We investigated the effect of different media components on the characteristics and productivity of Lentinula edodes cultivated in sawdust. Ten substrates were used for this study. Soybean and cotton seed meal were unsuitable substrates as fungal mycelia did not grow to maturity during the incubation period. Lentinula edodes (NIFoS culture number 2462) was grown on the ten substrates, and soybean hull, mixed medium, cotton seed hull and corn grain showed greater effect on the productivity than wheat bran. However, wheat bran induced higher productivity in NIFoS 2778 than the other substrates. Sawdust medium with soybean meal produced smaller fruiting bodies than the other substrates. In contrast, corn meal media produced larger mushrooms than the other substrates. The external characteristics of the mushrooms varied based on the substrates upon which they were grown. This is not surprising given that the substrates differed with respect to carbohydrate and protein content, e.g., cotton seed and soybean meal contained a higher crude protein and crude ash than the other substrates tested.

In spite of extensive knowledge of the surface chemistry and the transport mechanism in filtration systems, there is still insufficient understanding of the physical characteristics of suspensions and the system components. Because of this, no filtration mechanisms are mathematically generalized to the full extent. The purpose of this paper is to propose experimental equations for the filtration process. using the tracer study in filter layer. Some of results are as follows. (1) The Volume of the specific deposit (${\sigma}$) in filtration was directly measurable using the tracer study without interrupting the filtration. (2) It was also confirmed that the head loss in filtration was greatly in fluenced by the micro-air babbles. (3) The correction coefficient(f) was introduced into the Kozeny-Carman equation in order to apply it for the clogging filter media. The coefficient(f) was experimentally obtained. (4) The total head loss was graphically generalized assuming mathematical filtration models I II (see fig. 7,8) (5) The total head loss was obtained from the filtration model in the field filtration conditions. Kefir is a traditional fermented milk in Caucasusian area and is made mainly of milk fermented with lactic acid bacteria and yeasts. Six typical kefir grains were selected from ten kefir grains collected from different locals in Korea. Kefir grains were gelatinous in texture and had various shapes of villi, grapes, leaves, hulled millets, and towels. To investigate predominant microflora of kefir grains, SPC, MRS, M17, Rogosa, and APT agar media were used for viable cell count MRS, SPC, and Rogosa media were most acceptable for bacterial cell counts of the selected kefir grains. From one or two of the SPC agar plates which contained around 25∼50 colonies, all grown colonies were isolated and identified. Most predominant bacteria was identified as Lactobacillus fermentum by API 50 CHL kit. The proportions of Lb. fermentum and Lb. brevis among the total identified bacteria were around 41~88% and M4%, respectively. To select the best preservation method for kefir grains, refrigeration, freezing, and freeze drying were compared. Freeze drying was found most suitable for the preservation of kefir grains, based upon their acid-producing activities and production of off-flavors.
Seedling blight of sesame (Sesamum indicum L.) is one of the most serious diseases in Korea. This study was performed to control seedling blight in the field where sesame has been cultured for 4 years of Industrial Crop Division at the Crop Experiment Station. The pathogenic fungi of seedling blight which is known Fusarium oxysporum f. vasinfectium and Rhizoctonia solani was inoculated to the wheat media and it was spreaded on the experimental field in the concentration of 109 of wheat media mixed with 10g of fine soil. Seed sterilization was the most effective method for seedling blight control for two year experiment. Seed sterilant Benlate-T was highly effective for con trolling seedling blight with infection rate of 4% and yield of 40.7kg per 10a comparing to the control with infection rate of 96% and grain yield of 6.4kg per l0a, the other chemicals, Busan 30, Vitathiram, Captan showed good effect to control seedling blight, but less effect to the Benlate-T. The spraying treatments on leaf surface 25 days after sowing were not effective to seedling blight.

