The design optimization of a cold-formed steel portal frame building is considered in this paper. The proposed genetic algorithm (GA) optimizer considers both topology (i.e., frame spacing and pitch) and cross-sectional sizes of the main structural members as the decision variables. Previous GAs in the literature were characterized by poor convergence, including slow progress, that usually results in excessive computation times and/or frequent failure to achieve an optimal or near-optimal solution. This is the main issue addressed in this paper. In an effort to improve the performance of the conventional GA, a niching strategy is presented that is shown to be an effective means of enhancing the dissimilarity of the solutions in each generation of the GA. Thus, population diversity is maintained and premature convergence is reduced significantly. Through benchmark examples, it is shown that the efficient GA proposed generates optimal solutions more consistently. A parametric study was carried out, and the results included. They show significant variation in the optimal topology in terms of pitch and frame spacing for a range of typical column heights. They also show that the optimized design achieved large savings based on the cost of the main structural elements; the inclusion of knee braces at the eaves yield further savings in cost, that are significant.
The rice belonging to Oryza sativa is not only has significant economic importance, for it is the major source of nutrition for about 3 billion all around the world. But also plays a vital role as a model organism, because it has a number of advantages to be a model plant, such as efficient transformation system and small genome size. Many methods and techniques have been conducted to attempt to distinguish different Oryza sativa species, such as amplified fragment length polymorphism (AFLP), random amplified polymorphic DNA (RAPD), simple sequence repeat (SSR) and so on. However, studies using sequence analysis of internal transcribed spacer (ITS), a region of ribosomal RNA has not been reported until now. This study was undertaken with an aim to understand the phylogenetic relationships among sixteen isolates of Oryza sativa collected from abroad and fifteen isolates collected from Korea, using ribosomal RNA (rRNA) internal transcribed spacer (ITS) sequences to compare the phylogeny relationships among different Oryza sativa species. The size variation obtained among sequenced nuclear ribosomal DNA (nrDNA) ITS region ranged from 515bp to 1000bp. The highest interspecific genetic distance (GD) was found between Sfejare 45 (FR12) and Anapuruna (FR15). Taebong isolate showed the least dissimilarity of the ITS region sequence with other thirty isolates. This consequence will help us further understanding molecular diversification in intra-species population and their phylogenetic analysis.

Genetic diversity of 47 East-Asian vegetable soybean was characterized by means of agro-morphological traits and RAPD markers. A field trial was conducted to evaluate 14 agro-morphological traits. To study RAPD-based DNA analysis, a total of sixty 10-mer random primers were screened. Of these, 23 polymorphic markers in 16 varieties used for screening. Among 207 markers amplified, 48 were polymorphic for at least one pairwise comparison within the 47 varieties. A higher differentiation level between varieties was observed by using RAPD markers compared to morphological markers. Correspondence analysis using both types of marker showed that RAPD data could fully discriminate between all varieties, whereas morphological markers could not achieve a complete discrimination. Genetic distances between the varieties were estimated from simple matching coefficients, ranged from 0.0 to 0.640 with an average of 0.295$\pm$0.131 for morphological traits and 0.042 to 0.625 with an average of 0.336$\pm$0.099 for RAPD data, respectively. Cluster analysis based on genetic dissimilarity of these varieties gave rise to 4 distinct groups. The clustering results based on RAPDs did not match with those based on morphological traits. Geographical distribution of most varieties in each of the groups were not well defined. The results suggested that the level of genetic diversity within this group of East-Asian vegetable soybean varieties was sufficient for a breeding program and can be used to establish genetic relationships among them with unknown or unrelated pedigrees.
The relationship of nine Medicago species belonging to four subgenera were analyzed by using SDS-PAGE and restriction fragment length polymorphism (RELP) methodologies. Sixty-eight bands of alcohol and salt soluble proteins and 85-133 RFLP markers were used to estimate the genetic distance among the species. These species were clustered together at around 0.1 to 0.4 level of distance for both kind of markers, indicating that Medicago species have a large genetic similarity. A combined cluster diagram, at a dissimilarity level of 0.3, differentiated nine species in four groups: group 1, M. littoralis , M. truncatulam, M.scutellata and M. rigidula; group 2, M. sativa ; group 3, M. lupulina ; group 4, M. orbicularis, M. radiata and M. minima. All of them, but except for M. minima. corrensponded to the existing four subgenera of the genus Medicago classified by Lesins and Lesins(1979).The most similar species were M. littoralis and M. trucatula and the most dissimilar one was M. lupulina. In separate cluster diagrams based on RFLP and protein markers, some differences were observed. In the case of RFLP or DNA markers, M. sativa (alfalfa) was distantly clustered with other Medicago species. But in the case of protein markers, M. sativa was closely clustered with M. scutellata, M. littorulis and M. truncatula.
Spatial autocorrelation was applied to microgeographic variations of Sasa borealis populations in Korea. Separate counts of each type of join (combination of genotypes at a single locus) for each allele, and for each distance class of separation, were tested for significant deviation from random expectations by calculating the Standard Normal Deviation. Moran's I was significantly different from the expected value in 25 of 150 cases $(16.7\%)$. Seven of these values $(4.7\%)$ were negative, indicating genetic dissimilarity among pairs of individuals in the ten distance classes. Populations of S. borealis are small in Korea, and are distributed with occasional cutting of seed-bearing stems used for sieves. Thus, artificial disturbance may contribute to the fact that the S. borealis population of Jirisan is unusual in lacking spatial genetic structure.
