Identification of small molecules that inhibit the histone chaperone Asf1 and its chromatin function

  • The eukaryotic genome is packed into chromatin, which is important for the genomic integrity and gene regulation. Chromatin structures are maintained through assembly and disassembly of nucleosomes catalyzed by histone chaperones. Asf1 (anti-silencing function 1) is a highly conserved histone chaperone that mediates histone transfer on/off DNA and promotes histone H3 lysine 56 acetylation at globular core domain of histone H3. To elucidate the role of Asf1 in the modulation of chromatin structure, we screened and identified small molecules that inhibit Asf1 and H3K56 acetylation without affecting other histone modifications. These pyrimidine-2,4,6-trione derivative molecules inhibited the nucleosome assembly mediated by Asf1 in vitro, and reduced the H3K56 acetylation in HeLa cells. Furthermore, production of HSV viral particles was reduced by these compounds. As Asf1 is implicated in genome integrity, cell proliferation, and cancer, current Asf1 inhibitor molecules may offer an opportunity for the therapeutic development for treatment of diseases.

Synthesis, morphology and electrochemical applications of iron oxide based nanocomposites

  • The development of hybrid systems comprising nanoparticles and polymers is an opening pathway for engineering nanocomposites exhibiting outstanding mechanical, optical, electrical, and magnetic properties. Among inorganic counterpart, iron oxide nanoparticles (IONP) exhibit high magnetization, controllable surface chemistry, spintronic properties, and biological compatibility. These characteristics enable them as a platform for biomedical applications and building blocks for bottom-up approaches, such as the layer-by-layer (LbL). In this regard, the present study is addressed to investigate IONP synthesised through co-precipitation route (average diameter around 7 nm), with either positive or negative surface charges, LbL assembled with sodium sulfonated polystyrene (PSS) or polyaniline (PANI). The surface and internal morphologies, and electrochemical properties of these nanocomposites were probed with atomic force microscopy, UV-vis and Raman spectroscopy, scanning electron microscopy, cross-sectional transmission electron microscopy, and electrochemical measurements. The nanocomposites display a globular morphology with IONP densely packed while surface dressed by polyelectrolytes. The investigation of the effect of thermal annealing (300 up to $600^{\circ}C$) on the oxidation process of IONP assembled with PSS was performed using Raman spectroscopy. Our findings showed that PSS protects IONP from oxidation/phase transformation to hematite up to $400^{\circ}C$. The electrochemical performance of nanocomposite comprising IONP and PANI were investigated in $0.5mol{\times}L^{-1}$ $Na_2SO_4$ electrolyte solution by cyclic voltammetry and chronopotentiometry. Our findings indicate this structure as promising candidate for potential application as electrodes for supercapacitors.

Brazing Characteristics of Zircaloy-4 Using Rapidly Solidified Amorphous Zr-Be Alloy Filler Metals (급속응고된 비정질 Zr-Be 합금 용가재를 이용한 Zircaloy-4의 브레이징 특성)

  • This study was conducted to investigate the brazing characteristics between Zircaloy-4 nuclear fuel cladding tubes and bearing pads with filler metals of amorphous $Zr_{1-x}Be_x$(0.3$\leq$x$\leq$0.5) binary alloy, in which they were produced in the ribbon form by the melt-spinning metod. The crystallization behavior, stability, hardness and micro-structure of brazed zone were examined by X-ray diffraction, differential scanning calorimetry, micro-Vickers hardness test, optical microscopy, and transmission electron microscopy. $Zr_{1-x}Be_x$(0.3$\leq$x$\leq$0.4) amorphous alloys were crystallized to $\alpha$-Zr with increasing the temperature, and the rest were transformed to ZrBe$_2$at higher temperatures. On the other hand, $Zr_{1-x}Be_x$(0.4$\leq$x$\leq$0.5) amorphous alloys were crystallized to $\alpha$-Zr and ZrBe$_2$, simultaneously. The thickness of the layer brazed with amorphous alloy was increased with increasing the beryllium content due to the higher diffusion of Be. The morphology of brazed layer with PVD Be filler metal showed dendrite while that brazed with amorphous alloys appeared globular. Micro-Vickers hardness of brazed zone increased as the beryllium content of filler metal was decreased.

