, however, caffeine somehow impairs cellular fitness by compromising cell wall integrity, inhibiting target of rapamycin (TOR) signaling and growth, and overriding cell cycle arrest caused by DNA damage. Among its multiple inhibitory targets, caffeine specifically interacts with phosphatidylinositol 3-kinase (PI3K)-related kinases causing radiosensitization and cytotoxicity via specialized intermediate molecules. Caffeine potentiates the lethality of cells in conjunction with several other stressors such as oxidants, irradiation, and various toxic compounds through largely unknown mechanisms. In this review, recent findings on caffeine effects and cellular detoxification schemes are highlighted and discussed with an emphasis on the inhibitory interactions between caffeine and its multiple targets in eukaryotic microorganisms such as budding and fission yeasts.

죽력의 Salmonella typhimurium 등에 대한 항세균 활성과 Model Food System에서의 생육억제 효과 (Antibacterial Activities of Bamboo Sap Against Salmonella Typhimurium and Inhibitory Effects in a Model Food System)

  • 동결 건조된 죽력을 물 또는 50% ethanol에 각각 용해시킨 시료(1 mg eq./disc)에 대하여 항세균 활성을 측정한 결과, 물에 용해시킨 죽력은 gram 양성균의 경우 L. monocytogenes이 15 mm의 저해환으로 가장 활성이 강하였고 B. subtilis는 활성이 전혀 나타나지 않았으며 Gram 음성균의 경우는 S. dysenteriae가 저해환이 22 mm로 가장 강한 활성을 보였다. 50% ethanol에 용해시킨 죽력은 gram 양성균의 경우 L. monocytodenes, 가 가장 강한 활성을 보였으나 B. subtilis는 활성이 전혀 없던 물에 용해시킨 경우에 비하여 활성이 크게 나타났다. Gram 음성균의 경우, S. dysenteriae가 23 mm저해 환으로 가장 강한 활성을 보였고 V. parahaemolyticus, V. vulnificus, E. coli O157:H7는 16 mm로 상대적으로 약한 활성을 보였다. 젖산균 2종의 경우는 물 또는 50% ethanol에 용해시킨 경우 모두 전혀 활성을 나타내지 않았다. 최소저해농도는 L. monocytogenes는 50% ethanol에 녹인 경우 MIC가 0.6 mg/disc로 가장 낮았고 물에 용해시킨 죽력의 경우 S. epidermides, S. dysenteriae, L. monocytogenes, Sal. typhimurium, V. parahaemolyticus가 각각 0.8 mg/disc의 MIC값을 나타내 50% ethanol에 용해시킨 경우에 비하여 MIC값이 높은 경향을 보였으며 다른 세균들은 1.0 mg/disc의 거의 유사한 MIC값을 보였다. Model food system에서의 저해활성은 물에 용해시킨 죽력 보다 50%, ethanol에 용해시킨 죽력이 대조구에 비하여 생육을 강하게 저해하였고 MIC가 낮은 이들 4가지 공시균주들이 모두 model food system상에서 상당히 강한 저해활성을 나타내 50% ethanol에 용해시킨 죽력은 적절한 농도에서 부패세균들의 생육 억제제로 사용 가능할 것으로 사료되었다.
