고환에서 발생한 종자세포암의 폐전이 절제 - 1예 보고 - (Pulmonary Metastasectomy from Testicular Germ Cell Tumor - A case report-)

  • 이 보고는 고환종자세포암 치료 후 폐전이를 폐절제술을 시행하여 성공적으로 치료한 증례이다. 항암치료 후 양전자방출단층촬영상 FDG의 유의한 섭취는 없었고 잔존종양을 원발 병소와 조직학적 비교를 위해 전이암을 제거하였다. 본 증례는 잔존종양에 생존세포가 관찰되지 않아 부가적인 항암치료는 시행하지 않았으며, 현재 술 후 6개월로 현재 재발이나 합병증 없이 잘 지내고 있다.

두개내 악성 생식세포종양의 압착도말 세포학적 소견 (Cytologic Features of Intracranial Germ Cell Tumors in Crush Preparation)

  • Intraoperative cytologic examination of intracranial tumors using crush preparation provides useful information in operative decision making. The diminutive nature of many biopsy specimens, particularly those obtained by stereotactic neurosurgical procedures emphasizes the importance of combining the cytologic smear method with conventional frozen section interpretation. The great advantage of the cytologic smear method resides in its suitability for the study of minute fragments of tissue, allowing retention of the majority of the specimen for optimal processing. We present the cytologic features of 3 cases of intracranial germ cell tumors(2 germinomas and 1 endodermal sinus tumor), using crush preparation during intraoperative diagnosis and compare them with histologic findings. The cytologic features of the germ cell tumors were similar to those of the respective gonadal counterparts. The cytologic differential diagnosis of both types of germ cell tumors is described.

  • Pluripotent stem cells have been generated from two embryonic sources. ES cells are generated from ICM of blastocyst stage embryos, and embryonic germ (EG) cells are generated from primordial germ cells (PGCs). Both ES and EG cells are pluripotent and present important characteristics such as high levels of alkaline phosphatase (AP) activity, multi-cellular colony formation, normal and stable karyotypes, continuously passaging ability, and the capability of differentiation into all three embryonic germ layers.   • 영남대학교 의과대학 부속병원에서 난소 악성 생식세포종으로 확진된 12례를 대상으로 CT 소견을 분석하여 다음과 같은 결론을 얻었다. 내배엽동 종양은 낭성 종괴내 일부 고형조직과 격막이 혼재되어 있는데 반하여 미성숙 기형종은 비교적 특징적인 지방조직과 석회화가 보였고, 미분화 세포종은 비록 1례지만 전형적인 낭성 종괴가 없는 고형 조직으로 충만하였다. 그러나 혼합 생식 세포종은 2종이상의 생식 세포종이 혼합된 종양이므로 특징적인 CT 소견없이 혼합된 종양들의 종류에 따라 다양하게 나타날 것으로 생각되었다. 또한 모든 종양들의 크기는 11cm에서 33cm의 비교적 컸다. 결론적으로 젊은 여성에서 큰 난소 종양이 있을 때 CT 영상에서 그 조직성상을 분석하면 그들의 감별진단이 가능하리라고 생각되었다.

Artificial gametes from stem cells

  • The generation of artificial gametes is a real challenge for the scientific community today. In vitro development of human eggs and sperm will pave the way for the understanding of the complex process of human gametogenesis and will provide with human gametes for the study of infertility and the onset of some inherited disorders. However, the great promise of artificial gametes resides in their future application on reproductive treatments for all these people wishing to have genetically related children and for which gamete donation is now their unique option of parenthood. This is the case of infertile patients devoid of suitable gametes, same sex couples, singles and those fertile couples in a high risk of transmitting serious diseases to their progeny. In the search of the best method to obtain artificial gametes, many researchers have successfully obtained human germ cell-like cells from stem cells at different stages of differentiation. In the near future, this field will evolve to new methods providing not only viable but also functional and safe artificial germ cells. These artificial sperm and eggs should be able to recapitulate all the genetic and epigenetic processes needed for the correct gametogenesis, fertilization and embryogenesis leading to the birth of a healthy and fertile newborn.

생쥐 정자세포의 분리와 체외 배양에 관한 연구 (In Vitro Culture Following Purfication of Mouse Spermatogenic Cells)