A Hyper Suprime-Cam View of the Interacting Galaxies of the M81 Group - Structures and Stellar Populations

  • Over the last decade, deep studies of nearby galaxies have led to the discovery of vast stellar envelopes that are often rich in substructure. These components are naturally predicted in models of hierarchical galaxy assembly, and their observed properties place important constraints on the amount, nature, and history of satellite accretion. One of the most effective ways of mapping the peripheral regions of galaxies is through resolved star studies. Using wide-field cameras equipped to 8 m class telescopes, it has recently become possible to extend these studies to systems beyond the Local Group. Located at a distance of 3.6 Mpc, M81 is a prime target for wide-field mapping of its resolved stellar content. In this talk, we present the detailed results from our deep wide-field imaging survey of the M81 group with the Hyper Suprime-Cam (HSC), on the Subaru Telescope. We report on the analysis of the structures, stellar populations, and metallicities of old dwarf galaxies such as NGC3077, IKN, KDG061, as well as young stellar systems such as Arp's Loop and Holmberg IX. Several candidates for yet-undiscovered faint dwarf galaxies and young stellar clumps in the M81 group will also be introduced. The peculiar galaxy NGC3077 has been classified as the irregular galaxy. Okamoto et al. (2015, ApJ 809, L1) discovered an extended halo structure with S-shape elongated tails, obvious feature of tidal interaction. With a help of numerical simulation by Penarrubia et al. (2009, ApJ 698, 222), we will demonstrate that this tidal feature was formed during the latest close encounters between M81, M82, and NGC 3077, which induced star formation in tidally stripped gas far from the main bodies of galaxies. It is not clear whether the latest tidal interaction was the first close encounters of three galaxies. If NGC3077 is still surrounded by the dark matter halo, it implies that NGC3077 has undergone the first tidal stripping by larger companions. Kinematic studies of inter galactic globular clusters and planetary nebulae would tell us the past history of tidal interaction in this group of galaxies.

Spermiogenesis in the Korean Striped Fleid Mouse Apodcmus agrarius coreae (한국산 등줄쥐(Apodemus agrarius coreae)에서의 정자변태)

  • In order to study the process of spermiogenesis of the korean striped field mouse Apodemus agrorfus coreae, the cell differentiation of seminiferous epithelium and morphological features of mature sperm in cauda epididymis was examined and the results are as follows: Spermiogenesis was divided, according to the features of cell structure; Golgi, cap, acrosome and spermiation phases were further subdivided into two steps of early and late phases respectively, and maturation phase has only one step. Hence, the spermiogenesis consists of nine phases. in the changes of the chromatin in nucleus, the chromatin granules began to be condensed in the cap phase and the condensation proceeded to form a globular of nucleus at the acrsome phase. Finally, the chromatin regularization was completed and perfect nucleus of sperm was formed at the maturation phase. Sperm head had the falciform, and the outer dence fibers of middle piece were arranged in a horseshoe fashion. The outer dence fiber number 1, 5, 6 and 9 was larger than other fiber number 2, 3, 4, 7, 8.

Formation and evolution of sub-galactic structures around dwarf galaxy-sized halos

  • We aim to investigate formation of satellite sub-galactic structures around isolated dwarf galaxies using cosmological hydrodynamic zoom simulations. For this, we modify a cosmological hydrodynamic code, GADGET-3, in a way that includes gas cooling down to T~10K, gas heating by universal reionization when z < 8.9, UV shielding for high density regions of $n_{shield}$ > $0.014cm^{-3}$, star formation in the dense regions ($n_H$ > $100cm^{-3}$), and supernova feedback. To get good statistics, we perform three different simulations for different target galaxies of the same mass of ${\sim}10^{10}M_{sun}$. Each simulation starts in a cubic box of a side length of 1Mpc/h with 17 million particles from z = 49. The mass of dark matter (DM) and gas particle is $M_{DM}=4.1{\times}10^3M_{sun}$ and $M_{gas}=7.9{\times}10^2M_{sun}$, respectively, thus each satellite sub-galactic structure can be resolved with more than hundreds or thousands particles. We analyze total 90 sub-galactic structures that have formed outside of the main halos but infall the main halos. We found that 1) mini halos that interact more with the other mini halos tend to accrete the more mass, 2) mini halos that interact more before the reionization tend to form more stars, 3) mini halos with the more interaction tend to approach closer to the galactic center and have the lower orbital circularity, 4) survivals even in the strong tidal fields evolve baryon dominated system, such as globular clusters.

