Quality and genetic relationship on dehulling characteristic in oat variety (국내 귀리품종 탈부특성에 따른 품질 및 유연관계 분석)

  • The consumption of oats (Avena sativa L.) with high nutritional utility is accelerating due to the increased consumers' demand for functional foods. In Korea, naked oats are used as food, while covered oats are used for animal feed. However, it is difficult to distinguish naked oats from covered oats when the husk is removed from the grains by a special process. The present study was carried out to investigate experimental methods that would be beneficial in the segregation of different types of oats after husk removal. Grain quality-related biochemical compounds were analyzed in a bid to differentiate the oat dehulling characteristics. In addition, 61 SSR markers were examined for genetic relationship and variety identification of oats using five naked and seven covered oat varieties. Results showed that, the contents of protein, lipid, and ${\beta}-glucan$ were not significantly different among the oat varieties and this could not be used as an index for distinguishing oats husk character. However, in the fatty acid composition ratio,, naked oats had a higher ratio of stearic acid (C18:0) and oleic acid (C18:1) than covered oats, and covered oats had a higher ratio of linoleic acid (C18:2) and linoleic acid (C18:3) than naked oats. The assessment of SSR marker genotype revealed that 33 polymorphic bands among 12 oat varieties and 1 variety could be distinguished through the combination of polymorphic markers thus indicating the usability of these markers for variety identification in oats.

Comparison of Normalization Methods for Defining Copy Number Variation Using Whole-genome SNP Genotyping Data

  • Precise and reliable identification of CNV is still important to fully understand the effect of CNV on genetic diversity and background of complex diseases. SNP marker has been used frequently to detect CNVs, but the analysis of SNP chip data for identifying CNV has not been well established. We compared various normalization methods for CNV analysis and suggest optimal normalization procedure for reliable CNV call. Four normal Koreans and NA10851 HapMap male samples were genotyped using Affymetrix Genome-Wide Human SNP array 5.0. We evaluated the effect of median and quantile normalization to find the optimal normalization for CNV detection based on SNP array data. We also explored the effect of Robust Multichip Average (RMA) background correction for each normalization process. In total, the following 4 combinations of normalization were tried: 1) Median normalization without RMA background correction, 2) Quantile normalization without RMA background correction, 3) Median normalization with RMA background correction, and 4) Quantile normalization with RMA background correction. CNV was called using SW-ARRAY algorithm. We applied 4 different combinations of normalization and compared the effect using intensity ratio profile, box plot, and MA plot. When we applied median and quantile normalizations without RMA background correction, both methods showed similar normalization effect and the final CNV calls were also similar in terms of number and size. In both median and quantile normalizations, RMA backgroundcorrection resulted in widening the range of intensity ratio distribution, which may suggest that RMA background correction may help to detect more CNVs compared to no correction.

The EST Study of the Peri-implanting Porcine Embryos (Peri-implanting 단계의 돼지배아 EST 연구)

  • A dramatic morphological change of embryos occurs at peri-implantation. Maternal and embryonic cross-talk during this period, initiated by signals from embryo(s), provides signals for maternal recognition of pregnancy and establishing and maintaining the pregnancy. However, the cellular, biochemical and genetic processes that direct embryo remodeling in mammalian species are not well studied or understood. In order to identify potential genes responsible for morphological change and cross-talk between embryo and uterus, an initial EST analysis was performed. A catalog of expressed genes (Transcriptome) from the d12 peri-implanting porcine embryos was constructed. Six clones were chosen from the initial ESTs for elucidation of their expression patterns during embryogenesis in early pregnancy. A number of these genes demonstrated unique expression profiles in a tissue, cell-type, and temporal fashion, indicating dynamic regulation of embryonic and endometrial gene expressions at different stages of pregnancy. Cross-talk between the embryo and endometrium of the pregnant uterus has provided a suitable micro-environment for the embryo's rapid and dramatic morphological changing process at the peri-implantation stage.

