Gas chromatographic determination of l-ephedrine and dl-methylephedrine (가스크로마토그라피에 의한 l-에페드린과 dl-메칠에페드린의 정량)

  • The determination method of l-ephedrine and dl-methylephedrine by gas chromatography was developed, using reactive alkaline precolumn packed with celite 545 containing KOH. Symmetrical peaks were achieved under the condition, inlet temperature, $180^{\circ}C$-$230^{\circ}C$; column temperature, $180^{\circ}C$- $160^{\circ}C$; carrier gas flow rate, 30ml/minute. The peaks of the salts coincided with those of bases. When this method was applied to preparations, using d-dimethylaminobenzaldehyde as inner standard, good results were obtained. The relative retention times of l-ephedrine and dl-methylephedrine to p-dimethyl aminobenzaldehyde were 0.50 and 0.65 respectively.

  • Several commercial cottonseed, corn and rapeseed oils were stored at $60^{\circ}C\;and\;70^{\circ}C$ with daily exposure of fluorescent light for 12 hours and evaluated their rancidity by headspace gas chromatographic analysis of pentanal and hexanal. The data of gas chromatographic analysis was compared with organoleptic flavor evaluation. For headspace gas chromatographic analysis, the volatile compounds were recovered by porous polymer trap and flushed into a fused silica capillary column at $250^{\circ}C$. Twenty-three GC peaks were identified on the basis of relative retention time of reference compounds and gas chromatography-mass spectrometry. The results showed that the contents of pentanal and hexanal were linearly increased during storage. A very simple linear relationship was found between organoleptic flavor scores and amounts of two volatile compounds with very high correlation coefficient. This results suggested the possible implication of pentanal and hexanal as an quality index for rancidity evaluation of cottonseed, corn and rapeseed oils.

  • Five samples of triglyceride compositions of sesame oil and decuticled sesame oil have been determined by a gas chromatographic analysis. A similar distribution pattern of triglycerides was found in these five sesame oils. It was noted that $C_{50},\;C_{52}\;and\;C_{54}$ were the major components in these samples. The results showed that contents of $C_{50},\;C_{52}\;and\;C_{54}$ triglyceride types in five sesame oils were within $3.0{\sim}4.5%,\;23{\sim}28%\;and\;68{\sim}74%,$ respectively.

  • A coil pyrolyzer and processor-controlled gas chromatograph were used for analysis of rubber for compounding ratio of the single and blend rubber vlucanizates. Variables such as sample size, pyrolysis temperature, time allowed for pyrolysis, the column packing material, its length and programmable temperature for gas chromatography were examined to obtain optimum condition for application to NR, BR and SBR blends. By application fixed conditions, three kinds of standard curves were finally obtained from thirty samples of blend vulcanizates which were prepared in the pilot plant, NIRI. It is possible to determine rubber composition and their ratio in NR, BR and SBR products by pyrolysis.

Dual Capillary Column System for the Qualitative Gas Chromatography: 2. Comparison between Splitless and On-Column Injection Modes

  • A dual capillary column system is described for the simultaneous analysis of a given sample and measurement of retention index (RI) and area ratio (AR) values of each peak on two capillary columns of different polarity, DB-5 & DB-1701 from a single injection. Both capillary columns were connected to either a splitless injector or an on-column injector via a deactivated fused-silica capillary tubing of 1 m length and a 'Y' splitter. Both injection modes allowed to measure RI and AR values with high reproducibility (<0.01% RSD) and high accuracy (<10% RE), respectively with the exception that the trace and high boiling solutes required the on-column mode for the accurate quantification and AR comparison. When the dual capillary column system in on-column injection mode was applied to the blind samples containing organic acids, each acid was positively indentified by the combined computer RI library search-AR comparison.

Multi-Pesticide Residue Method for Analysis of Organochlorine and Organophosphorus Pesticide

  • Pesticide residues were extracted with 70% acetone and transferred to dichloromethane. Extracts were applied to open-column chromatography with florisil and alumina-N, The final extract was analyzed by gas chromatography with electron-capture detector(GC/ECD) and nitrogen-phosphorus detector(GC/NPD). Recoveries of the 17 organochlorine pesticides were ranged from 60.8 to 84.9% and those of 15 organophosphorus pesticides, from 70.5 to 100.0%(except phosmet and azinphos-methyl). The minimum detectable levels of this analytical method were low (0.021-0.058 mg/kg).

