Effect of butanol fraction from Cassia tora L. seeds on glycemic control and insulin secretion in diabetic rats

  • Cassia tora L. seeds have previously been reported to reduce blood glucose level in human and animals with diabetes. In the present study, the effects of Cassia tora L. seed butanol fraction (CATO) were studied on postprandial glucose control and insulin secretion from the pancreas of the normal and diabetic rats. Diabetes was induced by an i.p. injection of Streptozotocin (55 mg/kg BW) into the male Sprague-Dawley rats. The postprandial glucose control was monitored during a 240 min-period using a maltose loading test. In normal rats, rats fed CATO (20 mg/l00 g BW/d) showed lower postprandial glucose levels in all the levels from 30 min up to 180 min than those in the control rats without CATO (p<0.05). In diabetic rats, those levels in the CATO group seemed to be lower during the $30{\sim}180$ min, but only glucose level at 30 min showed significant difference compared to that in the control group. Moreover, CATO delayed the peak time of the glucose rise in both normal and diabetic rats in the glucose curves. On the other hand, when CATO was administered orally to the diabetic rats for 5 days, 12 hr fasting serum glucose level was decreased in the diabetic rats (p<0.05). Degree of a decrease in 12 hr fasting serum insulin levels was significantly less in the diabetic CATO rats as compared to diabetic control rats. On the last day of feeding, P cells of the pancreas were stimulated by 200 mg/dL glucose through a 40 min-pancreas perfusion. Amounts of the insulin secreted from the pancreas during the first phase ($11{\sim}20$ min) and the second phase ($21{\sim}40$ min) in the CATO fed diabetic rats were significantly greater than those in the diabetic control group (p<0.05). These findings indicated that constituents of Cassia tora L. seeds have beneficial effect on postprandial blood glucose control which may be partially mediated by stimulated insulin secretion from the pancreas of the diabetic rats.

Antidiabetic Activities Analysis by Oral Glucose Tolerance Test in Rats

  • In this study, we conducted an oral glucose tolerance test (OGTT) so as to compare antidiabetic activities of general potatoes, purple-flesh potatoes, and potato pigments in rats at various concentration levels. After allowing the rats to abstain from food for 12 hours, 10%/20% general potato, purple-flesh potato, and potato extract was orally administered to rats at 100 and 500 mg/kg concentrations. The blood glucose level was measured after an hour. Then, immediately, 1.5 g/kg of sucrose was administered through the abdominal cavity and the blood glucose measured after 30, 60, 120, and 180 minutes. 20% purple-flesh potato group and 10% general potato group, both 100 and 500 mg/kg, showed a significant concentration-dependent decrease in blood glucose levels after 30 minutes. The 100 mg/kg potato pigment group also showed a statistically significant decrease after 30 minutes. In conclusion, administration of 10% general potato, 20% purple-flesh potato, and potato pigment can reduce blood glucose level in an OGTT using rats.

Blood glucose monitoring under the existence of other blood components by a portable type-NIR spectrometer.

  • Many interference in blood should be considered for non-invasive blood glucose level monitoring by near-infrared spectroscopy because blood glucose concentration is about 0.1% (w/v) in normal state. In this study, we investigated the influence of other blood components on blood glucose level monitoring by near-infrared spectroscopy. It carried out by newly developed portable type-NIR system (1100∼2200 nm). Spectrum features of NIR diffuse spectral data were investigated for some blood compnents powder such as hemoglobin, blood serum albumin, urea, uric acid, ascorbate, glucose, cholesterol and as adding glucose powder into other blood components powder mixture. 쌀눈기름의 급여가 KK 당뇨 마우스의 혈당 수준에 미치는 영향 (Supplementary effect of the rice germ oil on blood glucose in Diabetic KK Mice)

  • This study was carried out to investigate the supplementary effects of the rice germ oil compared with soy bean oil on blood glucose level of non-insulin dependent diabetic mice. Forty diabetic KK mice were fed two kinds of experimental diets with 20% lipid from soy bean oil as a control(CO) and rice germ oil(RG) for 8 weeks, respectively. Diet intake, body weight, organs weights and lipids levels of serum, liver and feces were measured. There was no significant difference in food and water intake, body weight gain and organs weights between experimental groups. The concentrations of fasting and random blood glucose were similar between CO and RG groups. There was no significant difference in blood glucose levels after glucose treatment during the glucose tolerance test between two groups. The levels of $HbA_{1c}$ as the index of blood glucose status, and insulin were similar in two groups. These results suggested that rice germ oil can't reduce blood glucose concentration of non-insulin dependent diabetic mice compared with soybean oil. But we need to investigate the hypoglycemic effect of rice germ oil by changing supplementary level and period.

