Isolation and Identification of the High-Glutathione Producing Saccharomyces cerevisiae FF-8 from Korean Traditional Rice Wine and Optimal Producing Conditions (전통 발효주로부터 glutathione 고함유 효모 Saccharomyces cerevisiae FF-8의 분리.동정 및 최적 생산조건)

  • In this study, strain of high-producing intracellular glutathione was isolated from Korean traditional rice wine. The isolated strain was identified as Saccharomyces cerevisiae based on the morphological, physiological and biochemical characteristics, and was designated as FF-8. The optimal condition for glutathione production by Saccharomyces cerevisiae FF-8 was obtained after cultivation with shaking for 72 hours in the YM medium. The optimal temperature, shaking rate and initial pH for the glutathione production were $30^{\circ}C$, 100 rpm and pH 6.0, respectively. The dry cell weight and glutathione concentration produced by Saccharomyces cerevisiae FF-8 were 5.2 g/l and 72.0 mg/l, respectively, under the optimal culture condition.

  • Chemical properties and physiological activities of the freeze-dried synnemata of Beauveria bassiana were examined. A proximate analysis showed that the synnemata consisted mainly of carbohydrate (49.86%), protein 11.36%), and a moisture content of 30.64%. It contained a low amount of crude ash (4.76%) and fat (3.38%). The carbohydrate was composed mainly of mannose (52.3%), galactose (31.5%), glucose (13.2%), and rhamnose (3%). Trace amounts of arabinose, xylose, and fructose were present. Major amino acids In the synnemata were glutamic acid, glycine, aspartic acid, arginine, threonine, alanine, valine, leucine, lysine, and aspartic acid with the amounts of 30.42, 25.22, 17.17, 15.12, 12.65, 15.23, 12.47, 11.47, 14.24, and 17.17 mg/g, respectively. Among extracts from the synnemata, the hot-water extract showed 67% of anticomplementary activity compared to that of the positive control, followed by ethyl acetate extract (17%) and methanol extract (15%). The hot-water extract also had anticoagulant activity with 55 sec of coagulating time and this fraction exhibited the most potent Intestinal immune system modulating activity. The methanol extract showed the highest inhibitory activity (25%) on the 12-O-tetradecanoyl phorbol-13-acetate-induced superoxide ($O_2^-$) generation, followed by hot-water extract (18%) and ethyl acetate extract (10%). The data in the present study indicate that the extract of Beauveria bassiana synnemata contains some healthful chemical ingredients and it could provide beneficial physiological activities. These features of the synnemata should be of interest to the food industry as well as other industrial fields.

  • Fatty liver is a common metabolic disorder of dairy cows during the transition period. Historically, the diagnosis of fatty liver has involved liver biopsy, biochemical or histological examination of liver specimens, and ultrasonographic imaging of the liver. However, more convenient and noninvasive methods would be beneficial for the diagnosis of fatty liver in dairy cows. The plasma metabolic profiles of dairy cows with fatty liver and normal (control) cows were investigated to identify new biomarkers using $^1H$ nuclear magnetic resonance. Compared with the control group, the primary differences in the fatty liver group included increases in ${\beta}$-hydroxybutyric acid, acetone, glycine, valine, trimethylamine-N-oxide, citrulline, and isobutyrate, and decreases in alanine, asparagine, glucose, ${\gamma}$-aminobutyric acid glycerol, and creatinine. This analysis revealed a global profile of endogenous metabolites, which may present potential biomarkers for the diagnosis of fatty liver in dairy cows.

  • These studies were carried out to investigate the properties of H-Y antigen purified by immunoaffinity chromatography using monoclonal H-Y antibody. Immunoaffinity column was prepared by the coupling of monoclonal antibody to the Aminolink Coupling Gel. Murine testis supernatant was applied onto the column and eluted by O.lM glycine-HCl buffer and 31${\mu}g$ of H-Y Ag was eluted from one testis. Purified H-Y Ag strongly reacted with Con A and lentil from 6 different kinds of lectins tested, which may indicate that sugar moiety of H-Y Ag is composed of glucose, mannose and their derivatives. Con A-sepharose affinity column was used to purified H-Y Ag based on that H-Y Ag is glycoprotein. The fraction eluted by 0.2M Me-${\alpha}$-D-mannoside from the column loaded with murine testis supernatant was identified to be H-Y Ag by dot blot test. Molecular weight of the purified H-Y Ag was estimated by Sepharose G-75 gel filtration and SDS-PAGE, and showing that it was about 67,000 dalton. In fluorescence test, the ratio of XY embryos and XX embryos was 1:1.

  • Gibberellic acid ($CA_3$) is used in many industries and constitutes the primary gibberellins produced by fungi and bacteria. However, there is no information on $CA_3$ production by Methylobacterium oryzae CBMB20, a novel plant growth promoting bacterium. We investigated the favorable carbon (C) and nitrogen (N) sources and ratios and cultural conditions, such as incubation temperature, pH of the culture medium, and incubation period for the maximum production of $CA_3$ by Methylobacterium oryzae CBMB20. Maximum $CA_3$ production was observed in ammonium mineral salt (AMS) broth supplemented with Na-succinate and $NH_4Cl$ as C and N sources, respectively. The maximum $CA_3$ production was found at the C/N ratio of 5:0.4 g $L^{-1}$. The highest $CA_3$ production was obtained when the bacterial culture was incubated at $30^{\circ}C$ for 96 h at pH 7.