Construction of a full-length cDNA library from Pinus koraiensis and analysis of EST dataset (잣나무(Pinus koraiensis)의 cDNA library 제작 및 EST 분석)

  • In this study, we report the generation and analysis of a total of 1,211 expressed sequence tags (ESTs) from Pinus koraiensis. A cDNA library was generated from the young leaf tissue and a total of 1,211 cDNA were partially sequenced. EST and unigene sequence quality were determined by computational filtering, manual review, and BLAST analyses. In all, 857 ESTs were acquired after the removal of the vector sequence and filtering over a minimum length 50 nucleotides. A total of 411 unigene, consisting of 89 contigs and 322 singletons, was identified after assembling. Also, we identified 77 new microsatellite-containing sequences from the unigenes and classified the structure according to their repeat unit. According to homology search with BLASTX against the NCBI database, 63.1% of ESTs were homologous with known function and 22.2% of ESTs were matched with putative or unknown function. The remaining 14.6% of ESTs showed no significant similarity to any protein sequences found in the public database. Gene ontology (GO) classification showed that the most abundant GO terms were transport, nucleotide binding, plastid, in terms biological process, molecular function and cellular component, respectively. The sequence data will be used to characterize potential roles of new genes in Pinus and provided for the useful tools as a genetic resource.

Optimal Design of Stator Shape for Cogging Torque Reduction of Single-phase BLDC Motor (단상 BLDC 전동기의 코깅토크 저감을 위한 고정자 형상 최적설계)

  • This paper proposes the optimal design of stator shape for cogging torque reduction of single-phase brushless DC (BLDC) motor with asymmetric notch. This method applied size and position of asymmetric notches to tapered teeth of stator for single-phase BLDC motor. Which affects the variation of the residual flux density of the permanent magnet. The process of optimal design included the extraction of the sampling point by using Latin Hypercube Sampling(LHS), and involved the creation of an approximation model by using kriging method. Also, the optimum point of the design variables were discovered by using the Genetic Algorithm(GA). Finite element analysis was used to calculate the characteristics analysis and cogging torque. As a result of finite element analysis, cogging torque were reduced approximately 39.2% lower than initial model. Also experimental result were approximately 38.5% lower than initial model. The period and magnitude of the cogging torque were similar to the results of FEA.

RESULTS FROM ADAPTABILITY TRIAL OF RAMBOUILLET SHEEP AND THEIR CROSSBREEDING WITH KAGHANIS. EFFECTS ON EWE MATING WEIGHT, WOOL PRODUCTION, LITTER SIZE AND LAMB GROWTH

  • In order to upgrade native sheep, Rambouillet (R) rams were mated to Kaghani (K) ewes to generate F1 ($R{\times}K$) crossbred ewes. Crossbred ewes were backcrossed to Rambouillet rams to produce B1 ($R{\times}F1$), B2 ($R{\times}B1$) and B3 ($R{\times}B2$) genotypes. Weaning weight of 2605 lambs and wool weight of 2378 mature ewe records, representing R, K, F1, B1, B2 and B3 genotypes, were analyzed to compare genetic variation among genotypes produced during upgrading process and identify genotypes of the highest performance. Performance of Rambouillets was also evaluated under semi-temperate climate. Data were adjusted for yearly variation considering Rambouillet as a control. Genotypes influenced lambs weaning weight (p<.01). B1 lambs were heaviest (18.4 kg) followed in order by B2, F1, B3, R and K lambs (18.3, 17.9, 16.9, 16.8 and 13.2 kg, respectively). The highest wool production was 2.5 kg from R ewes followed by B2 (2.3), B3 (2.3), F1 (2.0) and K (1.2) ewes (p < .01). Ewe mating weight, reproduction, growth and wool production of Rambouillets deteriorated significantly after the first decade of their importation. Compared with the first phase (1959-1971), ewe mating weight, litter size, birth weight, lamb weaning weight and wool production declined by 20, 23, 32 and 36%, respectively, in the second phase (1972-1988).

