Formulation and Sensory Evaluation of Granule and Tablet from Lotus Rhizome(Nelumbo nucifera Rhizome)

  • Nelumbo nucifera root(NNR) is used to clear summerheat(暑熱), bear Yang(陽) upwards and stop bleeding as mentioned in traditional Korean medicine. Also, it has been known that NNR is effective for lowering blood pressure and hyperlipidemia. The rhizome is considered to be nutritive, demulcent, diuretic and cholagogue and is used to treat piles, dyspepsia and diarrhea. An increasingly growing market for nutraceuticals and functional foods has triggered the study on natural sources for nutraceuticals, health foods and functional foods. But rhizome was inconvenient to formulate liquid dosage form(extract) by way of hot water because of its limited storage. Also the majority of the consumers have a complaint against the dosage. The purpose of this study was to develop the functional materials from NNR without side effects. We formulated the solid dosage form viz tablet and granule from the lotus root. Sensory evaluation was performed in terms of smell, taste, color and overall of lotus root and all colored forms(brown, dark brown, light green and yellow) of tablet and granule to evaluate the acceptability of the formulated tablets and granules. In sensory evaluation, among the formulated tablets and granules, light green granules obtained best score overall and yellow tablets showed the overall improved acceptability. In conclusion, lotus rhizome could be recommended as functional food. Further studies to clarify bioactive functions of Nelumbo nucifera in experimental animal model on atopic dermatitis are in progress.

  • The effect of granule size of dried red ginseng extract on physical characteristics on tablet such as hardness, color, yield and disintegration time was studied. A moisture proofing agent was found to be necessary to form granules without serious absorption of moisture. The result showed that decrease in particle size resulted increase of values in weight, hardness and disintegration time. The granule size of 60∼80 mesh was found to be optimum for proper color uniformity and other physical characteristics of tables.

  • The primary objective of this work is to find the optimal condition for the granule tablet formulation of alfuzosin-HCl that aims to achieve a sustained drug release. (Hydroxypropyl)methyl cellulose (HPMC) is one of the most widely used polymer as a drug formulation and therefore has been utilized in this study as an excipient. Alfuzosin-HCl granule tablet was developed using the various viscosities of HPMC and the effects of viscosity on drug release was investigated. Fourier transform-infrared (FTIR) and X-ray diffraction (XRD) were employed to investigate the chemical structure and crystallization of alfuzosin-HCl in the formulation. We prepared the granule tablet by a direct compression method and studied the release profile in the stimulated intestinal fluid (pH 6.8). As the viscosity of HPMC increased the release of alfuzosin-HCl decreased, demonstrating that controlled release of alfuzosin-HCl can be achieved by varying the viscosity of HPMC.

  • Spherical granules of anti-inflammatory enzyme were prepared by mixer granulation and the recovery rate of enzyme activity by processing was compared with tablet ones. In the enteric granule coating .processing, the effect of the amount of coating solution and the conentration of fatty alcohol on disintegration and stabilities on the accelerated conditions were also studied. Being prepared in non-pressure and non-aqueous condition, spherical granules of enzyme made better recovery of enzme activity than tablet ones by 10 times. Combined processing of both mixer granulation and enteric granule film coating provided the noble enteric coated granules, in the sense of disintegration and stabilities, was obtained from using 0.125% fatty alcohol in coating solution.

Influence of Compression and Granule Size on Dissolution Rate of Acetaminophen, Fenbufen, and Mefenamic Acid Tablets (정제(錠劑)의 과립도(顆粒度) 및 임계타정압(臨界打錠壓)이 용출속도(溶出速度)에 미치는 영향(影響))

  • Acetaminophen, fenbufen, and mefenamic acid tablets were formulated with the same additives, granulated to different sizes, ranging $30{\sim}200mesh$, and compressed by pressure of $0.5{\sim}2.0\;ton{\cdot}cm^{-2}$. The hardness and disintegration time of the three tablets were determined. The dissolution rates of the three tablets at critical point of pressure were investigated. The results showed that fenbufen, and mefenamic acid tablet had critical compression pressure of $1.0\;ton{\cdot}cm^{-2}$, but acetaminophen tablet had that of $1.5\;ton{\cdot}cm^{-2}$. Neverthless, three tablets were disintegrated within 5 min. Among the three tablets, mefenamic acid tablet followed zero order of dissolution and the rate constants were as fellows; $1.68 \;mg{\cdot}min^{-1}\;(30{\sim}50mesh),\;1.76mg{\cdot} min^{-1}\;(50{\sim}80\;mesh)\;and\;4.85\;mg{\cdot}min^{-1}\;(80{\sim}200mesh)$.

