약초 근권토양 내 다당 생성세균 분리 및 계통학적 특성 (Isolation and Phylogenetic Characteristics of Exopolysaccharide Producing Bacteria in a Rhizosphere Soil of Medicinal Herbs)

  • 국내에 자생하는 당귀, 삽주, 쇠무릎, 지모, 황기의 근권토양내 EPS 생성균주의 분포율을 조사한 결과 당귀로부터 분리된 균주의 56%가 EPS 생성 균주로 가장 높은 분포율을 나타내었다. 또한, 당귀 근계 (근권, 근면, 근 내부) 내 EPS 생성 세균의 밀도를 측정한 결과, 근권 토양 내에는 $9.0{\times}10^6$ CFU/$g{\cdot}soil$, 근면에는 $7.0{\times}10^6$ CFU/$g{\cdot}soil$, 그리고, 근 내부에는 $1.4{\times}10^3$ CFU/$g{\cdot}soil$로 확인되어, 다수의 EPS 생성 세균이 분포하고 있음이 확인되었다. 당귀 근권으로부터 분리된 EPS 생성세균은 Alphaproteobacteria (4 strains), Betaproteobacteria (6 strains), Firmicutes (2 strains), Actinobacteria (3 strains), 그리고 Bacteroidetes (1 strain) 계통군에 속하는 균주였다. 근면으로 부터 분리된 EPS 생성세균은 Alphaproteobacteria (7 strains), Betaproteobacteria (3 strains), Actinobacteria (2 strains), Bacteroidetes (3 strains), 그리고 Acidobacteria (1 strain) 계통군으로 나타났으며, 근 내부에서 분리된 EPS 생성세균은 모두 Bacteroidetes 계통군 Chitinophaga에 속하는 특징을 나타내었다. 약초 근권토양으로부터 분리된 EPS 생성세균 112균주중에서 Burkholderia caribiensis DR14 (1,547 mpa.s), Terriglobus sp. DRP35균주(2,136 mpa.s), Rhizobium hainanense SAP110균주(1,680 mpa.s)를 최우수 EPS 생성 균주로 선발하였다. 분리 정제된 EPS를 Bio-LC로 분석한 결과 glucose, galactose, mannose의 중성당과, galactosamine, glucosamine의 아미노당이 나타났다. 특히 Rhizobium hainanense SAP110 균주는 주요 중성당으로 glucose (60-89%)를 그리고 주요 아미노당으로 glucosamine (8.5%)을 생성하는 특징을 나타내었다.

A Monoclonal Antibody That Specifically Binds Chitosan In Vitro and In Situ on Fungal Cell Walls

  • We report the generation of the first monoclonal antibody that specifically binds to the polysaccharide chitosan. Mice were immunized with a mixture of chitosans, and hybridoma clones were screened for specific binders, resulting in the isolation of a single clone secreting a chitosan-specific IgM, mAbG7. In ELISAs, the antibody could bind to chitosans of varying composition, but demonstrated the highest affinity for chitosans with lower degrees of acetylation (DA) and very poor binding to chitin. We tested the ability of the antibody to bind to chitosan in situ, using preparations of fungal cell walls. Immunofluorescence microscopy confirmed that the antibody bound strongly to the cell walls of fungi with high levels of chitosan, whereas poor staining was observed in those species with cell walls of predominantly chitin or cellulose. The potential use of this antibody for the detection of fungal contamination and the protection of plants against fungal pathogens is discussed.

