냉습적(冷濕積)에 따른 은행나무종자내(種子內) 생장조정물질(生長調整物質)의 변화(變化) (Alteration of Endogenous Growth Substances in Cold-moist Stratified Seeds of Ginkgo biloba L.)

  • 휴면기간(休眠期間)이 긴 은행나무 종자(種子)의 냉습적처리(冷濕積處理)에 따른 생장촉진물질(生長促進物質)과 생장억제물질(生長抑制物質)의 증감(增減)을 구명(究明)하기 위하여 본(本) 연구(硏究)를 수행(遂行)하고 얻어진 결과(結果)를 요약(要約)하면 다음과 같다. 1. 무처리종자(無處理種子)는 배(胚), 배유(胚乳), 종피(種皮)의 각추출물(各抽出物)에서 생장억제물질(生長抑制物質)을 볼 수 있었으며, 특(特)히 종피(種皮)에서 가장 현저(顯著)하게 나타났다. 2. 냉습적처리종자(冷濕積處理種子)의 배(胚)와 종피(種皮)에서 생장억제물질(生長抑制物質)의 현저(顯著)한 감소(減少)가 있었으나, 생장촉진물질(生長促進物質)은 배부분(胚部分)에서 증가(增加)가 약간 나타났다. 3. 이상(以上)의 결과(結果)에서 은행나무 종자(種子)의 휴면(休眠)에는 종피(種皮), 배(胚), 배유(胚乳)의 각부분(各部分)이 생장억제물질(生長抑制物質)이 존재(存在)하며 발아(發芽)에 관계하는 것으로 추측(推測)된다. 4. 은행나무 종자(種子)와 발아촉진(發芽促進)과의 관계는 생장촉진물질(生長促進物質)의 증가(增加)보다는, 배(胚)와 종피(種皮)의 생장억제물질(生長抑制物質)의 감소(減少)에 영향된다고 추측(推測)된다. 5. 생장촉진물질(生長促進物質)인 Gibberellin은 자외선형광검사등검정(紫外線螢光檢査燈檢定)에서 Rf 0.26, 에서 나타났고, 생장억제물질(生長抑制物質)인 Abscisic acid는 Rf 0.62, Rf 0.70, Rf 0.78에서 각각(各各) purple, gray, blue의 형광색(螢光色)으로 나타났다.

은행종실유의 all $cis-{\Delta}^{5,11,14}-C_{20:3}$ 지방산 존재에 관한 연구 (Studies on the Presence of all $cis-{\Delta}^{5,11,14}-C_{20:3}$ Fatty Acid in the Seed Oils of Ginkgo)

  • The fatty acid, all $cis-{\Delta}^{5,11,14}-C_{20:3}$, in the Gingko nuts oils, was isolated and, purified by urea-adduct method, silver ion silica gel chromatography and HPLC equipped with reversed phase ${\mu}-Bondapak$ $C_{18}$ column. Its structural elucidation was conducted by IR and $^1H$-, $^{13}C$-NMR technique. The fatty acid composition of seed oils mainly consists of linoleic acid(37.73%), vaccenic acid(18.30%), oleic acid(15.18%), palmitic acid(3.37%), palmitoleic acid(3.37%) and ${\Delta}^5$ NMDB fatty acids(8.50%) in which all $cis-{\Delta}^{5,11,14}-C_{20:2}$ predominates.

은행 외종피로부터 살선충 물질의 순수 분리와 활성 (Purification Nematicidal Substance and Nematicidal Activity from Ginkgo biloba L. Outer Seedcoat)

