Effects of Gibberellic Acid and Gibberellin Biosynthesis Retardants on Ethylene Production, Batatasins, and Free Sugars in Dormant Tubers of Chinese Yam

  • Gibberellic acid did not affect ethylene production, whereas gibberellin biosynthesis inhibitors triggered ethylene production in dormant tubers. Gibberellic acid did not induce sprouting of dormant tubers, however, treatment of gibberellin biosynthesis retardants enhanced sprouting rates. Sprouting rate in ancymidol-treated tubers was highest among gibberellin biosynthesis retardants. Sprouting rate of tubers treated with ancymidol increased to $91.4\%$. Batatasin-III content in $GA_3$ treated tuber was increased in the highest concentration $(30{\mu}g\;I^{-1})$. Tubers treated with mepiquat chloride, Batatasin-I was increased steadily, but contents of Batatasin-III and V showed dramatic decrease at the $ 1,000{\mu}g\;I^{-1})$ concentration. This infers that gibberellin biosynthesis retardants play key roles in promoting breaking dormancy on dormant tubers of Chinese yam.

Productivity test on some screened strains of Gibberella fujikuroi(Saw.) (Gibberellin 산류 생산균주의 분리와 생산성검토)

  • Gibberella fujikuroi(imperfect stage Fusarium moniforme) a soil fungi is well known as the producer of plant growth regulator Gibberellins. The present work was planned for the isolation of the active strains of Gibberella fujikuroi from the native paddy soils. Twenty two strains were isolated from the infected rice seedlings collected from four local areas. Pyongtaek, Yesan, Tangjin and Sunchon and screened through the activity test for the production of Gibberellins. The strains P-105, Y-14 and T-58 yielded higher activity than the others isolated and the referred strain IAM-8048. The strains Y-5, Y-7, T-54 and S-152, however, were less promotive or rather inhibitory in the growth of rice seedlings. Six different kinds of culture media developed by Cross, Raulin-Thom, Borrow, West, Stodola and Kurosawa respectively were compared with each other for the production of Gibberellins and the best result was obtained with Raulin-Thom's media(glucose 16% and $NH_4NO_3$ 0.24%).

Effects of Gibberellin Biosynthetic Inhibitors on Oil, Secoisolaresonolodiglucoside, Seed Yield and Endogenous Gibberellin Content in Flax

  • Flax (Linum usitatissimum L.) has been used for the edible oil in Korea. The evaluate the effect of plant growth retardants on flaxseed yield, oil content, and gibberellin level, chloromequat chloride (CMC), paclobutrazol (PBZ), and prohexadione-Ca (PHC) were used as plant growth retardants in this study. These plant growth retardants were foliar-sprayed to flax plant at 55 days after seeding. The concentrations of each plant growth retardant were as followed: CMC 250, 500, 1000 ppm, PBZ 40, 80, 160 ppm and PHC 500, 1000, 2000 ppm. PHC treatment to forming bolls was more stimulated than CMC and PBZ. The highest ripened seed rate was observed in PHC treatment at 2000ppm. The high see yields were obtained in PHC treatment following PBZ and CMC, in turn. Seed yield that significantly increased in PBZ and PHC was found to be increased 12.4 ~ 23.9% as compared to the control. The PHC showed higher flaxseed production and oil yield than that of CMC and PBZ. The results obtained in the present study suggest that higher concentration of plant growth retardant (PHC) increased flaxseed yield and oil content. The optimal concentration of PHC treatment was observed in 2000 ppm. It concludes that the foliar application of PHC 2000 ppm may be useful for the increasing oil and higher seed production in flax plants.

Phoma herbarum as a New Gibberellin-Producing and Plant Growth-Promoting Fungus

  • Endophytic fungi are known for the production of valuable metabolites, but information on the gibberellin production capacity of this group is limited. We isolated 9 endophytic fungi from the roots of salt-stressed soybean plants and screened them on waito-c rice, in order to identify plant growth promoting fungal strains. The fungal isolate TK-2-4 gave maximum plant length (20.35 cm) promotion in comparison with wild-type Gibberella fujikuroi (19.5 cm). In a separate experiment, bioassay of TK-2-4 promoted plant length and biomass of soybean cultivar Taegwangkong. The TK-2-4 culture filtrate was analyzed for the presence of gibberellins, and it was found that all physiologically active gibberellins, especially $GA_4$ and $GA_7$, were present in higher amounts ($GA_1$, 0.11 ng/ml; $GA_3$, 2.91 ng/ml; $GA_4$, 3.21 ng/ml; and $GA_7$, 1.4 ng/ml) in conjunction with physiologically inactive $GA_9$ (0.05 ng/ml), $GA_{12}$ (0.23 ng/ ml), $GA_{15}$ (0.42 ng/ml), $GA_{19}$ (0.53 ng/ml), and $GA_{20}$ (0.06 ng/ml). The fungal isolate TK-2-4 was later identified as a new strain of Phoma herbarum, through the phylogenetic analysis of 28S rDNA sequence.