Two-hundred and fifty-five strains of actinomycetes isolated from soil samples were screened for their antagonistic activities against four well-known wood decay fungi (WDF), including a brown rot fungus, Gloeophyllum trabeum and three white rot fungi Donkioporia expansa, Trametes versicolor, and Schizophyllum commune. A dual culture assay using culture media supplemented with heated or unheated culture filtrates of selected bacterial strains was used for the detection of their antimicrobial activity against four WDF. It was shown that Streptomyces atratus, S. tsukiyonensis, and Streptomyces sp. greatly inhibited the mycelial growth of the WDF tested compared with the control. To evaluate the biocontrol efficacy of S. atratus, S. tsukiyonensis, and Streptomyces sp., wood blocks of Pinus densiflora inoculated with three selected Streptomyces isolates were tested for weight loss, compression strength (perpendicular or parallel to the grain), bending strength, and chemical component changes. Of these three isolates used, Streptomyces sp. exhibited higher inhibitory activity against WDF, especially G. trabeum, as observed in mechanical and chemical change analyses. Scanning electron microscopy showed that cell walls of the wood block treated with Streptomyces strains were thicker and collapsed to a lesser extent than those of the non-treated control. Taken together, our findings indicate that Streptomyces sp. exhibits the potential to be used as a biocontrol agent for wood decay brown rot fungus that causes severe damage to coniferous woods.
This study was carried out to investigate the management characteristics and growth performance of L. edodes from the cooling stage to incubation. Bags of different heights and weights are available for bagging. When the medium size of $17{\times}13cm$ was used and the size of the inoculation hole was changed from 1/3 to 2/3, the browning period was shortened to 30 days. Mycelial growth was evaluated according to the cooling temperature after sterilization. It was observed to be the highest at 122 mm/15 days at $10^{\circ}C$ and 114 mm/15 days and 117 mm/15 days at $15^{\circ}C$ and $20^{\circ}C$, respectively. The contamination rate of the sawdust media before inoculation was measured as 0, $4.5{\times}10$, $1.3{\times}10^2$, $4.0{\times}10^3cfu$ at $5^{\circ}C$, $10^{\circ}C$, $15^{\circ}C$, and $24^{\circ}C$ respectively. The average of $1.6{\times}10^8$ colony forming units (cfu) of microorganisms was observed in the sawdust that had been piled for six months outdoors. In summer, the sawdust has to be used immediately after mixing. The sterilized medium had an average of $4{\times}10^3cfu$ of microorganisms at $24^{\circ}C$ and $1.3{\times}10^2cfu$ at $15^{\circ}C$. After 15 days of inoculation in vitro, the growth conditions of the sawdust was the best at 132 mm, followed by grain and liquid. When inoculated with liquid spawn, the moisture content of the substrate should be adjusted between 50% and 55% in advance.
Thirty species of wild yeasts were ioslated and identified from two hundred strains collected from flowers by enrichment techniques using steptomycin. Among them, twnety three species could assimilate methanol and three species, Candida incommunis, cryptococcus aerius and Hansenula ciferrii which showed good biomass yield were selected. These three species assimilated methanol as carbon and energy source without mixture of vitamin and yeast extract. The species grown on methanol media were confirmed to have all essential amino acids in their cellular constituents. The content of total amino acids are as followings ; Candida incommunis : 42.5%, Hansenula ciferrii : 39.9%. Of the essential amino acids lysine and threonine which are usually lacking in grain protein were as much as flour's. The experiments on the growth conditions for the higher biomass yield showed the result that the optimum concentration of methanol ans temeprature wre defined as 1.3% and $30^{\circ}C$ in each species.
