Purification and Characteristics of Chitosanase from Bacillus sp. HW-002

  • Chitosanase from Bacillus sp. HW-002 was purified with CM-cellulose column chromatography, and HPLC with DEAE- TSK gel and YMC-pack Diol 120. The purified enzyme appeared as a single band on SDS-polyacrylamide gel. The molecular weight of the enzyme was estimated to be about 46 kDa on SDS-polyacrylamide gel, and was estimated to be about 23 kDa by GFC. The optimal pH of chitosanolytic activity was about pH 5.5-6.0, and the purified enzyme was most stable at pH 5.0. The optimal temperature of chitosanolytic activity was $65^{\circ}C$ and the enzyme was stable at $45^{\circ}C$ for 1 h. Chitosan was the most favorable substrate among various $\beta$-glucan. UVmax of the purified enzyme was 195 nmand was not noted around 280 nm. The main product of enzyme reaction with chitosan was chitobiose.

임상간호사의 근거기반실무 신념과 연구 관련 행동의도에 따른 온 요법 간호 (Heat Application According to Nurse's Belief on Evidence-Based Practice and Behavioral Intention Related to Research)

  • Purpose: This study was done to compare heat applications by nurses in accordance to belief in evidence-based practice and behavior intention related to research. Methods: A cross-section survey design was used. Participants were 228 nurses from 5 institutions who completed the research questionnaire. Data were collected during July and August 2015. Results: The nurses reported that duration of heat applications was 2.5 minutes to 90 minutes. Frequently used heat application devices, in order of frequency, were rubber bag, gel pack and red clay pack. Most of the nurses (78%) responded that advice from colleagues was the most frequently used basic evidence for heat application. There was a statistically significant difference for the necessity of heat application practice guidelines between the high evidence-based practice belief cluster and the low cluster ($x^2$=15.39, p<.001). Conclusion: There were difficulties in providing consistent nursing interventions because of practical differences and absence of evidence-based guidelines for heat application. The researchers recommend that basic studies with various instruments be conducted and proper practice guidelines developed for heat application.

  • To prolong the shelf-life of strawberry, samples were treated with gel packs containing slow-released chlorine dioxide($ClO_2$) gas at 3~7 ppm for 6 days at room temperature. The weight loss and decay ratio as well as changes in pH, color and texture properties of the treated samples were investigated. The weight of the control and $ClO_2$ gas treated samples decreased slightly, but the weight of the control changed faster than those of the $ClO_2$ gas treated samples during the storage period. The decay ratio of control was higher than those of the $ClO_2$ gas treatments since 4 days of storage. The pH and acidity in the control and in the $ClO_2$ gas treated samples were no differences during storage period. The lightness of strawberry decreased during storage, but there was no difference in lightness among the treatments even when storage time was extended. The redness and yellowness of the control showed higher change than those of the $ClO_2$ gas treatments during 6 days. The firmness of the control changed more rapidly than those of the $ClO_2$ gas treatments during 6 days. Especially, the samples treated 3 and 5 ppm $ClO_2$ gas were the least changed. And the scores for appearance, firmness and overall acceptance control and 7 ppm $ClO_2$ gas treatment decreased more rapidly than those of 3 and 5 ppm treatment.

  • To prolong the shelf-life of chicken breast meat, samples were treated with gel packs containing slow-released chlorine dioxide ($ClO_2$) gas at 7~15 ppm for eight days at $4^{\circ}C$. The microbial, physicochemical properties and sensory evaluation of the treated samples were investigated. The total number of bacteria in the control increased during storage and showed 6.78 log CFU/g on the 8th day of storage, but $ClO_2$ gas treatments showed 6.24~6.58 log CFU/g at the same time. The initial pH of chicken breast meat was 6.00 and gradually increased during storage. And $ClO_2$ gas treatments did not show any significant difference from the control during storage period, but maintained a generally lower pH than that of the control. The lightness, redness, and yellowness during storage were not significantly different between the control and the 7~10 ppm $ClO_2$ gas treatments. However, as the storage period was increased, the redness of 15 ppm $ClO_2$ gas treatment was reduced. The cooking loss and shear force were not different between the control and $ClO_2$ gas treatments during the storage period. Volatile basic nitrogen (VBN) increased in the control from the 6th day of storage and 23.80 mg% in the 8th day of storage. However, VBN of $ClO_2$ treatments showed lower than that of the control. In the change of sensory evaluation during storage, 10 ppm $ClO_2$ treatment showed the highest preference in odor, appearance and overall acceptance during storage period.