Effects of Paclobutrazol and Prohexadione-Ca on Seed yield, and Content of Oils and Gibberellin in Flax Grown in a Greenhouse

  • We investigated the effect of paclobutrazol (PB) and prohexadione-Ca (PC) on seed yield, and the content of oils and endogenous gibberellin in flax grown in a greenhouse. Flax seedlings were cultivated in a greenhouse with average ambient temperature ranging from $35^{\circ}C$ to $38^{\circ}C$ during the growing period, and then PB and PC were applied on the leaves. The height of flax plants was reduced significantly by the foliar application of PB and PC. The highest ripening seed rate recorded was 94.5% with 2000 ppm PC; a 10.3% increase compared with that of the control. Both PB and PC markedly increased the fructification of flax seed. The application of two plant growth retardants resulted in higher seed production in flax seedlings grown in open field than in those grown in a greenhouse. The highest oil content was observed in flax treated with PC, whereas, the lowest oil content was observed in flax treated with PB. The oil content and ratio of unsaturated fatty acids were higher in flax seedlings grown in greenhouse than in those grown in open field. The level of two endogenous bioactive gibberellins ($GA_1$ and $GA_4$) in flax seeds decreased with the application of PB and PC. The flax plants treated with PB and PC presented higher secoisolariciresinol diglucoside content than that of the control. The optimal concentration of PC was found to be 2000 ppm. In conclusion, the foliar application of 2000 ppm PC might be useful in increasing oil content and seed production in flax plants.

Isolation and Identification of Endophytic Bacteria with Plant Growth Promoting Activity and Biocontrol Potential from Wild Pistachio Trees

  • In this study, samples were collected from the leaves and stems of healthy wild Pistachio trees (Pistacia atlantica L.) from various locations of Baneh and Marivan regions, Iran. In total, 61 endophytic bacteria were isolated and grouped according to phenotypic properties. Ten selected isolates from each group were further identified by partial sequencing of the 16S rRNA gene. Based on the results, isolates were identified as bacteria belonging to Pseudomonas, Stenotrophomonas, Bacillus, Pantoea and Serratia genus. The ability of these isolates was evaluated to phytohormone production such as auxin and gibberellin, siderophore production, phosphate solubilization, atmospheric nitrogen fixation, protease and hydrogen cyanide production. All strains were able to produce the plant growth hormone auxin and gibberellin in different amounts. The majority of strains were able to solubilize phosphate. The results of atmospheric nitrogen fixation ability, protease and siderophore production were varied among strains. Only Ba66 could produce a low amount of hydrogen cyanide. The results of biocontrol assay showed that Pb78 and Sp15 strains had the highest and lowest inhibition effects on bacterial plant pathogens, Pseudomonas syringae pv. syringae Pss20 and Pseudomonas tolaasii Pt18 under in vitro condition. Pb3, Pb24 and Pb71 strains significantly promote root formation on carrot slices. To our knowledge this is the first report of the isolation of endophytic bacterial strains belonging to Pantoea, Bacillus, Pseudomonas, Serratia and Stenotrophomonas genus from wild pistachio trees with plant growth promoting potential and biocontrol activity.

Isolation of a Gibberellin-producing fungus (Penicillium sp. MH7) and Growth Promotion of Crown Daisy (Chrysanthemum coronarium)

  • Plant growth promoting fungi (PGPF) are well known for the production of useful secondary metabolites. However, limited information is available on the gibberellin (GA) production capacity of PGPF of endophytic origin. In the current study, 15 fungal endophytes were isolated from the roots of Crown daisy, and then screened on Waito-c rice, in order to identify plant growth promoting fungi. The fungal isolate MH7 significantly increased the shoot length (12.1 cm) of Waito-c in comparison with control treatment (7.9 cm). In a separate experiment, the culture filtrate (CF) of MH7 significantly promoted the growth attributes of Crown daisy. The MH7 CF was analyzed for gibberellins and it contained all physiologically active gibberellins ($GA_1$, 1.37 ng/ml; $GA_3$, 5.88 ng/ml; $GA_4$, 8.62 ng/ml; and $GA_7$, 2.05 ng/ml) in conjunction with physiologically inactive $GA_9$ (0.83 ng/ml), $GA_{12}$ (0.44 ng/ml), $GA_{15}$ (0.74 ng/ml), $GA_{19}$ (1.16 ng/ml), and $GA_{20}$ (0.98 ng/ml). The CF of MH7 produced higher amounts of $GA_3$, $GA_4$, $GA_7$, $GA_9$, and $GA_{12}$ than wild-type Fusarium fujikuroi, which was used as a control for GA production. The fungal isolate MH7 was later identified as a new strain of Penicillium on the basis of its morphological characteristics and phylogenetic analysis of the 188 rDNA sequence.

Amylase Production by Continuous Cultures of Aspergillus oryzae and its Mutants (Aspergllus oryza와 그 변이주의 연적배양에 의한 amylase 생성에 관한 연구)

  • Irradiation with high doses of gamma rays induced the reduction of mycelial weight and anaylase activity, and increased relative amylase activity in surface cultures. Biphase in growth curves was shown in aeration-agitation cultures but the behavior of the first phase of growth could be eliminated by replacing the amylasehydrolysed starch substrates, so that enzyme production was shortened ca. 40 hours and relative amylase activity was increased about 3 times higher before onset of autolysis. In the effect of gibberellin on amylase production, the positive stimulation was appeared to only surface culturs of the liquid medium and the negative effect to shake-cultures in a mutant. Trials of various continuous culture were resulted not only the approalch to the value of amylase activity in surface cultures of liquid medium, but also higher productivity than in batch cultures. The culture-degeneration was observed in two-stage continuous culture, but did not appear in continuous elevation culture.
