Synthesis and Characterization of Water Soluble Fluorescent Copper Nanoparticles

  • The electrostatic interaction between emerging quantum-confined nanostructures with plasmonic structures is crucial for future biological applications. Water-soluble green fluorescent copper nanoparticles (Cu-NPs) were fabricated. We demonstrate that L-ascorbic acid is considered as a key to precisely control small Cu-NPs and the capability of the surface ligands, while cetyltrimethylammonium bromide is used as a stabilizing agent controls the particle growth, and stabilizes the nanoparticles. Water-soluble green fluorescent Cu-NPs are tunable through modification of the reaction periods.

Enigma of Small Peptides Ubiquitin and SUMO in Plants

  • Post-translational covalent modifications by small molecules or peptides remodel target proteins. One such modification, made by ubiquitin or small ubiquitin-related modifier (SUMO), is a rapidly expanding field in cell signaling pathways. Ubiquitin attachment controls the turnover and degradation of target proteins while SUMO conjugation regulates their activity and function. Recent studies report many examples of cross-talk between ubiquitin and SUMO pathways, indicating that the boundary is no longer clear. Here, we review recent progress concerning how ubiquitin and SUMO participate in new regulatory roles in plant cell, and how ubiquitination and sumoylation control plant growth and development.

Isolation and characterization of LHT-type plant amino acid transporter gene from Panax ginseng Meyer

  • A lysine histidine transporter (LHT) cDNA was isolated and characterized from the roots of Panax ginseng, designated PgLHT. The cDNA is 1,865 bp with an open reading frame that codes for a protein with 449 amino acids and a calculated molecular mass of 50.6 kDa with a predicted isoelectric point of 8.87. Hydropathy analysis shows that PgLHT is an integral membrane protein with 9 putative membrane-spanning domains. Multiple sequence alignments show that PgLHT shares a high homology with other plant LHTs. The expression profile of the gene was investigated by real-time quantitative polymerase chain reaction during various chemical treatments. PgLHT was up-regulated in the presence of abscisic acid, salicylic acid, methyl jasmonate, NaCl, and amino acids. To further explore the function of PgLHT gene, full-length cDNA of PgLHT was introduced into P. ginseng by Agrobacterium rhizogenes A4. The overexpression of PgLHT in the hairy roots led to an obviously increase of biomass compared to the controls, and after addition of the amino acids, the overexpressed-PgLHT hairy roots grew more rapidly than untreated controls during early stage of the culture cycle. The results suggested that the PgLHT isolated from ginseng might have role in the environmental stresses and growth response.

  • The factors involved in the initial neoplastic transformation and subsquent growth of uterine fibroid are poorly understood. The reduction in uterine fibroid volume associated with the chronic administration of the mechanisms mediating the decrease in fibroid volume in GnRH-a treated patients are poorly defined. The purpose of this study was to determine the proliferating cell nuclear antigen(PCNA) in fibroid from-women pretreated with GnRH analogue(GnRH-a) compared with controls. Tissue was obtained from 16 premenopausal women with uterine fibroid who received GnRH-a(D-Trp6-GnRH) intramusculary every 28 days for four injections. The mean proliferating index(PI) in patients with uterine fibroids was $2.25{\pm}0.9$, and in controls was $8.82{\pm}1.8$(P<0.001). The proliferating index was not corrleated with the reduction of fibroid volume. In this clinical study, although hypoestrogenism may be the main factor that reduce the volume of fibroid, other factors are also considered to be involved in that process. And the regrowth of uterine fibroid may be affected by increased production of PCNA after stopping GnRH-a.

Effect of Porcine Somatotropin and Insulin on Prenatal Survival and Uteroplacental and Umbilical Cord Development in Gestating Gilts