A Comparative Study of the Concentration of Salivary and Blood Glucose in Normal and Diabetic Subjects

  • Advantage of saliva analysis are the ease of sample collection and that samples can be collected more frequently with much less stress on the patient. The objective of the present study was to comparatively evaluate the concentrations of saliva and fasting serum glucose in both normal and diabetic subjects. The mean salivary glucose level in diabetic patients was $15.66{\pm}17.1$ mg/dl and $1.78{\pm}1.72$ mg/dl (P = 0.0006) in the control group. The mean fasting serum glucose level in diabetic patients was $202.12{\pm}66.91$ mg/dl, while that in the control group was $94.21{\pm}14.97$ mg/dl (P < 0.0001). The 0.95 degree of correlation between salivary and fasting serum glucose could be demonstrated. The concentration of salivary and fasting serum glucose was not significant different betweeen the measurements for male and female. In the oral glucose tolerance test (75g), the glucose concentration in saliva progressively increased during the first 30 minutes of the test and then progressively decreased, reaching at minutes 120 ~ 180 lowest point as like fasting serum glucose concentration. We can conclude that salivary glucose concentration was significantly higher in the diabetic subjects and that there was significant correlation between salivary and fasting serum glucose concentration. Measurement of salivary glucose could be a useful test having good correlation between salivary and fasting serum glucose concentration.

글루코만난과 유청칼슘의 혼합물이 흰쥐의 혈청콜레스테롤 및 혈당에 미치는 영향 (Effect of Mixed Glucomannan and WheyCalcium on the Serum Cholesterol and Blood Glucose in Rats)

  • 이 연구는 글루코만난과 유청칼슘의 혼합물이 $1\%$-Cholesterol을 식이한 후 혈청콜레스테롤에 미치는 영향과 $70\%$-Sucrose를 식이한 후 혈당에 미치는 영향을 알아보았다. $1\%$-Cholesterol을 식이한 쥐를 일반식이군(ND) 비교하였으며, 고콜레스테롤식이군(HCD)에 글루코만난식이군(HCDG)과 글루코만난과 유청칼슘군식이군(HCDGW)으로 나누어 관찰하였다. 또한 $70\%$-Sucroe를 식이한 쥐를 일반식이군(ND) 비교하였으며, 고혈당식이군(HGD)에 글루코만 난식이군(HGDG)과 글루코만난과 유청칼슘군식이군(HCDGW)으로 나누어 관찰하였다. 이를 비교 조사한 결과를 요약하면 다음과 같다. 1. 혈청콜레스테롤수치는 HCD>HCDG>HCDGW>ND의 순서로 감소하였다. 2. HCD의 혈청콜레스테롤증가량에 비하여HCDC는 약 $23.84\%$ HCDCW는 약 $42.38\%$ 감소하였다. 3. 혈당수치는 HGD>HGDG>HGDGW>ND의 순서로 감소하였다. 4. HGD의 혈당증가량에 비하여 HGDG는 약 $6.63\%$ HGDGW는 약$36.14\%$ 감소하였다.

Effects of soybean supplementation on blood glucose, plasma lipid levels, and erythrocyte antioxidant enzyme activity in type 2 diabetes mellitus patients