A portion of ribosomal internal transcribed spacer (ITS) 2 was sequenced from the samples of Pseudo nitzschia (P. deticatissima, P. multiseries, P. pungens and P. subfraudulenta) to investigate the genetic characteristics by measuring tile magnitude of genetic diversity and the degree of similarity coefficient using random amplified polymorphic DNAs (RAPD)-PCR patterns. The phylogenetic trees inferred from the genetic distance analyses showed the placement of P. delicatissima formed a quite long distance from p. P. multiseries, P. pungens, and even P. subfraudulenta. The phylogenetic tree from RAPD patterns showed that P. multiseries and P. pungens had dissimilarity coefficient of 0.31, while P. delicatissima and three species of Pseudo-nitzschia had that of 0.81. It is likely thought that the genetic position of P. delicatissima formed far from P. multiseries, P. punges, and P. subfraudulenta. These results imply that ITS2 region is expected to support a useful molecular characters for recognizing at the species level and for even discriminating P. multiseries from P. pungens. RAPD method also will be used to differentiate the species of Pseudo-nitzschia in a short time.
In order to estimate the genetic relationships among four taxa of Korean Symplocaceae and their regional populations, RAPDs analysis was performed. The length of the amplified DNA fragments ranged from 150 to 1,900 bp. Ninety-two RAPD bands were scored for 11 primers and the genetic distance was calculated with by Nei-Li's genetic dissimilarity. Deciduous and evergreen groups were clearly separated in the UPGMA analysis. Symplocos coreana was clustered as a distinct group, and S. sawafutagi and S. tanakana were also clustered at specific level, respectively. Zoysiagrass, especially Jungji implicating medium-leaf type zoysiagrass has been widely used in South Korea recently. This study was carried out to classify 36 selected medium-leaf type zoysiagrass accessions compared to 5 basic zoysiagrass species using RAPDs. Morphological characteristics such as leaf width, leaf angle, leaf sheath length, existance of trichomes and stolon color were measured as useful characteristics for identification of species in Zoysia genus. Nineteen RAPD markers were identified using 8 selected random primers. The dissimilarity coefficient of variants ranged from 0 to 0.736. Three zoysiagrass groups were clustered by dissimilarity coefficient analysis. Group 1 consisted of Z. japonica and some US varieties including 'Zenith' and 'Meyer'. Group 2 consisted of Z. sinica, Z. macrostachya and Korean commercial varieties such as 'Anyang', 'Samdeock', and 'Pyeongdong' medium-leaf type grasses. Group 3 was genetically distinct from Group 1 and Group 2, and included Z. matrella and Z. tenuifolia. 'Anyang', 'Samdeock', and 'Pyeongdong' medium-leaf type zoysiagrasses showed very close genetic relationship with Z. sinica and Z. macrostachya.
Fusarium head blight is one of the most important and most common diseases of winter wheat. In order to better understanding this disease and to assess the correlations between different factors, 30 cultivars of this cereal were evaluated in a two-year period. Fusarium head blight resistance was evaluated and the concentration of trichothecene mycotoxins was analysed. Grain samples originated from plants inoculated with Fusarium culmorum and naturally infected with Fusarium species. The genetic distance between the tested cultivars was determined and data were analysed using multivariate data analysis methods. Genetic dissimilarity of wheat cultivars ranged between 0.06 and 0.78. They were grouped into three distinct groups after cluster analysis of genetic distance. Wheat cultivars differed in resistance to spike and kernel infection and in resistance to spread of Fusarium within a spike (type II). Only B trichothecenes (deoxynivalenol, 3-acetyldeoxynivalenol and nivalenol) produced by F. culmorum in grain samples from inoculated plots were present. In control samples trichothecenes of groups A (H-2 toxin, T-2 toxin, T-2 tetraol, T-2 triol, scirpentriol, diacetoxyscirpenol) and B were detected. On the basis of Fusarium head blight assessment and analysis of trichothecene concentration in the grain relationships between morphological characters, Fusarium head blight resistance and mycotoxins in grain of wheat cultivars were examined. The results were used to create of matrices of distance between cultivars - for trichothecene concentration in inoculated and naturally infected grain as well as for FHB resistance Correlations between genetic distance versus resistance/mycotoxin profiles were calculated using the Mantel test. A highly significant correlation between genetic distance and mycotoxin distance was found for the samples inoculated with Fusarium culmorum. Significant but weak relationships were found between genetic distance matrix and FHB resistance or trichothecene concentration in naturally infected grain matrices.
A genetic variation within 29 strains of F. velutipes was analyzed by internal transcribed spacer (ITS) sequence analysis and random amplified polymorphic DNA (RAPD). Seven hundred and twenty base pairs were sequenced during the analysis of the ITS region, but no significant variation was observed among the 29 strains of F. velutipes. Sixteen out of 40 random primers amplified polymorphic RAPD fragment patterns. The polymorphic levels of RAPD bands by some primers (OPA-2,4,3,9,10,20) were very high in all 29 strains, with 3,030 fragments ranging between 200 and 2,000 bp. Intraspecific genetic dissimilarity of the 29 strains was calculated to range from 3.3% to 45% by Nei-Li's method using these 3,030 RAPD bands. The genetic variation among Korean strains was relatively high, with dissimilarities ranging between 17% and 38.6%. In the Neighbor-Joining analysis using the genetic dissimilarities based on RAPD, all 29 strains were classified into 5 clusters. Strains in each cluster showed specific characteristics according to their origin and strains. These results suggested that OPA and OPB primers could be used for developing molecular genetic markers and screening of unidentified (F. velutipes) strains.
