A Study on the Seed Germination and Seedling Density of Fraxinus mandshruica in Nursery (포지(圃地)에 있어서 들메나무의 종자발아(種字發芽) 및 묘목생육밀도(苗木生育密度)에 관한 연구(研究))

  • This study was carried out to compare the seed germination and seedling growth of Fraxinus mandshurica growing in natural forests of the interior of Korea. Three stimulation methods were used for germination and the seedlings were planted with various densities. The results obtained were summarized as follows: 1) It was suggested that the 80 seedlings per square meter were proper densities for their growth at a nursery bed. 2) Among the three stimulation methods, 19.9% of germination was shown by the moisture storage, 2.3% by cold and warm stratification, and 0.4% by cold stratification. 3) The 8.1% of the total seedlings showed secondary height growth.

  • Dental implants using titanium have greatly advanced through the improvement of designs and surface treatments. Nonetheless, the anatomical limits and physiological changes of the patient are still regarded as obstacles in increasing the success rate of implants further, even with the enhancement of implant products. So there have been many efforts to overcome these limits. The intrinsic potential for bone regeneration can be stimulated through adjuvant treatments with the continuous improvement of implant properties, and this can play an important role in achieving optimum osseointegration toward peripheral bone tissue and securing ultimate long-term implant stability in standard surgical procedures. For this purpose, various chemical, biological, or biophysical measures were developed such as bone grafts, materials, pharmacological agents, growth factors, and bone formation proteins. The biophysical stimulation of bone union includes non-invasive and safe methods. In the beginning, it was developed as a method to enhance the healing of fractures, but later evolved into Pulsed Electromagnetic Field, Low-Intensity Pulsed Ultrasound, and Low-Level Laser Therapy. Their beneficial effects were confirmed in many studies. This study sought to examine bone-implant union and its latest trend as well as the biophysical stimulation method to enhance the union. In particular, this study suggested the enhancement of the function of cells and tissues under a disadvantageous bone metabolism environment through such adjunctive stimulation. This study is expected to serve as a treatment guideline for implant-bone union under unfavorable circumstances caused by systemic diseases hampering bone metabolism or the host environment.

  • Purpose : The purpose of this experiment was to evaluate the pre-application effect of low frequency electrical stimulation(LFES) on VEGF expression of atrophic muscle and to determine the optimal pre-application period of LFES for prevent muscle atropy Methods : Twenty-five adult sprague-dawley rats were randomly assigned to weight bearing group, hindlimb suspension for 14 days group, hindlimb suspension with pre-application of LFES for 14 days group, hindlimb suspension with pre-application of LFES for 11 dsys group and, hindlimb suspension with pre-application LFES for 7 dsys group. 16Hz of Biphasic pulse current was applied to gastrocnemius for 15min per days. Results : VEGF were decreased expression in HSG groups, whereas VEGF were significantly increased in HS+ES14G, HS+ES11G, HS+ES7G groups Conclusion : LFES during the hindlimb suspension showed a positive effect in VEGF induction and early application is strongly encourage VEGF induction. This indicated that pre-application of LFES could prevent muscle atrophy.

Bioceramic Effects to Enhance Secondary Metabolites Production in Tissue Culture of Some Medicinal Plants

  • We have investigated that a couple of soft ferrite ceramic powders having a spinal structure have shown the effect on growth and secondary metabolites production of some medicinal plants cultured in vitro. The addition of the ceramic powders as bare state to culture medium has stimulated the growth of Achyranthes japonica callus and plantlet, adventitious root of Hyoscyamus niger and Platycodon grandiflorum hairy root about 65, 75, 150 and 50%, respectively. Whereas Centella asiatica callus and plantlet, Scopolia parviflora hairy root, and Hyoscyamus albus adventitious root were not affected markedly. Moreover, the ceramic powder has enhanced the growth of H. niger adventitious roots even under conditions of irradiating alone without any direct contact between ceramic powder and media. Based on growth stimulation effect, the ceramic powders have enhanced the gross production of tropane alkaloid in H. niger adventitious root, and polyacetylene in P. grandiflorum hairy root about 35 and 30%, respectively.

  • The insulin-like growth factor (IGF) system consisting of IGF-I, IGF-II, IGF-receptors, and IGF-binding proteins (IGFBP) regulates the proliferation of a variety of cancer cell types. To examine whether a decrease in endogenous IGFBP-3 stimulates proliferation or inhibits differentiation, Caco-2 cells, a human colon adenocarcinoma cell line, were stably transfected with an anti-sense IGFBP-3 expression construct or pcDNA3 vector as control. Accumulation of IGFBP-3 mRNA and secretion of IGFBP-3 into serum-free conditioned medium, 9 days after plating, were significantly lower in Caco-2 cell clones transfected with anti-sense IGFBP-3 cDNA compared to the controls. The anti-sense clones grew at a similar rate to the controls for 8 days after plating, but achieved a higher final density between days 10 and 12. The levels of sucrase-isomaltase mRNA, a marker of enterocyte differentiation of Caco-2 cells, were lower in the anti-sense clones examined on day 9. In conclusion, proliferation of Caco-2 cells can be stimulated by lowering endogenously-produced IGFBP-3.

