In the larvae of the black soldier fly, Hermetia illucens, innate immunity mechanisms are activated in response to various pathogens and stimulants, resulting in the expression of antimicrobial peptides (AMPs). To induce the mass production of AMPs, H. illucens fifth instar larvae were immunized with five different kinds of bacteria. We isolated from the hemolymph of the H. illucens larvae after bacterial challenge, and their antimicrobial activities against Gram-positive bacteria (Staphylococcus aureus) and Gram-negative bacteria (Escherichia coli) were measured using the inhibition zone assay. Among these five different kinds of bacteria, the hemolymph of Bacillus subtilis-challenged H. illucens larvae showed the strongest antimicrobial activity against both Gram-positive bacteria and Gram-negative bacteria. The antimicrobial activity of the hemolymph of $1{\times}10^9cfu/ml$ B. subtilis-challenged H. illucens peaks at 24 hr at 48 hr post-infection and gradually declines with time. Moreover, the immunized hemolymph also showed strong antimicrobial activity against various poultry pathogens such as S. enteritidis, S. typhimurium, and S. pullorum. These results suggest that the expression of AMP genes in B. subtilis-challenged H. illucens is up-regulated by innate immune responses, and that B. subtilis-challenged H. illucens overexpressing AMPs may be useful as a feed additive in livestock diets to reduce the need for antibiotics.
In the present study, using biological information of bacteria and biochemical information of chlorine dioxide gas, Gram-positive bacteria, e.g., Alloiococcus otitis, Erysipelothrix rhusiopathiae, Staphylococcus caprae, Staphylococcus lentus, and gram-negative bacteria, e.g., Acinetobacter baumannii complex, Aeromonas salmonicida, Brucella melitensis, Oligella ureolytica were used whether a plastic kit to release ClO2 gas could inhibit their growth. Overall, chlorine dioxide gas showed about 99% inhibition of bacterial growth, with less than 10 CFU. However, it was found that Gram positive Alloiococcus otitis and Gram negative Aeromonas salmonicida had more than about 50 CFU. When comparing the results of experiments with several bacteria, it suggested that the concentration of chlorine dioxide gas would be at least 10 ppm to 400 ppm for the bacterial inhibition. The results of this study could be used as basic data to evaluate the clinical usefulness of chlorine dioxide gas. If this study helps with prior knowledge to help clinicians to recognize and prevent the presence of micro-organisms that cause infections in hospitals, it would be helpful for activities such as patient care as a convergence field. In the future, it is considered that the research results will be the basis for rapidly inhibiting the microbes infected with patients by utilizing data of the information of the microbes that are inhibited for chlorine dioxide gas.
A study was carried out to define the relationship between the N-acetyl-$\beta$-D-glucosaminidase(NAGase) levels and isolated pathogenic bacteria in 379 quarter fore milk of mastitis suspected samples collected in this clinics. All samples were tested the NAGase, California mastitis test(CMT), Somatic cell count(SCC) and bacterial culture. Except 111 from 379 samples, 268 bacteria-positive quarter fore milk samples were classified into the latent and mastitis infection group by SCC($500,000cells\;per\;m{\ell}$), and the mean NAGase levels($nmol/min/m{\ell}$) of each isolated pathogen in mastitis infection group were Staphylococcus aureus 3.067, Coagulase-negative staphylococci 4.083, Staphylococcus aureus 3.594, Str. uberis 3.513, Str. dysgalactiae 1.640, E coli 4.441 and gram negative rods 4.560, respectively. Most of the relationship between mean SCC and NAGase in each pathogen group were highly significant using a student t test(p<0.05). When the mastitis pathogens were classified into minor(Coagulase-negative staphylococci, Corynebacterium sp.) and major pathogen group(Staphylococcus aureus, Streptococcus agalactiae, Str. uberis, Str. dysgalactiae, gram negative rods), the NAGase levels were higher at major than minor pathogen group. On the other hand, when the mastitis milk samples were classified by SCC($500,000cells\;per\;m{\ell}$) and by the presence of pathogen(IDF scheme), the NAGase levels were also higher at the mastitis than latent infection. The possibility of combining SCC and NAGase data in order to give the more difinitive diagnosis is discussed.