Association of Angiotensin Converting Enzyme I/D and ${\alpha}$-actinin-3 R577X Genotypes with Growth Factors and Physical Fitness in Korean Children

  • This study analyzed the differences in aerobic and anaerobic exercise ability and growth-related indicators, depending on the polymorphism of the ACE and the ACTN3 genes, to understand the genetic influence of exercise ability in the growth process of children. The subjects of the study consisted of elementary school students (n=856, age $10.32{\pm}0.07yr$). The anthropometric parameters, physical fitness and growth factors were compared among groups of the ACE I/D or the ACTN3 R577X polymorphisms. There were no significant differences between the anthropometric parameters, physical fitness and growth factors for the ACE gene ID or the ACTN3 gene R577X polymorphism. However, the DD type of ACE gene was highest in the side step test (p<0.05), and the DD type was significantly higher than the II+ID type (p<0.05) in the early bone age. The combined group of the ACE gene II+ID and the ACTN3 gene XX type significantly showed lower early bone age (p< 0.05). This study did not find any individual or compounding effects of the polymorphism in the ACE I/D or the ACTN3 R577X polymorphisms on the anthropometric parameters, physical fitness and growth factors of Korean children. However, the exercise experience and the DD type of the ACE gene may affect the early maturity of the bones.

A New Species of Eudactylopus (Copepoda: Harpacticoida) from the South Coast of Korea Based on Morphological and Molecular Evidence

  • A new species of Eudactylopus Scott A., 1909 is described from the southern coast of Korea. The specimens were collected using a light trap set overnight at the entrance near a pier. Eudactylopus yokjidoensis n. sp. is similar to E. andrewi Sewell, 1940 and E. spectabilis (Brian, 1923) in two key respects: similar length of proximal and distal inner setae on female P2 enp-2, and modification of two subapical setae on male P2 endopod. However, E. yokjidoensis can be differentiated from the two species by following morphological characteristics: in females, the length ratio of cephalothorax/2nd-4th thoracic somites combined is smaller in E. yokjidoensis than other two species (1 : 0.8 vs. 1 : 1); antennule has nine segments (vs. 7-segmented in E. andrewi); P2 to P4 each bears a process in medial distal margin of basis, while it is just smooth in E. spectabilis; in males; the length ratio of cephalothorax to 2nd-4th thoracic somites combined is smaller in E. yokjidoensis than other two species (1 : 0.6 vs. 1 : 1 in E. andrewi and 1 : 0.8 in E. spectabilis); and P5 exopod has a comb-like innermost seta, while it is bipinnate seta in E. spectabilis. To prove the Korean species of Eudactylopus to be new, full descriptions of both sexes are given here, and the claim is supported by distinct genetic differences between E. yokjidoensis and E. spectabilis (22.3-22.7%) in the mitochondrial gene cytochrome oxidase subunit I(mtCOI) sequence.

Single Cell Oil-Recent Trends in Microbial Production and Utilization (미생물에 의한 단세포유지의 생산과 이용)

  • With the shortage of edible fats and oils and depletion of fossil fuels in many countries, microbial lipids is emerging as one of the most promising sources of fats and oils in the global market. Oleaginous microorganisms, also called single cell oils (SCOs), can accumulate lipids more than 25% in the cell volume. Triacylglycerols are the major storage lipids. SCOs offer several advantages for lipid production as follows: SCOs have short life span which would shorten production time, cultivation conditions are not affected by climate and place; the production process is easy to control. There are a number of oleaginous yeasts, molds, and microalgae. Furthermore, the lipid productivity of SCOs can be enhanced through strain improvement and the optimization of cultivation conditions. The new strains developed using recent advanced biotechnical methods showed greatly improved oleaginicity. Further, hydrolysates of lignocellulosic materials can be used as carbon sources for economic production of SCO.

A Study on Identification of Optimal Fuzzy Model Using Genetic Algorithm (유전알고리즘을 이용한 최적 퍼지모델의 동정에 관한연구)

  • A identification algorithm that finds optimal fuzzy membership functions and rule base to fuzzy model isproposed and a fuzzy controller is designed to get more accurate position and velocity control of wheeled mobile robot. This procedure that is composed of three steps has its own unique process at each step. The elements of output term set are increased at first step and then the rule base is varied according to increase of the elements. The adjusted system is in competition with system which doesn't include any increased elements. The adjusted system will be removed if the system lost. Otherwise, the control system is replaced with the adjusted system. After finished regulation of output term set and rule base, searching for input membership functions is processed with constraints and fine tuning of output membership functions is done.
