Effect of New Remedies Mainly Comprised of Hovenia dulcis Thunb on Alcohol Degradation and Liver Protection in Sprague Dawley Male Rats (헛개나무 열매를 주성분으로 제조한 새로운 처방이 알코올 분해 및 간 손상에 미치는 영향)

  • We investigated whether two-kinds of mixture (NHDT-1 and NHDT-2) mainly composed of Hovenia dulcis Thunb had beneficial actions for alcohol and acetaldehyde degradation in acute alcohol treatment and liver protection from fatty liver induced by chronic alcohol administration. In acute alcohol degradation experiment, serum alcohol and acetaldehyde concentrations exhibited lower 1, 3 and 5 hours after taking 3 g ethanol per kg body weight in NHDT-1 treated rats, but not NHDT-2 including ginseng. On the contrast to the acute effect on alcohol degradation, the long-term alcohol administration revealed that NHDT-2, not NHDT-1, protected the increase in serum concentration of aspartate aminotransferase, alanine aminotrasferase and ${\gamma}-triglyceride$ metabolism similar to the rats not consuming alcohol, leading to decreased triglyceride accumulation in blood and liver. In liver morphological study, NHDT-1 preserved the regular hepatocyte morphology, decreased fat accumulation and reduced sinusoidal leukocyte infiltration in hepatocytes. In conclusion, NHDT-1 plays an important role in alcohol and acetaldehyde degradation without protecting liver damage while NHDT-2 works as hepatocyte protector from alcohol mediated damage.

Effects of Red-ginseng Extracts on the Activation of Dendritic Cells (고려홍삼의 수지상세포 활성화 효과)

  • Ginseng is a medicinal herb widely used in Asian countries. Dendritic cells(DCs) play a pivotal role in the initiation of T cell-mediated immune responses, making them an attractive cellular adjuvant for use in cancer vaccines. In this study, we examined the effects of Red-ginseng(water extract, edible and fermented ethyl alcohol extract, crude saponin) on the DCs phenotypic and functional maturation. Immature DCs were cultured in the presence of GM-CSF and IL-4, and the generated immature DCs were stimulated by water extract, edible and fermented ethyl alcohol extract, crude saponin and LPS, respectively, for 24hours. The expression of surface co-stimulatory molecules, including MHC(major histocompatibility complex) class II, CD40, CD80 and CD86, was increased on DCs that were stimulated with crude saponin, but antigen-uptake capacity was decreased. The antigen-presenting capacity of Red-ginseng extracts-treated DCs as analyzed by allogeneic T cells proliferation and IL-2, $IFN-{\gamma}$ production was increased. Furthermore, $CD4^+$ and $CD8^+$ syngeneic T cell(OVA-specific) proliferation and $IFN-{\gamma}$ production was significantly increased. However, $CD4^+$ syngeneic T cell secreted higher levels of IL-2 in responding but not $CD8^+$ syngeneic T cell. These results indicate the immunomodulatory properties of Red-ginseng extracts, which might be therapeutically useful in the control of cancers and immunodeficient diseases through the up-regulation of DCs maturation.

  • We determined the structure of main saponin which extracted from the shoot of Aralia Elata. The results were as follows. 1. The main aglycons and suger of the total saponins of Nr2 sample were identified as oleanolic acid and hederagenin, and glucose, arabinose and rhamnose. A probable new aglycon was isolated and inferred as 1, 3-methylenedioxy-3-dehydroxyoleanolic acid. 2. One compound of Fh saponin (named as Elatoside $Fh_2$) which was obtained first in this species was elucidated as 3-O-$({\alpha}-L-arabinopyranosyl(1{\rightarrow}2)-{\beta}-D-gluco-pyranosyl)$-28-O-${\beta}-D-glucophyranosyl$ oleanolic acid on the basis of chemical and spectral evidence of IR, $^1H$, $^{13}C-NMR$ and MS.

Intake of Traditional Beverages in Female University Students (여대생들의 전통음료 섭취 실태에 관한 연구)

  • This study was conducted to investigate the actual conditions of intake of Korean traditional beverages in 273 female university students in Seoul, Korea. The subjects of this study consisted of 128 food and nutrition majors and 145 non-majors. Questionnaires were administered to the subjects who had an average age of 20 years. Two-hundred and nineteen students (80.2%) lived with their parents. Regarding the type of beverages subjects often drink, coffee was the most preferred (51.3%) overall, and non-majors (4.1%) were shown to drink traditional beverages more often. Regarding preference for traditional beverages, most subjects (72.5%) responded that they liked their own traditional beverage. However, for frequency of intake of traditional beverages, 48.7% answered they do not drink any traditional beverages. The reasons why subjects like traditional beverages were taste (71.7%) and health (26.3%), and the type of traditional beverages they often drink were Yulmu tea (35.9%), Sik hae (30.8%), and Yuja tea (10.6%), in order. Regardless of major, ginseng tea, jujube tea, mulberry-leaf tea, Chinese matrimony vine tea, ssanghwa tea, ginger tea, and omija tea were found to be satisfactory traditional beverages. Therefore, due to the low intake frequency, preference, and satisfaction of traditional beverages, new traditional beverages that can meet the standards of female students should be immediately developed.

