Antioxidantive, Phospholipase $A_2$ Inhibiting, and Anticancer Effect of Polyphenol Rich Fractions from Panax ginseng C. A. Meyer (한국산 인삼의 Polyphenol 분획물의 항산화, Phospholipase $A_2$ 및 암세포증식 억제효과)

  • The polyphenol fractions of Korean ginseng were purified using Sephadex LH-20, MCI gel, Bondapak $C_{18}$ TLC, and HPLC from the 60% acetone soluble fraction. Fraction I showed 48.16%, 79.71% and 43.55% inhibition at 150 ppm against lipid oxidation in the presence of copper ion, superoxide and hydrogen peroxidation. Electron donating abilities of fraction II showed 35.17% inhibition at 200 ppm. Fraction III showed 48.49% and 25% inhibition at 150 ppm against lipid oxidation in the presence of ferrous ion and hydroxy radical ion. The phospholipase $A_2$ inhibitory effect of fraction III was 48.9% at the concentration of $60\;{\mu}g/ml$. The cytotoxic effects of fraction II was the highest (73.29% at 0.25 mg/ml) among the tested polyphenol fractions.

Anti-cancer and Anti-microbial Effect of the Fraction Isolated from Pyrus ussuriensis Leaves (산돌배나무(Pyrus ussuriensis) 잎 분획물의 항암 및 항균활성에 관한 연구)

  • This study was conducted to confirm the application as ingredients of cosmetics through an examination of the function for anti-cancer and anti-microbial of the fraction isolated from Pyrus ussuriensis leaves. The dried leaf of P. ussuriensis were extracted with acetone-$H_{2}O$ (6:4, v/v), concentrated and fractionated with the upper layer of acetone on a separatory funnel. Each fraction was freeze dried, then a portion of acetone soluble powder was chromatographed on a Sephadex LH-20 column using a series of aqueous methanol as eluents and also used the MIC-gel using a series of aqueous methanol as developing solvent. The isolated compounds were identified by silica-gel TLC. The growth inhibition activity was measured using the MTT assay by the mouse meltioma (B16F10) cell. The cancer cell growth inhibition rate of fractions isolated from P. ussuriensis leaf was 80%. In anti-microbial activity test, the fraction of P. ussuriensis with 0.25 mg/disc resulted in the clear zone of 1.3 cm and 2 cm for Staphylococcus aureus and S. epidermidis of gram positive bacillus, respectively. In Escherichia coli of gram negative bacillus, the fraction with 0.5 mg/disc resulted in the clear zone of 1.1 cm~1.5 cm each fraction. From these results, we confirmed that acetate fraction of P. ussuriensis has a great potential as a natural ingredients with a anti-cancer and anti-microbial source.

Isolation of Antioxidative Substances from Browning Reaction Product Obtained from L-Ascorbic Acid Solution (L-Ascorbic acid 갈변반응물질 중 항산화성물질의 분리)

  • Properties of the antioxidative fraction isolated from browning reaction product (BRP) that were obtained from 2 M L-ascorbic acid (AsA) solution (adjusted to pH 7.0) by heating for 25 hrs at $85^{\circ}C$ were investigated. Both of dialyzable and nondialyzable fraction isolated from BRP showed antioxidative effect. Dialyzable fraction has stronger antioxidative activity than nondialyzable. Dialyzable fraction was divided into the three fractions (A, B, C) by gel filtration. Among these fractions, the fraction (A) that had the highest reducing power and lowest browning drgree had lowest antioxidative activity. The fraction (C) that had lowest reducing power and highest browning degree showed strongest antioxidative effect. In the UV-visible spectrum of these fractions, the maximum absorption wavelengths of fraction A and B were 266.1 and 257.4nm, respectively, and fraction C showed a weaker absorption peak at 274.8nm. Infrared (IR) spectrum results showed that all fractions (A, B, C) had both hydroxy and carboxylic groups, and fraction B and C had carboxylic ester group.

Isolation and Identification of Antimicrobial Compound from Dandelion(Taraxacum platycarpum D.) (민들레로부터 항균성 화합물의 분리 및 동정)

  • Antimicrobial activity of dandelion(Taraxacum platycarpum D.) was investigated. Methanol extract of dried dandelion was fractionated to hexane, chloroform, ethylacetate, butanol and aqueous fraction. Ethylacetate fraction among these fractions showed the highest inhibitory effect on the microorganisms such as B. subtilis, L. monocytogenes, S. aureus, E. coli and V. parahaemolyticus at $500\mu\textrm{g}/disc$. Ethylacetate fraction was further fractionated into 13 fractions by silica gel column and thin layer chromatography(TLC). The results showed that ethylacetate fractions No. 4, 5 and 6 had high antimicrobial activity. These were mixed again, re separated and five fractions were obtained. Among them, No. 2 fraction had the highest inhibitory effect on the microorganisms, which was then separated into six fractions. In the 3rd fractionation, No. 3 fraction was identified as benzoic acid by HPLC, $^{1}H-NMR$ and GC MS.

