소당환이 Streptozotocin으로 유발된 흰쥐의 당뇨에 미치는 영향 (Antidiabetic Effect of So-Dang-Hwan in Streptozotocin-induced Diabetic Rats)

  • Objectives : So-Dang-Hwan (SDH) is used as a traditional treatment of diabetes in oriental clinics in Korea. This study aimed to evaluate antidiabetic effect of SDH in streptozotocin (STZ)-induced diabetic rats. Methods : Diabetes was induced by i.p. injection of STZ (45 mg/kg) to Sprague-Dawley rats. Experimental animals (eight per group), were treated by oral administration of SDH (60 mg/kg body weight) and glibenclimide (1 mg/kg), a known antidiabetic drug for comparison, during 5 weeks. To veridy the effect of SDH, the levels of glucose, triglyceride, insulin, BUN and creatinine were measured in sera from experimental diabetic rats, and an oral glucose tolerance test (OGTT) was also performed. Results : SDH prevented body weight loss in diabetic rats. SDH exhibited at termination, a significant reduction in blood glucose levels in STZ-induced diabetic rats. SDH significantly reduced serum creatinine levels toward the normal levels. The OGTT results showed a significant improvement in glucose tolerance in rats treated with SDH. Conclusions : These data indicate that SDH treatment may improve glocose homeostasis in STZ-induced diabetes.

The Relationship between Family History and Hypertension by Serum Glucose Levels and Age in Korean Men and Women

  • Hypertension is associated with cardiovascular disease. The environmental and genetic factors can cause the development of hypertension. In this study, the relationship between hypertension and family history of hypertension in Koreans was analyzed in consideration of serum fasting blood glucose levels and age. The study subjects were 2,484 subjects who had a medical examination at a university hospital. The main statistical analysis method was multiple logistic regression analysis. Hypertension prevalence was 16.4% of all subjects, and subjects with a family history of hypertension were 23.5%. The risk of hypertension was 2.36 times higher in subjects with a family history of hypertension than subjects without a family history of hypertension. In addition, in the subjects with fasting blood glucose levels more than 120 mg/dL, the risk of hypertension was 4.44 times higher in subjects with a family history of hypertension compared with subjects without a family history of hypertension. The relationship between family history and hypertension was slightly higher in the older group than in the younger group. To assess the association between hypertension and family history, further cohort study is necessary in the future.

Mammary alveolar cell as in vitro evaluation system for casein gene expression involved in glucose level

  • Objective: Glucose is an essential fuel in the energy metabolism and synthesis pathways of all mammalian cells. In lactating animals, glucose is the major precursor for lactose and is a substrate for the synthesis of milk proteins and fat in mammary secretory (alveolar) epithelial cells. However, clear utilization of glucose in mammary cells during lactogenesis is still unknown, due to the lack of in vitro analyzing models. Therefore, the objective of this study was to test the reliability of the mammary alveolar (MAC-T) cell as an in vitro study model for glucose metabolism and lactating system. Methods: Undifferentiated MAC-T cells were cultured in three types of Dulbecco's modified Eagle's medium with varying levels of glucose (no-glucose: 0 g/L, low-glucose: 1 g/L, and high-glucose: 4.5 g/L) for 8 d, after which differentiation to casein secretion was induced. Cell proliferation and expression levels of apoptotic genes, Insulin like growth factor-1 (IGF1) receptor, oxytocin receptor, ${\alpha}S1$, ${\alpha}S2$, and ${\beta}$ casein genes were analyzed at 1, 2, 4, and 8 d after differentiation. Results: The proliferation of MAC-T cells with high-glucose treatment was seen to be significantly higher. Expression of apoptotic genes was not affected in any group. However, expression levels of the mammary development related gene (IGF1 receptor) and lactation related gene (oxytocin receptor) were significantly higher in the low-glucose group. Expressions of ${\alpha}S1-casein$, ${\alpha}S2-casein$, and ${\beta}-casein$ were also higher in the low-glucose treated group as compared to that in the no-glucose and high-glucose groups. Conclusion: The results demonstrated that although a high-glucose environment increases cell proliferation in MAC-T cells, a low-glucose treatment to MAC-T cells induces higher expression of casein genes. Our results suggest that the MAC-T cells may be used as an in vitro model to analyze mammary cell development and lactation connected with precise biological effects.