Helicobacter pylori is a Gram-negative bacterium which causes chronic gastritis and is associated with gastric ulcer, duodenal ulcer and gastric carcinoma. In the process of screening of antibacterial activities against H. pylori from soil microorganisms, fungus No. 60686 was isolated. After fermentation of No.60686, the antibacterial compound was isolated, purified and followed by extraction of mycelium with organic solvents, acetone and ethyl acetate, through silica gel chromatography, LH-20 gel chromatography and HPLC. As a result of the structural analyses of the compound by IR, $^1$H- and $^{13}$C-NMR, FAB/Mass spectrophotometer, the compound having the antimicrobial activity was identified as chaetoglobosin A ($C_{32}H_{36}N_2O_5$), a cytochalasan derivative. The antimicrobial activity of chaetoglobosin A was tested against Gram-positive and negative bacteria by paper disk method. Among the test strains of 9 Gram-positive bacteria and 18 Gram-negative bacteria containing 4 H. pylori strains, the growth of 4 H. pylori strains and 3 S. aureus strains (SG 511, 285 and 503) was only inhibited by chaetoglobosin A. Also it was shown that its growth inhibition against H. pylori strains was stronger than that against S. aureus strains at the treatment of the same concentration. Therefore it was concluded that chaetoglobosin A has a specific growth inhibition against H. pylori of the tested bacteria.
The mode of the combined effect of adipic acid and medium chain monoglycerides was investigated by using nine strain. Though monoglycerides alone had little antibacterial activity against gram negative strains, the combined use with adipic acid showed much higher activity against others of gram positive strains as well as gram negative strains. But exceptionally, it seemed difficult practically to prevent the growth of lactic acid bacteria completely by the combined use of adipic acid and monoglyceride. For yeast and mold, monoglycerides alone had a high activity but adipic acid had a little activity. In antibacterial mechanism, we thought that adipic acid acted on the cell envelope and then monoglyceride acted on the altered cell.

Antimicrobial finishing of textiles has been found to be an economical way to prevent (or treat) skin disorders. Hence, this research effort was aimed at elucidating the relationship between the molecular weight (MW) of chitosan and its antimicrobial activity upon six dermal reference microorganisms, as well as the influence of the interactions with cotton fabrics on said activity. Using 3 chitosans with different MWs, as well as two chitooligosaccharide (COS) mixtures, a relevant antimicrobial effect was observed by 24 h for the six microorganisms tested; it was apparent that the antimicrobial effect is strongly dependent on the type of target microorganism and on the MW of chitosan - being higher for lower MW in the case of E. coli, K. pneumoniae, and P. aeruginosa, and the reverse in the case of both Gram-positive bacteria. Furthermore, a strong antifungal effect was detectable upon C. albicans, resembling the action over Gram-positive bacteria. Interactions with cotton fabric resulted in a loss of COS activity when compared with cultured media, relative to the effect over Gram-negative bacteria. However, no significant differences for the efficacy of all the 5 compounds were observed by 4 h. The three chitosans possessed a higher antimicrobial activity when impregnated onto the fabric, and presented a similar effect on both Gram-positive bacteria and yeast, in either matrix. Pseudomonas aeruginosa showed to be the most resistant microorganism to all five compounds.