단세포전기영동법으로 평가한 흡연자의 백혈구 DNA손상 (Smoking Related DNA Damage in Human Lymphocytes Assessed by the Comet Assay)

  • The single cell gel electrophoresis (comet) assay is one of the useful tools for the study of genetic damage in humans exposed to environmental mutagens and carcinogens. This study was undertaken to evaluate the status of DNA damage in peripheral lymphocytes depending on their sex, age, smoking habits, and other factors in normal healthy Korean population. The 99 volunteers included in the study and out of these, 36 volunteers were smoker and 63 volunteers were non-smoker aged between 20-59 years. All individual answered a questionnaire that assessed their general information including smoking habits and the extent of the environmental tobacco smoke (ETS) exposure, and blood samples were obtained. There was a statistically significant difference in the extent of DNA damage between smoker and non-smoker (p<0.001). A significant difference was also observed between male and female (p<0.001) and amongst the different group of age (p<0.005), however, correlation analysis showed that only smoking habit was a significant factor for DNA damage. No significant effect of smoking duration, number of cigarettes smoking a day, SPY (smoke pack years) in smokers and environmental tobacco smoke exposure in non-smokers on the status of DNA damage was observed.

흰점박이꽃무지(Protaetia brevitarsis) 함유 음료의 in vitro 항산화 관련 생리활성효능 및 안전성 검증 (Antioxidant Activity and Safety Evaluation of Juice Containing Protaetia brevitarsis)

  • 본 연구에서는 흰점박이꽃무지 함유 음료의 총폴리페놀함량과 항산화활성(DPPH 라디칼 소거능, TRAP, ORAC assay, comet assay) 및 사용에 대한 안전성을 검증하기 위하여 ICR mouse를 사용하여 흰점박이꽃무지 무첨가군과 첨가군 단회 투여(2 g/kg) 시 나타나는 치사율, 체중, 생리적 증상, 장기무게, 생화학적 분석(triglyceride, total cholesterol, HDL-cholesterol, LDL-cholesterol, aspartate transaminase, alanine transaminase)을 통해 급성독성시험을 실시하였다. 흰점박이꽃무지를 함유한 호박, 알로에, 돼지감자음료 1 pack의 총 페놀함량은 66.35~100.98 mg/pack, DPPH 라디칼 소거능은 67.1~90.0%/1 pack, TRAP 값은 $1.46\sim1.54{\mu}M$/trolox equivalent(TE)를 보였으며, 모든 음료의 ORAC 값은 1500보다 높았다. 모든 음료는 인체 임파구에서 세포독성을 보이지 않았으며, $H_2O_2$에 대한 DNA 손상에 대한 보호효과는 유의적이었다. 흰점박이꽃무지 첨가/무첨가 음료의 ICR mouse 암수 각각에 경구투여(2 g/kg) 후 14일까지 치사된 실험동물은 관찰되지 않았으며, 체중, 생리적, 병리적 및 생화학적 분석에서 대조군과 비교 시 그룹들 간의 유의한 변화는 관찰되지 않았다. 그러므로 2 g/kg의 흰점박이꽃무지 첨가/무첨가 음료를 ICR mouse에 단일 경구투여시 독성이 없는 것으로 판명되었다. 본 연구에서 규명된 흰점박이꽃무지 함유 음료의 항산화활성과 안전성은 산업적측면에서 약용 및 식용 자원으로 적극적인 활용을 기대할 수 있을 것으로 사료된다.

  • 배추김치의 dichloromethane 획분의 수율은 약 1.66% 이었으며, 이로부터 silica gel column chroamtography, Sepadex LH-20 column chromatography, Sep-pack catridge 등을 이용하여 활성물질을 분리 정제하였고, IR, EI-MS, GC-MS. $^{1}H-NMR,\;^13C-NMR$로 구조를 확인하여 분자량 226인 3-(4'-hydroxyl-3',5'-dimethoxyphenyl)propionic acid로 밝혀다. 비추김치 활성성분의 DPPH 유리기 소거능은 vitamin C에 비해 3.4배 높았고, LDL 산화억제 효과는 대조군에 비해 현저히 높아 인체에서 지질저하 효과 및 동맥경화 억제 효과가 있을 것으로 예상된다.