Preparation of Aminosiloxane-grafted Poly(imidesiloxane) Copolymer and its Morphology and Adhesive Properties in Film (아미노실록산이 그래프팅된 폴리(이미드실록산) 공중합체 제조와 필름 모폴로지 및 점착 특성 연구)

  • Polyimide (PI) containing carboxyl functional group was prepared and reacted with diaminosiloxane during high temperature film casting. The morphology of resulting film was observed by using transmission electron microscopy (TEM) and energy dispersive X-ray spectroscopy (EDX), which revealed that globular 100 nm-sized PI domains and continuous polysiloxane phase were formed. X-ray photoelectron spectroscopy (XPS) study indicated that air-film interface mainly consisted of polysiloxane blocks. Poly(imidesiloxane) thin layer was thermostable until $400^{\circ}C$ and its pressure- sensitive adhesive property was retained up to $300^{\circ}C$. The comparative experiments revealed that grafting between carboxyl groups of polyimide and aminosiloxane was crucial for formation of microdomain structure and pressure-sensitive adhesive property.

Effect of Arp2/3 Complex on Sperm Motility and Membrane Structure in Bovine

  • Sperm capacitation refers to polymerization of filamentous (F)-actin from globular (G)-actin. While the role of actin-related protein 2/3 (Arp2/3) complex in actin polymerization is well appreciated, the underlying mechanism(s) and its relationship with capacitation are poorly understood. Therefore, to evaluate the potential role of Arp2/3 complex on capacitation, bovine spermatozoa were incubated with multiple doses (1, 10 and $100{\mu}M$) of CK-636, an inhibitor of Arp2/3 complex with heparin. The cellular localization of the Arp2/3 complex in spermatozoa was identified by immunohistochemistry, whereas western blot was also applied to detect the protein tyrosine phosphorylation of sperm proteins. Additionally, sperm motility and kinematic parameters were evaluated using a computer-assisted sperm analysis system. CK-636 resulted in significant changes in the ratio of Arp2/3 complex localization between acrosome and equatorial region of the spermatozoa. Short-term exposure of spermatozoa to $100{\mu}M$ of CK-636 significantly decreased sperm motility, however a non-detectable effect on protein tyrosine phosphorylation was observed during capacitation. On the basis of these results, we propose that Arp2/3 complex is associated with morphological changes during capacitation and compromised sperm motility.

Morphological and Molecular Characterization of Toxocara apodemi (Nematoda: Ascarididae) from Striped Field Mice, Apodemus agrarius, in Korea

  • Adult ascarid worms from the field mice, Apodemus agrarius, were observed with a light and scanning electron microscope, and molecularly analized with 18S rRNA gene. In the scanning electron microscope, 3 prominent labia were present in the anterior end of male and female worms, but the interlabia and gubernaculum were absent. Scanning electron micrographs showed cervical alae as vestigial organs that looked like a slightly uplifted superficial sewing stitch. Total 6 pairs of post-cloacal papillae were observed on the tail of the male worms. The tail of female worms was blunt and conical shape with a spine-like structure, mucron. The eggs were sub-globular, coated with the albuminous layer and 73 by 82 ㎛ in average size. The superficial pits of T. apodemi egg (mean 8.6×6.7 ㎛) are obviously bigger than those of Toxocara spp. The partial sequence of 18S rRNA showed the sequence homology of Toxocara canis (99.6%), Toxocara cati (99.4%), Toxascaris leonina (99.4%), and Toxocara vitulorum (99.2%). Conclusively, it was confirmed that ascarid nematodes, Toxocara apodemi, recovered from striped field mice in Korea are taxonomically conspecific relationship with genus Toxocara and genetic divergence from other Toxocara species.

An Optimized Method for the Construction of a DNA Methylome from Small Quantities of Tissue or Purified DNA from Arabidopsis Embryo

  • DNA methylation is an important epigenetic mechanism affecting genome structure, gene regulation, and the silencing of transposable elements. Cell- and tissue-specific methylation patterns are critical for differentiation and development in eukaryotes. Dynamic spatiotemporal methylation data in these cells or tissues is, therefore, of great interest. However, the construction of bisulfite sequencing libraries can be challenging if the starting material is limited or the genome size is small, such as in Arabidopsis. Here, we describe detailed methods for the purification of Arabidopsis embryos at all stages, and the construction of comprehensive bisulfite libraries from small quantities of input. We constructed bisulfite libraries by releasing embryos from intact seeds, using a different approach for each developmental stage, and manually picking single-embryo with microcapillaries. From these libraries, reliable Arabidopsis methylome data were collected allowing, on average, 11-fold coverage of the genome using as few as five globular, heart, and torpedo embryos as raw input material without the need for DNA purification step. On the other hand, purified DNA from as few as eight bending torpedo embryos or a single mature embryo is sufficient for library construction when RNase A is treated before DNA extraction. This method can be broadly applied to cells from different tissues or cells from other model organisms. Methylome construction can be achieved using a minimal amount of input material using our method; thereby, it has the potential to increase our understanding of dynamic spatiotemporal methylation patterns in model organisms.