미생물 생육에 대한 무 추출물의 억제 및 촉진 효과

  • Extracts active on the growth of food-born microorganisms were prepared from radish by using acetone, methanol and water as the solvents. Strains of bacteria and yeast, such as L. brevis, L. fermentum, L. faecalis, L. mesenteroides, B. subtilis, S. flexneri, S. aureus and C. albicans. Addition of 1 $m\ell$ of acetone extract exhibited the strongest growth effect on L. brevis. The water extracts turned out to stimulate growth of all Lactic acid bacteria. Especially, it was observed that the same material produced the highest stimulatory effect(by about 40-50 times) on L. faecalis and L. fermentum and L. mesenteroides. Also, it was proved to have a weak stimulate effect on C. albicans, S. flexneri and S. aureus. But, on the contrary to this, when applied to B. subtilis it showed growth stimulation by about 10 times.

  • The study was carried out to investigate the change of bacterial growth in vitro according to polarity, current intensity and time, to prepare the basic data for electrotherapy and clinical research. The Gram positive Staphylococcus aureus and Gram negative Escherichia coli 1mm infect wound were cultured in Trypticase Soy Brath and Trypticase Soy Agar. The results were as followings. 1. The current stimulated group was changed in bacterial growth according to polarity, current intensity and time respectively. 2. The bacteriolytic effect revealed in the anode but the inhibitory effect of bacterial growth revealed in the cathode. 3. The lumber of E. coli reduced after 6-hours but the numbers of S. aureus reduced after 2 hours in Trypticase Soy Brath. 4. The anode showed acid reaction and cathode showed alkaline reaction in Trypticase Soy Agar.

Fractionated Aged Black Garlic Extracts Enhance Growth of Anti-My-10 Hybridoma Cells and Production of IgG1 Antibody

  • Aged black garlic (ABG) was extracted with 20% ethanol and water (crude extracts) and fractionated into three categories (>10, 3-10, and <3 kDa). The effect of crude extract supplements on anti-My-10 hybridoma cell growth and IgG1 antibody production was investigated in suspension culture with a chemically defined protein-free medium. We observed that supplementation of ABG to the cell culture medium stimulated anti-My-10 hybridoma cell growth and production of IgG1 antibody, particularly with fractionated ABG of low molecular weight. The stimulation depended upon the concentration and the size of the fractionated ABG. We also found that the growth-promoting activity was not correlated with high antibody production. These results suggest that fractionated ABG is a novel and promising alternative as an animal cell culture supplement.

Cytotoxicological Effect of Tebufenozide, an Insect Growth Regulator (IGR): Stimulation of Filamentous Actin Reorganization and Enhancement of Hsp27 Expression in Drosophila Kc Cells

  • The cytotoxicological responses to insect growth regulator (IGR), using tebufenozide as ecdysteroid mimic, were investigated in Drosophila Kc cells. Treatment of Kc cells with tebufenozide showed significant growth inhibition and striking morphological changes including aggregation and elongation of the cells. In order to understand the cellular mechanism underlying the response of Drosophila cells to tebufenozide, immunofluorescence microscopy was performed. We found that treatment of Kc cells with tebufenozide enhanced the reorganization of f-actin and stimulated the expression of hsp27. These data suggest a possible association of filamentous actin (f-actin) and hsp27 in the cytotoxicological mechanisms of growth regulators in Drosophila cells.

  • The effects of various dehiscent application such as dehiscent materials (big chaffs, vermiculite etc.), growth regulators and agricultural chemicals (plant protector.) on stimulation of dehiscence and shortening of dehiscent period were investigated Results obtained were as follows : 1. The moisture content of endosperm and seed coat at 10 day after dehiscent application amounts between 40% and 50%. 2. Endosperm diameter was increased with time of stratification, and the embryo growth showed in linear function, 3. Non-dehiscent seed showed also normally development of embryo, and the property of dehiscence dependent from physico-chemical nature of ginseng seed coat. 4. The best dehiscent materials were big chaffs and followed vermiculite, sand and sand with big chaffs. 5. The effect of dehiscence of ginseng seed showed higher activity in fungi than in bacteria in general. 6. Agricultural chemicals ( plant Protector) reduced the dehiscent rate of ginseng seed 7. The best timing of dehiscent treatment was between August 1 and August 10 but the smaller amount of dehiscent rate after August 10 dehiscent appllication indicated that big chaffs and growth regulator treatment may be controlled shortening of dehiscent period of ginseng seed.
