This paper reviews current status of weed control practices in herbicide-resistant crops to examine weed management strategies in cope with cropping herbicide-resistant crops in the near future. Herbicide-resistant crops were rapidly adopted weed management technologies due to broad-spectrum weed control without crop injury. Transgenic glyphosate-resistant cultivars in soybean, corn, canola, and cotton were adopted to manage weeds at lower cost in a simplified weed management system. Dual stack crops with glyphosate and glufosinate resistance were developed to control glyphosate resistant weeds in corn, soybean and cotton. New multiple herbicide-resistant crops with resistance to glyphosate and glufosinate, acetolactate synthase (ALS) inhibitors, synthetic auxin herbicides, 4-hydroxyphenyl pyruvate dioxygenase (HPPD) inhibitors or acetyl Coenzyme A carboxylase (ACCase) inhibitors will expended the utility of existing herbicide technologies to manage the evolution of resistant weeds. However, herbicide resistant crops alone cannot solve weed problems and thus studies on diverse weed managements using an array of alternating herbicides of mode of action, mechanical, and cultural practices are needed for integrated weed management systems in the future.
From the soil environment which is treated with herbicides. we isolated strain having high resistance against glyphosate. After being identified. the isolated strain was turned out to be Pseudomc~nu.c~c ~paciua nd to have intense tolerance lo the 10 mM glyphosate. The isolated strain shows slow, growth rate about twenty hours in glyphosate comparing with that in inorganic phosphate. As a result of confirming the p~~sitioonf glyphosate resistant gene. it was proved to exist in chromosome. After cloning it into E coli C600, transformants E. coli THC-101 and plasmid pGR19 were obtained.
This paper provides the current status of weed science and prospects for the development of weed science based on the research trends presented at the 7th International Weed Science Conference in 2016. Approximately 520 researchers from 59 countries, including Korea, participated in the conference and presented 625 papers in nine research areas. Major research topics were herbicide resistance, weed ecology, weed management in agricultural and non-agricultural lands, herbicide spray technology, and non-chemical weed control. Studies on herbicide resistance presented more than 30% of all papers presented. Particularly, resistance to non-selective herbicides, such as glyphosate and glufosinate-ammonium, and non-target sites of resistance mechanisms were the main subjects of the herbicide resistance research area. Moreover, the conference focused on research concerning herbicide resistant weeds of staple crops of the world (corn, wheat, and rice). Arylex was introduced as a new compound which has a mode of herbicidal action similar to synthetic auxin. Three compounds being developed as HPPD inhibitors were studied for ways to reduce their toxicity and tested as mixed with safeners. Additionally, parasitic weeds, which are not native to Korea, are an expanding research subject in the world. Although 45 years have passed since the first report of herbicide resistance in 1970, herbicide resistance remains a serious problem in most intensive cropping systems of the world and will continue to be a major area of study in the future.
This study describes isolation and identification of a soil bacterium which is degradable of phosphinothricin and improvement of the isolated strain by using mutagenesis and spheroplast fusion. The experiment was performed to search for a possibility of development of a new strain which is both PPT-degradable and glyphosate-resistant by using interspecies cell fusion between the PPT-degrading bacterium. Pseudornonu.\ puucimohlis and a glyphosate -resistant strain, Pseudornonu.~ cc,pucicl. Auxotrophic mutants were obtained by the treatement of P. puucimohili.\ with ethylmethanosulfate, and used to cell fusion. Lysozyme and EDTA were used to spheroplast formation and regeneration rates :)f the spheroplast were 6.5'%1 in P. pauc.irnohili.\ and 8.8% in P.ci,j~u[,i(lr, espctively. Polyethylenglycol 5.000 was used to cell fusion as fusogen. The fusant\ulcorner F1. F2. F\ulcorner and F4 werc- obtained by the intra- and interspecies cell fusion. The fusant Fl of intraspecies cell fusion was higher to the wild type by 1 I'%l in PPT degrading ability, and the fusant F3 of inierspesis cell fusion developed plyphosatc-resistant and PPI-dcgrading ability which were propertics of two parental strains.