  • This study was conducted to manufacture new product containing high soluble polysaccharide from Hericium erinaceus extract. The new products were prepared for powder, granulate and tablet form by spray drying process. The high soluble powder form was mixed with sub-ingredients and then granulated form by granulator and molded as tablet form by a press molder respectively. Moisture content of products was following order; spray dried powder > granule > tablet Total sugar content of produce was observed to 19.14% in spray dried powder, 37.58% in granulate and 35.76% in tablet. The hue angle of products was $85.5^{\circ}$ in spray dried powder, $95.37^{\circ}$ in granulate and $94.67^{\circ}$ in tablet. The absorption capacity was higher in powder, but the product type did not affect the color.

  • Osmotic granule system which is one of the drug delivery systems has been developed to improve manufacturing process and other problems of tablet osmotic systems. It consists of water swellable seed layer, nifedipine drug layer, and drug release controlled membrane layer and manufactured by fluidized bed coater. The granule size and mombrane thickness can be controlled by various amounts of seed and coating solution, respectively. It could be observed that the morphology of osmotic granule was different at each coating step as well as type of coating solution. The bigger the size of granule, the slower the release rate was observed due to decreasing the total specific surface wed of granule. Also, it was observed that the increase of membrane thickness was caused to retard the dissolution of nifedipine due to decreasing the water absorption rate. The drug solubility for dissolution media is greatly affected to nifedipine release. From these results, we assured that osmotic granule can be fabricated by fluidized bed coating methods, and the appropriate release profile could be controlled by the controlling of bead size, membrane thickness and dissolution media.

  • To improve the type error of osmotic tablet which is one of the drug delivery system, osmotic granule could be manufactured by fluidized bed coating. It has drug layer containing different amount of osmogant and is coated with membrane including different types of binder. We confirmed that the morphology of osmotic granule was different at each coating step. The more mont of osmotic agent, the faster drug release was observed due to increasing the driving force for drug release from osmotic granule. And drug release from osmotic granule coated with membrane using different types of binder was differed by solubility of binders to water. The formation of pore in membrane was confirmed by SEM and DSC Membrane using water soluble binder released more amount of drug. From these results, we assured that difference of osmotic pressure between the inside and the outside of granule and porosity of membrane have an effect on drug release from osmotic granule.

Optimization of the experimental conditions for determination of roxithromycin in bulk and dosage forms

  • Roxithromycin (RXT), which is an antibiotic used to treat respiratory tract and urinary infections, is official in Korean Pharmacopoeia (KP) and is marketed in various dosage forms including tablet, granule, suspension, and tablet for suspension in Korea. This study presents how a universal and reliable method to quantify RXT in bulk drug and formulations was developed. Effects of factors including column type, buffer concentration, type and concentration of organic solvent, buffer pH, and type and concentration of mobile phase additive, were examined, and some categorical or crucial factors including the types of column, organic solvent, mobile phase additive and the buffer pH were optimized by one-factor-at-a-time approach. Subsequently, concentrations of the buffer and additive and column temperature were optimized by response surface methodology using Box-Behnken design aiming to acquire the RXT peak of good shape. The optimized method employed a Phenomenex Gemini $5{\mu}$ C18 110A ($150{\times}4.60mm$, $5{\mu}m$) maintained at $30^{\circ}C$ with the mobile phase consisting of 25 mM phosphate buffer (pH 6.0) with 0.3 % tetrabutylammonium hydroxide and methanol at a ratio of 37:63 (v/v). Method validation results showed that the developed method was linear, precise, and accurate. Compared to the compendial methods in KP 10 that exhibited a significant tailing of the RXT peak despite using unfavorably high buffer concentrations and were not harmonized among bulk drug and formulations, this method could be universally applied to RXT bulk drug and marketed products in various dosage forms and thus was adopted in KP 11.
