Several enzyme immobilization methods has been compared for immobilization of pepsin. Carboxymethyl cellulose and diethylaminoethyl cellulose were activated with Hcl and with NaOH, and were used for immobilization of pepsin. Sepharose-4B was activated cyanogen bromide, and was used for immobilization of pepsin. Porous glass beads were derivatized with 3-aminopropyitrlethoxysilane and with succinicanhydride, and were used for immobilization of pepsin. The results abtained were summarized as follow, 1. 10 mg/gr. dry bead and 15mg/gr. dry bead of pepsin were absorbed to CM-cellulose and DEAE-cellulose, 20 mg/gr. dry bead and 27 mg/gr. dry bead were coupled to CM-cellulose and DEAE-cellulose with glutaraldehyde respectively. Enzyme yields were 22% and 24% of soluble pepsin. 2. 16 mg/gr. dry bead of pepsin was attached to cyanogen bromide activated sepharose-4B, 19mg/gr. dry bead was cross linked to the activated bead with glutaraldehyde. Immobilized enzyme activity was 23% of soluble pepsin. 3. 40 mg/gr. dry bead of pepsin was conjugated to the derivatized glass beads. Immobilized enzyme activity was 45% of soluble pepsin.
Despite significant effects on macroscopic migration and distribution of CO2 injected during geological sequestration, only limited information is available on wettability in microscopic scCO2-brine-mineral systems due to difficulties in pore-scale observation. In this study, a micromodel had been developed to improve our understanding of how scCO2 flooding and residual characteristics of porewater are affected by the wettability in scCO2-water-glass bead systems. The micromodel (a transparent pore structure made of glass beads and glass plates) in a pressurized chamber provided the opportunity to visualize scCO2 spreading and porewater displacement. CO2 flooding followed by fingering migration and dewatering followed by formation of residual water were observed through an imaging system. Measurement of contact angles of residual porewater in micromodels were conducted to estimate wettability in a scCO2-water-glass bead system. The measurement revealed that the brine-3M NaCl solution-is a wetting fluid and the surface of glass beads is water-wet. It is also found that the contact angle at equilibrium decreases as the pressure decreases, whereas it increases as the salinity increases. Such changes in wettability may significantly affect the patterns of scCO2 migration and porewater residence during the process of CO2 injection into a saline aquifer at high pressures.
A new exposure system was developed to generate p-dinitrobenzene (p-DNB) containing atmosphere. A glass column was filled with small glass beads coated with the chemical. The p-DNB containing medium was heated to a temperature beyond the boiling point of p-DNB. A stream of air flow was forced to pass through the column and let it mixed with fresh air before introducing into an inhalation chamber. The concentration of p-DNB in the chamber air was measured by direct assaying the air directly and by sampling the chemical using a microfilter installed in the chamber.

Modified product Consistency Test (M-PCT) has been developed as an alternative to other existing methods in determining the leachability of glass. M-PCT, the leaching method, is a hybrid of MCC-l and PCT, but can provide quicker sample preparation. Larger diameter glass sample (1.0-2.0 mm) than in the PCT method can be used so that the glass beads are more easily produced and cleaned. From the M-PCT, the total mass loss (ML) of glass, the normalized elemental release rate (NLi), pH value of leachate have been obtained. For some selected glasses in which leaching rates have been known, their chemical durablility have been tested using the M-PCT method. The results are compared to the literature data for the glasses. It is found that M-PCT method is reasonable and suitable in determining the leachability of Low and Intermediate level Radioactive Waste glass form, such as the pH, elemental loss and total mass loss.

In the automotive industry, such as scrap metal and plastic scrap process is being recycled. Although the glass beads are used as road paving or other additives and processing crushing, recycling is known that there are limits. The utilization of waste glass was evaluated as a concrete admixture by using powder characteristics and chemical composition of the glass. As a result of using waste-glass powder as an admixture, it is difficult to expect the pozzolanic effect, but it is found that it can increase the fluidity of concrete and ensure the durability performance in the appropriate amount range.

In this study, a series of experiments was conducted on the relative degradation of commonly known endocrine-disrupting compounds such as bisphenol A (BPA) and $17{\alpha}$-ethinyl estradiol (EE2) in a single-component aqueous solution using 28 and 580 kHz ultrasonic reactors. The experiments were conducted with three different types of model water: deionized water (DI), synthetic brackish water (SBW), and synthetic seawater (SSW) at pH 4, 7.5, and 11 in the presence of inert glass beads and humic acids. Significantly higher sonochemical degradation (93-97% for BPA) occurred at 580 kHz than at 28 kHz (43-61% for BPA), regardless of water type. A slightly higher degradation was observed for EE2 compared to that of BPA. The degradation rate of BPA and EE2 in DI water, SBW, and SSW after 30 min of ultrasound irradiation at 580 kHz increased slightly with the increase in pH from 4 (0.073-0.091 $min^{-1}$ for BPA and 0.081-0.094 $min^{-1}$ for EE2) to 7.5 (0.087-0.114 $min^{-1}$ for BPA and 0.092-0.124 $min^{-1}$ for EE2). In contrast, significant degradation was observed at pH 11 (0.149-0.221 $min^{-1}$ for BPA and 0.147-0.228 $min^{-1}$ for EE2). For the given frequencies of 28 and 580 kHz, the degradation rate increased in the presence of glass beads (0.1 mm and 25 g) for both BPA and EE2: 0.018-0.107 $min^{-1}$ without beads and 0.052-0.142 $min^{-1}$ with beads for BPA; 0.021-0.111 $min^{-1}$ without beads and 0.054-0.136 $min^{-1}$ with beads for EE2. A slight increase in degradation of both BPA and EE2 was found as the concentration of dissolved organic carbon (DOC, humic acids) increased in both SBW and SSW: 0.107-0.115 $min^{-1}$ in SBW and 0.087-0.101 $min^{-1}$ in SSW for BPA; 0.111-0.111 $min^{-1}$ in SWB and 0.092-0.105 $min^{-1}$ in SSW for EE2. After 30 min of sonicating the humic acid solution, DOC removal varied depending on the water type: 27% (3 mg $L^{-1}$) and 7% (10 mg $L^{-1}$) in SBW and 7% (3 mg $L^{-1}$) and 4% (10 mg $L^{-1}$) in SSW.

Carbohydrate-free caseinomacropeptide (CMP) was isolated from the sweet whey powder by a precipitation method using 12% trichloroacetic acid. The yield of carbohydrate-free CMP was 2.7 g from 100 g sweet whey powder. The electrophoretic pattern and the amino acid analysis of CMP showed that isolated CMP was quite pure, indicating the precipitation with 12% trichloroacetic acid was very effective for isolating carbohydrate-free CMP from the sweet whey powder. Trypsin, covalently immobilized on pore glass beads by carbodiimide (EDC) method, was 20mg per 1g glass beads. CMP was almost completely hydrolyzed by soluble trypsin in 24hr, but not by immobilized trypsin. The tryptic hydrolysates were fractionated on a Bio-Gel P 4 column $(1.5{\times}120\;cm)$and separated peptides were tested for their capacities to inhibit platelet aggregation using a aggregometer. The hydrolysate obtained from CMP after 24hr digestion by immobilized trypsin showed the highest activity.