  • The purpose of this study was to investigate the effect of soybean on blood glucose and lipid concentrations, and antioxidant enzyme activity in type 2 diabetes mellitus(DM) patients. We divided patients into two groups and fed them, respectively, a basal diet(control group) and a basal diet with 69 g/d of soybean(soybean group) for 4 weeks. Pills with roasted soybean powder were provided to the soybean supplementation group three times a day. Macronutrients intake except dietary fiber was similar between the two groups. No significant differences were observed in dietary intakes or body weight before and after the supplementation. Energy composition ratio of C:F:P was 65:19:16 in the control group, 64:20:16 in the soybean group. The blood parameters of subjects before supplementation, such as fasting blood glucose, postprandial glucose level, total cholesterol, triglyceride, LDL-cholesterol and $HbA_1C$ were not different between the two groups. After supplementation, fasting blood glucose(p<0.001), postprandial glucose level(p<0.001) and serum triglyceride level(p<0.05) were significantly reduced in the soybean group in comparison with the control group. The total cholesterol level was not significantly different between the control and the supplemented group after 4 weeks of treatment. TBARS levels of the soybean group were not significantly different from those of the control group. The activities of catalase(p<0.01) and glutathione peroxidase(p<0.05) were significantly higher in the soybean group compared to the control group. The results of this study suggest that soybean supplementation would be helpful to control blood glucose and serum lipid in diabetic patients. Also, soybean showed an antioxidant activity that may contribute to enhance the effect of antioxidant defense. This activity contributes to protection against oxidative damage in type 2 DM patients. Soybean may have potential use in the disease management of patients with DM.

공복혈당과 당화혈색소에 의한 당뇨병 진단 비교 (A Comparison of Fasting Glucose and HbA1c for the Diagnosis of Diabetes Mellitus Among Korean Adults)

  • Objectives: The American Diabetes Association (ADA) has recently recommended the HbA1c assay as one of four options for making the diagnosis of diabetes mellitus, with a cut-point of $\geq$ 6.5%. We compared the HbA1c assay and the fasting plasma glucose level for making the diagnosis of diabetes among Korean adults. Methods: We analyzed 8710 adults (age 45-74 years), who were not diagnosed as having diabetes mellitus, from the Namwon study population. A fasting plasma glucose level of $\geq$126 mg/dL and an A1c of $\geq$ 6.5% were used for the diagnosis of diabetes. The kappa index of agreement was calculated to measure the agreement between the diagnosis based on the fasting plasma glucose level and the HbA1c. Results: The kappa index of agreement between the fasting plasma glucose level and HbA1c was 0.50. Conclusions: The agreement between the fasting plasma glucose and HbA1c for the diagnosis of diabetes was moderate for Korean adults.

케톤식이로 발생한 체중감소 및 내당능장애에 침치료 효과 (Effects of Acupuncture Treatment on Weight Loss and Glucose Intolerance Induced by a Ketogenic Diet)

  • Objectives : This preliminary study investigated the effect of acupuncture treatment on short-term ketogenic diet-induced weight loss and glucose intolerance in mice. Methods : Six-week old male C57BL/6J mice were randomly assigned to 3 groups: normal, KD, and KD+ACU. All the mice except normal group were fed with ketogenic diet formula for 7 weeks and mice in KD+ACU group received acupuncture treatment three times a week. Body weights were measured three times a week, and glucose level was measured on week 1,3,5, and 7. Ketone level was measured on week 3,5, and 7. Results : Ketogenic diet showed short-term weight loss effect, however, acupuncture treatment did not affect on the weight loss. Ketone level was increased in KD fed mice compared to normal diet fed mice and the level was decreased in KD+ACU group on week 3. However, the change was not significantly different compared to KD group on week 7. Glucose intolerance was improved in KD+ACU group compared to KD group. Conclusions : Acupuncture treatment was effective in relieving glucose intolerance, and the results suggest that combining acupuncture treatment with ketogenic diet may complement each therapeutic intervention by improving glucose intolerance but not effecting on weight loss. This study provides meaningful evidence as a preliminary study of acupuncture treatment on ketogenic diet.

당뇨병 환자의 혈당 및 임상에 미치는 알로에의 효과 (Effects of Aloe vera Treatment on Blood Glucose Level and Clinical Chemistry in Diabetic Patients)

  • In order to study the effects of Aloe vera treatment on blood glucose level and clinical chemistry in diabetic patients, eight diabetic patients were administered orally with 800 mg of Aloe vera three time a day for three months. The high levels of blood and urine glucose in diabetic patients were significantly reduced by administration of Aloe vera. The increased plasma triglyceride concentration was also significantly reduced by Aloe vera treatment. A little amount of urine bilirubin, hematuria, nitrite, urobilinogen, protein and ketone bodies were detected before treatment, but not detected after Aloe vera treatment. But other blood parameters of clinical chemistry values were not affected by Aloe vera treatment. These data suggest that Aloe vera can be effective in the treatment of the diabetic patients.