  • 은행 종자와 잎은 많은 연구와 개발이 이루어졌으나, 외종피는 알러지 등의 부작용으로 사용을 기피해왔다. 본 연구에서는 버려지는 은행 외종피로부터 작물재배에 많은 피해를 주는 선충을 방제할 수 있는 천연물을 찾고자 했다. 은행 외종피 유기용매추출물의 H2O 분획물과 EtOAc 분획물에서 살선충 활성을 확인하였고, EtOAc 분획물에서 살선충 활성이 가장 높게 나타났으며, EtOAc 분획물을 0.1 mg/mL 농도 처리에서 74%의 살선충 활성이 나타났다. 4차례의 TLC를 실시하여 살선충 물질을 순수 분리하였으며, prep-TLC 에서 Rf 0.5에 single spot을 확인하였고, HPLC 분석에서 Rt 8.609에서 single peak를 확인하였다. 순수 분리한 물질을 GB4-3으로 명명하고, 살선충 활성을 조추출물과 농도별 비교하였다. 은행 외종피 조추출물 10 ㎍/mL 처리 후 18시간 후에 약 45%의 살선충 활성을 나타냈고, 20 ㎍/mL 처리한 곳에서는 55%의 활성을 보였다. 은행 외종피 추출물로부터 순수 분리한 물질 GB4-3을 20 ㎍/mL 처리한 곳에서는 18시간 후에 약 69%의 살선충 활성을 나타나 살선충제 개발 후 보소재로서 가치가 있는 것으로 판단된다. 부식선충에 대한 살선충 활성과 식물기생 선충에 대한 살선충 활성의 유의성은 선행 보고에서 볼 수 있으나, 은행 외종피를 이용한 선충방제제 개발을 위해서는 식물기생성 선충류에 대한 살선충 활성을 조사할 필요가 있다. 농산물재배와 식물생육에 심각한 피해를 주는 선충 방제를 위한 친환경적 방제제의 개발은 절실히 필요하다. 이를 위해서는 식물이나 미생물 등의 생물소재를 이용하거나 천연소재의 부산물을 이용 또는 이들을 혼용한 혼합제제로 개발한다면 상승효과를 기대할 수 있을 것으로 판단한다.

태음인(太陰人) 마황정천탕(麻黃定喘湯)의 기원, 변천과정 및 구성 원리 (Reconsideration on the Origins, Changes, and Constructive Principle of Mahwangjeongchun-tang)

  • Objectives This paper was aimed to review the preceding research on the origins, changes and constructive principle of Mahwangjeongchun-tang. Method Based on Jema Lee's works such as Dongyisusebowon and Dongmuyoogo, we analyzed contents related to Mahwangjeongchun-tang and its modification. Moreover, we reviewed physiological and pathological pecularities of Taeeumin symptomatology and applied the theory to induce the principle of the modified Mahwangjeongchun-tang in Sasang medicine. Results and Conclusion 1) The origins of Mahwangjeongchun-tang might be Mahwang-tang, Jungchun-tang and Saengmaek-san. 2) Mahwangjeongchun-tang had been changed in composition when it moved from Manbyonghoichun to Dongyisusebowon. 3) Mahwangjeongchun-tang consists of a) Platycodi Radix (桔梗), Liriopis Tuber(麥門冬) and Scutellariae Radix(黃芩) which directly or indirectly enhance lung function, b) Ephedra Sinica(麻黃) which affects circulation of lung companion, c) Prunus Armeniaca(杏仁) and Ginkgo Biloba(白果) which affect wiwan's upward circulatory force, d) Radish Seed (蘿葍子) which affects small intestine's digesting effect, e) Mori Cortex Radicis(桑白皮) and Farfarae Flos(款冬花) which affect cough and asthma.

Effect of Phytogenic Feed Additives in Soybean Meal on In vitro Swine Fermentation for Odor Reduction and Bacterial Community Comparison

  • The effect of different phytogenic feed additives on reducing odorous compounds in swine was investigated using in vitro fermentation and analyzed their microbial communities. Soybean meal (1%) added with 0.1% different phytogenic feed additives (FA) were in vitro fermented using swine fecal slurries and anaerobically incubated for 12 and 24 h. The phytogenic FAs used were red ginseng barn powder (Panax ginseng C. A. Meyer, FA1), persimmon leaf powder (Diospyros virginiana L., FA2), ginkgo leaf powder (Ginkgo biloba L., FA3), and oregano lippia seed oil extract (Lippia graveolens Kunth, OL, FA4). Total gas production, pH, ammonianitrogen ($NH_3$-N), hydrogen sulfide ($H_2S$), nitrite-nitrogen ($NO_2{^-}$-N), nitrate-nitrogen ($NO_3{^-}$-N), sulfate (${SO_4}^{--}$), volatile fatty acids (VFA) and other metabolites concentration were determined. Microbial communities were also analyzed using 16S rRNA DGGE. Results showed that the pH values on all treatments increased as incubation time became longer except for FA4 where it decreased. Moreover, FA4 incubated for 12 and 24 h was not detected in $NH_3$-N and $H_2S$. Addition of FAs decreased (p<0.05) propionate production but increased (p<0.05) the total VFA production. Ten 16S rRNA DGGE bands were identified which ranged from 96 to 100% identity which were mostly isolated from the intestine. Similarity index showed three clearly different clusters: I (FA2 and FA3), II (Con and FA1), and III (FA4). Dominant bands which were identified closest to Eubacterium limosum (ATCC 8486T), Uncultured bacterium clone PF6641 and Streptococcus lutetiensis (CIP 106849T) were present only in the FA4 treatment group and were not found in other groups. FA4 had a different bacterial diversity compared to control and other treatments and thus explains having lowest odorous compounds. Addition of FA4 to an enriched protein feed source for growing swine may effectively reduce odorous compounds which are typically associated with swine production.