  • Twenty five cooking recipe with yam (Dioscorea japonica Thunb) have been developed and their hypoglycemic effects on diabetic patients were investigated. The results of the sensory evaluations revealed that the tested recipe were in good scores (overall acceptability scores 3.30-4.33) except Chojuli. The 10-panel members indicated that the yam cookings were more acceptable when they were cooked with other vegetables. Twenty seven diabetic patients were entered in a general hospital during the 2-month experimental period and they were allowed to consume the diet with yam cooking dish, 75 g of yam a day. The fasting blood glucose and 2-hour postprandial blood glucose levels were determined. The diabetic patients who consumed yam showed the lower blood glucose levels than those patients who were controlled with the hospital diet only. The hypoglycemic effects of the yam were more evident in the patients who were injected with insulin. The constant lowering effects of blood glucose levels were observed in the normal subjects who had taken the yam cookery, leading to that one subject who started with a low blood glucose level must stop the yam consumption after 10 days.

갈근탕을 투여하여 호전을 보인 제 2형 당뇨병 증례 1례 (A Case Report of Patient with Type 2 Diabetes Mellitus Treated with Galgeun-tang)

  • Objectives: The aim of this study was to explore and describe how fasting blood sugar (FBS), postprandial 2 h Glucose (PP2h), and glycated hemoglobin (HbA1c) of a patient diagnosed with type 2 diabetes mellitus can be reduced by treatment with Galgeun-tang herbal medicine. Methods: The patient was administered herbal medicine to reduce serum glucose levels. The prescribed herbal medicines included Galgeun-tang and Galgeun-tang-gami. Results: The therapeutic outcomes were control of blood sugar and glycated hemoglobin (HbA1c) levels and decreased insulin administration. Conclusion: The herbal medicine, Galgeun-tang, appears to be a valid treatment for type 2 diabetes mellitus. Serum glucose (FBS/PP2hrs) and HbA1c were well controlled and insulin administration was decreased. Galgeun-tang was effective in controlling the daily glucose levels in a patient with type 2 diabetes mellitus.

트레드밀 운동이 SD계 흰 쥐의 혈당수준에 미치는 단기간의 효과 (The Short Period Effects of Treadmill Exercise on Glucose Levels in SD Rats)

  • Purpose: Hyperglycemia is associated with an risk of cardiovascular disease, mortality, diabetes mellitus and musculoskeletal disorders. The purpose of this study was to analyse the effect of two different treadmill exercise on blood levels of glucose in SD rats. Methods: The experimental groups were divided into 2 groups. The exercise was performed in the treadmill for 20minutes with 15m/min(group I, n=20) and 25m/min(group II, n=20) Blood samples were collected before exercise, 6day and 12days after treadmill exercise. Results: A significant difference was only at 12days on independent samples t-test for both groups. There were statistically significant difference between pre and 6days, 6days and 12days, pre and 12days on tests of pairwise comparisons for each groups. Both group were showed linear effects following treadmill exercise in 12days. Conclusion: These results suggest that a trend toward decrease in the levels of glucose, following treadmill exercise, were revealed in both group. Aerobic exercise related to a positive effect in control of glucose level.

Effects of Brazilin on Glucose Metabolism in Epididymal Adipocytes from Streptozotocin induced Diabetic Rats

  • Hypoglycemic mechanism of brazilin was investigated in the streptozotocin induced diabetic rats. Plasma glucose levels of diabetic rats were significantly ame]iorated by the treatment of brazilin, but there were no changes in plasma insulin levels. Brazilin increased insulin binding capacity to epididymal adipocytes, and Scatchard analysis revealed that this increase in insulin binding was not due to the increase of insulin binding sites but that of binding affinity. 2-Deoxyglucose uptake of epididymal adipocytes was significantly augmented by the intraperitoneal administration of brazilin and the same result was obtained in in vitro study. Glucose oxidation and lipogenesis in epididymal adipocytes were significantly enhanced by the treatment of bracilin.