  • The methanol extract of Thalictrum uchiyamai Nakai was fractionatedi nto four parts; (1) ether soluble nonphenolic fraction (2) phenolic tertiary bases fraction, (3) chloroform soluble tertiary bases fraction and (4) aqueous quaternary bases fraction. The fractions of this plant showed a total of twenty-four spots on the silica gel TLC. Ether soluble nonphenolic fraction and ether soluble phenolic fraction showed nine alkaloid spots respectively, chloroform soluble fraction showed three alkaloid spots and quaternary bases fraction showed three alkaloid spots. The antimicrobial activity of the four extracts was shown as follows; nonphenolic tertiary bases inhibited the growth of Micrococcus dutica at the concentration of $50\mu$g/ml, in phenolic tertiary bases $500\mu$g/ml concentration was inhibitory to Micrococcus dutica and to Staphyloccus aureus. In aqueous quaternary bases, $500\mu$g/ml concentration was inhibitory to Klebsiella pneumonae. Chloroform soluble tertiary fraction showed slight activity. All of bases showed reproducible activity against tested microorganisms.

  • Antimicrobial activity of Plantain(Plantago asiatica L.) was investigated. Methanol extract of dried Plantain was fractionated to hexane, chloroform, ethylacetate, butanol and aqueous fraction. Ethylacetate fraction among these fractions showed the highest inhibitory effect on the microorganisms such as B. subtilis, E. coli, and V. parahaemolyticus at 500 $\mu\textrm{g}$/disc. Ethylacetate fraction was further fractionated into 8 fractions by silica gel column and thin layer chromatography(TLC). The results showed that ethylacetate fractions No. 2 and 3 had the highest anti-microbial activity. They were mixed again, re-separated, and seven fractions were obtained. Among them, No.4 and 6 fraction had the highest inhibitory effect on the microorganisms, which were then separated into four fractions. In the 3rd fractionation, No.4 fraction was identified as hexadecanoic acid by HPLC, $^1$H-NMR and GC-MS.

  • The liquids of Lycopersicon esculentum were extracted with 70% aqueous MeOH and the concentrates were partitioned into EtOAc, n-BuOH, and H2O fractions. The repeated silica gel and octadecyl silica gel column chromatographies for the EtOAc fraction, whose activity was confirmed, led to isolation of one flavonol compound. The chemical structures of the compound were determined as quercetin (1) based on spectroscopic analyses including nuclear magnetic resornance, infrarad spectroscopy, and mass spectroscopy. Through this study, the antioxidant efficacy was confirmed by demonstrating that the L. esculentum fraction showing an increase in glutathione mean (GM) and a decrease in glutathione heterogeneity (GH) uniformly raises the intracellular glutathione (GSH) level.

  • Antimicrobial properties of Licorice(Glycyrrhizae radix L.) against food spoilage microorganism, Bacillus subtilis KCTC 1021 was investigated. Antibacterial activity of the essential oil was as equivalent as Potassium metabisulfite and myconazole. The licorice extracts was fractionated to hexane, chloroform, ethyl acetate, butanol and water fraction. Chloroform fraction showed the highest inhibitory effect on the Bacillus subtilis KCTC 1021. Chloroform fraction was further fractionated by silica gel column chromatography and thin layer chromatography(TLC). The antibacterial compound was isolated from their fractions and its chemical structures was identified as (R)-glabridin by ESI-MS, $^1H$-NMR and $^{13}C$-NMR.

Anti-Rheumatoid Arthritis Effect of the Kochia scoparia Fruits and Activity Comparison of Momordin Ic, its Prosapogenin and Sapogenin

  • MeOH extract of Kochia scoparia was fractionated into $CHCl_3-$, EtOAc- and BuOH extracts and the last fraction were hydrolyzed by 3%-NaOH ($MeOH-H_2O$) to compare the bioactivities on antinociceptive and anti-inflammatory effects. Silica gel column chromatography of BuOH fraction afforded a large amount of $3-Ο-{\beta}-D-xylopyranosyl {\;}(1{\rightarrow}3)-{\beta}-D-glucuronopyranosyl$ oleanolic acid (momordin Ic, 4) and that of acid hydrolysate of BuOH fraction gave $3-Ο-{\beta}-D-glucuronopyranosyl oleanolic$ acid (momordin Ib, 3), its 6'-Ο-methyl ester (2) and oleanolic acid (1). Silica gel column chromatography of alkaline hydrolysate afforded a large amount of 4. MeOH extract and both EtOAc- and BuOH fractions were active in the rheumatoidal rat induced Freund's complete adjuvant reagent (FCA) whereas $CHCl_3$ fraction was inactive. Compound 1 and 4 showed significant activities in the same assay but oleanolic acid 3-Ο-glucuronopyranoside (3) showed no activity. These fashions were also observed in carrageenan-induced edema of the rat and in the antinociceptive activity tests undertaken in hot plate- and writhing methods. These results suggest that momordin Ic and its aglycone, oleanolic acid, could be active principles for rheumatoid arthritis.

A Study of Glutathione S-transferase Inhibitors obtained from Allium cepa var. cepa Extract (양파 추출물에서의 글루타티온 전달효소 활성 저해제에 관한 연구)

  • In this study, an ethanol extract being obtained from Allium cepa var. cepa examins the inhibitory effects on the glutathione S-transferase and the separation had been done by silica-gel column chromatography using various eluents, such as ethyl acetate, methanol, and 50% methanol. A volume of column fraction was 50ml and evaporation has been performde by the rotary evaporator under reduced pressure. Each fraction is being examined by thin layer chromatography and the UV spectrum at 365 nm was used to investigate separation patterns of spots on thin layer chromatography. When the eluent was changed, the spot patterns showed another different pattern on thin layer chromatography, so on. Fractions showing similar pattern are combined and eventually, three fractions are obtained. Each fraction is testified to examine the inhibition effects on glutathione S-transferase. All of these showed inhibition effects on glutathione S-transferase. The GC-MS shows that each fraction contains more than 2 compounds.