국내(國內) 6개(個) 은행(銀杏)나무 식재지(植栽地)에 있어서 I-SSR 변이체(變異體)의 다양성(多樣性) (Diversity of I-SSR Variants in Gingko biloba L. Planted in 6 Regions of Korea)

  • 6개지역에 식재된 은행나무 182개체의 잎으로부터 DNA를 추출하여 I-SSR과 RAPD 표지자를 분석하였다. 15개 I-SSR primer를 사용하여 PCR을 수행해서 227개의 증폭산물 변이체를 확인하였고, 5개 RAPD primer를 사용하여 67개의 증폭산물 변이체를 확인했다. 6개 집단에 있어서 집단내의 유전적 다양성 정도는 유사한 것으로 나타났다(Shannon's Index, I-SSR : 0.35~0.40; 평균 0.38, RAPD : 0.31~0.38; 평균 0.35, 통합 : 0.35~0.40; 평균 0.37). 3개의 자료(I-SSR 표지자, RAPD 표지자 및 통합 자료)로부터 평가된 유전적 다양성 정도의 등급은 상호간에 일치하지 않았다. 유전적 다양성의 대부분이 집단내 개체간에 존재하는 것으로 나타났으며(I-SSR : 94.31%, RAPD : 93.62%, 통합 : 93.57%), 결과로써 집단간 분화정도는 상당히 낮게 나타났다. 수나무와 암나무 그룹간의 유전적 분화정도는 매우 낮았으며(I-SSR : 0.03, RAPD : 0.091, 통합 : 0.043), 수나무와 암나무들 동시에 채집한 3개 지역만을 대상을 분석했을 경우에는 약간의 변동이 있었다(I-SSR : 0.038, RAPD : 0.084, 통합 : 0.047). UPGMA법을 이용해서 분석한 집단간의 유전적 유연관계는 지리적 거리와 일치하지 않았는데, 이는 몇몇 지역들이 종자 공급원을 공유하고 있기 때문일 것으로 추정된다. I-SSR과 RAPD data를 성별로 재편성해서 유집분석을 수행한 결과, 모든 수나무 집단들과 암나무 집단들이 각각 별개의 성별 분지군을 형성하는 양상을 보였으나, 2개의 data를 통합해서 유집 분석을 수행했을 경우에는 성에 따른 명백한 분지군이 형성되지 않았다.

유기재배 단감 과수원의 병해충 종합관리기술 (Comprehensive Pest Management Techniques of Organic Cultivation Sweet Persimmon Orchards)

  • 본 연구는 국내 단감 유기재배 선도농가의 병해충 방제 실태조사, 주요 병해충에 대한 친환경 방제자재 선발, 수립한 방제시스템의 현장실증시험을 2013년부터 2015년까지 3년 동안 수행하였다. 유기재배 선도농가의 주 재배품종은 "부유"였으며, 병해충 방제를 연간 9~17회 유기농업 자재를 살포하였다. 피해가 가장 심한 병해충은 탄저병, 노린재, 감꼭지나방, 깍지벌레 등이었으며, 사용한 방제자재는 결정석회황합제, 황토유황, 유화제, 보르도액 그리고 마늘, 은행, 소나무를 이용한 자가제조 식물추출물제를 이용하였다. 탄저병 방제에 효율적인 자재는 결정석회황, 황토유황이었고, 마늘유+ 시트로넬라유제, 고삼종자 추출물+차추출물제가톱다리개미허리노린재와 독나방에 효과적이었다. 감꼭지나방 교미교란트랩을 60개/10a 설치하면 감 착과율을 30% 향상시켰다. 유기재배 단감에 연간 10회 방제하는 방제시스템의 현장실증시험 결과 70.7%의 과일을 수확할 수 있었다. 그러나, 수확기 즈음인 9월, 10월에는 약제를 살포할 수 없기 때문에 이때 가해하는 노린재류의 피해는 감소하지 않았으며, 추가적인 연구가 필요하다.