  • The optimum resolution condition of 11 organophosphorus pesticides by gas-liquid chromatography was studied using a capillary column. The injection modes and types of vaporization tubes were evaluated in relation to pesticide analysis. The best reproducibility of peak heights of pesticides was obtained by the split mode with the vaporization tube packed with quartz wool. A 25m SE-30 capillary column provided better resolution than SE-54 column for their optimum resolution. The column at $200^{\circ}C$ initially was heated by $5^{\circ}C/min\;to\;230^{\circ}C$ followed by $10^{\circ}C/min\;to\;270^{\circ}C$ and the temperatures were held constant for 5 minutes at $230^{\circ}C\;and\;270^{\circ}C$, respectively. The relative standard deviations of their retention times were less than 0.19%.

Gas Separations of Natural Zeolite by Chemical Treatments (화학처리에 의한 천연 Zeolite의 Gas 분리)

  • In the our country, especially in Yeongil and Wolsung area, abundant authigenic zeolites are found from the tuffaceous sediments and volcanic rocks of Miocene age showing wide variation in their mineralogy and abundance from horizon to horizon. The principal zeolite species identified are clinopti-lolite. mordenite. heulandite. ferrierite, and erionite. etc. Zeolite minerals are widely used in many countries in the following applications; (a) in air separation adsorption processes; (b)as desiccants; (c)in inorganic building materials; (d)in papermaking; (e)in fertilizers; (f)as soilconditioners-this application is based upon the ability of the zeolite to ion exchange with soil nutrients; (g)in the treatment of radioactive wastes; and (h)as adsorbents for toxic gases, etc. In the present paper, using natural zeolite mordenite treated with IN hydrochloric acid or IN sodium chloride solution as column packings, separation characteristics of argon, nitrogen, carbon monoxide, and methane gases have been studied by gas chromatography. By the use of mordenite treated with hydrochloric acid solution, the tailing peak of methane showed from untreated mordenite was satisfactorily reduced, although it was difficult to separate it from carbon monoxide with a column activated at $300^{\circ}C$. Using a column activated at $350^{\circ}C$, methane could be separated from carbon monoxide easily but only carbon monoxide eluted as a bad defined peak. Mordenite treated with sodium chloride solution was generally similar to chromatograms obtained by using the untreated mordenite. Both the above chemical treatments of mordenite had little effect on the separations of argon and nitrogen. The separations and the HETP values obtained from natural zeolite mordenite treated with continuously hydrochloric acid and sodium chloride solutions were almost identical with those obtained with synthetic molecular sieve 5A zeolite. On the other hand, the efficiency of column was good in the range 20~3Oml/min of the carrier helium gas rate.

Determination of barbiturates in plasma by gas chromatography-fralme photometric detector after N, N'-flame photometric detector after N, N'-dimethylthiomethyl derivatization

  • A specific and sensitive gas chromatographic (GC) procedure with the flame photometric detector (FPD) was developed for determination of barbiturates such as barbital, allobarbital, secobarbital, phenobarbital and thiopental in plasma. In order to evaluate the performance of the FPD, the results were campared with those of the flame ionization detector (FID). After extraction of barbiturates from plasma, the barbiturates were quantitatively N, N-dimethylthiometyl (MTM)-derivatized with methylthiomethyl chloride in 1, 8-diazabicyclo [5, 4, 0] undec-7-ene catalyst. The data indicate that the FPD is about 4 times more sensitive than the FID for barbiturates, although it is less reproducible. The FPD also produced chromatogram with less back ground for extracted plasma sample. The FPD also produced chromatogram with less background for extracted plasma sample. The minimum detectable amount of MTM-thiopental on 3% OV-225 column was 4, 4fmol and that of other MTM-barbiturate was about 10.0fmol.

  • The enantiomers of five amino acids (alanine, valine, threonine, leucine and phenylalanine) could be separated by gas chromatography with optically active (S)-5-isopropyll-$N^3$-phenyl-2-thiohydantoinic stationary phase, which prepared from L-valine and phenylisothiocyanate. Gas chromatographic separations on methylesterificated and N-trifluoroacetylated amino acids have been conducted in isothermal at several column temperatures (180~190, 200, $210^{\circ}C$). The separation factors were 1.29 (alanine, $190^{\circ}C$), 1.35 (valine, $190^{\circ}C$), 1.33 (threonine, $190^{\circ}C$), 1.17 (leucine, $190^{\circ}C$) and 1.05 (phenylalanine, $190^{\circ}C$) and D-isomers eluted prior to L-isomers in every instance. The result of this experiment shows that this stationary phase can be used for the separation of the other amino acids enantiomers.