This study was conducted to develop an antibiotics marker-free potato (Solanum tuberosum L., cv. Taedong valley) plant having resistance against two herbicides. Agrobacterium tumefaciens strain EHA105, harboring a binary vector plasmid pCAMBIA3300 containing bar gene under the control of a promoter CaMV35S and linked CP4-EPSPS genes driven by CaMV35S promoter, was used in the current study. The leaf segments of newly bred potato variety (cv. Taedong Valley) was co-cultured with Agrobacterium. Then, the regenerated individual shoots were excised and transferred to potato multiplication medium supplemented with 0.5 mg/L phosphinothricin. The shoots were rooted in MS medium without hormone and obtained putative transgenic plant E3-6. Integration of target genes into the E3-6 plant and their expression was confirmed by PCR, Southern analysis, and ELISA test. The tissue necrosis test on young leaf blade and shikimic acid accumulation test using the tissue of E3-6 plant were conducted to investigate the resistance to glufosinate-ammonium and glyphosate, respectively. The transgenic plants (E3-6) simultaneously showed a high resistance to both herbicides. The same results were surely obtained also in the whole plants foliar-treated with alone or mixture of two herbicides, glufosinate-ammonium and glyphosate.
This study is the first comprehensive report on the molecular cloning, structural characterization, sequence comparison between wild and mutant types, copy number in the genome, expression features and activities of a gene encoding 5-enolpyruvylshikimate-3-phosphate synthase (EPSPS) in Korean lawn grass ($Zoysia$$japonica$). The full length cDNA of the EPSPS from Korean lawn grass ($zj$EPSPS) obtained from a 3' and 5' RACE method was 1540 bp, containing a 1176 bp ORF, a 144 bp leader sequence (5' UTR) and a 220 bp 3' UTR, which was eventually decoded 391 amino acid residues with a molecular mass of 41.74 kDa. The Southern blot detection of the $zj$EPSPS showed that the gene exists as a single copy in the Korean lawn grass genome. Sequence comparison of the $zj$EPSPS gene demonstrated that the glyphosate-tolerant mutant (GT) having a Pro-53 to Ser substitution in the gene seems to have a preferred binding activity of the enzyme to phosphoenol pyruvate(PEP) over glyphosate, which allows the continuous synthesis of aromatic amino acids in the shikimate pathway. From the Northern blotting analysis, the $zj$EPSPS was found to be highly expressed, with continuous increase until 36 hours after 0.5% glyphosate treatment in both wild and mutant samples, but 1.5-fold higher EPSP synthase activity was observed in the tolerant mutant when exposed to the glyphosate treatment. The molecular information of the $zj$EPSPS gene obtained from this study needs to be further dissected to be more effectively applied to the development of gene-specific DNA markers and zoysiagrass cultivars; nevertheless, the glyphosate-tolerant mutant having the featured $zj$EPSPS gene can be provided to turfgrass managers for weed problems with timely adoptable management options.
Along with the worldwide rapid increase of the cultivation area and commercial production of genetically modified (GM) crops, the amount of GM grains imported to Korea has also been increasing. Roundup-Ready soybean (RRS) was introduced with 5-enolpyruvyl shikimate-3-photphate synthase (EPSPS) gene derived from Agrobacterium CP4 to confer the resistance to herbicide, glyphosate. In this study, we tried to develop PCR-based analytical method to detection the presence of RRS among non-GM soybeans. In order to detect RRS specifically, oligonucleotide primers were specifically designed based on the nucleotide sequence of EPSPS transgene. Qualitative PCR method was established and its specificity and accuracy were confirmed by analysing the nucleotide sequence of PCR DNA fragments. Bioassay was also conducted by spraying glyphosate at seedling stage. Survived individuals showed obvious resistance to Roundup Ready, however all of non-GM seedlings died in two weeks after spray. Conclusively, the highly selective detection systems for RRS were successfully established by both PCR using specific primers to EPSPS transgene and bioassay using the herbicide resistance of RRS. In addition to, the imported soybean showed to be mixed to several varieties regarding to 100-seed weight and hilum color.