Antioxidant and Nicotine Degradation Effects of Medicinal Herbs

  • Antioxidant activity and nicotine degradation activity (NDA) of 21 medicinal herbs were determined by using a 1,1-diphenol-2-picrylhydrazyl (DPPH) method and a PLC/PRF5 human liver cell line method, respectively, to develop an anti-smoking aid. The highest and lowest antioxidant activities represented by $IC_{50}$/ value were 30 $\mu$g/mL of Eugenia caryophyllus and 3,270 $\mu$g/mL of Panax ginseng C. A. Meyer, respectively. Antioxidant activity of Eugenia caryophyllus was equal to 38.0$\pm$1.2 mg VCEAC(vitamin C equivalent antioxidant capacity)/ g herb. The highest and lowest NDA values were 1.81 of Astrgalus membranaceus Bunge and 1.01 of Raphani seed and Lespedeza tomentosa Sieb, respectively. Eleven medicinal herbs with high antioxidant activity and/or NDA were selected to make an herbal tea. The herbal tea had high antioxidant activity (50 $\mu$g/mL $IC_{50}$/ and 22.4$\pm$1.4 mgVCEAC/g) and NDA (1.243). The medicinal herb tea could help smokers quitting smoking by degrading and exhausting nicotine accumulated in body and removing reactive oxygen species.

Oxidation of fatty acid may be enhanced by a combination of pomegranate fruit phytochemicals and acetic acid in HepG2 cells

  • We investigated whether the combination of phytochemicals and acetic acid in the form of fruit vinegar provides an additive effect on changes of mRNA levels related to fatty acid oxidation in human hepatocyte (HepG2). Among the seven fruit vinegars (Rubuscoreanus, Opuntia, blueberry, cherry, red ginseng, mulberry, and pomegranate) studied, treatment of HepG2 with pomegranate vinegar (PV) at concentrations containing 1 mM acetic acid showed the highest in vitro potentiating effect on the mRNA expression levels of peroxisome proliferator-activated receptor ${\alpha}$, carnitinepalmitoyl transferase-1, and acyl-CoA oxidase compared to the control group (P < 0.05). Reversed-phase liquid chromatography in combination with quadrupole time-of-flight mass spectrometry analysis revealed four potential compounds (punicalagin B, ellagic acid, and two unidentified compounds) responsible for altered gene expression in HepG2 cells treated with PV as compared with the others. Further investigations are warranted to determine if drinking PV beverages may help to maintain a healthy body weight in overweight subjects.

Effect of mucilage from yam on activation of lymphocytic immune cells

  • The immunostimulating activities of mucilage fraction from yam were investigated. The proliferation of BSA-primed lymph node cells was enhanced between 4.1- to 10.9-fold compare to control, when cultured with 1 to $25{\mu}g/mL$ of yam-mucilage fraction. It showed strong immunostimulating activity than ginseng extract and as remarkable as Bifidobacterium adolescentis M101-4 known as a positive immunostimulator. Mitogenicity to lymph node cells was fully induced by concanavalin A and lipopolysaccharide. The proliferation of splenocytes and Peyer's patch cells was enhanced between 5.0- to 14.1-fold and 2.4- to 6.4-fold, respectively, when cultured with 1 to $25{\mu}g/mL$ of yam-mucilage fraction. It enhanced the production of cytokines such as tumor necrosis $factor-{\alpha}$ and IL-6 in the culture of RAW 264.7 macrophage cells. In the culture of lipopolysaccharide-stimulated RAW 264.7 cells, production of cytokines was as similar as compared to controls. In unstimulated RAW 264.7 cells, both tumor necrosis $factor-{\alpha}$ and IL-6 production were enhanced between 15.6- to 60.1-fold and 2.3- to 9.1-fold, respectively. Mucilage fraction from yam is expected to be a safe immunopotentiator to maintain the host immunity and develop a physiologically functional food.

Manufacture and Quality Characteristics of Korean Traditional Gugija(Lycii fructus) Tagju (전통 구기자 탁주의 제조 및 품질 특성)

  • To develop a new traditional Gugija tagju, various koji were prepared using several starch sources along with Aspergillus awamori var. kawachi, and then their effects on the quality of the tagju were investigated. Ethanol contents(11.1${\sim}$13.8%) were not significantly different among the Gugija tagju made from the various types of koji, whereas the tagju made using non-glutinous rice koji had the best total acceptability. The addition of 1% Gugija extracts into the mash increased its total acceptability and anti-hypertensive angiotensin I-converting inhibitory activity, and the optimal fermentation period for brewing the Gugija tagju was 10 days.

  • This study aims to substitute the emulsifier and gelifier for the poly phosphates with regard to diminishing the foreign phosphates in diet and production cost. It is desirable to introduce the application of theory for melting phenomenon in this investigation since the rate of peptisation is very representative index during the conversion of Ca-paracaseinate to Na-paracaseinate in processed cheese. It will be impossible to substitute completely for polyphosphates but partial substitution could be possible if one consider the organoleptic quality of typical processed cheese.

Study on the Amounts of Kitchen Wastes and Method for Reduction of Its Wastes Disposed in Korean Style Restaurants (전국 한식제공 음식업소에서의 음식물 쓰레기량 실태 파악 및 감소방안에 관한 연구)

  • This research was conducted to survey amounts of kitchen wastes disposed and to find method for reduction of the kitchen wastes. Questionnaires were developed. Survey was conducted in Korean style restaurants 1,292 in nationwide and recovery rate of questionnaires is 97.52%. Amounts of kitchen wastes disposed were calculated using programme developed in this research institute. As a results, wastes of food disposed in prepreparation phase large quantities is the vegetables. Wastes of fruits, fishes and shellfishes were disposed in large quantities, too. Amounts of wastes of boiled chicken with ginseng, Jeon-Gol, and rosted beef ribs in dishes were much due to refuse portion as bone. Wastes of kitchen are composed of food wastes 44%, dishes wastes 56% in nationwide in Korean style restaurants. Amounts of kitchen wastes disposed in Korean style restaurants are 28.33 kg in large city, 24 kg in small city and rural, respectively. Methods for the effective reduction of kitchen wastes in restaurants were suggested. Also, it is possible to utilize the kitchen waste as fertilizer or feed for animals but economic feasibility could cause problems.