Separation of Glutathione by Ion Exchange Chromatography (이온교환 크로마토그래픽을 이용한 Glutathione 분리)

  • Glutathione(L-$\gamma$ -glutamyl-L-cysteinylglycine, GSH) produced by microbial enzymes was separated by a liquid chromatography. In order to select a resin which would bind GSH efficiently, a batch adsorption experiment was carried out with GSH solution and various resins at pH 8.0 GSH bound to Q-sepharose and QAE-sephadex among anion exchange resins, but the latter was found not to be suitable because of the reduction of resin volume at high salt concentration. Preliminary experiments using a standard solution were carried out to separate GSH. GSH and $\gamma$ -glutamylcysteine were separated from the other constituents by applying step gradient of salt(NaCl) concentration. GSH was successfully separated from $\gamma$ -glutamylcysteine by applying Tris buffer containing 35mM NaCl. Chromatographic separation behaviors for the enzymatic product was similar to that for the standard solution. Separation yields of GSH from the standard solution and enzymatic product solution were 72.6% and 84.4%, respectively.

  • The relative permeabilities, distribution coefficients and diffusion coefficient of some salts which are important components in blood plasma through a poly(HEMA) membrane were measured. The crosslinker which was used for preparing the membrane was tetraethylene glycol dimethacrylate(TEGDMA), the weight percentage of the latter was about 2.8. We found that the diffusion coefficients ($D_m$) of the solutes decrease exponentially with increasing molecular weight, and also that $D_m$'s decrease linearly (except urea) with cylindrical radius (a). These facts were explained by a sieve pore flow model. The relative permeability and diffusion coefficient of urea at various temperature were larger than those of other solutes such as glycine, ${\beta}$-alanine, D-glucose, saccharose and maleic acid. The result indicates that the poly(HEMA) membrane might be suitable for hemodialysis application.

  • The caterpillar-shaped Chinese medicinal mushroom (DongChongXiaCao) looks like a worm in the winter and like a grass in the summer. The fruiting body has been regarded as popular folk or effective medicines used to treat human diseases such as asthma, bronchial and lung inflammation, and kidney disease. The fruiting bodies of Paecilomyces tenuipes that formed on the living silkworm (Bombyx mon) host were used in this examination. This study was carried out to investigate the proximate composition, soluble sugar, amino acid and fatty acid profiles, and contents of the bioactive ingredient including adenosine and D-mannitol in the fruiting-bodies. The moisture content was 57.56%. Soluble sugars found were glycerol, glucose, mannitol and sucrose, and the contents exceeded $24\;mgg^{-1}dry$ weight. Total free amino acid content was $17.09\;mgg^{-1}dry$ weight. Arginine, glycine, proline and tyrosine were main amino acids. The content of oleic acid in fatty acids was high. Adenosine was more abundant in fruiting bodies than corpus.

Production of Glutathione by yeast and Process Monitoring (효모에 의한 글루타치온의 생산과 공정 모니터링)

  • In this work the production of glutathione (GSH) by yeast Saccharomyces cerevisiae and the monitoring of the process were studied. In shaking culture the production of GSH was high at initial pH value of 4 and at temperature of 30$^{\circ}C$. But when L-cysteine was added to the culture medium at the beginning of the cultivation, the productivity of GSH was low. In case 0,5% (v/v) of L-cysteine, glycine and glutamic acid were introduced to the culture medium in the exponential cell growth phase, high concentration of GSH (about 90 mg/L) was produced in the bioreactor. A fed-batch operation with stepwise glucose feeding strategy allowed to produce 102 mg/L of GSH. The cultivation processes were on-line monitored by a 2-dimensional fluorescence sensor. A few off-line data such as cell growth, cystein concentration, phosphate concentration and GSH productivity could be well correlated to the fluorescence intensity of some combinations of excitation and emission wavelengths.

Nutritional Characteristics of Juvenile Black Rockfish Sebastes schlegeli Fed a Diet of Fig Leaf Extract (무화과 잎 추출물 첨가 사료를 급이한 조피볼락(Sebastes schlegeli)치어의 영양학적 특성)

  • The present study investigated the effect of diets supplemented with different levels (0, 1, 3, and 5 %) of fig leaf ethanol extract (FLEE) on the nutritional composition of black rockfish Sebastes schlegeli. Fish ($10.05{\pm}0.44g$) were fed to apparent satiation twice daily for 8 weeks. Adding FLEE decreased crude lipid levels and increased crude protein and ash. Among the three organic acids in the whole body of black rockfish, lactic acid was predominant, followed by citric acid and oxalic acid. Five free sugars were found in all groups. Fucose and glucose were the dominant free sugars in the FLEE-added group. The abundant fatty acids in the FLEE-added group were C16:0, C18:1-cis (n9), and C22:6n-3. The major amino acids in the samples were glutamic acid, aspartic acid, glycine, leucine, alanine, lysine, and arginine. The abundant free amino acids in the FLEE-added group were taurine, glutamic acid, alanine, leucine, and arginine.