  • To improve the shelf-life of Centella asiatica, Centella asiatica was treated with gel packs containing slow-released chlorine dioxide (ClO2) gas at 3-5 ppm for 20 days at 4℃. The weight loss rate, as well as the changes in pH, color, and texture of the treated samples, were investigated. The weight of the control and ClO2 gas-treated samples was decreased during the storage period. The change in weight of the control was slightly faster than that of the samples treated with 3 and 4 ppm ClO2 gas. The pH of the control and the ClO2 gas treated samples were decreased during the storage period and there was no significant difference between the control and ClO2 gas treated samples. Concerning color (lightness, redness, and yellowness) changes of Centella asiatica during the storage period, there was no significant difference between the control and ClO2 gas treated samples. The change in shear force in the leaf and stem of Centella asiatica during the storage period was slightly lower in the 4 ppm ClO2 gas treated samples (in the leaf) compared to the control and 3 and 4 ppm ClO2 gas treated samples (in the stem) compared to the control and 5 ppm ClO2 gas treated sample.

전통 메주로부터 분리한 Bacillus subtilis PCA 20-3 유래 Protease 의 정제 (Purification of a Protease Produced by Bacillus subtilis PCA 20-3 Isolated from Korean Traditional Meju)

  • 재래식 메주로부터 분리한 Bacillus subtilis PCA203이 생산하는 protease를 분리 정제하였다. 먼저 효소 생산용 배지$(0.2%\;soytone,\;2%\;soluble\;starch,\;0.1%\;(NH_4)_2SO_4,\;0.1%\;CaCl_2,\;0.01%\;yeast\;extract,\;0.1%\;K_2HPO_4,\;0.1%\;KH_2PO_4)$를 이용하여 $30^{\circ}C$에서 20시간 배양한 다음, 원심분리하여 상징액을 분획한 후, 80% 포화 황산암모늄에 의한 염석과 CM Sephdex C-50 및 Sephadex G-100을 이용하여 비활성도 76.0 unit/mg, 수율 2.7%, 정제배수 7.6배로 효소를 정제하였다. 정제 단백질의 YMC-pack protein-RP column chromatography에 의한 순도검증에서 순도가 95% 이상인 것으로 나타났다. SDS-PAGE 분석에서 주 밴드의 분자량은 약 31.5 kDa이었고 아미노산 조성은 alanine, glycine, serine, valine의 함량이 많았으며 분자량 31,500 Da를 기준으로 하였을 경우 본 protease의 잔기수는 321잔기였다. RP-HPLC로 분획한 main peak의 N-terminal amino acid sequence를 확인한 결과 $Val^1-Pro^2-Tyr^3-Gly^4-Val^5-Ser^6-Gln^7-Gly^8-Lys^9-Ala^{10}$인 것으로 밝혀졌다.

휴대용 심전도 기기와 직물형 전극을 이용한 생체정보 측정용 밀착 의복 개발 (Development of Tight-Fitting Garments with a Portable ECG Monitor to Measure Vital Signs)

  • A Holter monitor is used for ECG monitoring of ambulatory daily life in hospital. However, the use of this apparatus causes skin allergies and discomfort in patients because of the attachment gel and tapes used to attach disposable electrodes to the skin. In this study, the development of tight-fitting clothing connected to a portable Holter monitor was proposed. In addition, the use of conductive fabrics as electrodes was proposed; this will enable the use of garments in u-health care for measuring ECG signals. The male subjects were university students in the ages of 20 to 24. Subjective wear sensations of the experimental garments were rated using seven Likert scales. A Likert type scale was used for the evaluation and a 7 point score indicates that it provided the best fit as a tight-fitting upper clothing. Clothing pressure was measured using an air-pack-type pressure sensor (model AMI 3037-2) at 4 locations (the conductive fabric electrode) As results, a male basic sloper for upper clothing was developed and that pattern was manipulated to the tight fit pattern by considering the reduction rate of the percentage stretch in the fabric. The developed tight-fitting garment was superior in terms of subjective sensation and 6t. The mean pressure of the garment with reduction rates of 40% in width and of 50% in length was 8.45gf/$cm^2$. A conductive fabric electrode was developed by considering the sewing method and the developed electrode was detected well. The ECG data were recorded for 13 hr 19 min 44 sec and the artifacts in the ECG signals were recorded for 9 hr 3 min 46 sec (total time: 22 hr 23 min 23 sec). The artifacts data were obtained during heavy activities.