Co-Zn ferrite thin films grown on thermally oxidized silicon wafers were fabricated by a sol-gel method. Magnetic and structural properties of Co-Zn thin films were investigated by using x-ray diffractometer (XRD), atomic force microscopy (AFM), auger electron spectroscopy (AES) and a vibrating sample magnetometer (VSM). Co-Zn ferrite thin films annealed at 400 $^{\circ}C$ presented have only a single phase spinel structure without any preferred crystallite orientation. Their surface roughness of Co-Zn ferrite thin films was shown as less than 3 nm and the grain size was about 40 nm for annealing temperatures over 600 $^{\circ}C$. A moderate saturation magnetization of Co-Zn ferrite thin films for recording media was obtained in this study and there is no significant difference of their magnetic property with those external fields of parallel and perpendicular to planes of the films. The maximum value of the coercivity was obtained as 1,900 Oe for Co-Zn ferrite thin film annealed at 600 $^{\circ}C$.
Populus. caspica L. is an Iranian indigenous poplar species which naturally distributed in the northern part of country. Unfortunately, overuse has removed many of the stems of better form, so that natural stands now usually appear small and crook. Therefore genetic variation for selection of new superior clone of this species is needed. Conventional hybridization system is currently used to induce genetic variation in poplar species but incompatibility barriers have been observed between them. In vitro ovule embryo culture was used to overcome incompatibility obstacle for interspecific hybridization between Populus caspica L. with Populus deltoids L.62/75. Female flowers of Populus caspica L. have artificially been pollinated with pollen grain of P. deltoides 62/75 in one direction using twig and pot crossing system. Ovaries at different ages (7, 14 and 21 days after pollination) were disinfected through 70% ethanol for 1 minute, 5% of sodium-hypochlorite solution for fifteen min followed by three time rising with sterile distil-water. Isolated ovaries were then transferred to MS hormone free medium containing 30 and 60 g/L sucrose for embryo development and germination. Collected data have been analyzed by two factorial experimental designs. The results indicated that there were significant differences between age of embryos for development and germination at ${\alpha}=0.01%$. Highest embryo germination (45%) was observed from 21 days old ovaries. No significant differences were observed between MS culture media containing 30 and 60 g/L for percentages of ovary-embryo germination and number of germinated embryo per ovary at ${\alpha}=0.05%$. Fourteen percentage of embryo germination obtained in MS medium supplemented with 60 g/L sucrose, while only 35% of isolated ovaries were able to germinate in MS containing 30 g/L sucrose. Induced plantlets in 4 cm height were transferred into pots containing soilless (1:1:1 peat, per lit and vermiculite) medium for acclimatization. After successful acclimatization, plants were delivered to nursery.
The weed population dynamics as affected by contrasting conventional tillege (CT) and no-tillage (NT) practices with a minimum herbicide application was studied in Athens, Georgia, U.S.A. Common chickweed (Stellaria media) was the most common spring weed while johnsongrass (Sorghum halepense), sicklepod (Cassia obtusifolia), and pigweed (Amaranthus retroflexus) accounted for 89∼97% of net production during summers of 1983 and 1984. Total weed production in summer of 1984 was 2∼5 times greater than that of 1983. Weed production was greater in NT plots than in CT plots in summer of 1983, but reverse was the case in summer of 1984. In spring, net production in NT plots was greater than that in CT plots, especially, in 1985. Species diversity was consistently higher in NT plots, but in the wet summer of 1984 the pattern was different, with higher diversity in CT plots. Weed species diversity was higher in the spring rye crop than in the summer grain sorghum crop. The larger but less diverse weed populations in summer of 1984 indicated that these populations experienced competitive exclusion. Under the favorable summer moisture conditions the three dominant species grew so vigorously and quickly as to exclude many less common species that were able to survive under the drier conditions in 1983. The three dominant species not only excluded other weeds in 1984 but also greatly reduced crop production. The perennial johnsongrass was equally successful, or even more so, in CT plots as in NT plots. Plowing did not kill johnsongrass rhizomes but tended to break them up, thus increasing the number of individual plants that appear after the plowing. It means that johnsongrass was not controlled by the plowing. In summer of 1983, a moderate amount of weedy growth was maintained with a minimum amount of gerbicide application in NT and CT plots. It is possible that a small mixed weed population would be beneficial by providing cover for predatory and parasitic arthropods, and by reducing soil temperature and moisture losses.