키틴 막 전극의 양이온에 대한 감응 연구 (Potentiometric Response of Chitin - based Membrane Electrode to various Metal cations)

  • 이온 운반 물질로 키틴(poly-[$1{\rightarrow}4$]-${\beta}$-acetyl-D-glucosamine)을 사용하고, 지지체(matrix)로 Poly(vinyl chloride)(PVC)를, 가소제로는 Dioctyl sebacate(DOS)를 사용하여 키틴 막 전극을 제작하였다. 얇은 조각 형태의 키틴을 막자사발로 갈아 100 메시(mesh)의 체에 거른 후, 이때 모아진 일정 크기의 미세 가루를 사용하였다. 키틴 막 전극을 지시 전극으로 사용하여 금속 양이온들의 감응전위를 알아본 결과, 특히 $Cd^{2+}$$Cu^{2+}$에 대한 감응전위를 기울기(mV/decade)는 바탕 전해질이 pH 4 acetate buffer에서 각각 34.9 mV/decade, 34.0 mV/decade로 다른 금속 양이온들에 비하여 크게 나타났다. 또한 pH 영향을 조사해 본 결과, pH 2~12 범위에서 전위값이 일정하게 유지되었다.

Chitin 유도체가 타피오카의 무증자 알콜발효에 미치는 영향 (Effect of Chitin Derivatives on Non-steamed Alcohol Fermentation of Tapioca)

  • Chitin 및 그 유도체(chitosan, glucosamine)의 YPD 배지에서 알콜발효력을 비교한 결과, 대조구에 비하여 모든 첨가구간에서 알콜수율이 높게 나타나는 경향이었으며, 특히 chitin 첨가구간에서 가장 높았다. 타피오카의 알콜발효에 미치는 영향은 chitin 함량 0.9%, 발효 120시간째에 알콜함량 13.6%로 가장 높게 증가되었다 Chitin 첨가는 알콜수율을 증가시키며 acetaldehyde와 methanol 함량 감소 효과가 있는 것으로 나타나, 앞으로 타피오카의 무증자 알콜발효에 chitin의 이용이 상당히 기대된다.

Novel Cationic Microbial Polyglucosamine Biopolymer from New Enterobacter sp. BL-2 and Its Bioflocculation Efficacy

  • A new bacterium BL-2 excreting a novel cationic polyglucosamine biopolymer was isolated from the spoiled leaves of Chinese cabbage and identified as Enterobacter sp. BL-2. The isolated Enterobacter sp. BL-2 was cultivated in pH-stat fed-batch culture using acetic acid as the feeding stock at pH 8.0, resulting in 17.11 g/l of cells and 1.53 g/l of an extracellular biopolymer after 72 h. The excreted biopolymer was purified by a three-step procedure, involving ethanol precipitation and deproteinizations, to a nearly homogeneous state, and its molecular weight was found to be 106 kDa. It was composed of glucosamine, rhamnose, and galactose at a molar ratio of 86.4:1.6:1.0, respectively, indicating a rarely found novel high-glucosamine-containing biopolymer. The FT-IR and $^{13}C-NMR$ spectra of the novel cationic polyglucosamine biopolymer PGB-l revealed a close identity with chitosan from crab shell. It can effectively flocculate various suspended solids, including kaolin clay, $Ca(OH)_2,\;Al_{2}O_3$, active carbon, microbial cells, and acidic dyes.

현미를 이용한 영지버섯쌀 생산 (A Mushroom-Rice(Ganoderma lucidum) development which uses the brown rice)

  • 현미를 이용하여 영지버섯의 균사성장에 적합한 고체배양 조건실험을 실시하였다. 냉침에 의하여 현미는 최종 수분함량이 31%로 10시간만에 hydration time에 도달하였으며, 온침의 경우 40분만에 65%의 수분함량이 되어, 균사배양에 적합한 수분함량에 도달하였다. 수분함량에 따른 균사성장속도 및 균사체량(glucosamine 함량) 측정에서는 수분함량이 65%일 때 균사성장속도 및 균사체량 측면에서 가장 양호한 결과를 나타내었다. 현미에 영지버섯 균사체를 배양한 결과 현미 자체에서 검출되지 않았던 trehalose가 검출되었으며, arabinose함량이 큰 폭으로 증가하였다. 일반미에 영지쌀을 첨가할 경우는 20% 첨가가 가장 기호도가 높은것으로 나타났다.