Contamination of bacterial infection is one of serious problems in in vitro culture system of root crops. From the contaminated tubes over 140 of petiole cultures of Pinellia ternata, a medicinal plant, 4 genera 8species 48 strains of bacteria, including Aeromonas and Pseudomonas, were isolated and identified and another 8 strains were not fully identified. Most of them were motile Gram positive bacteria as in common in early stage of in vitro cultures. Six strains of bacteria, 5 of Gram negative, including Enterobacter, and 1 of Gram positive, were identified from the embryonic axes cultures of tea plant. From the susceptibility test to pre-screened 5 antibiotics, all of the bacteria except for 2 species of Pseudomonas were susceptible to cefotaxime 60∼100mg/L. While 60mg/L erythromycin only was effective to Pseudomonas. Combination of erythromycin 20mg/L and cefotaxime 60mg/L totally suppressed the growth of all bacterial strains tested. Susceptibility test of bacteria from tea embryonic axes cultures showed similar results. Combination of erythromycin 35mg/L and cefotaxime 60mg/L was effective to 15 bacterial strains and partially effective to 1 unidentified.
In this study, Coffea arabica ethanol extract (CAE) was tested for possible functional cosmetic agent. Whitening effect was measured by tyrosinase inhibition assay, and anti-oxidant activity was checked by SOD-like activity. SOD-like activity of CAE showed $94.8{\pm}6.2%$ at $500{\mu}g/mL$. The anti-bacterial activities CAE was evaluated against three different gram-positive bacteria and six gram-negative bacteria including MRSA strains. CAE exhibited in vitro broad spectrum antimicrobial activities of gram-negative bacteria without antifungal activity. CAE was strong exhibited against MRSA CCARM3561. The tyrosinase and L-DOPA inhibitory activities of the CAE lower than those positive control arbutin. CAE reduced melanin contents of B16-F10 melanoma cell in a dose dependent manner and decrease about 89.2% at a concentration $100{\mu}g/mL$. These result highlight the potential of coffee extract as a naturally active and non-toxic antibacterial suitable for cosmetic applications.

This study was performed to evaluate the rodent repellency and antimicrobial activity of natural oils. Thirteen natural essential oils were evaluated for repellent efficacy against rodent feed intake suppression activity. Salvia sclarea (sage), Cinnamomum zeylanicum (cinnamon) Cymbopogon citratus (lemon grass), and Syzygium aromaticum (clove) showed the best repellent efficacy among tested natural oils. Antimicrobial activity against Gram-negative (four strains) and Gram-positive (four strains) bacteria were evaluated using four natural oils, which showed high efficacy on the rodent repellent test. Syzygium aromaticum showed antimicrobial activity against all tested bacteria and Cymbopogon citratus, Cinnamonum zeylanicum, and Salvia sclarea showed antimicrobial activity against some tested bacteria.
Purpose: We investigated trends in antibiotic pressure and the antibiotic susceptibility of gram negative bacteria isolated from Korean children over 10 consecutive years. Methods: From January 2004 to December 2013, the antibiotic susceptibility of Klebsiella pneumoniae, Escherichia coli, Pseudomonas aeruginosa, and Acinetobacter baumannii blood isolates obtained from children <18 years of age was determined according to the 2009 Clinical and Laboratory Standards Institute guidelines. Antibiotic consumption data were also analyzed. Results: The prevalence of K. pneumoniae, E. coli, P. aeruginosa, and A. baumannii bacteremia was 4.6, 3.5, 3.4, and 2.2 cases/1,000 blood cultures/year, respectively. In K. pneumoniae, resistance to the third and fourth cephalosporin did not increase significantly. However, carbapenem-resistant K. pneumoniae first appeared in 2010, and the resistance rate increased to 9% between 2012 and 2013. Resistance to 3rd and 4th cephalosporin increased from 10% to 50% in E. coli, and resistance to carbapenem rose abruptly from 11% to 71% in A. baumannii (P for trend <0.01). However, such an increase of resistance was not observed in P. aeruginosa. There is a positive correlation between the resistance rate of cefepime in E. coli and the consumption of cefepime (r=0.900, P=0.037). Conclusion: The significant burden of antibiotic consumption and the high prevalence of antibiotic resistance to gram negative pathogen isolated from bacteremic children were observed. Empirical antibiotics should be wisely selected, and continued efforts to decrease the overall antibiotic pressure are mandatory, especially in highly resistant situations.