  • Diabetes mellitus is an abnormally high glucose level in the bloodstream. Several pharmaceuticals are administered to diabetic patients to control their glucose levels. Early diagnosis and proper glycemic management are essential in this situation to prevent further progression and complications. Biosensor-based detection has progressed and shown potential in portable and inexpensive daily assessment of glucose levels because of its simplicity, low cost, and convenient operation without sophisticated instrumentation. This review discusses various systemic aspects of non-invasive glucose monitoring, including materials for monitoring and managing diabetes.

Streptozotocin으로 유발한 당뇨동물 모델에서 아프리칸 망고 추출물의 혈당 조절 효과 (Effect of African Mango (Irvingia gabonesis, IGOB 131TM) Extract on Glucose Regulation in STZ-Induced Diabetes)

  • 본 연구는 당뇨유발모델 쥐에서 아프리칸 망고(Irvingia gabonesis, IGOB $131^{TM}$) 추출물의 혈당 조절 효과를 알아보기 위해 SD계 수컷 흰쥐에 IGOB $131^{TM}$ 1%, IGOB $131^{TM}$ 2%(IGOB $131^{TM}$ 353 mg/kg body weight, IGOB $131^{TM}$ 706 mg/kg body weight)와 pinitol을 5주간 매일 강제 경구 투여하였다. 그리고 streptozotocin을 복강 주사하여 당뇨를 유발시킨 후 경구 당부하 검사(OGTT)를 실시하였다. 정상군에 비해 당뇨 유발군(DM, pinitol, IGOB $131^{TM}$ 1%, IGOB $131^{TM}$ 2%)의 체중 증가는 유의적으로 감소하였고, 당뇨 유발군 간의 유의적인 차이는 나타나지 않았다. 경구 당부하 검사 결과에서 DM군이 정상군에 비해 유의적으로 높았고, pinitol군, IGOB $131^{TM}$ 1%, IGOB $131^{TM}$ 2%군은 DM에 비해 각각 39.70%, 33.00%, 29.67% 유의적으로 감소하였다. 혈청 glucose 농도는 정상군에 비해 당뇨 유발군에서 유의적으로 높게 나타났고, pinitol, IGOB $131^{TM}$ 1%, IGOB $131^{TM}$ 2%군은 DM에 비해 유의적으로 감소하였다. 또한 혈청 insulin 농도는 정상군에 비해 당뇨 유발군에서 유의적으로 낮게 나타났고, pinitol, IGOB $131^{TM}$ 1%, IGOB $131^{TM}$ 2%군은 DM에 비해 유의적으로 증가한 것으로 나타났다. 따라서 아프리칸 망고 추출물이 STZ 유도 당뇨쥐의 혈청 insulin 함량을 개선시켜 혈청 glucose를 감소하는 효과가 있는 것으로 사료된다.

Regimen-related Mortality Risk in Patients Undergoing Peritoneal Dialysis Using Hypertonic Glucose Solution: A Retrospective Cohort Study

  • Objectives: The main purpose of this study was to quantify the risk of mortality linked to various regimens of hypertonic peritoneal dialysis (PD) solution. Methods: A retrospective cohort study of patients using home-based PD was carried out. The prescribed regimen of glucose-based PD solution for all patients, determined on the basis of their individual conditions, was extracted from their medical chart records. The primary outcome was death. The treatment regimens were categorized into 3 groups according to the type of PD solution used: original PD (1.5% glucose), shuffle PD (1.5 and 2.5% glucose), and serialized PD (2.5 and 4.5% glucose). Multivariate analysis (using the Weibull model) was applied to comprehensively examine survival probabilities related to the explanatory variable, while adjusting for other potential confounders. Results: Of 300 consecutive patients, 38% died over a median follow-up time of 30 months (interquartile range: 15-46 months). Multivariate analysis showed that a treatment regimen with continued higher-strength PD solution (serialized PD) resulted in a lower survival rate than when the conventional strength solution was used (adjusted hazard ratio, 2.6; 95% confidence interval, 1.6 to 4.6, p<0.01). Five interrelated risk factors (age, length of time on PD, hemoglobin levels, albumin levels, and oliguria) were significant predictors contributing to the outcome. Conclusions: Frequent exposure to high levels of glucose PD solution significantly contributed to a 2-fold higher rate of death, especially when hypertonic glucose was prescribed continuously.