F2 Gel Matrix - a Novel Delivery System for Immune and Gene Vaccinations

  • Exploiting the immune system to abolish cancer growth via vaccination is a promising strategy but that is limited by many clinical issues. For DNA vaccines, viral vectors as a delivery system mediate a strong immune response due to their protein structure, which could afflect the cellular uptake of the genetic vector or even induce cytotoxic immune responses against transfected cells. Recently, synthetic DNA delivery systems have been developed and recommended as much easier and simple approaches for DNA delivery compared with viral vectors. These are based on the attraction of the positively charged cationic transfection reagents to negatively charged DNA molecules, which augments the cellular DNA uptake. In fact, there are three major cellular barriers which hinder successful DNA delivery systems: low uptake across the plasma membrane; inadequate release of DNA molecules with limited stability; and lack of nuclear targeting. Recently, a polysaccharide polymer produced by microalgae has been synthesized in a form of polymeric fiber material poly-N-acetyl glucosamine (p-GlcNAc). Due its unique properties, the F2 gel matrix was suggested as an effective delivery system for immune and gene vaccinations.

Comparison of Properties of Polyclonal Anti-N-Acetylchitooligosaccharides and Anti-Chitooligosaccharides Antibodies Produced for ELISA

  • To develop the enzyme-linked immunosorbent assay (ELISA) for the analysis of N-acetylehitooligosaccharides (NACOS) and chitooligosaccharides (COS), specific antibodies (Abs) were produced, and their properties were compared. N-acetylehitohexaose (NACOS6), chitohexaose (COS6), and COS mixture (COSM) conjugated to bovine serum albumin (BSA) were used to immunize rabbits. By the use of specific Abs and NACOS6-horseradish peroxidase (HRP), COS6-HRP, and COSM-HRP conjugates, competitive direct ELISA (cdELISA) was developed. The detection limits of NACOS6 by the anti-NACOS6 Ab and COS6 by the anti-COS6 and the anti-COSM Abs in the cdELISAs were about 0.2, 2, ana 2 ng/ml (ppb), respectively. In the cdELISA, the anti-NACOS6 Ab was found to recognize NACOS3-NACOS6, but not N-acetyl-D-glucosamine (GlcNAc), NACOS2, and COSs; the anti-COS6 Ab recognized COS2-COS6 and COSM, but not glucosamine (GlcN) and NACOSs. The recognition pattern of the anti-COSM Ab was almost the same as that of the anti- COS6 Ab, except that the former recognized COS2 and COS3 slightly better than the latter.

우리나라 보건의료 발전을 위한 의료기술평가의 역할 (Roles of Health Technology Assessment for Better Health and Universal Health Coverage in Korea)

  • Health technology assessment (HTA) is defined as multidisciplinary policy analysis to look into the medical, economic, social, and ethical implications of the development, distribution, and use of health technology. Following the recent changes in the social environment, there are increasing needs to improve Korea's healthcare environment by, inter alia, assessing health technologies in an organized, timely manner in accordance with the government's strategies to ensure that citizens' medical expenses are kept at a stable level. Dedicated to HTA and research, the National Evidence-based Healthcare Collaborating Agency (NECA) analyzes and provides grounds on the clinical safety, efficacy, and economic feasibility of health technologies. HTA offers the most suitable grounds for decision making not only by healthcare professionals but also by policy makers and citizens as seen in a case in 2009 where research revealed that glucosamine lacked preventive and treatment effects for osteoarthritis and glucosamine was subsequently excluded from the National Health Insurance's benefit list to stop the insurance scheme from suffering financial losses and citizens from paying unnecessary medical expenses. For the development of HTA in Korea, the NECA will continue exerting itself to accomplish its mission of providing policy support by health technology reassessment, promoting the establishment and use of big data and HTA platforms for public interest, and developing a new value-based HTA system.