In order to elucidate the differences of protein profiles among soybean cultivars, the protein composition of three conventional domestic soybean cultivars and two imported ones including glyphosate-tolerant HS2906 was analyzed by total nitrogen measurement, amino acid analysis and PAGE/densitometry. There were no statistically significant differences in the levels of any amino acid, including aromatic amino acids, between glyphosale-tolerant soybean and the conventional soybean WS82. In the extraction of protein, the SDS/buffer system was more efficient than the defatting/water system. The SDS-PAGE/densitometry analysis showed that there was a similar profile of proteins among cultivars, although the amount of total protein ranged from 380.2 mg/g to 423.9 mg/g. In addition, there was no discernable difference of protein profile between glyphosate- tolerant soybean (total protein amount, 380.2 mg/g) and the conventional soybean WS82 (390.2 mg/g), although the amount of ${\beta}$-conglycinin (55 kDa) was lower in glyphosate-tolerant soybean. Meanwhile, the amount of 25 kDa protein was greater in domestic soybean cultivars than imported ones. Thus, normal PAGE/ densitometry method would be useful to analyze the difference in protein profiles of soybean proteins, and furthermore Evaluate the protein profile of proteins between GMO and conventional soybean.
This study was conducted to develop herbicide-resistance domestic maize plants by introducing the CP4 5-enol-pyruvylshikimate-3-phosphate synthase (CP4 EPSPS) gene using Agrobacterium tumefaciens-mediated immature embryo transformation. Immature embryos of five genotypes (HW1, KL103, HW3, HW4, HW7) were co-cultivated with strains Agrobacterium tumefaciens (strain C58C1) containing the binary vector (pCAMBIA2300) carrying Ubiquitin promoter-CP4 EPSPS gene and Cauliflower mosaic virus 35S (CaMV35S) promoter-nptll gene conferring resistance to paromomycin as a selective agent. The presence and expression of CP4 EPSPS transgene were confirmed by PCR, RT-PCR and Northern blot analysis, respectively. Also, the resistance to glyphosate in the transgenic maize ($T_1$) was analyzed by shikimate accumulation assay. The frequency (%) of paromomycin-resistance callus was 0.37, 0.03, 2.20, 2.37, and 0.81% in pure lines HW1, KL103, HW3, HW4 and HW7, respectively. EPSP transgene sequences were amplified in putative transgenic plants that regenerated from paromomycin-resistance calli of two inbred lines (HW3, HW4). Of them, RT-PCR and Northern blot analyses revealed that the transgene was only expressed in two transgenic events (M266, M104) of HW4 inbred line, and a mild glyphosate resistance of transgenic event (M266) was confirmed by the lower shikimate accumulation in leaf segments. These results demonstrate that transgenic maize with herbicide-resistance traits in Korean genotype can be genetically obtained.

The objective was to know if corn growers at Hongchcel County, Kangwon-do, where corn is the major agricultural product, control weeds effectively and use herbicides properly to control weeds. Sixty-eight corn growers at Hongchon were participated in this survey. Corn growers have noxious weeds, such as hairy carbgrass, common purslane, lambsquarters, Asiatic dayflower, mugwort and barnyard grass in their corn fields. Forty-four percents of the answered corn growers control weeds by hand weeding, and the others by a combination of physical and chemical methods. Corn growers have a preference of paraquat and alachlor for a pre-emergence treatment (94% of the answered) and of paraquat, glufosinate, glyphosate and fluazifop-P-butyl for a post-emergence treatment (87% of the answered). Corn growers did not select and use herbicides in a proper manner. fifty-four percent of the answered corn fevers selected herbicides by their own experience and 18 percent by neighbors recommendation. Herbicides were not selected and used in a proper manner by corn growers: 54 percent of the answered selected herbicides by the experience of growers and 18 percent by the recommendation of neighbors. In addition, a half of the answered applied over two-fold higher amounts of herbicides than the recommended rate. With these improper- selection and use of herbicides, 37 percent of the answered have experienced the crop damage and 42 percent the herbicide poisoning. We conclude that the development of the educational program for the proper selection and use of herbicides is needed for corn growers in Kangwon-do.