장수상황버섯 균사체를 이용한 고체 발효한약재의 대장암 세포성장 억제 활성 (Anti-Proliferative Activities of Solid-State Fermented Medicinal Herbs Using Phellinus baumii against Human Colorectal HCT116 Cell)

  • 본 연구에서는, 장수상황버섯(Phellinus baumii) 균사체를 이용한 한약재 고체발효 추출물의 암세포 성장억제 효과를 확인하고자 36종의 한약재 및 이들의 고체발효 한약재 추출물을 대상으로, 인간 대장암세포(HCT116) 성장억제능, 정상세포(HEK-293, 3T3-L1 및MC3T3-E1)에 대한 세포독성 및 인간 적혈구 용혈활성을 평가하였다. 선별된 36종의 한약재 추출물은 100 ${\mu}g/ml$ 농도에서, 사인, 천궁, 석곡, 백선피, 시체, 두충, 백과, 신이, 와송, 삼칠, 견우자, 원지 및 괄루인의 13종에서 암세포 성장률이 50% 이하를 나타내었으며, 균사체 추출물은 46.3%의 성장률을 나타내었다. 반면 25종의 발효한약재 추출물은 대부분 암세포 성장억제능의 변화가 미미하였으나, 백선피, 백과, 신이, 황부자, 산약, 현삼 및 괄루인은 암세포 증식억제능이 유의적으로 감소하였으며, 도인, 골쇄보, 구기자 및 패모에서는 암세포 성장억제능이 유의적으로 증가하였다. 특히 골쇄보, 구기자 및 패모 추출물의 대장암세포에 대한 $IC_{50}$는 각각 394, 917 및 149 ${\mu}g/ml$이었으나, 발효 후 각각 28, 85 및 80 ${\mu}g/ml$를 나타내었다. 또한 발효 골쇄보, 구기자 및 패모 추출물은 200 ${\mu}g/ml$ 농도까지 정상세포에서 미미한 세포독성을 나타내었다. 이러한 결과는 장수상황버섯 균사체를 이용한 한약재 고체발효가 새로운 항암 활성물질 개발 및 신규의 생리활성물질 생산에 이용될 수 있음을 제시하고 있다.

이비인후, 안, 치아 및 피부증상의 민간요법에 관한 고찰 (Study on the Herb Remedies of ENT, Eyes, Teeth and Skin Problems)