  • 본 연구는 생쥐의 고게정관내에 존재하는 분화단계의 정자세포를 발생단계별로 분리하여, 체외에서 단기간 배양체계를 확립하기 위하여 실시하였다. 8주령 이상된 생쥐의 정소로부터 정소막을 제거시킨 후, collagenase (1mg/ml), trypsin(2.5mg/ml)를 처리하여 곡세정관을 간질세포와 분리하여 배양액에 부유시켰다. 부유세포는 Celcep장치를 이용하여 세포크기와 밀도 차이에 의해 분화 단계별로 분리하였다. 회수된 세포의 균질성은 haematoxylin/eosin 또는 orcein으로 염색한 후, 광학현미경하에서 확인한 결과 약 85% pachytene spermatocyte와 round spermatid을 성공적으로 분리해냈다. 따라서 sedimentation velocity에 의해서 생쥐의 spermatogenic cell의 발달단계별 분리가 가능함을 알 수 있었다. 이러한 방법으로 분리된 pachytene spermatogenic cell들은 DMEM 배양액에서 6일 이상 배양한 결과 약 36%의 생존율을 보였다. 따라서, 분화단계별 정자 세포의 분리 및 배양체계의 확립은 웅성생식세포의 발생과정에 따른 생리 또는 분자생물학적 현상을 규명함은 물론 세포융합기술의 이용에 의한 형질전환동물 생산에의 응용을 통해 가축에 있어서의 형질전환 생산효율의 개선에 기여할 수 있으리라 사료된다.

Hoechst 33258 Staining을 이용한 웅성 생쥐 성세포의 간편 분류 (Simple Classification of Male Mouse Germ Cells using Hoechst 33258 Staining)

  • In the study for a differentiation and development of spermatogonial cells, the researchers should commonly require a simple, fast and reasonable method that could evaluate the developmental stage of male germ cells without any damage and also relentlessly culture them so far as a cell stage aiming at experimental applications. For developing the efficient method to identify the stage of sperm cells, the morphological characteristics of sperm cells were investigated by staining the cells with blue fluorescent dye Hoechst 33258, and a criterion for male germ cell classification was elicited from results of the previous investigation, then the efficiency of the criterion was verified by applying it to assort the germ cells recovered from male mice in age from 6 to 35 days. As morphological characteristics, spermatogonia significantly differed from spermatocytes in size, appearance and fluorescent patches of nucleus, and spermatids could also be distinguished from spermatozoa by making a difference in the volume and shape of nucleus and the shape and fluorescence of tail. Aforesaid criterion was applicable for classifying in vitro cultured sperm cells by verifying its efficiency and propriety for assorting the stages of testicular germ cells. However, the fluorescent staining showed that germ cells in mouse testis should be dramatically differentiated and developed at 21 days and 35 days of age, which were known as times of sexual puberty and maturity in male mice, respectively. In conclusion, the results indicated that this simple criterion for sperm cell classification using fluorescence staining with Hoechst 33258 may be highly efficient and reasonable for spermatogenesis study.

조류의 다능성 생식세포주 확립 및 분화 특성에 관한 연구

  • The use of pluripotent stem cells has tremendous advantages for various purposes but these cell lines with proven germ-line transmission have been completely established only in the mouse. Embryonic germ (EG) cell lines are also pluripotent and undifferentiated stem cells established from primordial germ cells (PGCs). This study was conducted to establish and characterize the chicken EG cells derived from gonadal primordial germ cells. We isolated gonadal PGCs from 5.5-day-old (stage 28) White leghorn (WL) embryos and established chicken EG cells lines with EG culture medium supplemented with human stem cell factor (hSCF), murine leukemia inhibitory factor (mLIF), bovine basic fibroblast growth factor (bFGF), human interleukin-11 (hIL-11), and human insulin-like growth factor-I (hIGF-I). These cells grew continuously for 4 months (10 passages) on a feeder layer of mitotically active chicken embryonic fibroblasts. These cells were characterized by screening with the Periodic acid-Shiff's reaction, anti-SSEA-1 antibody, and a proliferation assay after several passages. As the results, the chicken EG cells maintained characteristics of undifferentiated stem cells as well as that of gonadal PGCs. When cultured in suspension, the chicken EG cells successfully formed an embryoid body and differentiated into a variety of cell types when re-seeded onto culture dish. The chicken EG cells were injected into blastodermal layer at stage X and dorsal aorta of recipient embryo at stage 14 (incubation of 53hrs) and produced chimeric chickens with various differentiated tissues derived from the EG cells. The germline chimeras were also successfully induced by using EG cells. Thus, Chicken EG cells will be useful for the production of transgenic chickena and for studies of germ cell differentiation and genomic imprinting.

  • The testis is composed of four cell types like supporting cells, steroid-producing cells, connective tissue cells and germ cells. Apoptosis is a common phenomenon during spormatogenesis. Apoptosis of germ cells can also be induced by exposure to radiation. Previous studies have shown that most types of germ cells are rather radiosensitive while somatic cells in testis are much more radio-resistant. The somatic cells in testis are divided to mainly Sertoli and Leydig cells. Though somatic cells are more radio-resistant than germ cells, radiation can induce the impairment of their function. This damaged function of somatic cells may accelerates degeneration of germ cell indirectly. Tn the present study, we have examined the apoptotic effect of mouse testis and irradiation effect of steroidogenesis of Leydig cells after irradiation.
