Synthesis of Aesculetin and Aesculin Glycosides Using Engineered Escherichia coli Expressing Neisseria polysaccharea Amylosucrase

  • Because glycosylation of aesculetin and its 6-glucoside, aesculin, enhances their biological activities and physicochemical properties, whole-cell biotransformation and enzymatic synthesis methodologies using Neisseria polysaccharea amylosucrase were compared to determine the optimal production method for glycoside derivatives. High-performance liquid chromatography analysis of reaction products revealed two glycosylated products (AGG1 and AGG2) when aesculin was used as an acceptor, and three products (AG1, AG2, and AG3) when using aesculetin. The whole-cell biotransformation production yields of the major transfer products for each acceptor (AGG1 and AG1) were 85% and 25%, respectively, compared with 68% and 14% for enzymatic synthesis. These results indicate that whole-cell biotransformation is more efficient than enzymatic synthesis for the production of glycoside derivatives.

수삼에서의 아데노신 분리 및 동정 (Isolation and Identification of Adenosine in Fresh Ginseng)

  • The investigation of UV absorbing compounds in saponin fraction of Pnm.1. Kiairnk root was carried out by thin layer chromatography, semipreparative HVLC. l3C, 1H-NMR, mass spectrometry and chemical characteristics in searching plant for growth regulatony substances such as phenolic glycoside. Drying of fresh ginseng at 15 $^{\circ}C$ decreased not only number but also size of UV absorbing sports on TLC. One of the relatively large spots in fresh ginseng was isolated and identified as adenosine, which is subjected for growth stimulatory activity Detection of phenolic glycosides failed in dried root bolt was highly probable in fresh ginseng even with the insufficient amount of sample.

대두와 된장의 지용성 성분 비교 (Comparison of Lipid Constituents in soybean and Beanpaste)

  • Soybean and beanpaste have been used as ore of the important food in Korea. Beanpaste is made from soybean(seed of Glycine max $M_{ERRIL}$, Leguminosae) by traditional fermentation. It was expected that the secondary metabolites might be formed by the action of some microorganisms or enzymes on the constituents primarily contained in soybean during manufacturing or storage of beanpaste. A systematic investigation on the chemical constituents was carried out. As a result, sterol glycoside named daucosterin, lipo-daucosterin have been isolated from the lipid fraction soybean and their structures elucidated basis of chemical and physicochemical evidences. A ratio of composition of the lipid constituents in soybean and beanpaste were estimated.

(${\beta}-Galactosidase$를 이용한 Isofalvone 배당체의 비배당체화 연구)

  • 본 연구는 식물성 에스트로겐인 isoflavone 배당체의 ${\beta}-1,4\;glycoside$ 결합을 ${\beta}-galactosidase$로 분해하여 그 생리적 작용이 더욱 효과적인 aglycone화 하기 위한 조건을 연구하여 기능성식품개발에 응용토록 하는데 목적이 있다. 실험 결과, 모든 ${\beta}-galactosidase$${\beta}-1,4\;glycoside$ 결합의 분해력이 인정되지는 않았으며, 분해 가능성이 인정된 효소를 선택하여 실험한 결과 효소의 함량이 증가함에 따라 aglycone의 성분비율이 증대된 반면, 그 손실률 또한 증대되었고 배양시간이 길수록 aglycone의 성분비율이 증가하였으나 배양초기에 급격한 증가율을 보였으며, pH6과 7, 8에서 유사한 정도의 증가율을 보였고, 온도가 증가함에 따라 aglycone 성분비율이 증가되었지만 $40^{\circ}C$이상에서는 그 손실률이 크게 증가하였다. 결과적으로 판단해 볼 때, isoflavone을 aglycone화 하는데 있어 인체 내 유사조건에서 aglycone의 증가율이 높았으며 최적의 조건과 많은 차이를 나타내지 않았고 그 손실률 또한 최소화된 결과를 나타내어 isofalvone을 적용한 기능성 유제품을 연구하는데 있어 긍정적인 가능성을 보였다.

  • Scillascilloside E-3 (1) and E-1 (2) were isolated from the bulbs of Scilla scilloides. The cytotoxic activities of these compounds were tested against murine (B16/F-10, 3LL) and human cancer cell lines (MCF7, PC-3, HT29, LOX-IMVI, A549 and HT1080). These compounds exhibited a significant cytotoxic activities against all tested cancer cells. Futhermore, the contents of 1 and 2 in S. scilloides are 43.2 and 27.9 mg/kg, respectively.

박주가리 지상부로부터 Flavonol Glycoside 성분의 분리 (Flavonol Glycosides from the Aerial Parts of Metaplexis japonica)

  • Ten flavonol glycosides were isolated from the EtOAc fraction of the MeOH extract of Metaplexis japonica Makino. Structures of the flavonoids were elucidated on the basis of spectroscopic data and comparison with literature values. The flavonoids were found to be mostly common flavonol 3-glycosides. It is of interest that the sacchaide parts of the isolates were pairs of arabinosides, glucosides, galactosides, rutinosides and robinobiosides of kaempferol and quercetin. All of these compounds were isolated for the first time from this plant.

  • $\alpha$-Benzoyloxypaeoniflorin (1), a new antioxidant monoterpene $\alpha$-glycoside anomer was isolated from Paeonia suffruticosa along with known compounds, $\beta$-benzoyloxypaeoniflorin (2), paeonolide, paeoniflorin and mudanpioside H. The structure of 1 has been determined by comparing spectral data with those of $\beta$-Benzoyloxypaeoniflorin(2). Compound 1 exhibited moderately potent radical scavenging activity on DPPH radical.

  • 약 25년 생 잣나무 수피 추출물중 EtOAc용해분에 함유된 화합물을 단리 동정하였다. 화합물의 단리는 Sephadex-LH20과 TSK-gel HW-40F를 충전제로 한 Column chromatography로 하였다. 단리된 화합물의 구조동정은 $^1H{\cdot}^{13}C$-NMR, HMQC, HMBC등과 $FAB^+$ MS 등을 이용하여 결정하였다. 새로이 동정된 화합물은 Z-pinostilbenoside, E-desoxyrhaponticin, 그리고 E-resveratroloside 등이었다.

  • Antibacterial activity-guided fractionation of the $CHCl_3-MeOH$ (1:1) extract of Paeonia suffruticosa root bark furnished three monoterpene glycosides, 6-O-vanillyoxypaeoniflorin (1), mudanpioside-H (2), and galloyl-oxypaeoniflorin (3). Of the isolated compounds, compound 1 is a new compound. All isolated compounds showed broad, but moderate, antibacterial activity with minimum inhibitory concentration (MIC) values in the range of 100 to $500\;{\mu}g/mL$ against eighteen pathogenic microorganisms of concern for public health or zoonosis.