  • The intention of this study is to apprehend the con. tents and methods of herb remedies that are commonly conducted when there are health-problem cases of ENT, eyes, teeth and skin. Methods of this study are divided into two stages : 1) For a period of six months from December 1994 to June 1995, some 40 persons who are believed to be well versed with herb remedies have been randomly chosen, and we made a survey on herb remedies by symptoms: and 2) we have endeavored to make their grounds evident through the studies on literatures with the focus on the basic data collected. Their results are as follows: 1) When one feels a pain in ears, such herb remedies are employed as pouring the vapor into ears, which is made by steaming Alaska pollack, or as applying or wiping with the juices of radish or the ginkgo, or' alum. Applying the radish juice is effective for sterilization and fever removal: and applying the ginkgo juice is effective for cleanliness. But, plastering alum, sesame oil or castor oil, or pouring the vapor of Alaska pollack into ears are perhaps effective but do not have any pharmacologic grounds. 2) When one bleeds at the nose, such kinds of herb remedies are applied as stimulating nose or head with cold water, pressing nose or ridge of nose, or filling up nares with mashed mugworts. In addition, they have utilized garlic or leeks. Such methods as stimulating with cold water or just pressing nose and ridge of nose is based on reasonable grounds, i.e. vasoconstriction and vascular compression ; and applying mashed garlic on the sole of foot is good for the circulation of Qui ; and the use of mugworts and leeks is based upon the pharmacological function of hemostasis. 3) When one feels a sore throat such kinds of herb remedies are employed as gargling or rinsing throat with brine, drinking hot gruel or water, or drinking the juice of mugwort, radish, ginger or Chinese quince. Gargling with brine or drinking the juice of mugwort, radish or ginger is based upon the pharmacological function of pain alleviation, fever removal, and detoxication. 4) When a boil is formed in mouth, such herb remedies are applied as spreading honey, brine or alum water, and taking gall nut, Chinese matrimony vine, lotus root, etc, for drugs. Spreading honey, brine or water that is made by infusing gallnut, Chinese matrimony vine, lotus root is based upon such functions as hematosis, astriction, antibacterial, and antiphlogistic, Alum, eggplant and licorice are said to be effective, but their pharmacological effects have no grounds. 5) When one has conjunctivitis such herb remedies are commonly applied as irrigation with brine and dropping breast milk in eyes. Moreover, such other drugs are used as plantain. shepherd's purse, and purslane, etc. The use of brine, breast milk, plantain, shepherd's purse and purslane is based upon such functions as sterilization, antiphlogistic, disinfection and pain relieving. Eriocaulon sieboldianum, bean stem, bean pod and narcissus leaves are said to be effective, but their pharmacological action have no basis. When one has a stye, such herb remedies are applied as extracting eyelashes, stimulating by a massage of middle finger, third finger or big toe, as well as sear ing with a heated bamboo comb that is fine-toothed. Other than these, plantain and nightshade's nuts are used as drugs for it. Extracting eyelashes corresponds with exclusing suppurative node and draining the stye of pus ; and the use of plantain is based upon disinfection: and nightshade's nuts are said to be effective, however, their pharmacological action has no grounds. 6) For a treatment of toothache, such herb remedies are commonly employed as rinsing mouth with brine and holding cold water or gasoline in the mouth ; and as the drugs that are believed to be effective have been Welsh onion, ginger and castor-oil, plant, etc. The use of Welsh onion is based upon pain killing, antiinflammatory actions, and the use of ginger is based upon detoxication and disinfection ; and seeds of castor-oil plants are said to be effective, but they have no pharmacological basis. 7) When one has hives, such herb remedies are commonly applied as rubbing burned straw in affected parts, exposing to its smoke, rubbing with salt, sweeping down with a broom, and spreading and drinking boiled water of trifoliate orange. The use of cassia tora seeds, walnut, aloe and radish is said to be effective. The use of cassia tor a seeds has the functions of intestinal order, anti-paralysis, etc. The use of walnut has resulted in an increase of blood by invigorating spirits ; and the use of aloe is based upon disinfection, antibiotic, anti-salt, antihistamine and detoxication action. But, the effects of radish juice and straw's smoke have no pharmacological grounds. 8) When one gets a boil, such herb remedies are commonly used as applying a plaster, paste of flour mixed with yolk, soy sauce or honey, as well as spreading pounded elm tree. Other remedies that have been said to be effective are ; heating with mugwort, brine, wild rocambole, aloe, onion, squid's bone, etc. The use of mugwort is based upon pain killing, astringent antiinflammatory and tranquility. Wild rocambole is based upon the generation and maintenance functions of cell-joining textures ; elm tree upon antiphlogistic ; aloe upon fever removal and antiphlogistic ; onion on pain killing, fever removal, antiphlogistic and tranquility ; squid's bone on astriction: and brine or vinegar on sterilization. Pine resin and gardenia seed are said to be effective, but they have no pharmacological basis. 9) When one cuts his skin, such herb remedies are commonly employed as spreading mugwort's juice or squid's bone powder, or pressing the wounds. In addition, kalopanax, onion and fine soil are employed. The use of mugwort, kalopanax and squid's bone is based upon such functions as hemostasis, sedation, pain killing, antibacterial ; and fine soil is said to be effective, but it has no pharmacological basis. 10) When one suffers from whitlow, such herb remedies are commonly utilized as heating with boiled soy sauce, spreading soybean paste, or dipping into eggs, etc. Other drugs that have been employed are onion root, brine, eggplant, potato, loach, etc. The use of onion is based upon pain killing and antiphlogistic functions ; and that of brine upon antiphlogistic function. The use of soy sauce or soybean paste, fomentation, eggplant, potato and loach is said to be effective, but it has no pharmacblogic ground. 11) For the treatment of frostbite, such herb remedies are commonly used as dipping the affected part into frozen soybean sack, using boiled water of eggplant stem, garlic caulis, onion, hot pepper, caulis. Onion is based upon antiphlogistic and tranquility actions garlic upon disintection, metabolic exacerbation, tonic and aphrodisiac actions and the use of eggplant and hot pepper is based upon help blood circulation, dissolution and excretion of waste matters in vein. 12) For the treatment of burn, such herb remedies or drugs are commonly used as cleansing with Korean gin, spreading eggs, cleansing with cold water and soap water ; and as brine, cactus, moss, soybean paste, oil, etc. The cleansing with Korean gin, cold water, soap water, brine, vinegar is based upon cleaning and sterilizing functions ; and the use of cucumber is based upon nu. trition provision, and strengthening of resisting power by adjustment of metabolism. The use of potato, cactus, moss, oil and eggs is said to be effective, but their phamacological functions are not clarified. In view of the above results, we can realize that the drugs that have been employed in herb remedies are quite diverse. However, in regard to majority of herb remedies that have been employed by symptoms, the pharmacological functions of their drugs have not been clarified, and they are merely known as effective. Furthermore, they have not been recorded in the literature as yet ; and we confirm that there have been many herb remedies that were executed without the proper knowlege of their effects. It is now our view that the results of this survey may be utilized for consulting data in regard to the use of herb remedies.