  • Pregnant Yorkshire gilts were utilized to investigate the efficacy of exogenous administration of pST and/or insulin in enhancing prenatal piglet survival, uteroplacental and umbilical cord growth and development. Gilts were randomly assigned in a $2{\times}2$ factorial arrangement to four treatment combinations consisting of either daily i.m. injections of 5 mg pST (P, n=23); 0.50 IU/kg of insulin (I, n=23); combination of pST and insulin (P+I, n=23); or 1 ml of saline as control (C, n=23) from gestation Day 30 to 70. All gilts were sacrificed on gestation d 113 to evaluate piglet survival and uteroplacental or umbilical cord development Uteri were longer (346.3 vs 325.7 cm; p<0.05), and heavier (3122.8 vs 2940.7 g; p<0.05) in insulin treated gilts. Only placental macroscopic surface area was enhanced by maternal insulin injections (p<0.05) Incidence of umbilical cord abnormalities were low (14.3%), and they were independent of maternal treatment, occurring more in short cords than in long ones (21 vs 12%; p<0.05). A 6% increase in cord length (53.2 vs 48.6 cm; p=<0.05) was observed in piglets from treated gilts compared with controls. Significant sex differences (in favour of males) were observed in piglet weight, crown rump length and for most umbilical or placental parameters. Gilt weight gains from breeding to Day 113 of gestation were 10% and 15% greater in pST and insulin treated gilts compared with controls. These data indicate that prepartum injections of pST and/or insulin to gestating gilts seem to have a beneficial effect on uteroplacental or umbilical cord development and promote conditions conducive for perinatal piglet survival.

Promoter Methylation Status of Two Novel Human Genes, UBE2Q1 and UBE2Q2, in Colorectal Cancer: a New Finding in Iranian Patients

  • Background: The ubiquitin-proteasome system (UPS) degrades a variety of proteins which attach to specific signals. The ubiquitination pathway facilitates degradation of damaged proteins and regulates growth and stress responses. This pathway is altered in various cancers, including acute lymphoblastic leukemia, head and neck squamous cell carcinoma and breast cancer. Recently it has been reported that expression of newly characterized human genes, UBE2Q1 and UBE2Q2, putative members of ubiquitin-conjugating enzyme family (E2), has been also changed in colorectal cancer. Epigenetics is one of the fastest-growing areas of science and nowadays has become a central issue in biological studies of diseases. According to the lack of information about the role of epigenetic changes on gene expression profiling of UBE2Q1 and UBE2Q2, and the presence of CpG islands in the promoter of these two human genes, we decided to evaluate the promoter methylation status of these genes as a first step. Materials and Methods: The promoter methylation status of UBE2Q1 and UBE2Q2 was studied by methylation-specific PCR (MSP) in tumor samples of 60 colorectal cancer patients compared to adjacent normal tissues and 20 non-malignant controls. The frequency of the methylation for each gene was analyzed by chi-square method. Results: MSP results revealed that UBE2Q2 gene promoter were more unmethylated, while a higher level of methylated allele was observed for UBE2Q1 in tumor tissues compared to the adjacent normal tissues and the non malignant controls. Conclusions: UBE2Q1 and UBE2Q2 genes show different methylation profiles in CRC cases.

A new biomarker for the early diagnosis of ovarian torsion: SCUBE-1

  • Objective: Prompt diagnosis and management are essential for saving the adnexal organs from infarction in cases of ovarian torsion (OT). This study aimed to determine the diagnostic significance of signal peptide, complement C1r/C1s, Uegf, and Bmp1 (CUB), and epidermal growth factor-like domain-containing protein-1 (SCUBE-1) levels in cases of OT, an emergent ischemic condition, and the relationship of SCUBE-1 with oxidative stress parameters. Methods: This prospective study was conducted among 15 OT patients and 20 age- and gravidity-matched healthy women. SCUBE-1 serum concentrations were determined by using enzyme-linked immunosorbent assays. In addition, oxidative stress was evaluated by measuring the serum levels of advanced oxidation protein products (AOPP), ferric reducing ability of plasma (FRAP), and glutathione (GSH). Results: The SCUBE-1 titers were significantly higher in the patients with OT than in the controls (p=0.008). In addition, serum FRAP and GSH levels were significantly lower in the OT patients than in the controls (p<0.001 for both). Serum AOPP levels were higher in the OT patients, but this trend was not statistically significant (p>0.05). Furthermore, there were no correlations between SCUBE-1 levels and age, gravidity, parity, cyst size, and AOPP, FRAP, or GSH levels (p>0.05). Conclusion: We believe that SCUBE-1 may be a promising biomarker for the early diagnosis of OT.

미백제가 법랑질 명도 변화에 미세경도에 미치는 영향 (The Effect of Tooth Whitener on the Color and Microhardness of Human Enamel in Vitro)

  • Human teeth vary widely in color. Practitioner and patients are concerned with preventing and correcting discolored or dark teeth to achieve and maintain stain-free, white teeth. Tooth brushing cannot alter tooth color but it can remove adhering films and stains. The esthetics of natural dentition can be improved by bleaching and this process can be applied to intrinsically and extrinsically stained teeth. The need for a brighter, more attractive smile has made rapid growth in the market for tooth whiteners. There is no doubt these products work as whiteners, at least on mild to moderate stains, but the safety of these products are unclear. In this experiment, the effect of tooth whitener application on the color and microhardness of extracted human enamel was measured. RMS, RMT and NWT were used as tooth whiteners, and tooth paste(ETQ) and hydrogen peroxide solution(HPO) were used as controls. 35 caries-free extracted human molars were embedded and polished with the exposed enamel diameter of 4 mm. The tooth whiteners and control agents were applied according to the manufacturers' instructions or clinically simulated procedures for eight weeks, and measurements were repeated every two weeks. Value(L*) difference was measured using Differential Colorimeter(Model TC-6FX, Denshoku Co., Japan), and microhardness was measured using microhardness tester(Mitsuzawa Seiki Co., Japan). The results were as follows; 1. After application of agents for eight weeks, the Vickers hardness increased significantly in the ETQ, RMS and RMT application group(p〈0.01), and that decreased significantly in NWT application group(p〈0.01), but in HPO application group there was no significant change. The change in microhardness was greatest in NWT application group(p〈0.01). 2. After application of tooth whiteners and controls for eight weeks, the value change of toothpaste application group was significantly lower than those of other agents groups(p〈0.01), and there was no significant difference in value(L*) change among tooth whitener groups(p〉0.01). 3. The application of tooth paste and paste type tooth whitener made gradual value change, but hydrogen peroxide gel type tooth whitener and hydrogen peroxide solution made rapid value change during initial application period.

Effects of Bisphenol A on Sex Differentiation and Gonadal Development of Medaka, Oryzias latipes

  • A study on the effects of bisphenol A (BPA) on sex differentiation and gonadal development in medaka, Oryzias latipes, was investigated by histological examination. The fish were exposed to aqueous solutions of BPA at nominal concentrations of 50, 100, and 200 $\mu\textrm{g}$/L from newly-hatched larvae stage to 70 d. The ovaries of female fish were composed of oocytes at the chromatin nucleolus and peri-nucleolus stages at 20 d after the exposure. The testes contained a number of spermatogonia and spermatocytes at 30 d. In the process of sex differentiation. gonadal development was not different in all experimental groups until 30 d after the exposure. At 70 d after the exposure, however, advanced development of oocytes in the ovary and inhibition of spermatogenesis in the testis were observed in the BPA-treated groups compared to the non-treated controls. More females than males were identified in the 50 and 100 $\mu\textrm{g}$/L BPA-treated groups, in comparison to the 200 $\mu\textrm{g}$/L BPA-treated group and non-treated controls. Medaka exposed to 200 $\mu\textrm{g}$/L BPA were bigger compared to other experimental groups. The present study suggests that BPA may lead to problems in either mating or sexual behavior due to the difference in growth and disparity of sexual maturation between male and female fish.

Effect of agricultural materials of traditional agriculture on control of rice blast (Pyricularia oryzae)

  • The objective of this research was to determine controlling effects on rice blast (Pyricularia oryzae) in rice plants by using plant extracts from different extraction methods (water, boiling water, fermentation, and ethanol) from 38 agricultural materials of traditional agriculture. Rice blast was completely suppressed by 3% ethanol extracts in Rheum palmatum roots, and suppressed 97% and 77% by 10% ethanol extracts in onion bulb and pine tree leaves, respectively in a laboratory test. However, other agricultural materials showed low effect on suppression of rice blast. Additionally, in a seedling test, rice injury of two cultivars (Ilmibyeo and Hopyoungbyeo) against rice blast was reduced 40-71%, 29-63%, and 23-63% by 5 and 10% ethanol extracts in Rheum palmatum roots, onion bulb, and pine tree leaves, respectively, compared with non-treated controls. Rice injury of two cultivars (Ilmibyeo and Hopyoungbyeo) against rice blast was reduced by 21-55%, 23-46%, and 5-39% in response to Rheum palmatum roots, onion bulb, and pine tree leaf applications at 100, 200 and $400g/m^2$ at 0 day after seeding, respectively, compared with non-treated controls. Rice plants did not show any leaf injuries and growth reduction after treatments of the Rheum palmatum roots, onion bulb, and pine tree leaf extracts or soil application. Thus, the above materials may be used for controlling rice blast in organically produced rice fields.
